    Abstract: Qufengtongluo (QFTL) decoction is an effective treatment for diabetic nephropathy (DN). However, the underlying molecular mechanism is still unclear. In this study, we try to investigate whether QFTL decoction acts via inhibiting PI3K/Akt signaling pathway. Twenty-four GK rats were randomly divided into 3 groups: blank group, sham-operated group, and QFTL group. After model establishment, rats in QFTL group were given QFTL decoction by gavage, while the rest were given pure water. During the 8-week intervention, 24 hr urinal protein was measured every 2-3 weeks. After intervention, kidneys were removed for pathological smear, quantitative real-time PCR, and western blotting to detect expression levels of p-PI3K, p-Akt, PTEN, TGF- β , PI3K mRNA, Akt mRNA, PTEN mRNA, and TGF- β mRNA. QFTL group showed a slighter degree of renal fibrosis in Masson and PASM staining and a greater reduction of 24 hr urinal protein than blank group. Compared to blank group, expression levels of p-PI3K, p-Akt, PI3K mRNA, and Akt mRNA were lower in QFTL group, while expression levels of PTEN and PTEN mRNA were higher. Besides, TGF- β was downregulated by QFTL decoction. In conclusion, this study suggests that QFTL decoction might inhibit PI3K/Akt signaling pathway via activating PTEN and inhibiting TGF- β .

    Abstract: Using the freeze-thaw cycle test chamber, the red sandstone samples are subjected to cyclic freeze-thaw tests. The physical properties, static mechanical properties of freeze-thaw damage rocks, and the compressional wave velocity at specific axial pressure are measured using conventional physical tests and uniaxial compression tests. The mechanical properties of freeze-thaw damage rocks under dynamic and static loading were studied using Hopkinson pressure bar which can exert axial pressure. The studies show that, with the increase of freeze-thaw cycles, the surface layer of the rock sample undergoes spalling phenomenon, the weight gradually decreases, the sample compactness becomes worse, there are microcracks between the cemented particles, and the compressive strength and elastic modulus decrease. Under the static loading, the longitudinal wave velocity of freeze-thaw damaged samples change significantly compared with that of samples without freeze-thaw. The freeze-thaw damage degree, axial pressure, and strain rate are coupled with each other, which together affect the dynamic mechanical properties of samples, and make the variation of mechanical parameters, such as dynamic peak strength and dynamic elastic modulus of rock. The combined action of freeze-thaw damage and axial pressure weakens the strain rate effect of samples, but when the incident wave of SHPB test is same, the dynamic strength and elastic modulus of freeze-thaw damaged samples are reduced compared with those without freeze-thaw. Combining with strain equivalence principle, the constitutive relation of freeze-thaw damage of red sandstone under dynamic and static combined loading can reflect the influence of coupling damage of axial pressure and freeze-thaw, dynamic impact parameters, and other factors, which are in good agreement with the test results.

    Abstract: Background . Soluble lectin-like oxidized low-density lipoprotein receptor-1 (sLOX-1) may be a potential biomarker of coronary artery disease (CAD) and stroke. Objective . We aimed to investigate the association and prognostic value of elevated sLOX-1 concentrations with regard to long-term major adverse cardiovascular and cerebrovascular events (MACCEs) in patients with CAD undergoing primary percutaneous coronary intervention (PCI). Methods . A total of 1011 patients were enrolled. Serum sLOX-1 concentrations were detected by the enzyme-linked immunosorbent assay (ELISA). Patients were followed for 2 years. Multivariate Cox regression and Kaplan-Meier survival curve were explored to assess the association between sLOX-1 and MACCEs. A receiver operating characteristic (ROC) curve was used to evaluate the diagnostic efficacy of sLOX-1. Results . Two-year MACCEs were associated with serum sLOX-1 concentrations (HR 1.278, 95% CI 1.019-1.604, P = 0.034 ), left main disease (HR 2.938, 95% CI 1.246-6.925, P = 0.014 ), small-caliber stents used (HR 2.207, 95% CI 1.189-4.095, P = 0.012 ), and total stent length (HR 1.057, 95% CI 1.005-1.112, P = 0.030 ). Serum sLOX-1 concentration ≥ 1.10   ng / ml had maximum sensitivity and specificity in predicting the occurrence of 2-year MACCEs ( P 〈 0.001 ). Patients with higher serum sLOX-1 concentrations showed a significantly higher incidence of MACCEs in the Kaplan-Meier curve ( P 〈 0.001 ). The combination of any of the risk factors identified in multiple Cox regression was associated with a stepwise increase in MACCE rate ( P 〈 0.001 ). Conclusions . High baseline serum sLOX-1 concentration predicts 2-year MACCEs and shows an additional prognostic value to conventional risk factors in patients after primary PCI. sLOX-1 determination might play a complementary role in the risk stratification of patients with CAD treated with PCI.

