    Abstract: Background. Chinese patent medicines are increasingly used clinically, and the prescription drug monitoring program is an effective tool to promote drug safety and maintain health. Methods. We constructed a prescription drug monitoring program for Chinese patent medicines based on knowledge graphs. First, we extracted the key information of Chinese patent medicines, diseases, and symptoms from the domain-specific corpus by the information extraction. Second, based on the extracted entities and relationships, a knowledge graph was constructed to form a rule base for the monitoring of data. Then, the named entity recognition model extracted the key information from the electronic medical record to be monitored and matched the knowledge graph to realize the monitoring of the Chinese patent medicines in the prescription. Results. Named entity recognition based on the pretrained model achieved an F1 value of 83.3% on the Chinese patent medicines dataset. On the basis of entity recognition technology and knowledge graph, we implemented a prescription drug monitoring program for Chinese patent medicines. The accuracy rate of combined medication monitoring of three or more drugs of the program increased from 68% to 86.4%. The accuracy rate of drug control monitoring increased from 70% to 97%. The response time for conflicting prescriptions with two drugs was shortened from 1.3S to 0.8S. The response time for conflicting prescriptions with three or more drugs was shortened from 5.2S to 1.4S. Conclusions. The program constructed in this study can respond quickly and improve the efficiency of monitoring prescriptions. It is of great significance to ensure the safety of patients’ medication.

    Abstract: Cancer is a complex disease residing in various tissues of human body, accompanied with many abnormalities and mutations in genomes, transcriptome, and epigenome. Early detection plays a crucial role in extending survival time of all major cancer types. Recent advances in microarray and sequencing techniques have given more support to identifying effective biomarkers for early detection of cancer. MicroRNAs (miRNAs) are more and more frequently used as candidates for biomarkers in cancer related studies due to their regulation of target gene expression. In this paper, the comparative analysis is used to discover miRNA expression patterns in cancer versus normal samples on early stage of eight prevalent cancer types. Our work focuses on the specific miRNAs biomarkers identification and function analysis. Several identified miRNA biomarkers in this paper are matched well with those reported in existing researches, and most of them could serve as potential candidate indicators for clinical early diagnosis applications.

    Abstract: Background . Helicobacter pylori ( H. pylori ) is a common human pathogen, which is closely correlated with gastric cancer (GC). However, the mechanism of H. pylori -related GC has not been elucidated. This study aimed to explore the role of H. pylori infection in GC and find biomarkers for early diagnosis of H. pylori -related GC. Methods . We identified differentially expressed microRNAs (DEMs) and genes (DEGs) from the Gene Expression Omnibus (GEO) dataset, constructed microRNA-(miRNA-)mRNA expression networks, analyzed the function and signal pathway of cross-genes, analyzed the relations between cross-genes and GC prognosis with the Cancer Genome Atlas (TCGA) data, and verified the expression of cross-genes in patients with H. pylor i infection. Results . 22 DEMs and 68 DEGs were identified in GSE197694 and GSE27411 dataset. 16 miRNAs and 509 genes were involved in the expression network, while the cross-genes of the network were mainly enriched in MAP kinase (MAPK) signaling pathway and TGF-beta signaling pathway. Patients with higher expression of hsa-miR-196b-3p, CALML4, or SMAD6 or lower expression of PITX2 or TGFB2 had better outcomes than those with lower expression of hsa-miR-196b-3p, CALML4, or SMAD6 or higher expression of PITX2 or TGFB2 ( P 〈 0.05 ). Patients with H. pylori infection had a higher expression of hsa-miR-196b-3p and CALML4 than those without H. pylori infection ( P 〈 0.05 ). Conclusion . The study of miRNA-mRNA expression network would provide molecular support for early diagnosis and treatment of H. pylori -related GC.

    Abstract: Background. Pyroptosis has been shown to be involved in the overall process of atherosclerosis. This study was aimed at investigating pyroptosis-related gene expression patterns in atherosclerosis and their diagnostic significance. Methods and Results. In GSE100927, fifty-four pyroptosis-related genes were identified. Between atherosclerotic plaques and normal samples, the expression patterns of pyroptosis-related genes were significantly different. In order to construct a pyroptosis-related risk score signature (PRSS), the least absolute shrinkage and selection operator (LASSO) was combined with multivariate logistic regression to screen twelve genes. The diagnostic efficiency of the PRSS performed well in GSE43292, as shown by the results of receiver-operating characteristics (ROCs). Consensus clustering identified two expression patterns of pyroptosis-related genes in different statuses of atherosclerotic plaque in GSE163154. The biological behavior of the different clusters was examined by the gene set variation analysis (GSVA). The Kyoto Encyclopedia of Genes and Genomes (KEGG) and Gene Ontology (GO) enrichment analyses revealed that the differentially expressed genes (DEGs) in the two clusters were enriched in the immune response. The Cytoscape software was used to construct protein-protein interaction (PPI) networks for hub gene screening. Following that, the Drug Gene Interaction Database (DGIdb) was utilized to find 47 possible medicines and chemical compounds that interact with hub genes in atherosclerotic plaques. Conclusion. The results of this study showed that pyroptosis-related genes contribute to the progression of atherosclerosis and may serve as biomarkers in clinical diagnosis as well as novel therapeutic targets for the treatment of AS.

    Abstract: In order to prevent the occurrence of water inrush accidents, it is particularly important to predict the mine water inrush, especially the accurate prediction of the mine water inflow in the coal mining process. Mine water inflow is caused by mining disturbance, groundwater flows into mining panels along the mining-induced fissures, and a depression cone of water table is formed in certain range around the mining sites. Mining panel is usually regarded as an irregular “virtual large diameter well” so as to calculate mine water inflow in China. However, in the mining process, the area, shape, and cross section of goafs are constantly changing, so is the water inflow. Therefore, the central position and influence radius of the “virtual large diameter well” in the spatial and temporal distribution are in a dynamic process of continuous movement and expansion, rather than being confined to a static position. This paper firstly analyzes the formation mechanism of coal mine roof water inflow and the errors and defects in the calculation of mine water inflow by virtual large diameter well (VLDWM). Then, based on the theory of steady flow and combined with the dynamic change process of goaf area and depression cone of water table during mining activities, this paper proposes an improved method and puts forward a concept of dynamic virtual large diameter well (DVLDWM) and establishes a theoretical model of the central position and influence radius of the “mining large diameter well” moving forward. The first (periodic) caving step is taken as the calculation unit and generalized as the “dynamic large diameter well,” which is used to calculate the dynamic water inflow in the process of mine advancing. Taking the No.7208 mining panel in Zhangshuanglou Coalmine in Xuzhou City, Jiangsu Province, as the study area, the mine water inflow was calculated dynamically by using the DVLDWM. The results show that the mine water inflow calculated is similar to the actual mine water inflow of the No.7208 mining panel observed, thus proving the reliability and credibility of the DVLDWM.
