    Abstract: Hyperglycemia-induced oxidative stress and fibrosis play a crucial role in the development of diabetic cardiomyopathy (DCM). Tetrahydrocurcumin (THC), a major bioactive metabolite of natural antioxidant curcumin, is reported to exert even more effective antioxidative and superior antifibrotic properties as well as anti-inflammatory and antidiabetic abilities. This study was designed to investigate the potential protective effects of THC on experimental DCM and its underlying mechanisms, pointing to the role of high glucose-induced oxidative stress and interrelated fibrosis. In STZ-induced diabetic mice, oral administration of THC (120 mg/kg/d) for 12 weeks significantly improved the cardiac function and ameliorated myocardial fibrosis and cardiac hypertrophy, accompanied by reduced reactive oxygen species (ROS) generation. Mechanically, THC administration remarkably increased the expression of the SIRT1 signaling pathway both in vitro and in vivo , further evidenced by decreased downstream molecule Ac-SOD2 and enhanced deacetylated production SOD2, which finally strengthened antioxidative stress capacity proven by repaired activities of SOD and GSH-Px and reduced MDA production. Additionally, THC treatment accomplished its antifibrotic effect by depressing the ROS-induced TGF β 1/Smad3 signaling pathway followed by reduced expression of cardiac fibrotic markers α -SMA, collagen I, and collagen III. Collectively, these finds demonstrated the therapeutic potential of THC treatment to alleviate DCM mainly by attenuating hyperglycemia-induced oxidative stress and fibrosis via activating the SIRT1 pathway.

    Abstract: Objective. To identify the biological function and metabolic pathway of differential metabolites in follicular fluid of senile patients with kidney qi deficiency undergoing in vitro fertilization-embryo transfer (IVF-ET) and observe the effect of kidney-invigorating herbs on IVF outcomes in senile patients. Methods. A total of 95 women undergoing IVF treatment were recruited and divided into three groups, including 34 cases in the treatment group (the senile patients with kidney qi deficiency after the intervention of Chinese medicine), 31 cases in the experiment group (the senile patients with kidney qi deficiency of no intervention of Chinese medicine), and 30 cases in the control group (young women with infertility due to male factor). The three groups of women were treated with long protocol ovarian hyperstimulation; the treatment group was given Qi-Zi-Yu-Si decoction on the day of HCG downregulation. Their IVF clinical outcomes were observed. The metabolites changes of kidney qi deficiency syndrome were analyzed in follicular fluid metabolomics using liquid chromatography-mass spectrometry (UPLC-MS/MS). Results. The syndrome score of kidney qi deficiency syndrome in the treatment group was significantly improved after treatment ( P 〈 0.01 ). Compared with the experiment group, the available embryo rate and implantation rate were increased, and the difference was statistically significant ( P 〈 0.05 ). Progesterone, indoleacrylic acid, 2-propenyl 1-(1-propenylsulfinyl) propyl disulfide, N-acetyltryptophan, decanoylcarnitine, 20a-dihydroprogesterone, testosterone acetate, eicosatrienoic acid, 1H-indole-3-carboxaldehyde, choline, phosphorylcholine, and tryptophan were downregulated in the treatment group. Through pathway analysis, glycerophospholipid metabolism and steroid hormone biosynthesis were regulated in senile patients with kidney qi deficiency after Qi-Zi-Yu-Si decoction intervention. Conclusion. Qi-Zi-Yu-Si decoction can effectively improve the IVF outcome and clinical symptoms of senile patients. Follicular fluid metabolites were significantly changed in senile infertile women with kidney qi deficiency, and the mechanism by which kidney-invigorating herbs improve IVF treatment outcomes may be related to glycerophospholipid metabolism and steroid hormone biosynthesis. This study was registered in the Chinese Clinical Trials Registry Platform (ChiCTR1800014422).

