    Abstract: Current data is scarce regarding the function of noncoding RNAs (ncRNAs) such as microRNAs (miRNAs) and long noncoding RNAs (lncRNAs) in the interferon- (IFN-) mediated immune response. This is a comprehensive study that analyzes the lncRNA and miRNA expression profiles of the type I IFN and type II IFN in porcine alveolar macrophages using RNA sequencing. There was a total of 152 overexpressed differentially expressed (DE) lncRNAs and 21 DE miRNAs across type I IFN and type II IFN in porcine alveolar macrophages. Subsequent lncRNA-miRNA-mRNA network construction revealed the involvement of 36 DE lncRNAs and 12 DE miRNAs. LncRNAs such as the XLOC_211306, XLOC_100516, XLOC_00695, XLOC_149196, and XLOC_014459 were expressed at a higher degree in the type I IFN group, while XLOC_222640, XLOC_047290, XLOC_147777, XLOC_162298, XLOC_220210, and XLOC_165237 were expressed at a higher degree in the type II IFN group. These lncRNAs were found to act as “sponges” for miRNAs such as miR-34a, miR-328, miR-885-3p, miR-149, miR-30c-3p, miR-30b-5p, miR-708-5p, miR-193a-5p, miR-365-5p, and miR-7. Their target genes FADS2, RPS6KA1, PIM1, and NOD1 were found to be associated with several immune-related signaling pathways including the NOD-like receptor, Jak-STAT, mTOR, and PPAR signaling pathways. These experiments provide a comprehensive profile of overexpressed noncoding RNAs in porcine alveolar macrophages, providing new insights regarding the IFN-mediated immune response.

    Abstract: When brain injury happens, endogenous neural stem cells (NSCs) located in the adult subventricular zone (SVZ) and subgranular zone (SGZ) are attacked by ischemia/reperfusion to undergo cellular apoptosis and death before being induced to migrate to the lesion point and differentiate into mature neural cells for damaged cell replacement. Although promoting antiapoptosis and NSC survival are critical to neuroregeneration, the mechanism has yet been elucidated clearly. Here in this study, we established an in vitro oxygen-glucose deprivation (OGD)/reoxygenation model on NSCs and detected glucose-regulated protein 78 (GRP78) involved in apoptosis, while in the absence of GRP78 by siRNA transfection, OGD/reoxygenation triggered PI3K/Akt, ERK1/2, and NF- κ B/p65 activation, and induced NSC apoptosis was attenuated. Further investigation, respectively, with the inhibitor of PI3K/Akt or ERK1/2 demonstrated a blockage on GRP78 upregulation, while the inhibition of NF- κ B rarely affected GRP78 induction by OGD/reoxygenation. The results indicated the bidirectional regulations of GRP78-PI3K/Akt and GRP78-ERK1/2 and the one-way signalling transduction through GRP78 to NF- κ B/p65 on NSC survival from OGD/reoxygenation. In conclusion, we found that GRP78 mediated the signalling cross talk through PI3K/Akt, ERK1/2, and NF- κ B/p65, which leads to antiapoptosis and NSC survival from ischemic stroke. Our finding gives a new evidence of GRP78 in NSCs as well as a new piece of signalling mechanism elucidation to NSC survival from ischemic stroke.

    Abstract: Objectives . To evaluate the effectiveness and safety of a Ginkgo biloba extract for patients with early diabetic nephropathy. Methods . Randomised controlled trials (RCTs) conducted on adults with early diabetic nephropathy which used Gingko biloba extract were included. The major databases were searched, and manufacturers of Gingko biloba products were contacted for information on any published or unpublished studies. Two authors independently extracted the data from the included studies. Data analysis was conducted using Review Manager 5.0 software. Results . Sixteen RCTs were included. Ginkgo biloba extract decreased the urinary albumin excretion rate (UAER), fasting blood glucose (FBG), serum creatinine (SCR), and blood urea nitrogen (BUN). The extract also improved hemorheology. The methodological quality in the included studies was low. The explicit generation of the allocation sequence was described in only 6 trials. None of the included trials were confirmed to use blinding. Three studies had observed adverse events. One study using angiotensin-converting enzyme inhibitor (ACEi) reported mild cough in both groups. No serious adverse effects were reported. Conclusions . Gingko biloba extract is a valuable drug which has prospect in treating early diabetic nephropathy, especially with high UAER baseline level. The safety for early diabetic nephropathy is uncertain. Long-term, double-blinded RCTs with large sample sizes are still needed to provide stronger evidence.

