    Abstract: Type 2 diabetes (T2D) is the most prevalent metabolic disease, and many people are suffering from its complications driven by hyperglycaemia and dyslipidaemia. Nuclear receptors (NRs) are ligand-inducible transcription factors that mediate changes to metabolic pathways within the body. As metabolic regulators, the farnesoid X receptor (FXR) and the liver X receptor (LXR) play key roles in the pathogenesis of T2D, which remains to be clarified in detail. Here we review the recent progress concerning the physiological and pathophysiological roles of FXRs and LXRs in the regulation of bile acid, lipid and glucose metabolism and the implications in T2D, taking into account that these two nuclear receptors are potential pharmaceutical targets for the treatment of T2D and its complications.

    Abstract: The influence of acid solution and immersion time on the physicomechanical properties of sandstone is investigated. Uniaxial compression tests on sandstone samples are conducted to determine the variations of relative mass, deformation, and strength characteristics of sandstone subjected to different pH sulfuric acid corrosion values. The changes of pH and Mg 2+ and Ca 2+ concentration of immersion solutions are monitored during soaking. The corrosion mechanism of sandstone attacked by the acid solution is discussed with the results of SEM tests. Based on the nondestructive CT scanning test, the damage variables of acid-corroded sandstone are deduced, and the damage degree of sandstone is quantitatively analyzed. The results indicate that the deformation characteristics of sandstone samples under acid attack are characterized by the softening of rock, and the softening degree gradually increases with the increase of the acidity and the soaking time. The peak strength of sandstone samples declines as soaking time extends. The chemical effects lead to a large amount of dissolution of the rock mineral assemblage, resulting in the large-scale development of the pores inside the rock, which changes the macroscopic mechanical properties. The damage variables of acid corrosion sandstone based on CT numbers are deduced, and the quantitative relationship between damage variables and immersion time is established, which provides a basis for constructing a damage constitutive model of sandstone in the acidic environment.

    Abstract: This paper takes the Dizong tunnel engineering as its background. Combined with the on-site monitoring data, the wavelet packet program based on MATLAB was compiled to study the vibration response of the four-story masonry building in a typical southwestern mountainous area of China under the blasting load. The results showed that the maximum particle velocity increased to the 3rd floor and attenuation occurred on the 4th floor. The particle velocity in the z -direction was the largest and should be paid attention. The dominant frequency of the building showed a trend from high frequency to low frequency, the duration became short, and the acceleration decreased to the 4th floor. With the increase of the building floor, the main frequency domain of the building decreased and then gradually tended to the low-frequency domain. The high-frequency particle velocity gradually decreased, gathered to the low frequency, and developed from the dispersed multiband to the concentrated low-frequency band. The total energy value of vibration increased to the 3rd floor and then decreased to the 4th floor. The energy of the building was between 0 and 171.6 Hz. The higher the floor was, the more concentrated the energy was in the low-frequency domain.

    Abstract: Background . Mucin 5AC (MUC5AC) overproduction plays important roles in stone formation and recurrence of hepatolithiasis. We aim to investigate the involved mechanism and the potential target to block this process. Methods . 42 bile duct samples from hepatolithiasis and 15 normal bile duct samples from hemangioma patients were collected for detecting MUC5AC expression by immunohistochemistry. MUC5AC and phosphoepidermal growth factor receptor (pEGFR) expressions in human intrahepatic biliary epithelial cells (HIBECs) cultured with or without lipopolysaccharide (LPS) were detected by real-time PCR and western blot analysis. Transforming growth factor- α (TGF- α ) secretion in HIBECs was detected by ELISA. Results . MUC5AC was overexpressed in bile ducts of hepatolithiasis samples compared with bile ducts from hemangioma samples. LPS upregulated MUC5AC expression in HIBECs. LPS promoted EGFR activation, and inhibiting EGFR activation by AG1478 significantly decreased LPS-induced MUC5AC overexpression in HIBECs. Moreover, LPS increased TGF- α secretion, and inhibiting tumor necrosis factor- α converting enzyme (TACE), which has been implicated in ectodomain cleavage of TGF- α , significantly inhibited LPS-induced EGFR activation and subsequent MUC5AC overexpression in HIBECs. Conclusion . Our results suggested that LPS increases MUC5AC expression through the TACE/TGF- α /EGFR pathway in HIBECs. This new finding might give light to the prevention of stone formation and recurrence of hepatolithiasis.

    Abstract: The aim of this study was to explore cervical carcinoma and screen a suitable gene as the biomarker used for prognosis evaluation as well as pain therapy. Low expression levels of solute carrier family 24 member 3 (SLC24A3) was involved in the appearance and development of numerous malignancies. Nevertheless, the prognostic value of SLC24A3 expression with cervical squamous cell carcinoma and endocervical adenocarcinoma (CESC) patients remains uncertain. During the present study, SLC24A3 expression in CESC was retrieved from TCGA, GEO, and MSigDB databases. Based on TCGA and GEO profiles, we performed survival and difference analyses about SLC24A3 both in two GEO (GSE44001 and GSE63514) and TCGA-CESC cohorts (all p 〈 0.05 ), indicating that SLC24A3 was low expressed in tumors and associated with higher overall survival in CESC patients. Additionally, we programmed a series of analyses, including genomic profiling, enrichment analysis, immune infiltration analysis, and therapy-related analysis to identify the mechanism of the SLC24A3 in the process of cancer in CESC. Meanwhile, qRT-PCR was used to validate that the expression of SLC24A3 mRNA in Hela and SiHa cell lines was significantly lower than in PANC-1 and HUCEC cell lines. Our finding elucidated that the SLC24A3, a sodium-calcium regulator of cells, is an indispensable factor which can significantly influence the prognosis of patients with CESC and could provide novel clinical evidence to serve as a potential biological indicator for future diagnosis and pain therapy.

