    Kurzfassung: IL24 mRNA is known to have an apoptotic effect on cancer cells but not on noncancer cells. However, the expression level of the IL24 mRNA in head and neck squamous cell carcinoma (HNSCC) and its subgroups is rarely studied. In this study, the clinical implication of IL24 mRNA was evaluated in the common subgroups of HNSCC, including oral squamous cell carcinoma (OSCC), nasopharyngeal carcinoma (NPC), and laryngeal squamous cell carcinoma (LSCC) for analysis. Substantial IL24 mRNA expression data were calculated from several databases, such as the Gene Expression Omnibus (GEO), ArrayExpress, Sequence Read Archive (SRA), ONCOMINE, and The Cancer Genome Atlas (TCGA) databases. We ultimately collected a total of 41 microarrays and RNA-seq including 1,564 HNSCC and 603 noncancer tissue samples. IL24 mRNA was highly expressed in OSCC, LSCC, and NPC as shown by the separated standard mean difference (SMD), as well as HNSCC as a whole part (SMD = 1.47, 95% confdence interval (CI) = 1.24−1.70, P 〈 0.0001 ). In all subgroups, the IL24 mRNA upregulation had the ability to distinguish cancer from noncancer tissue with area under the curves (AUCs) of the summary receiver operating characteristic (sROC) higher than 0.85. In conclusion, IL24 mRNA may be used as a potential marker for cancer screening, and its clinical diagnostic value needs to be further studied. It also provides a new idea for the treatment of the IL24 gene in HNSCC and its subgroups in the future.

    Kurzfassung: Using the downhole annular flow measurement system to get real-time information of downhole annular flow is the core and foundation of downhole microflux control drilling technology. The research work of electromagnetic flowmeter in recent years creates a challenge to the design of downhole annular flow measurement. This paper proposes a design and optimization of annular flow electromagnetic measurement system for drilling engineering based on the finite element method. Firstly, the annular flow measuring and optimization principle are described. Secondly, a simulation model of an annular flow electromagnetic measurement system with two pairs of coil is built based on the fundamental equation of electromagnetic flowmeter by COMSOL. Thirdly, simulations of the structure of excitation system of the measurement system are carried out, and simulations of the size of the electrode’s radius are also carried out based on the optimized structure, and then all the simulation results are analyzed to evaluate the optimization effect based on the evaluation indexes. The simulation results show that optimized shapes of the excitation system and electrode size can yield a better performance in the annular flow measurement.

    Kurzfassung: Using downhole annular electromagnetic flow measurement to obtain annular flow in real-time is a foundation of microflow control drilling technology. The existing annular flow electromagnetic measurement method based on low-frequency rectangular excitation is affected by slurry interference and formation fluid invasion, which results in large noise generated on the electrode output signal. These noise causes the instability of the flow measurement system output and lower accuracy. Double-frequency rectangular wave excitation has the advantages of excellent zero-point stability attributed to low-frequency rectangular wave excitation and fast response speed with great ability to suppress slurry interference. First, the double-frequency rectangular wave excitation for annular flow electromagnetic measurement is researched, and its corresponding electromagnetic induction signal process is investigated. In order to verify the feasibility of downhole annular electromagnetic flow measurement, a flow verification platform for comparison of standard and detected parameters is established for simulation experiment, and the ability to suppress slurry interference and response speed of the technology in downhole slurry flow measurement are analyzed. The test results show that the downhole annular electromagnetic flow measurement based on double-frequency rectangular wave excitation can not only satisfy the requirements of regular electromagnetic flow measurement but also suppress the annular slurry interference effectively.

    Kurzfassung: Excessive compression, the main cause of intervertebral disc (IVD) degeneration, affected endogenous repair of the intervertebral disc. Pioglitazone (PGZ) is the agonist of peroxisome proliferator-activated receptor γ , which has been widely used in the treatment of diabetes mellitus. The present study aim at investigating whether pioglitazone has protective effects on compression-mediated cell apoptosis in nucleus pulposus mesenchymal stem cells (NP-MSCs) and further exploring the possible underlying mechanism. Our results indicated that the isolated cells satisfied the criteria of MSC stated by the International Society for Cellular Therapy. Besides, our research revealed that pioglitazone could protect cell viability, cell proliferation of NP-MSCs and alleviated the toxic effects caused by compression. The actin stress fibers was suppressed obviously under compression, and pioglitazone alleviated the adverse outcomes. Pioglitazone exerted protective effects on compression-induced NP-MSCs apoptosis according to annexin V/PI double-staining and TUNEL assays. Pioglitazone suppressed compression-induced NP-MSCs oxidative stress, including decreasing compression-induced overproduction of reactive oxygen species (ROS) and malondialdehyde (MDA), and alleviated compression-induced mitochondrial membrane potential (MMP) decrease. Ultrastructure collapse of the mitochondria exhibited a notable improvement by pioglitazone in compression-induced NP-MSCs according to transmission electron microscopy (TEM). Furthermore, the molecular results showed that pioglitazone significantly decreased the expression of apoptosis-associated proteins, including cyto.cytochrome c, Bax, cleaved caspase-9, and cleaved caspase-3, and promoted Bcl-2 expression. These results indicated that pioglitazone alleviated compression-induced NP-MSCs apoptosis by suppressing oxidative stress and the mitochondrial apoptosis pathway, which may be a valuable candidate for the treatment of IVD degeneration.

