    Abstract: Background. Rab22a is a member of the RAS superfamily, involved in early endosome formation and intracellular vesicle transport. Rab22a is significantly upregulated in a variety of malignant tumors. However, its function in thyroid cancer has never been addressed. Methods. The expression of Rab22a in paraffin sections of 101 patients was detected by immunohistochemical staining. By upregulating and downregulating the expression of Rab22a in thyroid cancer cell lines, the effect of Rab22a on cell proliferation, invasion, and migration was analyzed. Co-IP was employed, and the interaction between Rab22a and PI3Kp85α was shown. The function of Rab22a on PI3K/AKT/mTOR signaling and epithelial-mesenchymal transition (EMT) was further studied by western blot analysis. Results. Immunostaining showed that Rab22a was significantly overexpressed in thyroid cancer tissues but negative in adjacent normal tissues or nodular goiters. The proliferation, migration, invasion, and EMT in papillary thyroid carcinoma cell lines were enhanced upon Rab22a overexpression but inhibited after knocking down Rab22a. The co-IP assay demonstrated an interaction between Rab22a and PI3K85α, an effector of PI3K. We further found that Rab22a can activate the PI3K/AKT/mTOR signaling pathway. However, the ability of Rab22a to promote the proliferation, invasion, migration, and EMT of papillary thyroid carcinoma cells was significantly inhibited after being treated with LY294002, a PI3K inhibitor. Conclusions. Rab22a can promote the EMT process and enhance proliferation, migration, and invasion of papillary thyroid carcinoma cells by activating the PI3K/AKT/mTOR signaling pathway. Our study provides new pathological diagnosis clues and clinical treatment targets for thyroid cancer.

    Abstract: Respiratory diseases, always being a threat towards the health of people all over the world, are most tightly associated with immune system. Neutrophils serve as an important component of immune defense barrier linking innate and adaptive immunity. They participate in the clearance of exogenous pathogens and endogenous cell debris and play an essential role in the pathogenesis of many respiratory diseases. However, the pathological mechanism of neutrophils remains complex and obscure. The traditional roles of neutrophils in severe asthma, chronic obstructive pulmonary diseases (COPD), pneumonia, lung cancer, pulmonary fibrosis, bronchitis, and bronchiolitis had already been reviewed. With the development of scientific research, the involvement of neutrophils in respiratory diseases is being brought to light with emerging data on neutrophil subsets, trafficking, and cell death mechanism (e.g., NETosis, apoptosis) in diseases. We reviewed all these recent studies here to provide you with the latest advances about the role of neutrophils in respiratory diseases.

    Abstract: Parabolic trough systems are the most used concentrated solar power technology. The operating performance and optical efficiency of the parabolic trough solar collectors (PTCs) are different in different regions and different seasons. To determine the optimum design and operation of the parabolic trough solar collector throughout the year, an accurate estimation of the daily performance is needed. In this study, a mathematical model for the optical efficiency of the parabolic trough solar collector was established and three typical regions of solar thermal utilization in China were selected. The performance characteristics of cosine effect, shadowing effect, end loss effect, and optical efficiency were calculated and simulated during a whole year in these three areas by using the mathematical model. The simulation results show that the optical efficiency of PTCs changes from 0.4 to 0.8 in a whole year. The highest optical efficiency of PTCs is in June and the lowest is in December. The optical efficiency of PTCs is mainly influenced by the solar incidence angle. The model is validated by comparing the test results in parabolic trough power plant, with relative error range of 1% to about 5%.

    Abstract: Background and Objective . Asthma as a chronic heterogeneous disease seriously affects the quality of life. Incorrect identification for its clinical phenotypes lead to a huge waste of medical resources. Metabolomic technique as a novel approach to explore the pathogenesis of diseases have not been used to study asthma based on their clear defined inflammatory phenotypes. This study is aimed to distinguish the divergent metabolic profile in different asthma phenotypes and clarify the pathogenesis of them. Methods . Participants including eosinophilic asthmatics (EA, n=13), noneosinophilic asthmatics (NEA, n=16), and healthy controls (HC, n=15) were enrolled. A global profile of untargeted serum metabolomics was identified with Ultra Performance Liquid Chromatography–Mass Spectrometry technique. Results . Multivariate analysis was performed and showed a clear distinction between EA, NEA, and HC. A total of 18 different metabolites were recognized between the three groups based on OPLS-DA model and involved in 10 perturbed metabolic pathways. Glycerophospholipid metabolism, retinol metabolism, and sphingolipid metabolism were identified as the most significant changed three pathways (impact 〉 0.1 and -log( P ) 〉 4) between the phenotypes. Conclusions . We showed that the different inflammatory phenotypes of asthma involve the immune regulation, energy, and nutrients metabolism. The clarified metabolic profile contributes to understanding the pathophysiology of asthma phenotypes and optimizing the therapeutic strategy against asthma heterogeneity.

