    Abstract: As a calcium-regulated protein, CaMK II is closely related to cell death, and it participates in the development of pathological processes such as reperfusion injury, myocardial infarction, and oligodendrocyte death. The function of CaMK II activation in acute pancreatitis (AP) remains unclear. In our study, we confirmed that the expression of p-CaMK II was increased significantly and consistently in injured pancreatic tissues after caerulein-induced AP. Then, we found that KN93, an inhibitor of CaMK II, could mitigate the histopathological manifestations in pancreatic tissues, reduce serum levels of enzymology, and decrease oxidative stress products. Accordingly, we elucidated the effect of KN93 in vitro and found that KN93 had a protective effect on the pancreatic acinar cell necroptosis pathway by inhibiting the production of ROS and decreasing the expression of RIP3 and p-MLKL. In addition, we identified the protective effect of KN93 on AP through another mouse model induced by pancreatic duct ligation (PDL). Together, these data demonstrated that CaMK II participates in the development of AP and that inhibiting CaMK II activation could protect against AP by reducing acinar cell necroptosis, which may provide a new idea target for the prevention and treatment of AP in the clinic.

    Abstract: Background . Danshen ( Salvia miltiorrhiza Bunge) and its main active component Tanshinone IIA (TSA) are clinically used in China. However, the effects of TSA on acute pancreatitis (AP) and its potential mechanism have not been investigated. In this study, our objective was to investigate the protective effects of TSA against AP via three classic mouse models. Methods . Mouse models of AP were established by caerulein, sodium taurocholate, and L-arginine, separately. Pancreatic and pulmonary histopathological characteristics and serum amylase and lipase levels were evaluated, and changes in oxidative stress injury and the ultrastructure of acinar cells were observed. The reactive oxygen species (ROS) inhibitor N-Acetylcysteine (NAC) and nuclear factor erythroid 2-related factor 2 (Nrf2) knockout mice were applied to clarify the protective mechanism of the drug. Results . In the caerulein-induced AP model, TSA administration reduced serum amylase and lipase levels and ameliorated the histopathological manifestations of AP in pancreatic tissue. Additionally, TSA appreciably decreased ROS release, protected the structures of mitochondria and the endoplasmic reticulum, and increased the protein expression of Nrf2 and heme oxygenase 1 of pancreatic tissue. In addition, the protective effects of TSA against AP were counteracted by blocking the oxidative stress (NAC administration and Nrf2 knockout in mice). Furthermore, we found that TSA protects pancreatic tissue from damage and pancreatitis-associated lung injury in two additional mouse models induced by sodium taurocholate and by L-arginine. Conclusion . Our data confirmed the protective effects of TSA against AP in mice by inhibiting oxidative stress via the Nrf2/ROS pathway.

    Abstract: The analysis of human brain fMRI subjects can research neuro-related diseases and explore the related rules of human brain activity. In this paper, we proposed an algorithm framework to analyze the functional connectivity network of the whole brain and to distinguish Alzheimer’s disease (AD), mild cognitive impairment (MCI), and cognitively normal (CN). In other studies, they use algorithms to select features or extract abstract features, or even manually select features based on prior information. Then, a classifier is constructed to classify the selected features. We designed a concise algorithm framework that uses whole-brain functional connectivity for classification without feature selection. The algorithm framework is a two-hidden-layer neural network based on extreme learning machine (ELM), which overcomes the instability of classical ELM in high-dimensional data scenarios. We use this method to conduct experiments for AD, MCI, and CN data and perform 10-fold cross-validation. We found that it has several advantages: (1) the proposed method has excellent classification accuracy with high speed. The classification accuracy of AD vs. CN is 96.85%, and the accuracy of MCI vs. CN is 95.05%. Their AUC (area under curve) of ROC (receiver operating characteristic curve) reached 0.9891 and 0.9888, respectively. Their sensitivities are 97.1% and 94.7%, and specificities are 96.3% and 95.3%, respectively. (2) Compared with other studies, the proposed method is concise. Construction of a two-hidden-layer neural network is to learn features of the whole brain for the diagnosis of AD and MCI, without the feature screening. It avoids the negative effects of feature screening by algorithm or prior information. (3) The proposed method is suitable for small sample and high-dimensional data. It meets the requirements of medical image analysis. In other studies, its classifiers usually deal with several to dozens of feature dimensions. The proposed method deals with 4005 feature dimensions.

