    Abstract: Objective. The purpose of the study is to explore the effect of K5BW12O40, a polyoxometalate (POM), on the apoptosis of A549 cells and its underlying mechanism and to analyze the potential therapeutic effect of K5BW12O40 in non-small-cell lung cancer. Materials and Methods. A549 cells were treated with different concentrations of K5BW12O40 (0 mg/ml, 0.001 mg/ml, 0.01 mg/ml, and 0.1 mg/ml). The proliferation of A549 cells treated with different concentrations of K5BW12O40 was detected by MTT assay (3-(4,5-dimethylthiazol-2-yl)-2 and 5-diphenyl tetrazolium bromide). The apoptosis of A549 cells induced by K5BW12O40 was detected by flow cytometry. Western blot was used to detect the changes in Bax and caspase-3 protein levels in A549 cells induced by K5BW12O40. Results. As the dose of the K5BW12O40 increases, the cell viability of A549 cells gradually decreases. The results of flow cytometry showed that the apoptotic rate of A549 cells increased with the increase of K5BW12O40 concentration. Western blot results showed that the expression of the apoptosis marker protein caspase-3 was increased in the three groups treated with K5BW12O40 whereas the protein level of Bax did not change significantly in A549 cells treated with K5BW12O40. Conclusions. K5BW12O40 increases the apoptotic rate of A549 cells by upregulating caspase-3.

    Abstract: Objectives. We assessed the potential of glial cell line-derived neurotrophic factor (GDNF) as a useful biomarker to predict cirrhosis in chronic hepatitis B (CHB) patients. Methods. A total of 735 patients from two medical centers (385 CHB patients and 350 healthy controls) were included to determine the association of serum and tissue GDNF levels with biopsy-proven cirrhosis. The diagnostic accuracy of serum GDNF (sGDNF) was estimated and compared with other indices of cirrhosis. Results. We showed significantly higher levels of sGDNF in CHB patients with fibrosis (28.4 pg/ml vs. 11.6 pg/ml in patients without) and patients with cirrhosis (33.8 pg/ml vs. 23.5 pg/ml in patients without). The areas under receiver operating curve (AUROCs) of sGDNF were 0.83 (95% confidence interval (CI): 0.80–0.87) for predicting liver fibrosis and 0.84 (95% CI: 0.79–0.89) for cirrhosis. Findings from the serum protein level and hepatic mRNA expression were consistent. Using the best cutoff to predict cirrhosis, we categorized the patients into sGDNF-high and sGDNF-low groups. The sGDNF-high group had significantly larger Masson’s trichrome and reticulin staining-positive area, higher Scheuer score, and METAVIR fibrosis stage (all p 〈 0.001 ) but not steatosis. On multivariable regression, sGDNF was independently associated with cirrhosis with an odds ratio of 6.98 (95% CI: 1.10–17.94). Finally, we demonstrated that sGDNF outperformed AST to platelet ratio index, FIB-4, fibroscore, forn index, and fibrometer in differentiating F4 vs. F3. Conclusion. Using serum, tissue mRNA, and biopsy data, our study revealed a significant potential of sGDNF as a novel noninvasive biomarker for cirrhosis in CHB patients.

    Abstract: Endomorphins (EMs) have a very important bridge-function in cardiovascular, endocrinological, and neurological systems. This study is to investigate the effects of EMs on the synthesis and secretion of vasoactive substances induced by advanced glycation end products in primary cultured human umbilical vein endothelial cells (HUVECs). Firstly, HUVECs were stimulated with AGEs-bovine serum albumin (AGEs-BSA), bovine serum albumin (BSA), or both AGEs-BSA and EMs together, respectively. Then, HUVEC survival rate was calculated by MTT assay, the levels of NO, endothelial nitric oxide synthase (eNOS), and inducible nitric oxide synthase (iNOS) were detected by colorimetric analysis, and the contents of endothelin-1 (ET-1) were detected by ELISA. The mRNA levels of eNOS and ET-1 were measured by RT-PCR. The expression of p38 mitogen-activated protein kinase (p38 MAPK) was detected by immunofluorescence assay. The results showed that the mRNA expression and secretion of eNOS were significantly enhanced after incubation with EMs compared to those with AGEs-BSA, while the secretion of NO and iNOS, mRNA expression, and secretion of ET-1 had opposite changes. The fluorescence intensity of p38MAPK in nuclear was decreased after pretreatment with EMs compared to incubation with AGEs-BSA. Conclusion . The present study suggests that EMs have certain protection effect on AGEs-BSA-induced injury in HUVEC.

