    Abstract: MicroRNAs (miRNAs) are deregulated in various vascular ailments including abdominal aortic aneurysm (AAA). MiR-103 is involved in vascular, metabolic, and malignant diseases, but whether it participates in the pathogenesis of AAA remains elusive. ADAM10 plays a vital role in the formation of aneurysm, but whether miRs modulate its activity during AAA formation is totally unknown. In this study, we detected the significantly increased protein expression of ADAM10 in angiotensin II induced murine AAA specimens, while the mRNA expression of ADAM10 was similar between AAA and control, suggesting the posttranscriptional regulation. The ADAM10 specific inhibitor GI254023X dramatically reduced the macrophage infiltration of murine abdominal aorta. Bioinformatic predictions suggest that ADAM10 is the target of miR-103a/107 but the binding site is exclusive. At the cellular level, miR-103a-1 suppressed the protein expression of ADAM10, while antisense miR-103a-1 increased its expression. Particularly, with the progression of murine AAA, the mRNA expression of miR-103a/107 substantially decreased and the protein expression of ADAM10 greatly increased. Together, our data afford the new insight that miR-103a inhibited AAA growth via targeting ADAM10, which might be a promising therapeutic strategy to alleviate AAA.

    Abstract: Objectives . To systematically evaluate the efficacy and safety of sinomenine preparation (SP) for treating ankylosing spondylitis (AS). Methods . Clinical randomized controlled trials (RCTs) of SP for treating AS were systematically identified in six electronic databases including PubMed, Embase, Cochrane Library, China National Knowledge Infrastructure (CNKI), Chinese Scientific Journal Database (VIP), and Wanfang Databases from the inception up to 31 October 2019. Cochrane’s risk of bias tool was used to assess the methodological quality and Review Manager 5.3 software was used to analyze data. Results . A total of 12 RCTs involving 835 patients were finally included. According to interventions, RCTs were divided into two types. The intervention in 10 RCTs was SP combined with conventional pharmacotherapy (CPT) versus CPT and that in 2 RCTs was SP alone versus CPT. The results of the meta-analysis showed that, compared with CPT alone, SP combined with oral CPT has better improvement in BASDAI (WMD = −1.84, 95% CI [−3.31, −0.37], P = 0.01 ), morning stiffness time (WMD = −13.46, 95% CI [−16.12, −10.79], P 〈 0.00001 ), the Schober test (WMD = 1.26, 95% CI [0.72, 1.80], P 〈 0.00001 ), the occipital wall test (WMD = −0.55, 95% CI [−0.96, −0.14], P = 0.009 ), the finger-to-ground distance (WMD = −3.28, 95% CI [−5.64, −0.93], P = 0.006 ), 15 m walking time (WMD = −8.81, 95% CI [−13.42, −4.20], P = 0.0002 ), the C-reactive protein (CRP) (WMD = −1.84, 95% CI [−3.24, −0.45], P = 0.01 ), and the total effective rate (RR = 1.10, 95% CI [1.01, 1.20], P = 0.03 ). Besides, it also showed that oral SP alone may be more effective in improving morning stiffness time (WMD = −31.89, 95% CI [−34.91, −28.87], P 〈 0.00001 ) compared with CPT alone. However, this study cannot provide evidence that loading the injectable SP based on CPT can significantly increase the efficacy due to the insufficient number of studies included. In terms of adverse events, there was no statistically significant difference between the experimental group and the control group. Conclusions . This study shows that oral SP may be effective and safe in the treatment of AS. Due to the low methodological quality of the included RCTs and the limitations of the meta-analysis, it is still necessary to carry out more multicenter, large-sample, and high-quality RCTs to further verify the conclusions. The review protocol was registered on PROSPERO ( CRD42018099170 ), and the review was constructed following the PRISMA guidelines (Annex 1).