    Abstract: In this study, an analytical method was developed for the rapid determination of 21 pesticides used in ginseng cultivation. All pesticides covered by this method have been registered by 2020 in China for use on ginseng. The extracts were cleaned up using zirconium-oxide-modified silica (Z-Sep) and primary secondary amine (PSA). The combination of Z-Sep and PSA provided good recovery for all analytes and the cleanest matrix background out of a number of PSA-based sorbent combinations, as indicated by high-performance liquid chromatography (HPLC) and gas chromatography (GC). Instrumental analysis was completed in 5 min using the ultrahigh-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS). The linearity (r  〉  0.99) for all analytes was satisfactory over the calibration range of 0.002–0.1 μg mL−1. Intraday recoveries (n = 5) at ginseng-spiked levels of 0.02, 0.05, 0.1, and 1 mg kg−1 ranged between 72% and 119%, with the corresponding relative standard deviations (RSDs), were less than 19%, while the interday recoveries (n = 15) ranged between 77% and 103%, and RSDs were less than 22%. Limits of quantitation (LOQs) ranged between 0.02 and 0.05 mg kg−1 for all 21 pesticides. This is a seminal study using Z-Sep for the efficient cleanup of ginseng samples, and it could present a practical method for future monitoring of pesticide residues in ginseng produced in China.

    Abstract: Purpose . To assess whether renin-angiotensin system inhibitor (RASI) utilization is associated with age-related macular degeneration (AMD) prevalence among hypertensive patients. Methods . A US population-based, cross-sectional study was conducted. 3,023 hypertensive participants aged 40 years and older with gradable retinal images and ascertained RASI usage in the National Health and Nutrition Examination Survey (NHANES), 2005–2008, were finally enrolled into the study. RASI usage was obtained by interview, and AMD was determined through retinal image assessment. We performed multivariable analyses to assess the relationship between utilization of RASIs and AMD prevalence. We also took drug treatment duration into account, in order to better understand the effects of RASIs. Results . Multivariable logistic regression analyses revealed that AMD prevalence had no significant association with RASI usage but was inversely correlated with RASI treatment duration (odds ratio (OR) = 0.87, 95% confidence interval (CI) = 0.78–0.98, p = 0.02 ). Long-term usage ( 〉 5 years) of RASIs was significantly associated with not only reduced overall risk of AMD (OR = 0.23, 95% CI = 0.14–0.38, p 〈 0.001 ) but also lower propensity to have early (OR = 0.23, 95% CI = 0.14–0.37, p 〈 0.001 ) and late (OR = 0.25, 95% CI = 0.07–0.87, p = 0.03 ) AMD. Furthermore, long-term RASI users were less prone to develop soft drusen (OR = 0.67, 95% CI = 0.45–0.99, p = 0.04 ) and geographic atrophy (GA) (OR = 0.39, 95% CI = 0.22–0.71, p = 0.003 ). Conclusions . Evidence supporting that RASI utilization could directly protect against AMD in hypertensive patients was still insufficient, but long-term RASI treatment seemed to be beneficial for both early and late AMD, implicating a promising therapeutic approach that RASIs might offer for AMD prevention and management.

    Abstract: Purpose . This meta-analysis was conducted to determine the potential association between adiposity and glaucoma incidence. Materials and Methods . A comprehensive literature search was performed in PubMed and ISI Web of Science. A meta-analysis was conducted using STATA software. Results . Fifteen eligible studies involving 2,445,980 individuals were included to investigate the association between adiposity and glaucoma incidence. The relative risks (RRs) were pooled with 95% confidence intervals (CI) by using a random-effects model. The pooled RR between adiposity and elevated intraocular pressure (IOP) was 1.73 (95% CI, 1.18–2.54), whereas that between adiposity and open-angle glaucoma (OAG) was 0.97 (95% CI, 0.83–1.13). The pooled RR between abdominal adiposity and glaucoma was 1.28 (95% CI, 1.15–1.41), whereas that between general adiposity and glaucoma was 1.09 (95% CI, 0.87–1.37). Results of subgroup analysis by sex indicated the association between adiposity and glaucoma in the female group (RR, 1.31; 95% CI, 1.05–1.64), but not in the male group (RR, 1.11; 95% CI, 0.77–1.60). The pooled RR of cohort studies and cross-sectional studies were 1.00 (95% CI, 0.84–1.20) and 1.22 (95% CI, 0.89–1.66), respectively. Conclusions. Adiposity has a higher risk of elevated IOP, and abdominal adiposity has a positive association with glaucoma, especially in female patients.