    Abstract: The statistical characteristics of precipitation play important roles not only in flood and drought risk assessments but also in water resource management. This paper implements a statistical analysis to study the spatial and temporal variability in precipitation in the upper reaches of the Hongshui River basin (UHRB), southwestern China, by analysing time series of daily precipitation from 18 weather stations during the period of 1959 to 2015. To detect precipitation concentrations and the associated patterns, three indices, the precipitation concentration index (PCI), precipitation concentration degree (PCD), and precipitation concentration period (PCP), were used. The relationships between the precipitation concentration indices (PCI, PCD, and PCP) and geographic variables (latitude, longitude, and elevation), large-scale atmospheric circulation indices, and summer monsoon indices were investigated to identify specific dependencies and spatial patterns in the precipitation distribution and concentration. The results show that high PCI values were mainly observed in the northeastern portion of the basin, whereas low PCI values were mainly detected in the southwest. The Mann-Kendall test results demonstrate that the majority of the UHRB is characterized by nonsignificant trends in the PCI, PCD, and PCP from 1959 to 2015. The PCP results reveal that rainfall in the UHRB mainly occurs in summer months, and the rainy season arrives earlier in the eastern UHRB than in the western UHRB. Additionally, the PCD results indicate that the rainfall in the western UHRB is more dispersed throughout the year than that in the eastern UHRB. Compared with other geographical factors, longitude is the most important variable that governs the spatial distribution and variations in annual precipitation and the precipitation concentration indices. Due to a combination of topography, the Indian subtropical high, and monsoon weakening, precipitation may be more concentrated in one period, especially in the eastern part of the basin, which increases the risk of drought.

    Abstract: SAHA (suberoylanilide hydroxamic acid or vorinostat) is the first nonselective histone deacetylase (HDAC) inhibitor approved by the US Food and Drug Administration (FDA). SAHA affects histone acetylation in chromatin and a variety of nonhistone substrates, thus influencing many cellular processes. In particularly, SAHA induces selective apoptosis of tumor cells, although the mechanism is not well understood. A series of microarray experiments was recently conducted to investigate tumor cell-selective proapoptotic transcriptional responses induced by SAHA. Based on that gene expression time series, we propose a novel framework for detailed analysis of the mechanism of tumor cell apoptosis selectively induced by SAHA. Our analyses indicated that SAHA selectively disrupted the DNA damage response, cell cycle, p53 expression, and mitochondrial integrity of tumor samples to induce selective tumor cell apoptosis. Our results suggest a possible regulation network. Our research extends the existing research.

    Abstract: Herb-pairs are the basic units of composition in Chinese herbal formulae, where the bridge linking Chinese medicine and prescription consists of two Chinese medicine herbs. The Suanzaoren-Wuweizi herb-pair (SWHP) is commonly used as a sedative or tranquilizer. SWHP has been demonstrated to exert an antianxiety effect in animal models of anxiety. However, little information about its mechanism is available and the effects of SWHP have not been investigated. This study examined the effects of SWHP on ameliorating anxiety-like behaviors by regulating endocannabinoids system (ECS)—brain-derived neurotrophic factor (BDNF)—extracellular regulated protein kinases (ERK) signaling pathway expression, induced by restraint stress (RS) procedures. The antianxiety effects of SWHP on RS rats were then examined through the open-field test (OF) and the elevated plus maze test (EPM). The concentration of BNDF, ERK1/2, p-ERK1/2, cAMP-response element binding protein (CREB), and p-CREB expression in the prefrontal cortex and hippocampus of the rats was then measured by western blot. The number of positive cells of CB1 and CB2 in the rats’ hippocampus CA1 region was measured by immunohistochemistry. These results gave compelling evidence that SWHP could modify anxiety-like behaviors of RS rats through regulation of the ECS-BDNF-ERK signaling pathway. Our study demonstrated that SWHP improved anxiety-like behaviors in RS rat models by regulating the ECS-BDNF-ERK signaling pathway. The findings indicate that SWHP may have a therapeutic application in the RS model of anxiety disorder, which proposes a potential new direction for research into anxiety disorders regarding mechanisms and the development of novel antianxiety drugs.