    Abstract: The photon-assisted magnetic recording utilizes the ultrafast laser to excite the spin wave in the magnetic nanostructures and accordingly switch its magnetization state. Here, by means of micromagnetic simulation, the motion of magnetic skyrmions, a topologically protected chiral magnet with few nanometer size, induced by the spin wave is studied. It is found that the magnetic skyrmion can move in the same direction of spin-wave propagation, which is first accelerated and then decelerated exponentially. The magnetic skyrmion motion originated from the robust coupling of the spin waves with the skyrmion, through the SW’s linear momentum transfer torque acting on the skyrmion. Besides amplitude, the reflectivity of the spin wave by skyrmion has tremendous impact on the velocity of skyrmion motion. The skyrmion velocities are mainly determined by the reflectivity, when the spin-wave amplitude is almost identical. Our results give guidance for the design and development of spin-wave control spintronics.

    Abstract: Objectives. To systematically collate, appraise, and synthesize the current evidence on the Xuebijing injection (XBJI) for sepsis. Methods. Eight databases were searched for systematic reviews (SRs) or meta-analyses (MAs) on XBJI for sepsis. Assessing the Methodological Quality of Systematic Reviews-2 (AMSTAR-2), Preferred Reporting Item for Systematic Reviews and Meta-Analyses (PRISMA), and Grading of Recommendations, Assessment, Development, and Evaluation (GRADE) methods were used to assess the methodological quality, reporting quality, and evidence quality of the enrolled studies, respectively. Results. Out of the 13 studies that were included, all studies were rated critically low quality based on AMSTAR-2 results. Based on the results obtained from PRISMA, all studies were reported to be over 80%, while the GRADE system yielded three outcome measures rated high-quality, 16 were of moderate quality, and the rest were of low or critically low quality. Conclusions. The combination of XBJI and Western medicine (WM) showed significant synergy for the treatment of sepsis compared to WM alone. However, this conclusion should be treated with caution since the quality of the SRs/MAs providing the evidence was relatively low.

    Abstract: Objectives . The aim of this study was to investigate the relationship between serum vitamin D and insulin resistance in Chinese subjects without diabetes mellitus. Methods . Serum 25(OH)D was measured in 897 individuals with normal glucose tolerance (NGT). Oral glucose tolerance tests (OGTTs) were conducted to exclude cases with diabetes, impaired fasting glucose (IFG), and impaired glucose tolerance (IGT). Metabolic parameters were measured and compared between the highest and lowest 25(OH)D quartiles. The relationship between serum 25(OH)D and homeostatic model assessment-insulin resistance (HOMA-IR) was analyzed. Results . Indexes, such as HOMA-IR, FINS, and SBP, were negatively correlated with serum 25(OH)D concentrations. Compared with the lowest quartile, individuals in the highest group had decreased Lg (HOMA-IR), Lg (FINS), and SBP. Pearson correlation analyses showed that serum 25(OH)D was negatively associated with age, BMI, Lg (HOMA-IR), and Lg (FINS). Multivariate linear regression analysis confirmed the negative correlation of Lg (HOMA-IR) and 25(OH)D. Conclusions . This study showed that serum 25(OH)D could be regarded as an independent predictor of insulin resistance for subjects without diabetes mellitus in China. Adequate vitamin D supplementation may improve multiple metabolic disturbances.

    Abstract: A method for classification of the tunnel wall rock is established based on the rough set theory and unascertained measurement theory. The saturated uniaxial compressive strength, rock mass integrity index, structural surface condition, seepage measurement of groundwater, and the angle between the hole axis and main structural surface are selected as the evaluation indexes. The problem of weight coefficients for these evaluation indexes is converted into that of significance estimating on the attributes in the rough set theory. The proposed method is verified by the wall rock data of Yuanyanghui tunnel. The results show that the proposed method has an excellent performance in good agreement with the practical situation of wall rock and is feasible to classify the wall rock. Finally, the proposed method is applied to Xihualing tunnel of Zhu-Yong Expressway. The results are basically the same as those from Delphi-ideal point method, set pair analysis method, and the actual situation, which proves that the rough set theory and unascertained measurement theory are effective for classification of the wall rock.

    Abstract: Hepatic ischemia-reperfusion injury (IRI) is the most common cause of liver damage leading to surgical failures in hepatectomy and liver transplantation. Extensive inflammatory reactions and oxidative responses are reported to be the major processes exacerbating IRI. The involvement of Yes-associated protein (YAP) in either process has been suggested, but the role and mechanism of YAP in IRI remain unclear. In this study, we constructed hepatocyte-specific YAP knockout (YAP-HKO) mice and induced a hepatic IRI model. Surprisingly, the amount of serum EVs decreased in YAP-HKO compared to WT mice during hepatic IRI. Then, we found that the activation of YAP increased EV secretion through F-actin by increasing membrane formation, while inhibiting the fusion of multivesicular body (MVB) and lysosomes in hepatocytes. Further, to explore the essential elements of YAP-induced EVs, we applied mass spectrometry and noticed CD47 was among the top targets highly expressed on hepatocyte-derived EVs. Thus, we enriched CD47+ EVs by microbeads and applied the isolated CD47+ EVs on IRI mice. We found ameliorated IRI symptoms after CD47+ EV treatment in these mice, and CD47+ EVs bound to CD172α on the surface of dendritic cells (DCs), which inhibited DC activation and the cascade of inflammatory responses. Our data showed that CD47-enriched EVs were released in a YAP-dependent manner by hepatocytes, which could inhibit DC activation and contribute to the amelioration of hepatic IRI. CD47+ EVs could be a potential strategy for treating hepatic IRI.