    Abstract: The bulb tubular unit has advantages over other units in the low head, large discharge, and tidal energy. The development and utilization of low head and ultralow head hydraulic resources can increase the runner’s energy parameters, make up for the defects of large size and low speed of the unit, and reduce the construction investment. The bulb tubular turbine’s operation stability and structural strength are compared and analyzed by the orthogonal test method to improve the perfomance of bulb tubular turbine. The results show that the optimal design scheme can be obtained when the number of guide vane is 16, the distance between the guide vane and the blade is 1995 mm, the hub ratio is 0.31, the relative twist angle of the blade is 27°, and the cascade density is 0.72. After optimization, the bulb tubular turbine’s efficiency and output are increased by 5.72% and 2.86% at a low flow rate, respectively. The vortex at the tailpipe is reduced, and the blade work effect and the cavitation performance are well. The maximum pressure pulsation amplitude in the flow passage is reduced by 73.15%, the maximum blade deformation is reduced by 39.59%, and the maximum blade static stress is reduced by 13.16%. Finally, the reliability of numerical simulation results is verified by the model test.

    Abstract: Intervertebral disc degeneration (IDD) is the leading cause of low back pain (LBP). However, effective therapeutic drugs for IDD remain to be further explored. Inflammatory cytokines play a pivotal role in the onset and progression of IDD. Dihydroartemisinin (DHA) has been well reported to have powerful anti-inflammatory effects, but whether DHA could ameliorate the development of IDD remained unclear. In this study, the effects of DHA on extracellular matrix (ECM) metabolism and cellular senescence were firstly investigated in nucleus pulposus cells (NPCs) under tumor necrosis factor alpha (TNFα)-induced inflammation. Meanwhile, AKT agonist sc-79 was used to determine whether DHA exerted its actions through regulating PI3K/AKT and NF-κB signaling pathways. Next, the therapeutic effects of DHA were tested in a puncture-induced rat IDD model. Finally, we detected the activation of PI3K/AKT and NF-κB signaling pathways in clinical degenerative nucleus pulposus specimens. We demonstrated that DHA ameliorated the imbalance between anabolism and catabolism of extracellular matrix and alleviated NPCs senescence induced by TNFα in vitro. Further, we illustrated that DHA mitigated the IDD progression in a puncture-induced rat model. Mechanistically, DHA inhibited the activation of PI3K/AKT and NF-κB signaling pathways induced by TNFα, which was undermined by AKT agonist sc-79. Molecular docking predicted that DHA bound to the PI3K directly. Intriguingly, we also verified the activation of PI3K/AKT and NF-κB signaling pathways in clinical degenerative nucleus pulposus specimens, suggesting that DHA may qualify itself as a promising drug for mitigating IDD.

    Abstract: The understanding of F− concentration in groundwater in humid areas is limited although there are lots of research on high-fluoride groundwater in arid areas. In this paper, with controlling factors of F− concentrations in humid areas as the focus, 130 groundwater samples, obtained from four subsystems in Northwest Xingguo County, Jiangxi Province, China, were investigated to demonstrate the controlling factors of F− concentrations in humid areas. According to analytical results, the following hydrogeochemical characteristics of the fluorine in humid mountainous areas were determined: (1) F− concentration is positively correlated with total dissolved solids (TDS), Ca2+, HCO 3 − , and pH; (2) the groundwater features a high flow rate and low TDS; (3) the equilibrium constant of CaF2 is less than its solubility product constant, and the fluorine-bearing minerals in rocks are in a dissolved state; and (4) the dissolved fluoride-bearing minerals constitute the main sources of F− in the groundwater. Fluorine mainly comes from groundwater fluorine-bearing minerals in metamorphic rocks. Moreover, the low F− concentration in the groundwater mainly results from the fast flow rate of groundwater. Fluoride in groundwater has great potential hazards in humid areas.

