    Abstract: Dax1(also known as Nr0b1) is regarded as an important component of the transcription factor network in mouse embryonic stem cells (ESCs). However, the role and the molecular mechanism of Dax1 in the maintenance of different pluripotency states are poorly understood. Here, we constructed a stable Dax1 knockout (KO) cell line using the CRISPR/Cas9 system to analyze the precise function of Dax1. We reported that 2i/LIF-ESCs had significantly lower Dax1 expression than LIF/serum-ESCs. Dax1KO ES cell lines could be established in 2i/LIF and their pluripotency was confirmed. In contrast, Dax1-null ESCs could not be continuously passaged in LIF/serum due to severe differentiation and apoptosis. In LIF/serum, the activities of the Core module and Myc module were significantly reduced, while the PRC2 module was activated after Dax1KO. The expression of most proapoptotic genes and lineage-commitment genes were drastically increased, while the downregulated expression of antiapoptotic genes and many pluripotency genes was observed. Our research on the pluripotent state-dependent role of Dax1 provides clues to understand the molecular regulation mechanism at different stages of early embryonic development.

    Abstract: Objective. As a first-line treatment for non-small cell lung cancer (NSCLC), the efficacy of chemotherapy is still unsatisfactory. Moxibustion has been shown to improve the side effects of radiotherapy and chemotherapy and regulate immune function. This study aimed to explore the antitumor effects and potential mechanisms of combinatorial cisplatin and moxibustion treatment for NSCLC by targeting the tumor microenvironment. Methods. Lewis lung cancer (LLC)-bearing mice were induced and treated with cisplatin or/and moxibustion at ST36 (Zusanli), and the growth, weight, and area of the tumor were evaluated. The numbers of various T cell subsets and myeloid cells in the tumor were assessed by flow cytometry, and the gene expression of related markers and cytokines was detected with real-time quantitative polymerase chain reaction (RT-qPCR). In addition, the tumor vascular structure was investigated using CD31 and α-smooth muscle actin (α-SMA) immunofluorescence staining. The expression of the vascular endothelial growth factor (VEGF) and hypoxia-inducible factor-1α (HIF-1α) was detected by immunohistochemical staining. Results. Both cisplatin and moxibustion inhibited LLC tumor growth and reduced both the tumor area and weight, with the combinatorial therapy showing superior outcomes. Moxibustion upregulated the infiltration of CD4+ T cells and Th1 cells in the tumor, and the combinatorial therapy increased the proportion of CD8+ cytotoxic T cells (CTLs), CD4+T cells, Th1, Th9 cells, and M1 macrophages, as well as the expression of Cd69, Ifng, and Cd86 mRNA. The combinatorial therapy improved vascular normalization by increasing both the microvessel density (MVD) and pericyte coverage (α-SMA area density) and inhibiting the expression of the VEGF. Conclusions. Combinatorial cisplatin and moxibustion treatment inhibited the LLC tumor growth by mechanisms related to the improvement of the tumor immune microenvironment and vascular normalization, providing an effective combinatorial therapy beneficial for patients with NSCLC.

    Abstract: Background . Steroid-induced osteonecrosis of the femoral head is a relatively serious condition which seriously reduces patient quality of life. However, the pathogenesis of steroid-induced ONFH is still unclear. In recent years, more scholars have found that the pathogenesis of steroid-induced ONFH is related to susceptibility factors such as MMPs / TIMPs system. The main purpose of this study is to investigate the correlation between MMP2 and MMP10 gene polymorphisms and steroid-induced ONFH in Chinese Han population. Methods . Six SNPs in MMP2 and two SNPs in MMP10 were genotyped using Agena MassARRAY RS1000 system from 286 patients of steroid-induced ONFH and in 309 healthy controls. The association between MMP2 and MMP10 polymorphisms and steroid-induced ONFH risk were estimated by the Chi-squared test, genetic model analysis, haplotype analysis, and stratification analysis. The relative risk was estimated by odd ratios (ORs) and 95% confidence intervals (CIs). Result . We found that the minor TG allele of rs470154 in MMP10 was associated with an increased risk of steroid-induced ONFH (OR = 1.45, 95% CI, 1.03 – 2.05, p = 0.032). In the genetic model analysis, we found that rs2241146 in MMP2 gene and rs470154 in MMP10 gene showed a statistically significant association with increased risk of steroid-induced ONFH. The six SNPs (rs470154, rs243866, rs243864, rs865094, rs11646643, and rs2241146) showed a statistically significant association with different clinical phenotypes. Conclusion . Our results verify that genetic polymorphisms of MMP2 and MMP10 contribute to steroid-induced ONFH susceptibility in the population of Chinese Han population, and our study provides new insights into the role that MMP2 and MMP10 plays in the mechanism of ONFH.