    Abstract: Background. With an enormous amount of research concerning kidney cancer being conducted, various treatments have been applied to its cure. However, high recurrence and metastasis rates continue to pose a threat to the survival of patients with kidney renal clear cell carcinoma (KIRC). Methods. Data from The Cancer Genome Atlas were downloaded, and a series of analyses were performed, including differential analysis, Cox analysis, weighted gene coexpression network analysis, least absolute shrinkage and selection operator analysis, multivariate Cox analysis, survival analysis, and receiver operating characteristic curve and functional enrichment analysis. Results. A total of 5,777 differentially expressed genes were identified from the differential analysis. The Cox analysis showed 1,853 significant genes ( P 〈 0.01 ). Weighted gene coexpression network analysis revealed that 226 genes in the module were related to clinical parameters, including Tumor-Node-Metastasis (TNM) staging. Least absolute shrinkage and selection operator and multivariate Cox analyses suggested that four genes (CDKL2, LRFN1, STAT2, and SOWAHB) had a potential function in predicting the survival time of patients with KIRC. Survival analysis uncovered that a high risk of these four genes was associated with an unfavorable prognosis. Receiver operating characteristic curve analysis further confirmed the accuracy of the risk score model. The analysis of clinicopathological parameters of the four identified genes revealed that they were associated with the progression of KIRC. Conclusion. The gene expression model consisting of CDKL2, LRFN1, STAT2, and SOWAHB is a promising tool for predicting the prognosis of patients with KIRC. The results of this study may provide insights into the diagnosis and treatment of KIRC.

    Abstract: In ultrasonic ranging system, the time-of-flight (TOF), which acquired by the determination of two special points of ultrasonic echo, is important for accuracy and speed. The published calculation of TOF processing methods is too enormous to affect the real-time performance. To improve its measurement accuracy and speed, we proposed a new algorithm called “moving sine-wave fitting” to calculate the envelope of ultrasonic echo for each calibration point. Then, the absolute feature point (AFP) corresponding to half of the envelope peak can be defined and used to find out the relative feature point (RFP); the linear relationship between the RFPs and the known distances should be obtained referring to calibration points. At last, the distance on the measure can be acquired by using linear interpolation according to the above relationship. The measurement experiments have been performed on a set of distances, which shows a good measurement resolution that the overall measurement accuracy is 0.513 mm, and the linear correlation coefficient between the RFPs and the obtained distances is R2 = 0.806.

    Abstract: Background . We report a modified transplantation surgical technique for CHD with dextrocardia which is rare and surgically challenging. Methods. From January 2015 to May 2018, 5 patients with end-stage CHD with dextrocardia underwent heart transplantation at our institute. They were 10, 29, 13, 15, and 22 years old, respectively; 3 of them had dextroversion, and the other 2 had mirror-image dextrocardia and post-TCPC. The atrial-atrial anastomosis was performed first between the donor’s upper-left PVO and the recipient’s lower-left PVO. The apex thereby rotated approximately 90° clockwise (to the right). The end-to-end donor and recipient aortas, vena cava, and pulmonary arteries were then anastomosed. Results . The cold ischemic time of the donor heart was 284.6 ± 108.3   min , and the CPB time was 190.2 ± 43.8   min . The postoperative X-ray showed the apex on the right. Four patients were successfully discharged, and the follow-up times were 47 months, 36 months, 12 months, and 12 months. One post-TCPC patient died because of pneumonia and hypoxia at 59 postoperative days. Conclusions . Heart transplantation with dextrocardial CHD is rare. A 90° rotation at the left atrial level, aortic end-to-end anastomosis, and vena cava reconstruction by vascular prosthesis or systemic atrial cuff is a simple and effective surgical strategy.

    Abstract: Rock burst is a typical dynamic disaster in deep underground coal mining. Based on the support problems of the deep roadways in fully mechanized caving face 401111 of Hujiahe Coal Mine suffering from rock burst in Shaanxi Province of China, the failure law and influencing factors of the surrounding rock of the roadway are analyzed. The results show that the deformation of surrounding rock in the roadway shows the characteristics of elastic, plastic transformation, rheology, and expansion. At the same time, it has the typical characteristics of deep roadway, such as the fast deformation speed, long duration, asymmetric deformation, and large loose broken area of surrounding rock. Based on the principle of “strengthening support in shallow zones” and “deep pressure relief in deep zones” in the surrounding rock, the control scheme of surrounding rock in the return roadway of fully mechanized caving working face 401111 is proposed by taking the large diameter pressure relief and deep hole blasting as the main means of pressure relief. The practice shows that the surrounding rock of the return roadway is relatively stable after the implementation of the new scheme, which shows that the design of the new support scheme is reasonable and reliable. It is of great significance for the stability control of surrounding rock of the mining roadway suffering from rock burst.

    Abstract: In traditional Chinese medicine theory, blood stasis syndrome (BSS), characterized by blood flow retardation and blood stagnation, is one of the main pathologic mechanisms and clinical syndromes of cardiovascular diseases (CVDs). Rhodiola wallichiana var. cholaensis injection (RWCI) is made from dry roots and stems of RWC via the processes of decoction, alcohol precipitation, filtration, and dilution. Studies indicated the extracts of RWC could alleviate CVDs; however, the mechanism had not been illustrated. In the present study, the acute blood stasis rat model was established to investigate the pathogenesis of BSS and the therapeutic mechanism of RWCI against BSS. Hemorheological parameters (whole blood viscosity and plasma viscosity) and inflammatory factors (TNF- α and IL-6) were used to evaluate the success of the BSS rat model and RWCI efficacy. 14 and 33 differential metabolites were identified from plasma and urine samples using the metabolomics approach based on ultrahigh-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry. The results of multivariate analysis displayed that there were significant separations among model, control, and treatment groups, but the high-dose RWCI treatment group was closer to the control group. 9 perturbed metabolic pathways were related to BSS’s development and RWCI intervention. 5 metabolic pathways (arachidonic acid metabolism, linoleic acid metabolism, alpha-linolenic acid metabolism, retinol metabolism, and steroid hormone biosynthesis) showed apparent correlations. These differential metabolites and perturbed metabolic pathways might provide a novel view to understand the pathogenesis of BSS and the pharmacological mechanism of RWCI.