    Kurzfassung: The goal of the scheduling is to arrange operations on suitable machines with optimal sequence for corresponding objectives. In order to meet market requirements, scheduling systems must own enough flexibility against uncertain events. These events can change production status or processing parameters, even causing the original schedule to no longer be optimal or even to be infeasible. Traditional scheduling strategies, however, cannot cope with these cases. Therefore, a new idea of scheduling called inverse scheduling has been proposed. In this paper, the inverse scheduling with weighted completion time (SMISP) is considered in a single-machine shop environment. In this paper, an improved genetic algorithm (IGA) with a local searching strategy is proposed. To improve the performance of IGA, efficient encoding scheme, fitness evaluation mechanism, feasible initialization methods, and a local search procedure have been employed in the paper. Because of the local improving method, the proposed IGA can balance its exploration ability and exploitation ability. We adopt 27 instances to verify the effectiveness of the proposed algorithm. The experimental results illustrated that the proposed algorithm can generate satisfactory solutions. This approach also has been applied to solve the scheduling problem in the real Chinese shipyard and can bring some benefits.

    Kurzfassung: The motive of this article is to present the case study of patients to investigate the association between the ultrasonographic findings of lower extremity vascular disease (LEAD) and plaque formation. Secondly, to examine the association between the formation of coronary artery and carotid artery atherosclerosis in patients with type 2 diabetes mellitus. 124 patients with type 2 diabetes (64 males and 60 females with the age group 25-78 years) are considered for the research studies who have registered themselves in the Department of Endocrinology and Metabolism from April 2017 to February 2019. All participants have reported their clinical information regarding diabetes, alcohol consumption, smoking status, and medication. The blood samples from subjects are collected for measurement of HbA1c, total cholesterol, triglycerides, HDL-c, and LDL-c levels. Two-dimensional ultrasound has been used to measure the inner diameter, peak flow velocity, blood flow, and spectral width of the femoral artery, pop artery, anterior iliac artery, posterior tibial artery, and dorsal artery and to calculate the artery stenosis degree. Independent factors of atherosclerosis are determined by multivariate logistic regression analysis. The results are evaluated within the control group and it is found that there is no significant impact of gender, age, and body mass index ( P 〉 0.05 ) on the lower extremity vascular diseases. Those with smoking, alcohol consumption, hypertension, and dyslipidemia have higher positive rate ( P 〈 0.05 ). The type 2 diabetes mellitus group has higher diastolic blood pressure and lower triglyceride ( P 〈 0.05 ). Diastolic blood pressure, HbA1C, total cholesterol, HDL-c, and LDL-C are not remarkably dissimilar between the type 2 diabetes mellitus group and the control group ( P 〉 0.05 ). Compared with the control group, the type 2 diabetes mellitus group has higher frequency of lower extremity vascular diseases in the dorsal artery than in the pop artery ( P 〈 0.05 ). The blood flow of type 2 diabetes mellitus group is found to be lower than that of the control group, especially in the dorsal artery ( P 〈 0.05 ). The blood flow velocity of the dorsal artery is accelerated ( P 〈 0.01 ). Among 117 patients of type 2 diabetes mellitus (94.35%) with a certain degree of injury, there are 72 cases of type I carotid stenosis (58.06%), 30 cases of type II carotid stenosis (24.19%), and 15 cases of type III carotid stenosis (12.10%). Out of 108 subjects in the control group, there are 84 cases of type 0 carotid stenosis (77.78%), 19 cases of type I carotid stenosis (17.59%), 5 cases of type II carotid stenosis (4.63%), and 0 case of type III carotid stenosis (0.00%). Compared with the control group, carotid stenosis is more common in patients with type 2 diabetes mellitus ( P 〈 0.05 ). Age, smoking, duration of diseases, systolic blood pressure, and degree of carotid stenosis are found to be associated with atherosclerosis. The findings suggest that the color Doppler ultrasonography can give early warning when applied in patients with carotid and lower extremity vascular diseases to delay the incidence of diabetic macroangiopathy and to control the development of cerebral infarction, thus providing an important basis for clinical diagnosis and treatment.

    Kurzfassung: International crash data indicate that roadside characteristics contribute to more than half of all roadside accidents involving serious injury or death. Therefore, research on roadside safety is urgently needed. Based on the vehicle departure speed, pavement height (i.e., the difference between pavement elevation and ground elevation), slope gradient, and horizontal curve radius, this study uses PC-Crash simulation software to carry out tests of trucks and cars exiting a road. A chi-squared automatic interaction detection (CHAID) decision tree is used to explore the causative mechanism of vehicle rollover, and the concept of a “safe slope” to ensure that vehicles do not roll over is proposed. Aiming at straight and curved sections, discriminant functions of vehicle rollover and nonrollover are fitted through Bayesian discriminant analysis, and safe slope calculation models for trucks and cars are then constructed. Based on the obtained safe slope models, calculation methods for the safe slope and the roadside clear zone width involving different traffic compositions are proposed by calibrating the lateral distance from the final position of nonrollover vehicles to the road edge. The results show that the factors affecting vehicle rollover are, in descending order of importance, the slope gradient, pavement height, vehicle type, departure speed, and horizontal curve radius. For a section with a large proportion of cars, the slope gradient should not be steeper than 1:3.5. The horizontal curve radius should not be less than 600 m for a section with a large proportion of trucks and a slope gradient steeper than 1:3.5 or shallower than 1:2.5. Additionally, for a section with a pavement higher than 0.5 m and a slope gradient steeper than 1:2.5, the operating speed limit should be lower than 60 km/h. These research results have theoretical value and practical significance to improve the driving safety level and reducing the risk of roadside accidents.
