    Abstract: Hyperlipidemia and its associated obesity, hepatic steatosis, and NAFLD are worldwide problems. However, there is no ideal pharmacological treatment for these. Therefore, the complementary therapies that are both natural and safe have been focused. Healthy foods, such as fruit vinegar, may be one of the best choices. In this study, we made a special medicinal fruit vinegar, Schisandra fruit vinegar (SV), and examined its lipid-lowering effects and the underlying mechanisms in a high-fat diet rat model. The results showed that SV significantly reduced the body weight, liver weight, liver index, the serum triglyceride (TG), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), free fatty acid (FFA), aspartate aminotransferase (AST), alanine aminotransferase (ALT), and malondialdehyde (MDA), increased the content of high-density lipoprotein cholesterol (HDL-C) and the activity of superoxide dismutase (SOD), upregulated the expressions of peroxisome proliferative activated receptor (PPAR- α ), peroxisomal acyl-coenzyme A oxidase 1 (ACOX1), and carnitine palmitoyltransferase 1 (CPT1) proteins, increased the contents of key component of antioxidant defense NF-E2-related factor 2 (NRF2) and its downstream heme oxygenase-1 (HO-1) protein, and downregulated the expression of Kelch-like ECH-associated protein 1 (KEAP1). These results suggest that SV has weight loss and lipid-lowering effects in HFD rats, which may be related to its upregulation of the expressions of β -oxidation -elated PPAR- α , CPT1, and ACOX1 and the regulation of the expressions of antioxidant pathway-related KEAP1-NRF2-HO-1. Therefore, all these data provide an experimental basis for the development of SV as a functional beverage which is safe, effective, convenient, and inexpensive.

    Abstract: Black rice anthocyanins (BRAs) have extremely high nutritional value and health care effects. This study investigated the intervention effect of BRAs on type 2 diabetes mellitus (T2DM) and the regulation effect on intestinal microbiota imbalance in T2DM rats. This study established successfully a T2DM model in a high-fat and high-glucose diet combined with streptozotocin (STZ). BRAs intervention reduced significantly the fasting blood glucose level of T2DM rats, improved the glucose tolerance of rats, reduced the blood lipid level and inflammation state, and repaired liver, oxidative stress, and other injuries. In addition, BRAs’s intervention enhanced the expression of phosphoinositol 3-kinase (PI3K)/protein kinase B (AKT), activated the expression of adenosine 5’-monophosphate-activated protein kinase(AMPK), and the downstream acetyl-CoA carboxylase (ACC) and carnitine palmitoyl transferase (CPT1) in the liver. 16S rRNA sequencing showed that BRAs significantly decreased the abundances of Bifidobacterium and Clostridiaceae_Clostridium, and promoted the abundances of Akkermansia and Lactobacillus. Accelerate the recovery of gut microbiota diversity. BRAs play an antidiabetic role by regulating the PI3K/AKT signaling pathway and intestinal microbiota in T2MD rats.

    Abstract: To study the effect of saturation on the freeze-thaw damage of fractured rocks in cold regions, five prefabricated oval double-hole red sandstones with different saturations were prepared. Three-dimensional images of rock fractures were obtained by CT technology. The freeze-thaw damage mechanism and critical saturation of the red sandstone were explored through the fracture propagation evolution and quantitative characterization of the pore structure. The experimental results show that the pore size distribution can reflect the complex pore structure. Pores can be divided according to their sizes: small pores, mesopores, and macropores; mesopores account for the largest proportion and more than 70% of the total, and the proportion of mesopores in high-saturation sandstone (90% or 100%) increases under the action of freeze-thaw cycle. This is also accompanied by a small increase in the proportion of macropores. An 80% critical saturation of the sandstone was obtained. In addition, prefabricated cracks make the ice separation mechanism more likely to occur. The low-saturation sandstone mainly damaged the fracture area during freeze-thaw cycles, while the prefabricated cracks provided a good seepage channel for high-saturation sandstone. The corresponding rock bridge area eventually demonstrated a connectivity trend. This study more realistically reflects the freeze-thaw damage of actual rock masses in cold regions and provides a theoretical basis for the prediction of rock mass engineering disasters in cold regions.

    Abstract: The South China Sea is China’s largest marginal sea area, and it is rich in oil and gas mineral resources; thus, estimating its sea level changes is of practical significance. Based on linear and nonlinear sea level change characteristics, this paper decomposes 1992–2019 monthly mean sea level anomaly time series in the South China Sea into trend, seasonal, and random terms. This paper compares the seasonal autoregressive integrated moving average (SARIMA) and Prophet models for estimating the trend and seasonal terms and the long short-term memory (LSTM) and radial basis function (RBF) models for estimating random terms, and the more suitable models were selected. A Prophet-LSTM combined model was developed based on the accuracy results. This paper uses the combined model to study the effect of known data length on the experimental results and determines the best prediction duration. The results show that the combined model is suitable for short-term and medium-term estimations of 12–36 months. The accuracy at 36 months is 0.962 cm, which proves that the combined model has high application value for estimating sea level changes in the South China Sea.

