    Abstract: Heterogeneous nuclear ribonucleoproteins (HNRNPs) are reported to play a crucial role in the pathogenic process of multiple malignancies. However, the expression patterns and prognostic values of HNRNPs in pancreatic cancer (PC) are lacking. In this study, several public databases were explored to identify the commonly upregulated HNRNPs in PC. The clinical significance of HNRNPL (heterogeneous nuclear ribonucleoproteins L) in PC was analyzed. We further performed a series of experiments to elucidate the biological functions of HNRNPL. Bioinformatics analysis including pathway enrichment and interactors with HNRNPL was used to explain the potential mechanisms of HNRNPL in PC pathogenesis. Herein, we reported that HNRNPL was commonly overexpressed in public databases and that high expression of HNRNPL in PC was positively associated with aggressive disease and poor overall survival. Downregulation of HNRNPL suppressed the abilities of migration and epithelial mesenchymal transition of PC cells in vitro, while depletion of HNRNPL did not affect the proliferation rate of PC cells. We further showed that HNRNPL might combine with RNA-binding protein, PTBP1, and function as a part of the spliceosome to regulate alternative splicing of target genes in the occurrence and development of PC. HNRNPL could be employed as an innovative prognostic biomarker and therapeutic target for PC.

    Abstract: Purpose . To assess the consistency of anterior segment measurements obtained using a Sirius Scheimpflug/Placido photography-based topography system (CSO, Italy) and IOLMaster partial coherence interferometry (Carl Zeiss Meditec, Germany) in eyes with cataracts. Methods . A total of 90 eyes of 90 patients were included in this prospective study. The anterior chamber depth (ACD), keratometry (K), corneal astigmatism axis, and white to white (WTW) values were randomly measured three times with Sirius and IOLMaster. Concordance between them was assessed by calculating 95% limits of agreement (LoA). Results . The ACD and K taken with the Sirius were statistically significantly higher than that taken with the IOLMaster; however, the Sirius significantly underestimated the WTW values compared with the IOLMaster. Good agreement was found for Km and ACD measurements, with 95% LoA of −0.20 to 0.54 mm and −0.16 to 0.34 mm, respectively. Poor agreement was observed for astigmatism axis and WTW measurements, as the 95% LoA was −23.96 to 23.36° and −1.15 to 0.37 mm, respectively. Conclusion . With the exception of astigmatism axis and WTW, anterior segment measurements taken by Sirius and IOLMaster devices showed good agreement and may be used interchangeably in patients with cataracts.

    Abstract: Purpose. To compare between a new optical biometer (AL-Scan, Nidek Co., Aichi, Japan) and an anterior segment optical coherence tomographer (Visante AS-OCT, Carl Zeiss Meditec, Dublin, USA) for measuring central corneal thickness (CCT), anterior chamber depth (ACD), and aqueous depth (AD). Methods. Sixty-three eyes of 63 normal subjects were examined with AL-Scan and Visante AS-OCT in this prospective study. One eye per subject was measured three times with both devices to record their CCT, ACD, and AD. All procedures were performed by the same operator. Agreement between the two devices was assessed using paired t -tests, Bland-Altman plots, and 95% limits of agreement (LoA). Results. The mean CCT, ACD, and AD measured by AL-Scan were 538.59 ± 27.37   μ m, 3.70 ± 0.30  mm, and 3.16 ± 0.30  mm, respectively. The mean values obtained by the Visante OCT were 536.14 ± 26.61   μ m for CCT, 3.71 ± 0.29  mm for ACD, and 3.17 ± 0.29  mm for AD. The mean CCT by the AL-Scan was higher than that obtained by the Visante AS-OCT (difference = 2.45 ± 6.07   μ m, P 〈 0.05 ). The differences in ACD and AD measurements were not statistically significant. The 95% LoA of CCT, ACD, and AD were between −9.44 and 14.35  μ m, −0.15 and 0.12 mm, and −0.15 and 0.12 mm, respectively. Conclusions. Since these two devices were comparable for measuring CCT, ACD, and AD, their results can be interchangeably used in the clinic.

