    Abstract: In this paper, we study a task of slope collapse detection (SCD) for river embankment and formulate it as the tasks of motion detection and image recognition. Specifically, we introduce an SCD method based on motion detection and image recognition technologies to help inspector attendants detect the slope collapse. In this method, we use the foreground motion detection algorithm to identify the slope collapse of the scene of the river embankment. Since the moving targets in the foreground may not only be the slope collapse but also maybe some biology, we further use the image feature extraction and image recognition technology to recognize the foreground motion area, thus eliminating the influence of the biology on the detection results. Experimental results on the relevant scene data show that the proposed method can identify the slope collapse in real-time, and can effectively eliminate the motion interference of the biology, which has a high practical value.

    Abstract: The construction of municipal road engineering is a complex system engineering, and its internal components are interconnected and mutually restricted, and the relationship is intricate. The quality of its construction plan directly affects the realization of the project's quality, safety, environment, progress, cost, and other goals, and the project construction plan occupies an important position in the construction of the project. The selection of construction plans for municipal road projects involves a wide range of areas. According to the characteristics of specific municipal road engineering projects, it is very important to establish a scientific construction scheme intelligent generation model and select the optimal construction scheme suitable for the project from many feasible construction schemes, which has very important theoretical research value and practical application value. After analyzing the knowledge characteristics of road construction technology and the content of road construction schemes, this study decomposes the knowledge of road construction schemes into two parts: case characteristics and solutions. Then, according to the needs of case retrieval technology, the data storage form of each subdivision index is proposed and the value range is explained, and a complete structure form of the road construction scheme case database is formed. Combined with actual engineering cases, the feasibility and applicability of the intelligent generation method of the road construction scheme based on the KNN algorithm is confirmed, which provides a new idea for the automation of road construction scheme preparation and the improvement of the scheme application effect.

    Abstract: The purpose of this work was to investigate the porous polyetherimide scaffold (P-PEIs) as an alternative biopolymer for bone tissue engineering. The P-PEIs was fabricated via solvent casting and particulate leaching technique. The morphology, phase composition, roughness, hydrophilicity, and biocompatibility of P-PEIs were evaluated and compared with polyetherimide (PEI) and Ti6Al4V disks. P-PEIs showed a biomimetic porous structure with a modulus of 78.95 ± 2.30 MPa. The water contact angle of P-PEIs was 75.4 ± 3.39°, which suggested that P-PEIs had a wettability surface. Moreover, P-PEIs provides a feasible environment for cell adhesion and proliferation. The relative cell adhesion capability and the cell morphology on P-PEIs were better than PEI and Ti6Al4V samples. Furthermore, the MC3T3-E1 cells on P-PEIs showed faster proliferation rate than other groups. It was revealed that the P-PEIs could be a potential material for the application of bone regeneration.

    Abstract: Ferroptosis is a newly defined mode of programmed oxidative cell death. Knowledge of ferroptosis-related long noncoding (lnc) RNA in the tumor immune microenvironment of colon cancer is lacking. We systematically analyzed the correlations between ferroptosis-related lncRNAs and the tumor microenvironment, immune cell infiltration, and patient prognosis for 379 colon cancer samples in the Cancer Genome Atlas (TCGA). Using consensus clustering, we divided the 379 colon cancer patients into two subgroups (clusters 1 and 2) based on the differentially expressed ferroptosis-related lncRNAs. Cluster 1 was preferentially associated with longer overall survival, upregulated immune checkpoint inhibitor expressions, higher immunoscores, higher stromal scores, higher estimated scores, and distinct immune cell infiltration. Cancer- and metabolism-related pathways were enriched by gene set enrichment analyses. We constructed a prognostic signature of 15 ferroptosis-related lncRNAs (ZEB1-AS1, LINC01011, AC005261.3, LINC01063, LINC02381, ELFN1-AS1, AC009283.1, LINC02361, AC105219.1, AC002310.1, AL590483.1, MIR4435-2HG, NKILA, AC021054.1, and AL450326.1) and divided the patients into the high- and low-risk-score groups. The signature was validated using TCGA training and testing cohorts. The risk signature was an independent prognostic factor for predicting survival and excellently predicted the prognoses of patients with colon cancer. Moreover, the risk signature was related to immune characteristics. Chemosensitivity analyses showed that low-risk-score patients were more sensitive to sorafenib. In summary, our work revealed the important role of ferroptosis-related lncRNAs in the tumor microenvironment and immune cell infiltration and may help determine personalized prognoses and treatment for patients with colon cancer.