    Abstract: Gastric ulcer is one of the most common gastrointestinal diseases. Anwulignan (AN) is a major active component of Schisandra sphenanthera Rehd. This study was designed to evaluate the protective effect of AN against the acute gastric ulcer induced by HCl/ethanol in mice. The mice were given HCl/ethanol by gavage to establish an acute gastric ulcer model. Then, the serum and gastric tissue samples were taken for biochemical analyses. The results showed that the pretreatment with AN could significantly reduce the gastric ulcer index (GUI) and increase the ulcer inhibition rate, indicating that AN can protect against gastric ulcers. AN showed its antioxidant roles by decreasing the content of reactive oxygen species (ROS), malondialdehyde (MDA), and 8-hydroxydeoxyguanosine (8-OHdG) and increasing the activity of superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GSH-Px) and anti-inflammatory roles by decreasing the content of tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), interleukin-1β (IL-1β), and myeloperoxidase (MPO) and increasing the content of interleukin-2 (IL-2), interleukin-4 (IL-4), interleukin-10 (IL-10), prostaglandin E2 (PGE2), and nitric oxide (NO) in both serum and gastric tissue. Furthermore, AN also activated the NRF2/ARE signaling pathway and inhibited the MAPK/NF-κB signaling pathway. AN improves the acute gastric ulcer induced by HCl/ethanol in mice, which may be mainly through its antioxidant capacity and anti-inflammatory effect.

    Abstract: Pancreatic cancer remains one of the leading causes of cancer-related deaths worldwide. Pancreatic ductal adenocarcinoma (PDAC) is the most common type of pancreatic tumor. Many circular RNAs (circRNAs) have proven to play vital roles in the physiological and pathological processes of tumorigenesis; however, their biogenesis in PDAC remains unclear. In this study, the expression profiles of circRNAs from 10 PDAC tissues and their paired adjacent nontumor tissues were analyzed through RNA sequencing analysis. An enrichment analysis was employed to predict the functions of the differentially expressed circRNAs. Sequence alignment information and mRNA microarray projects were used to predict the RNA regulatory network. The knockdown of circRNAs by small interfering RNAs followed by wound healing and western blot assays was used to confirm their functions in a PDAC cell line. A total of 278 circRNAs were identified as differentially expressed in PDAC tissue. Of these, we found that hsa_circRNA_0007334 was significantly upregulated and may serve as a competing endogenous RNA to regulate matrix metallopeptidase 7 (MMP7) and collagen type I alpha 1 chain (COL1A1) by the competitive adsorption of hsa-miR-144-3p and hsa-miR-577 to enhance the expression and functions of MMP7 and COL1A1 in PDAC. In vitro experiments confirmed these results. The present study is the first to propose two regulatory pathways in PDAC: hsa_circRNA_0007334–hsa-miR-144-3p–MMP7 and hsa_circRNA_0007334–hsa-miR-577–COL1A1.

    Abstract: A calcium phosphate (CaP) coating on titanium surface enhances its biocompatibility, thus facilitating osteoconduction and osteoinduction with the inorganic phase of the human bone. Electrochemical deposition has been suggested as an effective means of fabricating CaP coatings on porous surface. The purpose of this study was to develop CaP coatings on a direct laser metal forming implant using electrochemical deposition and to investigate the effect of electrolytic concentration on the coating’s morphology and structure by X-ray diffraction, scanning electron microscopy, water contact angle analysis, and Fourier transform infrared spectroscopy. In group 10 −2 , coatings were rich in dicalcium phosphate, characterized to be thick, layered, and disordered plates. In contrast, in groups 10 −3 and 10 −4 , the relatively thin and well-ordered coatings predominantly consisted of granular hydroxyapatite. Further, the hydrophilicity and cell affinity were improved as electrolytic concentration increased. In particular, the cells cultured in group 10 −3 appeared to have spindle morphology with thick pseudopodia on CaP coatings; these spindles and pseudopodia strongly adhered to the rough and porous surface. By analyzing and evaluating the surface properties, we provided further knowledge on the electrolytic concentration effect, which will be critical for improving CaP coated Ti implants in the future.

    Abstract: Aim. To apply the network pharmacology method to screen the target of catalpol prevention and treatment of stroke, and explore the pharmacological mechanism of Catalpol prevention and treatment of stroke. Methods. PharmMapper, GeneCards, DAVID, and other databases were used to find key targets. We selected hub protein and catalpol which were screened for molecular docking verification. Based on the results of molecular docking, the ITC was used to determine the binding coefficient between the highest scoring protein and catalpol. The GEO database and ROC curve were used to evaluate the correlation between key targets. Results. 27 key targets were obtained by mapping the predicted catalpol-related targets to the disease. Hub genes (ALB, CASP3, MAPK1 (14), MMP9, ACE, KDR, etc.) were obtained in the key target PPI network. The results of KEGG enrichment analysis showed that its signal pathway was involved in angiogenic remodeling such as VEGF, neurotrophic factors, and inflammation. The results of molecular docking showed that ACE had the highest docking score. Therefore, the ITC was used for the titration of ACE and catalpol. The results showed that catalpol had a strong binding force with ACE. Conclusion. Network pharmacology combined with molecular docking predicts key genes, proteins, and signaling pathways for catalpol in treating stroke. The strong binding force between catalpol and ACE was obtained by using ITC, and the results of molecular docking were verified to lay the foundation for further research on the effect of catalpol on ACE. ROC results showed that the AUC values of the key targets are all 〉 0.5. This article uses network pharmacology to provide a reference for a more in-depth study of catalpol’s mechanism and experimental design.
