    Abstract: We report the clinical and genetic features of a Han Chinese boy who presented with disease suspect for Alagille syndrome (ALGS). Multiple genetic analyses (panel sequencing, multiplex-ligation-dependent probe amplification, and whole genome sequencing) failed to uncover a causative variant. Optical genomic mapping detected a reciprocal translocation between chromosomes 4 and 20, interrupting JAG1. Long-range polymerase chain reaction and targeted sequencing identified the exact breakpoints. Sanger sequencing and reanalysis of genome sequencing raw data further confirmed the result. This translocation is expected to generate aberrant JAG1 transcripts that lead to complete loss of JAG1 expression. This is the first t(4;20)(q22.1;p12.2) balanced translocation detected by optical genomic mapping and characterized at base-pair resolution in ALGS. Our approach permitted precise diagnosis and genetic counseling.

    Abstract: The Euler-Taylor-Galerkin flux-corrected transport (ETG-FCT) algorithm for the numerical solution of particle transport equations is described, based on the method developed by Lohner to solve conservation equations in fluid mechanics, and its application is extended to gas discharge problems. To improve the efficiency of computing and reduce numerical error, the nonuniform triangular mesh method is introduced, and the continuity equation is solved by ETG-FCT. The new contributions in this paper include the development of the ETG scheme and its application to rod-plate gap streamer discharge in 50~50% SF 6 /N 2 gas mixtures problems. Results are obtained: the spatial distributions of electron densities, positive ion densities, negative ion densities, photoelectron densities, and the electric field, respectively. The velocities of streamer propagations and the radius of streamer obtained from the proposed model are in good agreement with experimental and simulation results in literature. The results also prove that the ETG-FCT method is valid.

    Abstract: Beijing-Tianjin-Hebei is a typical developed region in China. The development of economy has brought lots of carbon emissions. To explore an effective way to reduce carbon emissions, we applied the Logarithmic Mean Divisia Index (LMDI) model to find drivers behind carbon emission from 2003 to 2013. Results showed that, in Beijing, Tianjin, and Hebei, economic output was main contributor to carbon emissions. Then we utilized the decoupling model to comprehensively analyze the relationship between economic output and carbon emission. Based on the two-level model, results indicated the following: ( 1 ) Industry sector accounted for almost 80% of energy consumption in whole region. The reduced proportion of industrial GDP will directly reduce the carbon emissions. ( 2 ) The carbon factor for CO 2 /energy in whole region was higher than that of Beijing and Tianjin but lower than that of Hebei. The impact of energy structure on carbon emission depends largely on the proportion of coal in industry. ( 3 ) The energy intensity in whole region decreased from 0.79 in 2003 to 0.40 in 2013 (unit: tons of standard coal/ten thousand yuan), which was lower than national average. ( 4 ) The cumulative effects of industrial structure, energy structure, and energy intensity were negative, positive, and negative, respectively.

    Abstract: In recent years, the energy efficiency of thermal power plant largely contributes to that of the industry. A thorough understanding of influencing factors, as well as the establishment of scientific and comprehensive diagnosis model, plays a key role in the operational efficiency and competitiveness for the thermal power plant. Referring to domestic and abroad researches towards energy efficiency management, based on Cloud model and data envelopment analysis (DEA) model, a qualitative and quantitative index system and a comprehensive diagnostic model (CDM) are construed. To testify rationality and usability of CDM, case studies of large-scaled Chinese thermal power plants have been conducted. In this case, CDM excavates such qualitative factors as technology, management, and so forth. The results shows that, compared with conventional model, which only considered production running parameters, the CDM bears better adaption to reality. It can provide entities with efficient instruments for energy efficiency diagnosis.

    Abstract: Under vertical load, the composite foundation may be damaged due to insufficient lateral restraint capacity. If the lateral restraint pile is added to the edge of the composite foundation, the lateral stability can be effectively enhanced. This new reinforcement scheme is called a combined composite foundation. For the combined composite foundation, the lateral restraint piles will bear a certain force when the composite foundation is loaded, and the force may cause the piles horizontal displacement, instability, or even failure. After preload on the composite foundation, the mechanical behavior of lateral restraint piles will be more sophisticated. To study the mechanical behavior of lateral restraint piles under such condition, a laboratory model test was conducted. The results are suggested as follows: Axial force distribution curves tend to be “S” shape with two peak values at the upper and lower parts, respectively. The negative axial force is distributed at the upper part, and the positive axial force is distributed at the lower part. The bending moment and lateral soil pressure distribution of piles appear with the regularity of “increase-decrease” along the depth. Such a phenomenon is associated with the expansion of a potential slip surface. In accordance with the relative location of the potential slide surface and the piles, the lateral restraint piles can fall into three sections. Different sections get different stresses. The reinforcement scopes have an impact on the stress of different lateral restraint piles. With the increase of the reinforcement scope, the axial force, soil pressure, and bending moment show the regularity of increase. The values of axial force, the lateral soil pressure, and the bending moment increase with load growth and become stable when the load is larger than 60 kN. This is associated with the piles in the composite foundation, which verifies the reinforcement effect to prevent the soil slide and the foundation deformation.