    Abstract: Fluid resuscitation could hardly be performed immediately after fatal hemorrhagic shock in outpatients. We investigated whether electroacupuncture (EA) at Zusanli (ST36) could prevent fatal hemorrhagic shock induced heart failure with delayed fluid resuscitation and whether the protective role of EA is related to the autonomic nervous system. Sixty Sprague Dawley rats were randomly divided into five groups ( n = 12 each): group of sham hemorrhagic shock (SHAM), group of EA, group of sham EA (SEA), group of delayed fluid resuscitation with EA (EA + DR), and group of delayed fluid resuscitation with SEA (SEA + DR). After blood loss for 6 hours, caspase-3 activity and positive rate of TUNEL in EA + DR group were significantly lower than in other hemorrhagic shock groups (e.g., versus SEA + DR: 0 . 156 ± 0.0 39 versus 0 . 301 ± 0.0 42 ; P 〈 0.05 ). Immediately EA treatment after the blood loss enhanced the protective effect of delayed resuscitation on the cardiac tissue of hemorrhagic shock rats. Considering the significant changes of epinephrine ( 137.8 ± 6.9  ng/L versus 98.6 ± 7.4  ng/L; P 〈 0.05 ) and acetylcholine ( 405 ± 8.6  pmol/L versus 341 ± 10.1  pmol/L; P 〈 0.05 ) after EA treatment (SEA + DR versus EA + DR), this cardiac protective effect may be related to regulation of the autonomic nervous system.

    Abstract: Immersed boundary-lattice Boltzmann method (IB-LBM) has become a popular method for studying fluid-structure interaction (FSI) problems. However, the performance issues of the IB-LBM have to be considered when simulating the practical problems. The Graphics Processing Units (GPUs) from NVIDIA offer a possible solution for the parallel computing, while the CPU is a multicore processor that can also improve the parallel performance. This paper proposes a parallel algorithm for IB-LBM on a CPU-GPU heterogeneous platform, in which the CPU not only controls the launch of the kernel function but also performs calculations. According to the relatively local calculation characteristics of IB-LBM and the features of the heterogeneous platform, the flow field is divided into two parts: GPU computing domain and CPU computing domain. CUDA and OpenMP are used for parallel computing on the two computing domains, respectively. Since the calculation time is less than the data transmission time, a buffer is set at the junction of two computational domains. The size of the buffer determines the number of the evolution of the flow field before the data exchange. Therefore, the number of communications can be reduced by increasing buffer size. The performance of the method was investigated and analyzed using the traditional metric MFLUPS. The new algorithm is applied to the computational simulation of red blood cells (RBCs) in Poiseuille flow and through a microchannel.

    Abstract: Objective . To observe the curative effect of VAWI on Xinjiang Uygur patients with silicosis fibrosis. Methods . After we diagnosed the 40 patients with the first phase of silicosis, we randomly divided them into two groups: the basic treatment group (group A, n = 20 ) and the VAWI group (group B, n = 20 ). At the same time, we selected the age-matched healthy patients ( n = 20 ). We applied the combined protein chip with SELDI-TOF-MS to carry out the serum analysis. The data were analyzed throughout data preprocessing, difference in PEAK screening, hierarchical cluster analysis, and Principal Component Analysis (PCA). We built decision tree model and predict the difference between the PEAK corresponding proteins. Results . The proteins peaks corresponding to name, predicted protein, and gene name were as follows: M2001_69, amyloid beta a4 protein, APP, and M2017_02, amyloid beta a4 protein, APP. The different expression of proteins in patients with silicosis was found before and after with VAWI treatment. The predicted proteins were as follows: M1982_50, amyloid beta a4 protein, APP; M3164_50, fibrinogen alpha chain frag, FGA; M3379_28, fibrinogen alpha chain frag, FGA; and so on. Conclusion . VAWI presented curative effect on patients with silicosis fibrosis via the alternation of proteins expression in serum.

