    Abstract: Oxidative stress plays an important role in the pathogenesis of liver diseases. N-Acetyl-serotonin (NAS) has been reported to protect against oxidative damage, though the mechanisms by which NAS protects hepatocytes from oxidative stress remain unknown. To determine whether pretreatment with NAS could reduce hydrogen peroxide- (H 2 O 2 -) induced oxidative stress in HepG2 cells by inhibiting the mitochondrial apoptosis pathway, we investigated the H 2 O 2 -induced oxidative damage to HepG2 cells with or without NAS using MTT, Hoechst 33342, rhodamine 123, Terminal dUTP Nick End Labeling Assay (TUNEL), dihydrodichlorofluorescein (H2DCF), Annexin V and propidium iodide (PI) double staining, immunocytochemistry, and western blot. H 2 O 2 produced dramatic injuries in HepG2 cells, represented by classical morphological changes of apoptosis, increased levels of malondialdehyde (MDA) and intracellular reactive oxygen species (ROS), decreased activity of superoxide dismutase (SOD), and increased activities of caspase-9 and caspase-3, release of cytochrome c (Cyt-C) and apoptosis-inducing factor (AIF) from mitochondria, and loss of membrane potential (ΔΨ m ). NAS significantly inhibited H 2 O 2 -induced changes, indicating that it protected against H 2 O 2 -induced oxidative damage by reducing MDA levels and increasing SOD activity and that it protected the HepG2 cells from apoptosis through regulating the mitochondrial apoptosis pathway, involving inhibition of mitochondrial hyperpolarization, release of mitochondrial apoptogenic factors, and caspase activity.

    Abstract: Hepatocellular carcinoma (HCC) is a worldwide malignant cancer with high incidence and mortality. Considering the high heterogeneity of HCC, clarifying molecular characteristics associated with HCC development could help improve patients’ outcomes. Pyroptosis is a novel form of cell death and is noted to be implicated in HCC pathogenesis whereas its molecular feature in HCC is unclear. Thus, we intended to clarify the molecular characteristic as well as the clinical significance of pyroptosis for HCC. A systematic bioinformatics analysis was conducted among 40 pyroptosis-related genes based on The Cancer Genome Atlas, the International Cancer Genome Consortium, and the Gene Expression Omnibus databases. A total of 12 HCC-associated pyroptosis-related genes (HPRGs) were identified to be overexpressed in HCC tissues and significantly connected to patients’ poor survival. Through consensus clustering based on the HPRGs’ expression, we found patients could be stratified into two distinctive pyroptosis subtypes, PyLow and PyHigh. The PyHigh group owned a notable lower survival rate and a higher high-grade proportion compared with the PyLow subtype. Besides, patients’ sensitivities to chemotherapeutic drugs also presented distinctive differences between the two subtypes. Indicated by pathway enrichment analysis and immune characteristic difference analysis, the distinctions between the pyroptosis subtypes may be related to tumor immunity. Further, a five-gene risk model composed of BAK1, CHMP4A, CHMP4B, DHX9, and GSDME was established. Subsequent analyses demonstrated that the model could credibly classify patients as low or high risk and was an independent prognostic indicator for HCC. Abnormal expressions of the five genes were validated by biological experiments and new bioinformatics analysis. In conclusion, this study recognized and verified two heterogeneous pyroptosis subtypes and a predictable prognosis model for HCC. Our work may help facilitate the clinical management and treatment of HCC and understand the functions of pyroptosis in oncology.

    Abstract: Objective. The endocrine function of bone in energy metabolism may be mediated by the osteocalcin (OC). We examined the association between OC and energy metabolism among Chinese postmenopausal women. Design and Setting. A cross-sectional cohort study enrolling 1635 participants was conducted using data from the Peking Vertebral Fracture study. Partial correlation analysis was performed to explore the correlation of OC, parathyroid hormone (PTH), or 25-hydroxyvitamin D (25(OH)D) with glycemic and lipid metabolic parameters. A logistic regression model was used to investigate the association of OC, PTH, or 25(OH)D with the prevalence of diabetes and dyslipidemia. Results. Serum levels of OC, PTH, and 25(OH)D were all positively correlated with serum cholesterol levels, whereas only OC was negatively associated with serum glucose level. In the logistic regression model, both OC and PTH were negatively associated with the prevalence of diabetes (odds ratio [OR], 95% confidence interval [95% CI] : 0.967, 0.948–0.986 for OC and 0.986, 0.978–0.994 for PTH). No significant association was found between 25(OH)D and diabetes. Both OC and 25(OH)D, rather than PTH, were associated with abnormalities of high cholesterol levels, such as hypercholesterolemia and high LDL-C levels. Further classifying the population based on the median value of OC and PTH, low OC and low PTH subgroup had the highest OR, 95% CI for diabetes (1.873, 1.287–2.737) and the lowest OR, 95% CI for hypercholesterolemia (0.472, 0.324–0.688) and for high LDL-C (0.538, 0.376–0.771). Conclusion. Among Chinese postmenopausal women, a lower serum level of OC was associated with a higher prevalence of diabetes and lower serum cholesterol levels, and a low PTH concentration could magnify these associations.