    Abstract: Pancreatic ductal adenocarcinoma (PDAC) is one of the highest-grade malignancies in the world. More effective biomarkers and treatment plans are necessary to improve the diagnosis rate and clinical outcome. The oncogenesis of PDAC is influenced by several factors, including chronic pancreatitis (CP). Keratin 8 (KRT8) is an important member of the keratin protein family and plays a role in regulating the cellular response to stress stimuli and mediating inflammatory reactions. However, the role of KRT8 in pancreatitis and PDAC is still poorly understood. Here we assessed the differentially expressed genes (DEGs) by bioinformatic methods with expression profiles available online for a caerulein-induced mouse model and human PDAC tissue. The prognostic value was evaluated by Kaplan–Meier analysis and Cox regression analysis. The diagnostic value was evaluated by Receiver Operating Characteristic analysis (ROC). The function of the genes was predicted by protein-protein interaction analysis, correlation analysis, and GO analysis. The conclusion was further validated in rat pancreatitis model, human tissue, and PDAC cell lines, including immunohistochemical staining (IHC), CCK-8 assay, wound healing assay, and flow cytometry. KRT8 was found to be upregulated in murine pancreatitis tissue, human CP tissue, and human PDAC tissue. High expression of KRT8 had a negative impact on the prognosis of PDAC patients. KRT8 was predicted to be involved in the regulation of the migration and viability of PDAC cells, which was validated in PDAC cell lines. Knockdown of KRT8 impaired the migration and proliferation and induced apoptosis in PDAC cell lines. In conclusion, keratin 8 is an inflammation-induced molecule and could serve as a diagnostic and prognostic marker for PDAC patients. More studies are needed for further validation from the perspective of precision and individualized medicine.

    Abstract: Terahertz time-domain spectroscopy (THz-TDS) systems are widely used to obtain fingerprint spectra of many different biomedical substances, and thus the identification of different biological materials, medicines, or dangerous chemicals can be realized. However, the spectral data for the same substance obtained from different THz-TDS systems may have distinct differences because of differences in system errors and data processing methods, which leads to misclassification and errors in identification. To realize the exact and fast identification of substances, spectral standardization is the key issue. In this paper, we present detailed disposal methods and execution processes for the spectral standardization and substance identification, including feature extracting, database searching, and fingerprint spectrum matching of unknown substances. Here, we take twelve biomedical compounds including different biological materials, medicines, or dangerous chemicals as examples. These compounds were analyzed by two different THz-TDS systems, one of which is a commercial product and the other is our verification platform. The original spectra from two systems showed obvious differences in their curve shapes and amplitudes. After wavelet transform, cubic spline interpolation, and support vector machine (SVM) classification with an appropriate kernel function, the spectra from two systems can be standardized, and the recognition rate of qualitative identification can be up to 99.17%.

    Abstract: Sewage discharge has become a key issue affecting the quality of the water environment, and how to effectively monitor and manage sewage discharge behavior has become a key factor to avoid water pollution and improve water quality. However, the current domestic sewage discharge monitoring system is not perfect, resulting in the lack of effective monitoring of enterprise sewage discharge by regulatory authorities, which provides an opportunity for enterprises to steal discharge. In the background of sewage treatment plant, the comprehensive design of sewage monitoring and alarm system is carried out based on the idea of physical information fusion. The design adopts a four-layer information physical architecture, which is divided into four parts: perception communication, fusion processing, push, and execution. In the fusion treatment part, the neural network intelligent algorithm is used to predict the dissolved oxygen, and the oxygen delivery is adjusted according to the predicted value to achieve accurate aeration and optimize the effluent quality. The push and execution parts adopt multiparameter monitoring to realize the smooth operation of equipment and ensure the system security. A new optimal control strategy of dissolved oxygen based on neural network is proposed. Through a large number of experiments and historical data, the intake index and dissolved oxygen value of the aeration tank under the condition of optimal outlet water are obtained as samples. According to the sample training, the BP neural network optimized by particle swarm optimization algorithm is adopted to achieve accurate prediction of dissolved oxygen under different inlet water conditions. The smooth operation of sewage treatment equipment is accomplished by the lower machine and the upper machine. In sewage treatment, each process section collects the equipment status in strict accordance with the order of sewage monitoring facilities. Then the communication network between the upper computer and the lower computer and the sensor is designed. The lower machine adopts PLC as the core, programming PLC through STEP7, and uses PID algorithm to control dissolved oxygen. The PC is developed in C language, so as to realize user login, real-time data display, over-limit fault alarm, report query, user management, etc. The PC integrates MATLAB neural network on the platform to predict dissolved oxygen through mixed programming quantity. The sewage alarm system based on improved artificial neural network is sensitive and has excellent performance. It provides a new idea for intelligent sewage detection and real-time monitoring.