    Abstract: Background . Acute coronary syndromes (ACSs) are clinically cardiovascular events associated with dyslipidemia in common. Single nucleotide polymorphisms (SNPs) and haplotypes in the APOA1/C3/A5 gene cluster are associated with diabetes and familial combined hyperlipidaemia (FCH). Little is known about whether the polymorphisms in these genes affect lipid homeostasis in patients with ACSs. The present paper aimed to examine these associations with 4 SNPs in the APOA1 −75 G 〉 A , the APOC3 −455 T 〉 C , and APOA5 −1131 T 〉 C , c.553 G 〉 T variant to ACSs in Chinese Han. Methods . Chinese Han of 229 patients with ACSs and 254 unrelated controls were analyzed. Four SNPs in APOA1/C3/A5 cluster were genotyped and lipid was determined. Results . Our data show that minor allelic frequencies of APOC3 −455 T 〉 C , APOA5 −1131 T 〉 C , and c.553 G 〉 T polymorphisms in patients with ACSs were significantly higher than control group ( P 〈 0.05 ). Furthermore, the 3 polymorphic sites were strongly of linkage disequilibrium, and minor alleles of 3 SNP sites had higher TG level than wild alleles ( P 〈 0.05 ), APOC3 −455C and APOA5 c.553T allele carriers also had lower level of HDL-C. Conclusions . The minor alleles of APOC3 −455 T 〉 C , APOA5 −1131 T 〉 C , and c.553 G 〉 T polymorphisms are closely associated with ACSs.

    Abstract: Therapeutic interventions in prediabetes are important in the primary prevention of type 2 diabetes (T2D) and its chronic complications. However, little is known about the pharmacogenetic effect of traditional herbs on prediabetes treatment. A total of 194 impaired glucose tolerance (IGT) subjects were treated with traditional hypoglycemic herbs (Tianqi Jiangtang) for 12 months in this study. DNA samples were genotyped for 184 mutations in 34 genes involved in drug metabolism or transportation. Multinomial logistic regression analysis indicated that rs1142345 ( A 〉 G ) in the thiopurine S-methyltransferase (TPMT) gene was significantly associated with the hypoglycemic effect of the drug ( P = 0.001 , FDR P = 0.043 ). The “G” allele frequencies of rs1142345 in the healthy (subjects reverted from IGT to normal glucose tolerance), maintenance (subjects still had IGT), and deterioration (subjects progressed from IGT to T2D) groups were 0.094, 0.214, and 0.542, respectively. Binary logistic regression analysis indicated that rs1142345 was also significantly associated with the hypoglycemic effect of the drug between the healthy and maintenance groups ( P = 0.027 , OR = 4.828 ) and between the healthy and deterioration groups ( P = 0.001 , OR = 7.811 ). Therefore, rs1142345 was associated with the clinical effect of traditional hypoglycemic herbs. Results also suggested that TPMT was probably involved in the pharmacological mechanisms of T2D.

    Abstract: Conventional methods for fault diagnosis typically require a substantial amount of training data. However, for equipment with high reliability, it is arduous to form a large-scale well-annotated dataset due to the expense of data acquisition and costly annotation. Besides, the generated data have a large number of redundant features which degraded the performance of models. To overcome this, we proposed a feature transfer scenario that transfers knowledge from similar fields to enhance the accuracy of fault diagnosis with small sample. To reduces the redundant information, data were filtered according to manifold consistency. Then, features were extracted based on CNN and feature transfer was conducted. For adequate fitness, the joint adaptation of conditional distribution and marginal distribution was used between the two domains. Minimum structural risk and MMD of adaptation were two indicators weighted for training the model. To test the efficiency of the model, we built an airborne fuel pump testbed, and contributed a new dataset that contained 15 categories of fault data, which serves as the small sample dataset in this research. Then the proposed model was applied in our experimental data. As a result, the fault diagnosis rate increases by 28.6% through our proposed model, which is more precise than other classical methods. The results of feature visualization further demonstrate that the features are more distinguished through the proposed method. All code and data are accessible on my GitHub.

