    Abstract: Detection of nonstructural protein (NS1) is an important diagnostic marker during acute phase of dengue infection. Not only for diagnostic purpose, the protein had important role in vaccine design as well, as a candidate for studying virus assembly and maturation. Various researchers employed different expression systems and strategies for recombinant NS1 protein production. Attempts to express NS1 protein in prokaryotic and yeast expression system result in formation of insoluble protein which needs to undergo refolding to attain native structural and functional forms. Here, we report the production of soluble NS1 protein in E. coli by using appropriate vector and employing suitable culture conditions to maximize protein production. Proteins were purified using metal affinity chromatography. SDS-PAGE and western blot analysis reveal the native structure of NS1 protein. Solid phase ELISA using the recombinantly expressed antigen with positive and negative dengue samples showed that the expressed protein retains its antigenic and immunological properties. To our knowledge, this is the first report on the successful production of functionally active recombinant dengue-2 NS1 protein production without undergoing any in vitro posttranslational modification process.

    Abstract: With the rapid rise of the Internet and electronics in people’s life, the data related to it has also undergone a mammoth increase in magnitude. The data which is stored in the cloud can be sensitive and at times needs a proper file storage system with a tough security algorithm. Whereas cloud is an open shareable elastic environment, it needs impenetrable and airtight security. This paper deals with furnishing a secure storage system for the above-mentioned purpose in the cloud. To become eligible to store data a user has to register with the cloud database. This prevents unauthorized access. The files stored in the cloud are encrypted with RSA algorithm and digital fingerprint for the same has been generated through MD5 message digest before storage. The RSA provides unreadability of data to anyone without the private key. MD5 makes it impossible for any changes on data to go unnoticed. After the application of RSA and MD5 before storage, the data becomes resistant to access or modifications by any third party and to intruders of cloud storage system. This application is tested in Amazon Elastic Compute Cloud Web Services.

    Abstract: Stir casting was utilized to generate the composites. The AA 2618–10wt% zirconium carbide 10wt% rice husk ash hybrid composite had the highest hardness value. In comparison to pure AA 2618, it had a 72% increase. The rule of mixture was used to compute theoretical density, whereas the Archimedes rule was utilized to evaluate real density. For AA 2618, AA 2618/zirconium carbide, and rice husk ash hybrid composites, simple carbide inserts were used for turning. Surface roughness, metal removal rate, and tool wear were among the numerous responses examined. RSM was utilized to analyze and optimize the test outcomes. Feed rate was the most critical issue for surface roughness and material removal rate. Tool wear was shown to be most strongly controlled by tool speed, whereas metal removal rate was found to be least significantly influenced by the weight percent of reinforcement.

    Abstract: Thirteen carbon materials were tested as sorbent layers in bar adsorbent microextraction (BAμE) to monitor hint amounts of 10 common pharmaceutical compounds (PhCs) in surface and groundwater matrices such as surface and groundwater, saltwater, spring water, and sewage. The persistence of trace amounts of three organophosphate insect repellent and cis and trans permethrin (PERM) in water quality matrices is suggested using bar adsorptive microextraction in conjunction with microliquid dissolution accompanied by significant volume injection-gas chromatography-mass spectroscopic analysis able to operate in the particular ion monitoring acquisition mode. Using BAμE to compare several sorbent coatings (five porous carbon and six polymers), it was discovered that activated carbon (AC2) was the optimum compromise among specificity and effectiveness. 17-estradiol, estrone, sulfamethoxazole, diclofenac, triclosan, gemfibrozil, 17-ethinylestradiol, mefenamic acid, and clofibric acid were chosen as system drugs to represent different treatment groups. Despite their lower porosity, statistics revealed that low-T-activated hydrochars, made from carbohydrates and a eutectic salt mixture at constant temperature (e.g., 180°C) and autogenerated pressures, could compete at the top level commercially carbonaceous materials in this purpose. These L-T-activated hydrochars had the best overall recovery (between 21.8 and 83.5 percent) for the simultaneous analysis of ten targeted PhCs with very different physical and chemical possessions, utilizing higher-efficiency liquid chromatography diode array identification.