    Abstract: The actual operating state of the wind turbine group is influenced by the wake effect and control mode; however, the current models cannot describe the actual operating state very well. A dynamic equivalent modeling method for a doubly fed wind power generator is proposed on the basis of ensuring the accurate description of the wind turbine group. As the clustering index, dominant variables are used in the hierarchical clustering algorithm, which are extracted by principal component analysis. Three dynamic equivalent models of 24 wind turbines are established using PSCAD software platform, which use 13 state variables, wind speed, and dominant variables as clustering indexes, respectively. Furthermore, the active power and reactive power output curves of wind farm are simulated in the case of the three-phase short-circuit fault on the system side or wind speed fluctuation, respectively. The simulation results demonstrate that it is reasonable and effective to extract slip ratio and wind turbine torque as clustering index, and the maximal relative error between the dominant variable equivalent model and 13-state-variable model is only 9.9%, which is greatly lower than that of the wind speed model, K-means clustering model, neural network model, and support vector machine model. This model is easy to implement and has wider application prospect, especially for characteristics analysis of large-scale wind farm connected to power grid.

    Abstract: Optical computed tomography technique has been widely used in pathological diagnosis and clinical medicine. For most of optical computed tomography algorithms, the relaxation factor plays a very important role in the quality of the reconstruction image. In this paper, the optimal relaxation factors of the ART, MART, and SART algorithms for bimodal asymmetrical and three-peak asymmetrical tested images are analyzed and discussed. Furthermore, the reconstructions with Gaussian noise are also considered to evaluate the antinoise ability of the above three algorithms. The numerical simulation results show that the reconstruction errors and the optimal relaxation factors are greatly influenced by the Gaussian noise. This research will provide a good theoretical foundation and reference value for pathological diagnosis, especially for ophthalmic, dental, breast, cardiovascular, and gastrointestinal diseases.

    Abstract: Hydrate formation risk is an important challenge in the development of deep-water gas field. Considering the characteristics of the Lingshui (LS) gas field in the South China Sea and the difference of well structures, a model for calculating wellbore temperature and pressure in deep-water gas production well is proposed and verified by the field data. Combining the hydrate equilibrium models with varied gas components, the prediction method of hydrate formation region in deep-water gas well in the South China Sea is obtained. The hydrate formation regions under different operating conditions for a deep-water gas well in the South China Sea were given by the proposed model. The results show that no hydrate formation risk exists in the production operation, but the risk exists in the shut-in and testing operations. Meanwhile, the determination of the hydrate inhibitor injection parameters during the testing operation is studied, and the influence of the inhibitors’ injection concentration and pressure on preventing gas hydrates is analyzed. This work provides useful advice for the prediction and prevention of hydrate formation risk in the development of deep-water gas fields, especially in the South China Sea.

    Abstract: Objective. To investigate the clinical evaluation of unilateral vertebroplasty for OVCF. Methods. A retrospective analysis was performed on 60 patients treated with PVP from January 2020 to December 2021. Patients were divided into two groups according to the treatment method, 30 patients in the PVP group received PVP and 30 patients in the PCVP group received PCVP. The VAS score, ODI score, bone cement dosage, and leakage were compared between the two groups preoperatively, immediately postoperatively, and 7 and 30 days postoperatively. Results. VAS scores in the PCVP and PVP groups before, immediately after, and 7 days after surgery were P 〉 0.05 , and the difference was not statistically significant; ODI score in group 1 before surgery was not statistically significant ( P 〉 0.05 ); bone cement injection volume in the PVP group was significantly higher than that in the PCVP group ( P 〈 0.05 ), and the difference was statistically significant; the difference in bone cement leakage between the two groups was not statistically significant ( P 〉 0.05 ). Conclusion. Under the same puncture conditions, the PCVP group used the method of injection while retreating to achieve a better bone cement dispersion effect by using less bone cement and achieving uniform dispersion of bone cement. It can relieve the patients’ back pain and improve the back function.

    Abstract: Asthma is a chronic inflammatory disease that cannot be cured. Maresin 1 (MaR1) is a specific lipid synthesized by macrophages that exhibits powerful anti-inflammatory effects in various inflammatory diseases. The goal of this study was to evaluate the effect of MaR1 on allergic asthma using an ovalbumin- (OVA-) induced asthma model. Thirty BALB/c mice were randomly allocated to control, OVA, and MaR1 + OVA groups. Mice were sacrificed 24 hours after the end of the last challenge, and serum, bronchoalveolar lavage fluid (BALF), and lung tissue were collected for further analysis. Western blotting was used to measure the protein level of IκBα, the activation of the NF-κB signaling pathway, and the expression of NF-κB downstream inflammatory cytokines. Quantitative real-time polymerase chain reactions (qRT-PCRs) were used to evaluate the expression levels of COX-2 and ICAM-1 in lung tissues. We found that high doses of MaR1 were most effective in preventing OVA-induced inflammatory cell infiltration and excessive mucus production in lung tissue, reducing the number of inflammatory cells in the BALF and inhibiting the expression of serum or BALF-associated inflammatory factors. Furthermore, high-dose MaR1 treatment markedly suppressed the activation of the NF-κB signaling pathway, the degradation of IκBα, and the expression of inflammatory genes downstream of NF-κB, such as COX-2 and ICAM-1, in the OVA-induced asthma mouse model. Our findings indicate that MaR1 may play a critical role in OVA-induced asthma and may be therapeutically useful for the management of asthma.
