    Abstract: Iron metabolism in inflammation has been mostly characterized in macrophages exposed to pathogens or inflammatory conditions. The aim of this study is to investigate the cross-regulatory interactions between M1 macrophage polarization and iron metabolism. Firstly, we characterized the transcription of genes related to iron homeostasis in M1 RAW264.7 macrophages stimulated by IFN- γ . The molecular signature of M1 macrophages showed high levels of iron storage (ferritin), a low level of iron export (ferroportin), and changes of iron regulators (hepcidin and transferrin receptors), which favour iron sequestration in the reticuloendothelial system and are benefit for inflammatory disorders. Then, we evaluated the effect of iron on M1 macrophage polarization. Iron significantly reduced mRNA levels of IL-6, IL-1 β , TNF- α , and iNOS produced by IFN- γ -polarized M1 macrophages. Immunofluorescence analysis showed that iron also reduced iNOS production. However, iron did not compromise but enhanced the ability of M1-polarized macrophages to phagocytose FITC-dextran. Moreover, we demonstrated that STAT1 inhibition was required for reduction of iNOS and M1-related cytokines production by the present of iron. Together, these findings indicated that iron decreased polarization of M1 macrophages and inhibited the production of the proinflammatory cytokines. The results expanded our knowledge about the role of iron in macrophage polarization.

    Abstract: Simulated moving bed (SMB) chromatographic separation is a new type of separation technology based on traditional fixed bed adsorption operation and true moving bed (TMB) chromatographic separation technology, which includes inlet-outlet liquid, liquid circulation, and feed liquid separation. The input-output data matrices were constructed based on SMB chromatographic separation process data. The SMB chromatographic separation process was modeled by utilizing two subspace system identification algorithms: multivariable output-error state-space (MOESP) identification algorithm and numerical algorithms for subspace state-space system identification (N4SID), so as to obtain the 3rd-order and 4th-order state-space yield models of the SMB chromatographic separation process, respectively. The model predictive control method based on the established state-space models is used in the SMB chromatographic separation process. The influence of different control indicators on the predictive control system response performance is discussed. The output response curves of the yield models were obtained by changing the related parameters so that the yield model parameters are optimized set meanwhile. Finally, the simulation results showed that the yield models are successfully controlled based on the each control period and given yield range.

    Abstract: Simulated moving bed (SMB) chromatographic separation technology is a new adsorption separation technology with strong separation ability. Based on the principle of the adaptive neural fuzzy inference system (ANFIS), a soft sensing modeling method was proposed for realizing the prediction of the purity of the extract and raffinate components in the SMB chromatographic separation process. The input data space of the established soft sensor model is divided, and the premise parameters are determined by utilizing the meshing partition method, subtractive clustering algorithm, and fuzzy C -means (FCM) clustering algorithm. The gradient, Kalman, Kaczmarz, and PseudoInv algorithms were used to optimize the conclusion parameters of ANFIS soft sensor models so as to predict the purity of the extract and raffinate components in the SMB chromatographic separation process. The simulation results indicate that the proposed ANFIS soft sensor models can effectively predict the key economic and technical indicators of the SMB chromatographic separation process.

    Abstract: Sepsis may lead to sleep deprivation, which will promote the development of neuroinflammation and mediate the progression of sepsis-associated encephalopathy (SAE). Senkyunolide I, an active component derived from an herb medicine, has been shown to provide a sedative effect to improve sleep. However, its role in sepsis is unclear. The present study was performed to investigate whether Senkyunolide I protected against SAE in a murine model of cecal ligation and puncture (CLP). Here, we showed that Senkyunolide I treatment improved the 7-day survival rate and reduced the excessive release of cytokines including TNF-α, IL-6, and IL-1β. A fear conditioning test was performed, and the results showed that Senkyunolide I attenuated CLP-induced cognitive dysfunction. Senkyunolide I treatment also decreased the phosphorylation levels of inflammatory signaling proteins, including p-ERK, p-JNK, p-P38, and p-P65, and the level of inflammatory cytokines, including TNF-α, IL-6, and IL-1β, in the hippocampus homogenate. Sleep deprivation was attenuated by Senkyunolide I administration, as demonstrated by the modification of the BDNF and c-FOS expression. When sleep deprivation was induced manually, the protective effect of Senkyunolide I against inflammatory responses and cognitive dysfunction was reversed. Our data demonstrated that Senkyunolide I could protect against sepsis-associated encephalopathy in a murine model of sepsis via relieving sleep deprivation.

