    Abstract: Automatic classification of ECG is very important for early prevention and auxiliary diagnosis of cardiovascular disease patients. In recent years, many studies based on ECG have achieved good results, most of which are based on single-label problems; one record corresponds to one label. However, in actual clinical applications, an ECG record may contain multiple diseases at the same time. Therefore, it is very important to study the multilabel ECG classification. In this paper, a multiscale residual deep neural network CSA-MResNet model based on the channel spatial attention mechanism is proposed. Firstly, the residual network is integrated into a multiscale manner to obtain the characteristics of ECG data at different scales and then increase the channel spatial attention mechanism to better focus on more important channels and more important ECG data fragments. Finally, the model is used to classify multilabel in large databases. The experimental results on the multilabel CCDD show that the CSA-MResNet model has an average F1 score of 88.2% when the multilabel classification of 9 ECGs is performed. Compared with the benchmark model, the F1 score of CSA-MResNet in the multilabel ECG classification increased by up to 1.7%. And, in the model verification on another database HF-challenge, the final average F1 score is 85.8%. Compared with the state-of-the-art methods, CSA-MResNet can help cardiologists perform early-stage rapid screening of ECG and has a certain generalization performance, providing a feasible analysis method for multilabel ECG classification.

    Abstract: A previous study showed that a downexpression of protease-activated receptor 4 (PAR4) is associated with the development of esophageal squamous cell carcinoma (ESCC). In this study, we explored the relationship between PAR4 activation and the expression of p16, and elucidated the underlying mechanisms in PAR4 inducing the tumor suppressor role in ESCC. ESCC cell lines (EC109 and TE-1) were treated with PAR4-activating peptide (PAR4-AP). Immunohistochemistry for DNA methyltransferase 1 (DNMT1) and histone deacetylase 2 (HDAC2) was performed in 26 cases of ESCC tissues. We found that DNMT1 and HDAC2 immunoreactivities in ESCC were significantly higher than those in adjacent noncancerous tissues. PAR4 activation could suppress DNMT1 and HDAC2, as well as increase p16 expressions, whereas silencing PAR4 dramatically increased HDAC2 and DNMT1, as well as reduced p16 expressions. Importantly, the chromatin immunoprecipitation-PCR (ChIP-PCR) data indicated that treatment of ESCC cells with PAR4-AP remarkably suppressed DNMT1 and HDAC2 enrichments on the p16 promoter. Furthermore, we demonstrated that activation of PAR4 resulted in an increase of p38/ERK phosphorylation and activators for p38/ERK enhanced the effect of PAR4 activation on HDAC2, DNMT1, and p16 expressions, whereas p38/ERK inhibitors reversed these effects. Moreover, we found that activation of PAR4 in ESCC cells significantly inhibited cell proliferation and induced apoptosis. These findings suggest that PAR4 plays a potential tumor suppressor role in ESCC cells and represents a potential therapeutic target of this disease.

    Abstract: MicroRNA (miRNA) has been widely suggested to play a vital role of in the pathogenesis of gestational diabetes mellitus (GDM). We have previously demonstrated that miR-657 can regulate macrophage inflammatory response in GDM. However, the role of miR-657 on M1/M2 macrophage polarization in GDM pathogenesis is not clear yet. This study is aimed at elucidating this issue and identifying novel potential GDM therapeutic targets based on miRNA network. miR-657 is found to be upregulated in placental macrophages demonstrated by real-time PCR, which can enhance macrophage proliferation and migration in vitro. Luciferase reporter assay shows the evidence that FAM46C is a target of miR-657. In addition, miR-657 can promote macrophage polarization toward the M1 phenotype by downregulating FAM46C in macrophages. The present study strongly suggests miR-657 is involved in GDM pathogenesis by regulating macrophage proliferation, migration, and polarization via targeting FAM46C. miR-657/FAM46C may serve as promising targets for GDM diagnosis and treatment.

    Abstract: Deciphering the factors causing damage to white matter fiber bundles and exploring new strategies to alleviate white matter injury (WMI) is a promising treatment to improve neurological impairments after intracerebral hemorrhage (ICH). Ferroptosis usually occurs at perihematomal region and contributes to neuronal death due to reactive oxygen species (ROS) production. Dexpramipexole (DPX) easily crosses the blood brain barrier (BBB) and exerts antioxidative properties by reducing ROS production, while the role of DPX in ferroptosis after ICH remains elusive. Here, our results indicated that ferroptosis played a significant role in WMI resulting from iron and ROS accumulation around hematoma. Further evidence demonstrated that the administration of DPX decreased iron and ROS deposition to inhibit ferroptosis at perihematomal site. With the inhibition of ferroptosis, WMI was alleviated at perihematomal site, thereafter promoting locomotion and motor coordination recovery in mice after ICH. Subsequently, the results showcased that the expression of glutathione peroxidase 4 (GPX4) and ferroptosis suppressing protein 1 (FSP1) was upregulated with the administration of DPX. Collectively, the present study uncovers the underlying mechanism and elucidates the therapeutic effect of DPX on ICH, and even in other central nervous system (CNS) diseases with the presence of ferroptosis.