    Abstract: Purpose. This study is aimed at exploring the effects of cognitive nursing combined with continuous nursing on postpartum mental state and rehabilitation. Methods. Totally, 124 puerperas admitted to our hospital from January 2019 to January 2020 were selected and divided into a research group and a control group according to different nursing methods, with 62 cases in each group. The control group received routine care, while the research group received cognitive nursing combined with continuous nursing on this basis. The mental state, rehabilitation indicators, quality of life, incidence of complications, and nursing satisfaction were compared between the two groups after intervention. Results. Before nursing, there was no statistically significant difference in the SAS and SDS scores between the two groups ( P 〉 0.05 ); after intervention, the SAS and SDS scores of the two groups were significantly reduced, and those of the research group were lower than those of the control group ( P 〈 0.05 ). After intervention, the time of the first breastfeeding, duration of lochia rubra, length of hospital stay, and score of uterine contraction pain of the research group were lower than those of the control group ( P 〈 0.05 ); the psychological function, physical function, material life, and social function scores of the research group were higher than those of the control group ( P 〈 0.05 ); the incidence of complications in the research group was 4.84%, lower than 20.97% in the control group ( P 〈 0.05 ); the nursing satisfaction of the research group was 96.77%, which was significantly higher than 83.87% in the control group ( P 〈 0.05 ). Conclusions. Cognitive nursing combined with continuous nursing can effectively improve the mental state, shorten the length of hospital stay, increase the perceived well-being, and promote the physical rehabilitation in puerperas, which is worth promoting in clinical practice.

    Abstract: In this study, we found that increased BRF2 protein expression was prevalent in NSCLC. Overexpression of BRF2 correlated with abnormal expression of E-cadherin, N-cadherin, and snail. Additionally, expression of BRF2 was found to be an independent prognostic factor in NSCLC patients. Furthermore, we showed that targeted knockdown of BRF2 expression could inhibit the migratory and invasive abilities of NSCLC cells and induced loss of the epithelial-mesenchymal transition of NSCLC cells. These results suggested that BRF2 overexpression in tumor tissues is significantly associated with the poor prognosis of NSCLC patients through promoting epithelial-mesenchymal transition (EMT) program.

    Abstract: Background and Aim . The incidence of nonalcoholic fatty liver disease (NAFLD) as a metabolic disease is increasing annually. In the present study, we aimed to explore the influence of NAFLD on the severity of acute pancreatitis (AP). Methods . The severity of AP was diagnosed and analyzed according to the 2012 revised Atlanta Classification. Outcome variables, including the severity of AP, organ failure (all types of organ failure), and systemic inflammatory response syndrome (SIRS), were compared for patients with and without NAFLD. Results . Six hundred and fifty-six patients were enrolled in the study and were divided into two groups according to the presence or absence of NAFLD. The non-NAFLD group contained 278 patients and the main etiology in this group was gallstone. The NAFLD group consisted of 378 patients and the main etiology was hyperlipidemia. The incidence of mild AP, moderately severe AP, and severe AP was 77.30%, 18.3%, and 4.3% in the non-NAFLD group and 58.2%, 33.9%, and 7.9% in the NAFLD group, respectively. There were significant differences between the two groups according to the severity of AP (P ≤ 0.001). In addition, the Ranson and BISAP scores as well as the incidence of SIRS and organ failure in the NAFLD group were higher than those in the non-NAFLD group (all P 〈 0.05). The patients were further divided into non-NAFLD, mild-NAFLD, and moderate-severe NAFLD (M+S-NAFLD) groups. The results showed that the severity of AP increased gradually from the non-NAFLD group to the M+S-NAFLD group. In addition, the incidence rates of SIRS and organ failure showed an upward trend with the aggravation of fatty liver severity. Multivariate logistic analysis showed that patients with NAFLD, especially those with M+S-NAFLD, had higher risks of SIRS and organ failure. Conclusions . Compared with non-NAFLD, NAFLD has a clinically relevant impact on the severity of AP and may be an early prognostic parameter for patients with AP.