    Abstract: Bioaugmentation with degrading bacteria can improve the treatment of nicotine-containing tobacco industrial wastewater effectively. However, the transient and extremely high feeding of pollutants may compromise the effectiveness of the bioaugmented reactors. The effect of transient nicotine shock loads on the performance of Pseudomonas sp. HF-1 bioaugmented SBRs were studied. The results showed that, under 500–2500 mg/L of transient nicotine shocks, all the reactors still could realize 100% of nicotine degradation in 4 days of recovery, while the key nicotine degradation enzyme HSP hydroxylase increased in expression. Though the dramatic increase of activities of ROS, MDA, SOD, and CAT suggested that transient nicotine shock loads could induce oxidative stress on microorganisms in activated sludge, a decrease to control level demonstrated that most of the microorganisms could resist 500–1500 mg/L of transient nicotine shock under the protection from strain HF-1. After 8 cycles of recovery, high ROS level and low TOC removal in high transient shock reactors implied that 2000–2500 mg/L of transient nicotine shock was out of its recovery of strain HF-1 bioaugmented system. This study enriched our understanding on highly efficient nicotine-degrading strain bioaugmented system, which would be beneficial to tobacco waste or wastewater treatment in engineering.

    Abstract: Objective : Particularly interesting new cysteine-histidine rich protein (PINCH) is an important component of the local adhesion complexes and upregulated in several types of malignancies, and involved in the incidence and development of tumours. PINCH expression is also independently correlated with poorer survival in patients with colorectal cancer. However, there is no study of PINCH in gastric cancer, therefore, the aim of this project was to investigate PINCH expression and its clinicopathological significance in gastric adenocarcinoma. Patients and methods : PINCH expression was immunohistochemically examined in normal gastric mucous ( n = 30) and gastric adenocarcinoma ( n = 73), from gastric cancer patients. Results : PINCH expression in the associated-stroma of gastric cancers was heterogeneous, and its positive rate (75%) was higher than that of normal gastric mucosa (43%, X 2 = 9.711, p = 0.002). The stronger staining was observed at the invasive edge of tumour when compared to the inner area of tumour. The rate of positive PINCH (88%) in the cases with lymph node metastasis was higher than that (52%) in the cases without metastasis ( X 2 = 11.151, p = 0.001). PINCH expression was not correlated with patients’ gender, age, tumour size, differentiation and invasion depth ( p 〉 0.05). Comclusion : PINCH protein might play an important role in the tumourigenesis and metastasis of gastric adenocarcinoma.

    Abstract: Background. To investigate whether intestinal mucosal barrier was damaged or not in chronic kidney disease progression and the status of oxidative stress. Methods. Rats were randomized into two groups: a control group and a uremia group. The uremia rat model was induced by 5/6 kidney resection. In postoperative weeks (POW) 4, 6, 8, and 10, eight rats were randomly selected from each group to prepare samples for assessing systemic inflammation, intestinal mucosal barrier changes, and the status of intestinal oxidative stress. Results. The uremia group presented an increase trend over time in the serum tumor necrosis factor-alpha, interleukin-6 (IL-6) and IL-10, serum D-lactate and diamine oxidase, and intestinal permeability, and these biomarkers were significantly higher than those in control group in POW 8 and/or 10. Chiu’s scores in uremia group were also increased over time, especially in POW 8 and 10. Furthermore, the intestinal malondialdehyde, superoxide dismutase, and glutathione peroxidase levels were significantly higher in uremia group when compared with those in control group in POW 8 and/or 10. Conclusions. The advanced chronic kidney disease could induce intestinal mucosal barrier damage and further lead to systemic inflammation. The underlying mechanism may be associated with the intestinal oxidative stress injury.

    Abstract: Background. Although traditional Chinese medicine (TCM) has good efficacy in the treatment of mild cognitive impairment (MCI), especially memory improvement and safety, its substance basis and intervention mechanism are particularly complex and unknown. Therefore, based on network pharmacology and data mining, this study aims to explore the rules, active ingredients and mechanism of TCM in the treatment of MCI. Methods. By searching the GeneCard, OMIM, DisGeNET and DrugBank databases, we obtained the critical targets associated with MCI. We matched the components and herbs corresponding to the important targets in the TCMSP platform. Using Cytoscape 3.7.2 software, we constructed a target-component-herb network and conducted a network topology analysis to obtain the core components and herbs. Molecular docking was used to preliminarily analyze and predict the binding activities and main binding combinations of the core targets and components. Based on the analysis of the properties, flavor and meridian distribution of herbs, the rules of herbal therapy for MCI were summarized. Results. Twenty-eight critical targets were obtained after the screening. Using the TCMSP platform, 492 components were obtained. After standardization, we obtained 387 herbs. Based on the target-composition-herb network analysis, the core targets were ADRB2, ADRA1B, DPP4, ACHE and ADRA1D. According to the screening, the core ingredients were beta-sitosterol, quercetin, kaempferol, stigmasterol and luteolin. The core herbs were matched to Danshen, Yanhusuo, Gancao, Gouteng and Jiangxiang. It was found that the herbs were mainly warm in nature, pungent in taste and liver and lung in meridian. The molecular docking results showed that most core components exhibited strong binding activity to the target combination regardless of the in or out of network combination. Conclusion. The results of this study indicate that herbs have great potential in the treatment of MCI. This study provides a reference and basis for clinical application, experimental research and new drug development of herbal therapy for MCI.