    Abstract: It is critical to develop a cost-effective detection kit for rapid diagnosis and on-site detection of severe fever with thrombocytopenia syndrome virus (SFTSV) infection. Here, an immunochromatographic assay (ICA) to detect SFTSV infection is described. The ICA uses gold nanoparticles coated with recombinant SFTSV for the simultaneous detection of IgG and IgM antibodies to SFTSV. The ICA was developed and evaluated by using positive sera samples of SFTSV infection ( n = 245 ) collected from the CDC of China. The reference laboratory diagnosis of SFTSV infection was based on the “gold standard”. The results demonstrated that the positive coincidence rate and negative coincidence rate were determined to be 98.4% and 100% for IgM and 96.7% and 98.6% for IgG, respectively. The kit showed good selectivity for detection of SFTSV-specific IgG and IgM with no interference from positive sera samples of Japanese encephalitis virus infection, Dengue virus infection, Hantavirus infection, HIV infection, HBV surface antigen, HCV antibody, Mycobacterium tuberculosis antibody, or RF. Based on these results, the ICS test developed may be a suitable tool for rapid on-site testing for SFTSV infections.

    Abstract: Cartilage dysplasia is one of the important reasons for the weakness of pig limbs and hooves. Porcine rickets with weak limbs and hooves bring huge economic losses to the pig industry. However, research on the development of pig cartilage is lacking. This study investigated the key genes and molecular mechanisms involved in cartilage development via an RNA-seq technique. Samples of proximal tibia cartilage were collected from three normal piglets with 1 day, 14 days, and 28 days of age, respectively, and then these samples were divided into two comparison groups (1-day vs. 14-day group, 14-day vs. 28-day group). Through the transcriptome analysis, 108 differentially expressed genes (DEGs), such as FORL2 , were obtained from 1-day vs. 14-day comparison group, and 3602 DEGs were obtained from 14-day vs. 28-day comparison group, including SOX9 , BMP6 , and MMP13 . The gene ontology (GO) functional and KEGG pathway enrichment revealed that many functions of DEGs were related to bone development. The pathways of DEGs from Day 1 vs. Day 14 were mainly enriched in mineral absorption, but the DEGs of Day 14 vs. Day 28 were enriched in osteoclast differentiation. Then, the expression patterns of six candidate genes were verified via qPCR. In conclusion, candidate genes affecting cartilage development in Yorkshire pigs were obtained by transcriptome analysis, and the clues showed that Day 14 to Day 28 is a more active and extensive period in cartilage developments, which played a key role in revealing the molecular mechanism of pig cartilage development basis, also compensating for vacancies in cartilage research.

    Abstract: The lining crack is common for the tunnel in the stage of operation, which has seriously influenced the service life and safety of tunnel engineering. It is a new trend to use computer vision to detect tunnel cracks over the past few years in China and foreign countries. By image processing technology and intelligent algorithm, the computer has a hominine visual perception system which understands, analyzes, and determines input image information, thus recognizing and detecting specific objectives. However, the effect of image recognition for tunnel crack now cannot satisfy the demands of practical engineering. SSD algorithm has been used when analyzing features of lining surface image, while comparison analysis has been made from image recognition results, error rate, and running time. The results indicate that the SSD algorithm can accurately and rapidly detect and mark the position of the tunnel crack. The tunnel information obtained from image recognition is subsequently imported into the team independently developed software GeoSMA-3D, which is useful for determining tunnel grade.

    Abstract: The present study aims to elucidate the problem of a rock mass structural plane with a range of damage degrees and the numerical model selection for analysis of a slope with multiple sliding surfaces. Based on the relative displacement between blocks, the dynamic strength reduction-discontinuous deformation analysis (hereinafter referred to as DSR-DDA) method is proposed for studying slopes with multiple sliding surfaces. The slope-slider classic case was used to test the displacement threshold. The model was applied to the stability analysis of multiple sliding surfaces of a high rock slope in the Fushun West Open-Pit Mine. The results show that when the displacement threshold is set to 1 mm, the error between the DSR-DDA results and the theoretical solution is within 0.5%, which satisfies the calculation requirements. The most dangerous slip surface in the Fushun West Open-Pit Mine slope was identified. Based on the numerical slope model after the first landslide, the position of the secondary slip surface was then identified. The failure mode is traction sliding failure, and the middle and lower oil shales play a key role in the slope stability. This study recommends that mining of the remaining oil shale should stop to avoid causing large-scale landslides in the upper part of the slope and landslides at the pit-city boundary.

    Abstract: To determine whether Tai Chi (TC) is effective in slowing cognitive decline in older populations with mild cognitive impairment (MCI), we performed a systematic review and meta-analysis of randomized controlled trials (RCTs) on Tai Chi and MCI. We searched eight electronic databases (PubMed, PsycINFO, Wanfang, Web of Science, MEDLINE, CNKI, EBSCO, and the Cochrane Central Register of Controlled Trials) for appropriate RCTs published up to August 2019. For those studies included, the data were extracted, methodological quality was evaluated, and then meta-analysis was performed using Review Manager software (version 5.3). A total of 11 of the studies were available for systematic review, which together included 1061 participants, met the inclusion criteria, and ten of these were included in the meta-analysis. For most RCTs, the methodological quality was moderate. The meta-analysis revealed that Tai Chi could significantly improve global cognitive function; memory and learning; mental speed and attention; ideas, abstraction, figural creations, and mental flexibility; and visuospatial perception. The present review adds to the evidence showing that Tai Chi is potentially beneficial in improving cognitive functions among elderly people with MCI. However, strictly designed and well-reported RCTs are required.