    Abstract: Stem cell survival after transplantation determines the efficiency of stem cell treatment, which develops as a novel potential therapy for several central nervous system (CNS) diseases in recent decades. The engrafted stem cells face the damage of oxidative stress, inflammation, and immune response at the lesion point in host. Among the damaging pathologies, oxidative stress directs stem cells to apoptosis and even death through several signalling pathways and DNA damage. However, the in-detail mechanism of stem cell survival from oxidative stress has not been revealed clearly. Here, in this study, we used hydrogen peroxide (H 2 O 2 ) to induce the oxidative damage on neural stem cells (NSCs). The damage was in consequence demonstrated involving the activation of heat shock protein 90 (HSP90) and NF- κ B/p65 signalling pathways. Further application of the pharmacological inhibitors, respectively, targeting at each signalling indicated an upper-stream role of HSP90 upon NF- κ B/p65 on NSCs survival. Preinhibition of HSP90 with the specific inhibitor displayed a significant protection on NSCs against oxidative stress. In conclusion, inhibition of HSP90 would attenuate NF- κ B/p65 activation by oxidative induction and promote NSCs survival from oxidative damage. The HSP90/NF- κ B mechanism provides a new evidence on rescuing NSCs from oxidative stress and also promotes the stem cell application on CNS pathologies.

    Abstract: The quality control process for sintered ore is cumbersome and time- and money-consuming. When the assay results come out and the ratios are found to be faulty, the ratios cannot be changed in time, which will produce sintered ore of substandard quality, resulting in a waste of resources and environmental pollution. For the problem of lagging sinter detection results, Long Short-Term Memory and Genetic Algorithm-Recurrent Neural Networks prediction algorithms were used for comparative analysis, and the article used GA-RNN quality prediction model for prediction. Through correlation analysis, the chemical composition of the sintered raw material was determined as the input parameter and the physical and metallurgical properties of the sintered ore were determined as the output parameters, thus successfully establishing a GA-RNN-based sinter quality prediction model. Based on 150 sets of original data, 105 sets of data were selected as the training sample set and 45 sets of data were selected as the test sample set. The results obtained were compared to the real value with an average prediction error of 1.24% for the drum index, 0.92% for the low-temperature reduction chalking index (RDI), 0.95% for the reduction index (RI), 0.40% for the load softening temperature T10%, and 0.43% for the load softening temperature T40%, with all within the running time thresholds. The study of this model enables the prediction of the quality of sintered ore prior to sintering, thus improving the yield of sintered ore, increasing corporate efficiency, saving energy, and reducing environmental pollution.

    Abstract: To the problem of large subjectivity in existing index screening methods, this paper uses principal component analysis and the rough set method to select faulty factors in a traction fault elevator, eliminate the unrelated and unimportant indices, and then set up 4 main indices: rated parameter, running environment, manufacturing maintenance condition, and man-machine factor; and 10 two-grade fingers, including rated load, use unit, and manufacturing unit, which constitute a traction failure elevator factor index system, provide a new train of thought and method for the research and analysis of traction fault elevators.

    Abstract: 1 H NMR-based metabolic profiling combined with multivariate data analysis was used to explore the metabolic phenotype of functional dyspepsia (FD) in stressed rats and evaluate the intervention effects of the Chinese medicine Weikangning (WKN). After a 7-day period of model establishment, a 14-day drug administration schedule was conducted in a WKN-treated group of rats, with the model and normal control groups serving as negative controls. Based on 1 H NMR spectra of urine and serum from rats, PCA, PLS-DA, and OPLS-DA were performed to identify changing metabolic profiles. According to the key metabolites determined by OPLS-DA, alterations in energy metabolism, stress-related metabolism, and gut microbiota were found in FD model rats after stress stimulation, and these alterations were restored to normal after WKN administration. This study may provide new insights into the relationship between FD and psychological stress and assist in research into the metabolic mechanisms involved in Chinese medicine.