    Abstract: With the rapid development of agricultural economy, people are paying more and more attention to how to apply high-efficiency technologies that save resources to improve agricultural production efficiency, so water-saving irrigation technology has gradually developed. China’s agricultural irrigation technology is relatively backward. In addition to the inappropriate irrigation methods and irrigation systems, problems such as siltation, seepage, and frost heave damage in irrigation canals have seriously affected the durability and service life of the canals. The backward irrigation technology seriously restricts the development of agricultural water-saving irrigation. Compared with large irrigation channels, the fabricated reinforced concrete small irrigation channels studied in this paper are less prone to frost heave damage and infiltration problems, and have the advantages of standardized production, simple transportation and installation, and convenient maintenance. In order to study the durability issues such as the basic characteristics, frost heave damage, and service life of fabricated irrigation channels, this paper takes the channel concrete and the formed channel as the research objects, and discusses the research on the properties of the channel concrete through theoretical research, numerical analysis, experiments, and other methods. Strength properties, water penetration resistance, cyanide ion penetration resistance and frost resistance; simulate the seasonal frost heave failure process of the channel; finally, on the basis of the test data, the service life aims to explore the safety and applicability of the fabricated reinforced concrete irrigation channel during the design use period.

    Abstract: Background. The isolation of tigecycline-resistant Acinetobacter baumannii in recent years has brought great difficulties to clinical prevention and treatment. Purpose. To explore the effect of efflux pump system and other resistance related gene mutations on tigecycline resistance in Acinetobacter baumannii. Methods. Fluorescence quantitative PCR was used to detect the expression levels of major efflux pump genes (adeB, adeJ, and adeG) in extensive drug-resistant Acinetobacter baumannii. The minimum inhibitory concentration (MIC) of tigecycline was detected by the broth microdilution testing and efflux pump inhibition experiment to assess the role of efflux pump in tigecycline resistance of Acinetobacter baumannii. Efflux pump regulatory genes (adeR and adeS) and tigecycline resistance related genes (rpsJ, trm, and plsC) were amplified by PCR and sequenced. By sequence alignment, tigecycline sensitive and tigecycline-insensitive Acinetobacter baumannii were compared with standard strains to analyze the presence of mutations in these genes. Results. The relative expression of adeB in the tigecycline-insensitive Acinetobacter baumannii was significantly higher than that in the tigecycline sensitive Acinetobacter baumannii (114.70 (89.53-157.43) vs 86.12 (27.23-129.34), P = 0.025 ). When efflux pump inhibitor carbonyl cyanide 3-chlorophenylhydrazone (CCCP) was added, the percentage of tigecycline-insensitive Acinetobacter baumannii with tigecycline MIC decreased was significantly higher than that of tigecycline-sensitive Acinetobacter baumannii (10/13 (76.9%) vs 26/59 (44.1%)), P = 0.032 ); the relative expression of adeB in the MIC decreased group was significantly higher than that in the MIC unchanged group (110.29 (63.62-147.15) vs 50.06 (26.10-122.59), P = 0.02 ); The relative expression levels of efflux pumps adeG and adeJ did not increase significantly, and there was no significant difference between these groups. One adeR point mutation (Gly232Ala) and eight adeS point mutations (Ala97Thr, Leu105Phe, Leu172Pro, Arg195Gln, Gln203Leu, Tyr303Phe, Lys315Asn, Gly319Ser) were newly detected. Consistent mutations in trm and plsC genes were detected in both tigecycline-insensitive and tigecycline-sensitive Acinetobacter baumannii, but no mutation in rpsJ gene was detected in them. Conclusion. Tigecycline-insensitive Acinetobacter baumannii efflux pump adeABC overexpression was an important mechanism for tigecycline resistance, and the mutations of efflux pump regulator genes (adeR and adeS) are responsible for adeABC overexpression. The effect of trm, plsC, and rpsJ gene mutations on the development of tigecycline resistance in Acinetobacter baumannii remains controversial.