    Abstract: With the high development of sports, football has attracted more and more attention from the public. However, the hot competition has made football players undergo high-intensity training, and the risk of injury caused by training is also increasing. Lateral incision knee ACL injury is one of the most common types of injuries in football players, which has a serious impact on the athlete’s physiology and daily training. The comprehensive, multi-level, convenient and fast medical system brought by the medical Internet of Things has become the growing demand of the medical industry. Based on the medical Internet of Things, this paper studies the rehabilitation nursing of football players’ ACL injury by lateral cutting and running combined with optical microscope. In this paper, 36 male and female football players were selected for group experiments, and the landing and peak torque indexes of the experimental group and the control group of male and female athletes were analyzed under the observation of an optical microscope. Group (P  〈 0.05), coxa valgus (P  〈 0.01), internal rotation and knee valgus angles were greater than those in the EM group. And the peak hip flexion angle and knee valgus moment in NF group were lower than those in EF group (P  〈 0.05). After 12 weeks of rehabilitation training, there was a significant difference in the Q/H joint contraction index between the REF group and the CON group (P  〈 0.05), and there was no statistical difference in the other groups. After 12 weeks of rehabilitation training, there was a significant difference in the Q/H joint contraction index between the REF group and the CON group (P  〈 0.05), and there was no statistical difference in the other groups. That is to say, after the systematic rehabilitation training proposed in this paper, the joint contraction performance of the hamstrings/quadriceps in the REF group has been significantly improved. This shows that the rehabilitation nursing under the optical microscope based on the medical Internet of Things has a good effect on the rehabilitation of the football player’s side-cut running knee joint ACL injury.

    Abstract: Higher sensitive transglutaminase autoantibody (TGA) assay will detect the onset of celiac disease (CD) autoimmunity earlier. In developing a nonradioactive assay for TGA, we utilized electrochemiluminescence (ECL) technology and compared it to a high-performance radioimmunoassay (RIA) currently being used to screen patients with type 1 diabetes (T1D) and genetically at-risk individuals for CD. We selected 183 T1D patients with 60 patients having received biopsy and analyzed 396 sequential samples from 73 young children longitudinally followed up with TGA seroconversion, with 27 undergoing biopsy. In addition, 112 age-matched healthy control subjects were included in the study. With the 99th percentile of specificity, the ECL assay detected significantly more TGA positivity among patients with T1D (133/183) than RIA (114/183) and more of the sequential samples (34%) from 73 children than RIA (18%). The TGA assay performed by ECL was positive in all 59 subjects with villous atrophy. Among 73 longitudinally followed up children, ECL assay had earlier detection of TGA on 34 children by a mean of 2.5 years. In conclusion, the new TGA assay by ECL has a higher sensitivity than the current RIA assay and may better predict the onset of CD.

    Abstract: Introduction. Systemic lupus erythematosus (SLE) is one of the systemic autoimmune diseases characterized by the polyclonal autoantibody production. The human homologue of the mouse double minute 2 (MDM2) is well known as the negative regulator of p53. MDM2 has been reported to be overexpressed in SLE animal model and to promote SLE. Since abnormally expressed proteins can induce autoimmune response, anti-MDM2 autoantibody was examined in SLE patients. Methods. Anti-MDM2 antibody in sera from 43 SLE patients and 69 healthy persons was investigated by ELISA. Positive samples were further confirmed by western blotting. The immunological feathers of anti-MDM2 positive sera were analyzed by indirect immunofluorescence assay. Anti-p53 was also investigated in SLE patients by ELISA, and the correlation of anti-MDM2 and anti-p53 was analyzed. Results. The presence of anti-MDM2 in SLE patients was 23.30%, much higher than normal healthy persons (4.30%). These anti-MDM2 positive sera present a nuclear staining pattern. The presence of anti-p53 in SLE patients was 39.50%, and the titer of anti-MDM2 was positively correlated with anti-p53 in SLE patients. Conclusions. Anti-MDM2 autoantibody was detected at high prevalence in SLE patients. The detection of anti-MDM2 in SLE patients should be clinically useful.