    Abstract: Purpose. To assess the repeatability and reproducibility of central corneal thickness (CCT) measurements by corneal dynamic Scheimpflug analyzer Corvis ST in normal eyes and compare the agreement with Pentacam rotating Scheimpflug System and ultrasound pachymetry. Methods. 84 right eyes underwent Corvis ST measurements performed by two operators. The test-retest repeatability (TRT), within-subject coefficient of variation (CoV), and intraclass correlation coefficient (ICC) were used to evaluate the intraoperator repeatability and interoperator reproducibility. CCT measurements also were obtained from Pentacam and ultrasound pachymetry by the first operator. The agreement between the three devices was evaluated with 95% limits of agreement (LoA) and Bland-Altman plots. Results. Corvis ST showed high repeatability as indicated by TRT ≤ 13.0  μ m, CoV 〈 0.9%, and ICC 〉 0.97. The interoperator reproducibility was also excellent. The CoV was 〈 0.9%, and ICC was 〉 0.97. Corvis ST showed significantly lower values than Pentacam and ultrasound pachymetry ( P 〈 0.001 ). The 95% LoA between Corvis ST and Pentacam or ultrasound pachymetry were −15.8 to 9.5  μ m and −27.9 to 12.3  μ m, respectively. Conclusions. Corvis ST showed excellent repeatability and interoperator reproducibility of CCT measurements in normal eyes. Corvis ST is interchangeable with Pentacam but not with ultrasound pachymetry.

    Abstract: To explore the influence of initial damage on the dynamic characteristics of rock mass, the Φ 50 mm split Hopkinson pressure bar (SHPB) test system was used, and the uniaxial impact compression tests on yellow sandstone specimens with different damage degrees were conducted, and then the variation law of mechanical properties of rock specimens with the initial damage was determined. The test results show that the dynamic stress-strain curve of rock specimens with initial damage can be roughly divided into compaction stage, elastic deformation stage, crack evolution stage, and strain-softening stage; the higher the initial damage degree of rock mass, the more significant the compaction stage. With the increase of the initial damage degree, the dynamic elastic modulus and peak stress of rock mass decrease gradually in a power number, while the peak strain of rock mass increases exponentially. With the increase of the initial damage degree, both the reflected energy ratio and the dissipated energy ratio decrease linearly, while the transmitted energy ratio increases linearly; the increasing rate of the transmitted energy ratio is greater than the decreasing rate of the reflected energy ratio.

    Abstract: Objectives. Human adenoviruses (HAdV) are classified as 7 HAdV species, and some serotypes in species B like HAdV 3, HAdV 7, HAdV 21, and HAdV 55 caused severe symptoms, even fatalities. Patients may be misdiagnosed and inadequately treated without reliable and practical methods for HAdV serotyping. Developing rapid, sensitive, and specific diagnostic methods for HAdV is critical. Methods. Detection methods were established based on a recombinase polymerase amplification (RPA) assay and lateral flow (LF) test. Specific target sequence was screened, targeting which, primers and probes were designed, synthesized, and screened for establishing assay with high amplification efficiency. Primer or probe concentrations and amplification time were optimized. Detection limit, sensitivity, and specificity were evaluated. Results and Conclusions. Simple, sensitive, and specific RPA-LF methods for detection of four serotypes of HAdV together or separately were established, which had detection limits of 10 to 280 copies/reaction comparable to real-time PCR without recognizing other pathogens. The sensitivity and specificity were 〉 92% and 〉 98%, respectively, evaluated by limited clinical samples. The detection can be completed in 25 min without requirement of any instrument except a constant temperature equipment, showing superior detection performance and promising for a wide use in the field and resource-limited area.

    Abstract: Purpose. To assess the reliability and comparability of measuring central corneal thickness (CCT) and thinnest corneal thickness (TCT) using a new Scheimpflug-Placido analyzer (TMS-5, Japan) and ultrasound (US) pachymetry. Methods. Seventy-six healthy subjects were prospectively measured 3 times by 1 operator using the TMS-5, 3 additional consecutive scans were performed by a second operator, and ultrasound (US) pachymetry measurements were taken. The test-retest repeatability (TRT), coefficient of variation (CoV), and intraclass correlation coefficient (ICC) were calculated to evaluate intraoperator repeatability and interoperator reproducibility. Agreement among the devices was assessed using Bland-Altman plots and 95% limits of agreement (LoA). Results. The intraoperators TRT and CoV were 〈 19  μ m and 2.0%, respectively. The interoperators TRT and CoV were 〈 12  μ m and 1.0%, respectively, and ICC was 〉 0.90. The mean CCT and TCT measurements using the TMS-5 were 15.97  μ m (95% LoA from −26.42 to −5.52  μ m) and 20.32  μ m (95% LoA from −30.67 to −9.97  μ m) smaller, respectively, than those using US pachymetry. Conclusions. The TMS-5 shows good repeatability and reproducibility for measuring CCT and TCT in normal subjects but only moderate agreement with US pachymetry results. Caution is warranted before using these techniques interchangeably.
