    Abstract: Background . p38 mitogen-activated protein kinase (MAPK) plays a crucial role in regulating signaling pathways implicated in inflammatory processes leading to diabetic nephropathy (DN). This study aimed to examine p38 MAPK activation in DN and determine whether beraprost sodium (BPS) ameliorates DN by inhibiting inflammation and p38 MAPK signaling pathway in diabetic rats. Methods . Forty male Sprague Dawley (SD) rats were randomly divided into the normal control group, type 2 diabetic group, and BPS treatment group. At the end of the 8-week experiment, we measured renal pathological changes and the activation of the p38 MAPK signaling pathway and inflammation. Result . After BPS treatment, renal function, 24-hour urine protein, lipid profiles, and blood glucose level were improved significantly; meanwhile, inflammation and the expression of p38 MAPK signaling pathway in the diabetic kidney were attenuated. Conclusions . BPS significantly prevented type 2 diabetes induced kidney injury characterized by renal dysfunction and pathological changes. The protective mechanisms are complicated but may be mainly attributed to the inhibition of the p38 MAPK signaling pathway and inflammation in the diabetic kidney.

    Abstract: Protein microarray-based approaches are increasingly being used in research and clinical applications to either profile the expression of proteins or screen molecular interactions. The development of high-throughput, sensitive, convenient, and cost-effective formats for detecting proteins is a necessity for the effective advancement of understanding disease processes. In this paper, we describe the generation of highly multiplexed, antibody-based, specific, and sensitive protein microarrays coupled with rolling-circle signal amplification (RCA) technology. A total of 150 cytokines were simultaneously detected in an RCA sandwich immunoassay format. Greater than half of these proteins have detection sensitivities in the pg/ml range. The validation of antibody microarray with human serum indicated that RCA-based protein microarrays are a powerful tool for high-throughput analysis of protein expression and molecular diagnostics.

    Abstract: Background. For locally advanced breast cancer (LABC) patients who remained unresectable after neoadjuvant systemic therapy (NST), radiotherapy (RT) is considered as an approach for tumor downstaging. In this study, we attempted to discuss the value of RT for patients with unresectable or progressive disease in the breast and/or regional nodes following NST. Methods. Between January 2013 and November 2020, the data for 71 patients with chemo-refractory LABC or de novo bone-only metastasis stage IV BC who received locoregional RT with or without surgical resection were retrospectively analyzed. Factors associated with tumor complete response (CR) were recognized using logistic regression. Locoregional progression-free survival (LRPFS) and progression-free survival (PFS) were calculated using the Kaplan–Meier method. The Cox regression model was applied to recognize the recurrence risk factors. Results. After RT, 11 patients (15.5%) achieved total cCR. Triple-negative subtype (TNBC) was associated with a lower total cCR rate compared with other subtypes ( p  = 0.033). 26 patients proceeded to surgery, and the operability rate was 36.6%. 1-year LRPFS and PFS were 79.0% and 58.0%, respectively, for the entire cohort. Surgical cases had an improved 1-year LRPFS ( p  = 0.015), but not 1-year PFS ( p  = 0.057), compared with definitive RT cases. Non-any cCR was the most prominent predictor of a shorter LRPFS ( p   〈  0.001) and PFS ( p  = 0.002) in the multivariate analysis. Higher TNM stage showed a trend toward a shorter LRPFS time ( p  = 0.058), and TNBC ( p  = 0.061) showed a trend toward a shorter PFS interval. Conclusions. This study demonstrated that RT was an effective tumor downstaging option for chemo-refractory LABC. For patients with favorable tumor regression, surgery following RT might bring survival benefits.

    Abstract: Background . Characteristics of mammographic density for Chinese women are understudied. This study aims to identify factors associated with mammographic density in China using a quantitative method. Methods . Mammographic density was measured for a total of 1071 (84 with and 987 without breast cancer) women using an automatic algorithm AutoDensity. Pearson tests examined relationships between density and continuous variables and t-tests compared differences of mean density values between groupings of categorical variables. Linear models were built using multiple regression. Results . Percentage density and dense area were positively associated with each other for cancer-free (r=0.487, p 〈 0.001) and cancer groups (r=0.446, p 〈 0.001), respectively. For women without breast cancer, weight and BMI (p 〈 0.001) were found to be negatively associated (r=-0.237, r=-0.272) with percentage density whereas they were found to be positively associated (r=0.110, r=0.099) with dense area; age at mammography was found to be associated with percentage density (r=-0.202, p 〈 0.001) and dense area (r=-0.086, p 〈 0.001) but did not add any prediction within multivariate models; lower percentage density was found within women with secondary education background or below compared to women with tertiary education. For women with breast cancer, percentage density demonstrated similar relationships with that of cancer-free women whilst breast area was the only factor associated with dense area (r=0.739, p 〈 0.001). Conclusion . This is the first time that mammographic density was measured by a quantitative method for women in China and identified associations should be useful to health policy makers who are responsible for introducing effective models of breast cancer prevention and diagnosis.