    Abstract: Here we aimed to develop a capillary electrophoresis-based high-throughput multiplex polymerase chain reaction (PCR) system for the simultaneous detection of nine pathogens in swine. Nine pairs of specific primers and a set of universal primers were designed; the multiplex PCR was established. The specificity and cross-reactivity of this assay were examined, and the detection limit was determined using serial 10-fold dilutions of plasmids containing the target sequences. The assay was further tested using 144 clinical samples. We found that the nine specific amplification peaks were observed, and the assay had a high degree of specificity, without nonspecific amplification. The simultaneous detection limit for the nine viruses reached 10000 copies μ L −1 when all of the premixed viral targets were present. Seventy-seven of the clinical samples tested positive for at least one of the viruses; the principal viral infections in the clinical samples were porcine circovirus type 2 and porcine reproductive and respiratory syndrome virus. This approach has much potential for further development of high-throughput detection tools for the diagnosis of diseases in animals.

    Abstract: In this study, a method using ultrahigh-performance liquid chromatography with photodiode array (UHPLC-PDA) was established and validated for the simultaneous quantification of 10 active components, including eight macamides and two glucosinolates, in Lepidium meyenii (maca). A gas chromatographic mass spectroscopy (GC-MS) method was used to determine the levels of three benzyl isothiocyanates and two sterols in maca. Liquid chromatographic separation was achieved on a Waters Acquity UHPLC HSS T3 column (2.1 mm × 100 mm, 1.8 μm) with gradient elution over 15 min. The mobile phase was (B) acetonitrile-(A) 10 mM aqueous ammonium phosphate, and the detection wavelength was 210 nm. The gas chromatographic separation was performed on an SH-Rxi-1 MS column, and the ionization mode was electron ionization (EI). Two methods were confirmed to have desirable precision (RSD  〈  1.58%), repeatability (RSD  〈  1.97%), stability (RSD  〈  1.76%), and good linearity (R2 ≥ 0.999) within the test range. The recoveries were in the range of 96.79–109.99%, with an RSD below 2.39%. We applied the established methods and successfully analyzed 15 compounds in maca processed under different drying conditions, providing a comprehensive reference for maca processing method of development. In summary, this study provided two rapid and effective methods for the quantification of 15 active components, which contributed to the in-depth maca quality control and provided a reference for the development of maca products.

    Abstract: CD147, also known as EMMPRIN or basigin, is a transmembrane glycoprotein receptor that activates matrix metalloproteinases and promotes inflammation. CD147 function is regulated by posttranslational modifications of which glycosylation has attracted the most attention. In this study, we demonstrated that glycosylated CD147 was the dominant form in heart tissue, and its levels were markedly elevated in response to transverse aortic constriction (TAC). Adeno-associated virus 9-mediated, cardiac-specific overexpression of wild-type CD147 in mice significantly promoted pressure overload-induced pathological cardiac remodeling accompanied by augmented oxidative stress and ferroptosis. By contrast, mutations of CD147 glycosylation sites notably weakened these detrimental effects of CD147. Mechanistically, CD147 exacerbated TAC-induced pathological cardiac remodeling via direct binding with the adaptor molecule TRAF2 and subsequent activation of TAK1 signalling, which was dependent on glycosylation of CD147. Collectively, our findings provide the first evidence that CD147 promoted pathological cardiac remodeling and dysfunction in a glycosylation-dependent manner through binding the adaptor protein TRAF2 and activating the downstream TRAF2-TAK1 signalling pathway. Thus, glycosylation of CD147 may be a potent interventional target for heart failure treatment.