    Abstract: Neural stem cells (NSCs) are self-regenerating cells, but their regenerative capacity is limited. The present study was conducted to investigate the effect of β -sitosterol-D-glucoside (BSSG) on the proliferation of hippocampal NSCs and to determine the corresponding molecular mechanism. Results of CCK-8 assay showed that BSSG significantly increased NSC proliferation and the effectiveness of BSSG was similar to that of basic fibroblast growth factor and epidermal growth factor. mRNA expression profiling showed that 960 genes were differentially expressed after NSCs were treated with BSSG. Among the 960 genes, IGF1 is considered as a key regulatory gene that functionally promotes NSC proliferation. MicroRNA (miRNA) expression profiling indicated that 30 and 84 miRNAs were upregulated and downregulated, respectively. miRNA-mRNA relevance analysis revealed that numerous mRNAs including IGF1 mRNA were negatively regulated by miRNAs with decreased expression, thereby increasing the corresponding mRNA expression. The increased expression of IGF1 protein was validated by ELISA. Picropodophyllin (PPP, an inhibitor of IGF-1R) inhibition test confirmed that the proliferation-enhancing effect depended on IGF1. This study provided information about BSSG as an efficient and inexpensive growth factor alternative, of which the effect is closely involved in IGF1.

    Abstract: The Zhijiadi Ag-Pb-Zn deposit is located in the central North China Craton. Fluid inclusions (FIs) studies indicate three types of FIs, including aqueous, aqueous-carbonic, and daughter mineral-bearing multiphase inclusions. The daughter minerals in FIs are mainly composed of marcasite, chalcopyrite, calcite, and dolomite. Microthermometric data show that the homogenization temperature and salinity of FIs decrease gradually from early to late stages. Homogenization temperatures from early to main to late stages span from 244 to 334°C, from 164 to 298°C, and from 111 to 174°C, respectively, while their salinities are 4.0–9.9 wt.% NaCl equiv., 0.5–12.7 wt.% NaCl equiv., and 0.2–8.8 wt.% NaCl equiv., respectively. Trapping pressures drop from 203–299 MPa (the early stage) to 32–158 MPa (the main stage). The dropping of pressure and temperature and mixing and/or dilution of ore-forming fluids result in the formation of ore deposit. Combined with C-O-S-Pb isotopic compositions, the initial ore-forming fluids and materials were likely derived from a magmatic system. As a whole, we proposed that this deposit belongs to medium-low temperature hydrothermal deposit related to volcanic and subvolcanic magmatism strictly controlled by the fault zones.

    Abstract: Suppression of milling chatter by disrupting regenerative effect is a well-known method to obtain higher cutting stability domain. In this paper, a dynamic model of the milling process with variable spindle speed and pitch angle considering helix angle and process phase difference is presented. Then, an updated semidiscretization method is applied to obtain the stability chart. After the effectiveness of the proposed method is confirmed by comparisons with the previously published works and the time-domain simulations, lots of analyses are conducted to deeply evaluate the influence of the helix angle, the process phase difference, and feed per tooth on milling stability. Results show that the change of helix angle can result in significant stability discrepancies for both high-speed and low-speed regions. Though the process phase difference has the randomness and immeasurability in the practical application, it has an important influence on the stability and will result in a periodic evolution of the stability with a period π. Also, its recommended values are given for the practical milling process.

    Abstract: The present study was carried out to evaluate the inhibitory effects of ginsenoside Rh2 on nuclear-factor- (NF-) κ B in lipopolysaccharide- (LPS-) activated RAW 264.7 murine macrophages. RAW 264.7 cells were pretreated with indicated concentrations of ginsenoside Rh2 for 1 h prior to the incubation of LPS (1  μ g/mL) for indicated time period. Ginsenoside Rh2 reduced CD14 and Toll-like receptor 4 (TLR4) expressions 24 h after LPS stimulation. Furthermore, ginsenoside Rh2 significantly inhibited TGF-beta-activated kinase 1 (TAK1) phosphorylation 30 min after LPS stimulation. Ginsenoside Rh2 was further shown to inhibit NF- κ B p65 translocation into the nucleus by suppressing I κ B- α degradation. Also, LPS increased mRNA expression of TNF- α and IL-1 α time-dependently, while TQ reduced TNF- α within 3 h and IL-1 α within 1 h. And we firstly found that pretreatment of ginsenoside Rh2 successively inhibited hypoxia-inducible factor- (HIF-) 1 α expression increased by LPS. In conclusion, ginsenoside Rh2 may inhibit LPS-induced NF- κ B activation and reduce HIF-1 α accumulation, suggesting that ginsenoside Rh2 may be considered as a potential therapeutic candidate for chronic inflammatory diseases.