    Abstract: In this paper, we analyze the location-following processing of the image by successive approximation with the need for directed privacy. To solve the detection problem of moving the human body in the dynamic background, the motion target detection module integrates the two ideas of feature information detection and human body model segmentation detection and combines the deep learning framework to complete the detection of the human body by detecting the feature points of key parts of the human body. The detection of human key points depends on the human pose estimation algorithm, so the research in this paper is based on the bottom-up model in the multiperson pose estimation method; firstly, all the human key points in the image are detected by feature extraction through the convolutional neural network, and then the accurate labelling of human key points is achieved by using the heat map and offset fusion optimization method in the feature point confidence map prediction, and finally, the human body detection results are obtained. In the study of the correlation algorithm, this paper combines the HOG feature extraction of the KCF algorithm and the scale filter of the DSST algorithm to form a fusion correlation filter based on the principle study of the MOSSE correlation filter. The algorithm solves the problems of lack of scale estimation of KCF algorithm and low real-time rate of DSST algorithm and improves the tracking accuracy while ensuring the real-time performance of the algorithm.

    Abstract: Purpose . This study examines factors associated with outcomes following salvage radiation and systemic therapy for breast cancer patients who developed isolated locoregional recurrence (ILRR) after mastectomy alone, while focusing on the prognostic significance of constructed biologic subtype in this setting. Methods and Materials . 269 postmastectomy patients in total treated for ILRR were included. Cumulative incidence of locoregional control (LRC), distant metastasis (DM)-free survival (DMFS), disease-free survival (DFS), and overall survival (OS) were calculated using Kaplan-Meier method. For statistical analysis, biologic subtypes were constructed from hormonal receptors (Rec) and HER2, consisting of Rec+/HER2-, Rec+/HER2+, Rec-/HER2+, and Rec-/HER2-. The association of clinic-pathological and treatment-related parameters with outcomes was evaluated using a Cox regression model. Results . At a median follow-up of 65 months, 56 (20.8%) patients failed to secure LRC after radiotherapy, and 165 patients (61.3%) developed DM. Overall, the actuarial 5-year LRC, DMFS, DFS, and OS rate was 77.3%, 45.6%, 43.9%, and 66.8%, respectively. Multivariate analysis revealed that constructed biologic subtype represented the most significant prognostic factor for any outcome. Compared to patients with Rec+/HER2- disease, those with Rec-/HER2- had significantly poorer 5-year LRC (84.2% versus 58.3%, HR = 4.36, P 〈 0.001) and worse survivals including 5-year DMFS (63.0% versus 15.8%, HR = 4.28, P 〈 0.001), DFS (59.7% versus 13.6%, HR=3.92, P 〈 0.001), and OS (87.8% versus 22.3%, HR = 8.55, P 〈 0.001). Other factors associated with reduced LRC were no radical surgery and involved field irradiation alone, whereas factors associated with poor survivals included positive nodes at primary diagnosis and regional recurrence. Conclusions . Constructed biologic subtypes remained to be predictive of both disease control and survivals after salvage radiation for postmastectomy ILRR. Notably, Rec-/HER2- patients were demonstrated to be at high risk of locoregional failure and subsequent DM and tended to have worse survivals despite salvage therapies.

    Abstract: Based on the simple ocean data assimilation (SODA) data, this study analyzes and forecasts the monthly sea surface height anomaly (SSHA) averaged over South China Sea (SCS). The approach to perform the analysis is a time series decomposition method, which decomposes monthly SSHAs in SCS to the following three parts: interannual, seasonal, and residual terms. Analysis results demonstrate that the SODA SSHA time series are significantly correlated to the AVISO SSHA time series in SCS. To investigate the predictability of SCS SSHA, an exponential smoothing approach and an autoregressive integrated moving average approach are first used to fit the interannual and residual terms of SCS SSHA while keeping the seasonal part invariant. Then, an array of forecast experiments with the start time spanning from June 1977 to June 2007 is performed based on the prediction model which integrates the above two models and the time-independent seasonal term. Results indicate that the valid forecast time of SCS SSHA of the statistical model is about 7 months, and the predictability of SCS SSHA in Spring and Autumn is stronger than that in Summer and Winter. In addition, the prediction skill of SCS SSHA has remarkable decadal variability, with better phase forecast in 1997–2007.