    Abstract: Phytosterols (PS) have been shown to regulate cholesterol metabolism and alleviate hyperlipidemia (HLP), but the mechanism is still unclear. In this study, we investigated the mechanism by which PS regulates cholesterol metabolism in high-fat diet (HFD) mice. The results showed that PS treatment reduced the accumulation of total cholesterol (TC), triglycerides (TG), and low-density lipoprotein cholesterol (LDL-C) in the serum of HFD mice, while increasing the serum levels of high-density lipoprotein cholesterol (HDL-C). Compared with HFD mice, PS not only increased the antioxidant activity of the liver but also regulated the mRNA expression levels of enzymes and receptors related to cholesterol metabolism. The hypolipidemic effect of PS was abolished by antibiotic (Abx) intervention and reproduced by fecal transplantation (FMT) intervention. The results of 16S rRNA sequencing analysis showed that PS modulated the gut microbiota of mice. PS reduced the relative abundance of Lactobacillus and other bile salt hydrolase- (BSH-) producing gut microbiota in HFD mice, which are potentially related to cholesterol metabolism. These findings partially explain the mechanisms by which PS regulates cholesterol metabolism. This implies that regulation of the gut microbiota would be a potential target for the treatment of HLP.

    Abstract: This paper addresses the problem of open gaps caused by differential settlement in the process of constructing sluice buildings in soft soil beach areas, combined with the construction of sluice and supporting facilities in a reclamation project. First, the change rules for the shear strength and compression modulus of soft soil under different consolidation degrees are studied by theoretical analysis. Then, an interaction model for soft soil and pile soil is established using the geotechnical finite element analysis software MIDAS/GTS NX. The change rules for the vertical and horizontal ultimate bearing capacities of a single pile with the degree of soil consolidation are studied. On this basis, a three-dimensional numerical analysis model of drainage sluice, seawall, cofferdam, and foundation soil is established, and the relationship between the degree of soil consolidation and the development of structural gaps caused by differential settlement is obtained. The research results show that the bearing capacity of a single pile increases greatly with the consolidation of soil around the pile and that the gap width between the structures in the project decreases with increasing consolidation. This paper provides a theoretical basis for the prediction of pile bearing capacity in the preliminary design stage and the evaluation and calculation of differential settlement of intersecting buildings in soft soil beach areas.

    Abstract: Introduction . Gene signature has been used to predict prognosis in melanoma patients. Meanwhile, the efficacy of immunotherapy was correlated with particular genes expression or mutation. In this study, we systematically explored the gene expression pattern in the melanoma-immune microenvironment and its relationship with prognosis. Methods . A cohort of 122 melanoma cases with whole-genome microarray expression data were enrolled from the Gene Expression Omnibus (GEO) database. The findings were validated using The Cancer Genome Atlas (TCGA) database. A principal component analysis (PCA), gene set enrichment analysis (GSEA), and gene oncology (GO) analysis were performed to explore the bioinformatic implications. Results . Different gene expression patterns were identified according to the clinical stage. All eligible gene sets were analyzed, and the 8 genes (GPR87, KIT, SH3GL3, PVRL1, ATP1B1, CDAN1, FAU, and TNFSF14) with the greatest prognostic impact on melanoma. A gene-related risk signature was developed to distinguish patients with a high or low risk of an unfavorable outcome, and this signature was validated using the TCGA database. Furthermore, the prognostic significance of the signature between the classified subgroups was verified as an independent prognostic predictor of melanoma. Additionally, the low-risk melanoma patients presented an enhanced immune phenotype compared to that of the high-risk gene signature patients. Conclusions . The gene pattern differences in melanoma were profiled, and a gene signature that could independently predict melanoma patients with a high risk of poor survival was established, highlighting the relationship between prognosis and the local immune response.