    Abstract: As one of the most catastrophic dynamic hazards in underground coal mines, coal bursts have been a major safety concern around the world for many years. Although the coal bursts can occur in all cases of hard to soft coal if the right stress environment is created, the occurrence of coal bursts is closely related to the intrinsic mechanical properties of coal, such as the bursting proneness. In this study, a total of 27 coal specimens are selected in the open literature studies to obtain a group of fundament data, such as the mechanical parameters, four bursting proneness indices, stress-strain curves, and their geological conditions where the specimens were taken. The relationship between bursting proneness indices and the cohesion of the coal specimens is established by numerically fitting the stress-strain curves and theoretically deduction. By taking into account the coal heterogeneity, eight probability distribution functions are employed to assignment nonuniform cohesion to the numerical model and to study the influence of heterogeneity on bursting proneness. The results reveal that the coal cohesion, which combines the common advantages of the four proneness indices, can be used as bursting proneness index. In the research of heterogeneity, the coal bursting proneness will decrease with the increasing of cohesion scatter degree. The larger the cohesion scatter degree increase is, the lower the bursting proneness will be. The failure of coal specimen is more and more severe with the decrease of cohesion scatter degree. In addition, this paper provides two methods for assigning heterogeneous parameters to the numerical model. The contours of shear strain rate and plastic state between homogeneous and heterogeneous coal specimens are compared to study the failure types of coal specimens and to reveal the mechanism of violent failure in coal bursts.

    Abstract: To solve the problems of surface deformation and destruction of buildings caused by urban mining and realise the coordinated development of mining cities, the solid backfilling method was used to extract coal resources beneath the buildings of Tangshan. Based on surface deformation monitoring data of the continuously operating reference station (CORS) system for the past 5 years, the surface deformation process caused by solid backfilling was analysed. The final results revealed a maximum surface subsidence of 66 mm in the T zone coal area and 31 mm in the F zone area. Furthermore, the surface control effects of the caving method and the solid backfilling method were compared and analysed, and it was shown that solid backfilling could meet the surface building setup requirements. Moreover, based on the probability integral method, the effects on surface deformation due to the surface length of the F zone, compression ratio, and coal pillar width were analysed, and the effects on the prediction results due to the subsidence factor, tangent of the major effective angle, and offset distance of the inflection point were studied. The results showed that the compression ratio is the main factor controlling the surface deformation and that it should be kept above 80% for solid backfilling of urban mines. The subsidence factor should be 0.82, and the tangent of the major effective angle should be 2.15 when the surface subsidence of solid backfilling is to be predicted. This paper provides a technical reference for the realisation of urban mining with solid backfilling.

    Abstract: In this paper, the strain rate effect on mechanical properties, failure modes, acoustic emission (AE) characteristics, and energy mechanism of the karst limestone was analyzed based on uniaxial compression tests with different strain rates (5 × 10 −6 –5 × 10 −4 /s). The results showed that the peak strength increased linearly and peak strain increased quadratically with the logarithm value of the strain rate. Moreover, the strain rate effect on elastic modulus was not significant. Under low strain rates, the rock was damaged seriously, AE signals appeared continuously, and the cumulative number of AE signals was high. Under high strain rates, the total quantity of the macroscopic cracks decreased, but the crack length extended with better coalescence. The AE peak significantly increased under high strain rates, while the cumulative AE activity significantly reduced. The energy evolution of the karst limestone failure process had significant stage characteristics, and the strain energy ratio presented an S-shape. The maximum value of the elastic strain energy at peak stress showed a linear relationship with the logarithm value of the strain rate.

    Abstract: Background. Mortality after percutaneous coronary intervention (PCI) in ST-elevation myocardial infarction (STEMI) patients with cardiogenic shock (CS) remains high. However, the real-world risk factors for mortality in these patients are poorly defined. Objective. The aim of this study is to establish a clinical prognostic nomogram for predicting in-hospital mortality after primary PCI in STEMI patients with CS. Methods. This retrospective, multicenter, observational study included STEMI patients with CS who underwent PCI at 39 hospitals in Hebei Province from January 2018 to December 2019. A multivariate logistic regression model was used to identify the factors associated with in-hospital mortality. These factors were then incorporated into a nomogram and its performance was evaluated by discrimination, calibration, and clinical utility. Results. This study included 274 patients, among whom 179 died in hospital. Sex, random blood glucose on admission, ejection fraction after PCI, no-reflow, and intra-aortic balloon pump (IABP) were independently associated with in-hospital mortality (all P 〈 0.05 ). In the training set, the nomogram showed a C-index of 0.819, goodness-of-fit of 0.08, and area under the receiver operating characteristic curve (AUC) of 0.819 (95%CI = 0.759–0.879). In the testing set, the C-index was 0.842, goodness-of-fit was 0.585, and AUC was 0.842 (95%CI = 0.715–0.970). The results indicate that the nomogram had good discrimination and good prediction accuracy and could achieve a good net benefit. Conclusion. We established and validated a nomogram that provided individual prediction of in-hospital mortality for STEMI patients with CS after PCI in a Chinese population.

    Abstract: Imaging methods for gastrointestinal diseases were based on X-ray imaging until the 1970s, but the development of fiberoptic endoscopy in the 1980s has replaced X-ray imaging. Endoscopy can directly observe the location, size, scope, and color of lesions and obtain pathological results through biopsy, while ligation and other treatments can be performed on polyps and other lesions. Studies have shown that multilayer spiral computed tomography (CT) examination after standardized gastrointestinal preparation and full use of the advantages of various 3D postprocessing reconstruction techniques are of great clinical value in the detection of gastrointestinal diseases, determination of the nature of lesions, localization of lesions, and staging of gastrointestinal malignancies and can make up for the shortcomings of fiberoptic endoscopy, and various 3D postprocessing reconstruction modes have their own advantages and disadvantages. Among them, conventional CT cross-sectional images are the basic images for the diagnosis of various gastric testicular lesions. Axial images, especially thin-layer axial images, can detect the absolute majority of lesions, but there are limitations in observing the anatomical position of lesions, invasion of surrounding tissues, lymph node metastasis, vascularity, and determination of the stage of malignant tumors.