    Abstract: Caveolin-1 and flotillin-1 are considered as markers of lipid rafts which can be regarded as sorting platforms for targeted transport of transmembrane proteins and are involved in fundamental cellular events such as signal transduction, cell adhesion, lipid/protein sorting, and human cancer. We addressed caveolin-1 and flotillin-1 expression in 90 human hepatocellular carcinoma (HCC) and adjacent noncancerous tissues (ANT) samples by SDS-PAGE and immunoblotting with specific antibodies. Significant caveolin-1 and flotillin-1 overexpression was found in HCC tissues compared to ANT and was confirmed by immunohistochemistry. Raft-associated Akt signaling pathway components involved in the regulation of cell survival were altered by western blotting in HCC microdomain-enriched subcellular fractions purified from paired HCC and ANT samples. Our results demonstrated that the activity of raft-associated but not total membrane Akt determines its cellular functions. Lipid rafts differ in different types of tissues, which allows for the possibility of tissue-type-specific targeting for cell survival.

    Abstract: Calorie restriction (CR) is a nongenetic intervention with a robust effect on delaying aging in mammals and other organisms. A mild stimulation on mitochondrial biogenesis induced by CR seems to be an important action mode for its benefits. Here, we reported that a component isolated from Rhodiola rosea L ., salidroside, delays replicative senescence in human fibroblasts, which is related to its stimulation on mitochondrial biogenesis by activating SIRT1 partly resulted from inhibition on miR-22. Salidroside increased the mitochondrial mass that accompanied an increment of the key regulators of mitochondrial biogenesis including PGC-1 α , NRF-1, and TFAM and reversed the mitochondrial dysfunction in presenescent 50PD cells, showing a comparable effect to that of resveratrol. SIRT1 is involved in the inducement of mitochondrial biogenesis by salidroside. The declined expression of SIRT1 in 50PD cells compared with the young 30PD cells was prevented upon salidroside treatment. In addition, pretreatment of EX-527, a selective SIRT1 inhibitor, could block the increased mitochondrial mass and decreased ROS production induced by salidroside in 50PD cells, resulting in an accelerated cellular senescence. We further found that salidroside reversed the elevated miR-22 expression in presenescent cells according to a miRNA array analysis and a subsequent qPCR validation. Enforced miR-22 expression by using a Pre-miR-22 lentiviral construct induced the young fibroblasts (30PD) into a senescence state, accompanied with increased senescence-related molecules including p53, p21, p16, and decreased SIRT1 expression, a known target of miR-22. However, salidroside could partly impede the senescence progression induced by lenti-Pre-miR-22. Taken together, our data suggest that salidroside delays replicative senescence by stimulating mitochondrial biogenesis partly through a miR22/SIRT1 pathway, which enriches our current knowledge of a salidroside-mediated postpone senility effect and provides a new perspective on the antidecrepitude function of this naturally occurring compound in animals and humans.

    Abstract: Background. Visceral adiposity plays a key role in the development of insulin resistance (IR), so surrogate index that can indicate visceral obesity may have higher predictive value for IR. This study aimed to establish and validate a new predictive model including indicator of visceral obesity for IR. Methods. The study population consisted of two cohorts. The derivation cohort was a group of 667 patients with newly diagnosed type 2 diabetes and the population undergoing a routine health checkup was the validation cohort. The predictive model was established by the logistic regression analysis. Its value for predicting IR was compared with other surrogate indices by the receiver operating characteristic curve. Results. The odds ratio (OR) of age, visceral fat area (VFA), triglyceride (TG), fasting plasma glucose (FPG), and alanine aminotransferase (ALT) for IR was 1.028 (95% CI, 1.008–1.048) ( P 〈 0.01 ), 1.016 (95% CI, 1.009–1.023) ( P 〈 0.001 ), 1.184 (95% CI, 1.005–1.396) ( P 〈 0.05 ), 1.334 (95% CI, 1.225–1.451) ( P 〈 0.001 ), and 1.021 (95% CI, 1.001–1.040) ( P 〈 0.05 ). The formula of the predictive model was (0.0293 × age + 1.4892 × Ln VFA + 0.4966 × Ln TG + 2.784 × Ln FPG + 0.6906 × Ln ALT)/2. The area under the curve was the largest among all the previously reported predictors. Conclusions. This study established and validated a predicting model for IR and confirmed its predictive value in comparison with other surrogate indicators, which will offer a simple and effective tool to measure IR in future large population studies.
