    Abstract: This study was conducted to evaluate the effect of dietary choline levels on growth performance, liver histology, lipid metabolism and related gene expression of hybrid grouper (♀ Epinephelus fuscoguttatus × ♂ E. lanceolatus) fed high-lipid diets. The fish (initial body weight 6.86 ± 0.01   g ) were fed diets containing different choline levels (0, 5, 10, 15, and 20 g/kg, named D1, D2, D3, D4, and D5, respectively) for 8 weeks. The results shows that (a) dietary choline levels had no significant effect on the final body weight (FBW), weight gain rate (WGR), and specific growth rate (SGR) compared with the control group ( P 〉 0.05 ); (b) with the increase in choline levels, triglyceride (TG), total cholesterol (TC), lipoprotein lipase (LDL), and high-density lipoprotein (HDL) of serum, all showed a tendency to increase and then decrease, and the maximum values were obtained in the D4 group ( P 〈 0.05 ); (c) TG and TC in the liver showed a tendency to decrease first and then increase with the dietary choline levels and reached the minimum value in D3 and D4 groups, respectively ( P 〈 0.05 ); (d) with increased choline levels, hepatic lipase (HL), lipoprotein lipase (LPL), adipose triglyceride lipase (ATGL), and fatty acid synthetase (FAS) in the liver all showed a tendency to increase and then decrease, and the maximum values were obtained in the D4 group, except ATGL that the maximum value was obtained in the D3 group ( P 〈 0.05 ); (e) oil red staining showed that dietary choline significantly reduced hepatic lipid deposition; (f) choline supplementation significantly upregulated the expression of lipolytic genes lipoprotein lipase (lpl), hormone-sensitive lipase (hsl), and peroxisome proliferator-activated receptor α (pparα) and inhibited the expression of lipid synthesis genes fatty acid synthetase (fas), acetyl-CoA carboxylase α (accα), and peroxisome proliferator-activated receptor γ (pparγ). In summary, choline can promote lipid metabolism of hybrid grouper by regulating lipid metabolism-related enzyme activity and gene expression and mitigating the negative effects of high-lipid diets.

    Abstract: The purpose was to investigate the preventive and restorative functions of Lactobacillus pentosus of hybrid grouper (♀ Epinephelus fuscoguttatus×♂ E. lanceolatu) flesh due to ingestion oxidized fish oil (OFO) diet. Basal diets were formulated using OFO as main lipid source and 1.0 × 10 9  CFU/g of L. pentosus was added to basal diet as experimental diet. Control group (OFO: 60 d basal diet), prevention group (OFOP: 30   d   experimental   diet + 30   d   basal   diet ), and restoration group (OFOR: 30   d   basal   diet + 30   d   experimental   diet ) were set up for grouper experiment with triplication. The results show that compared with the OFO group, the OFOP and OFOR groups significantly decreased muscle lipid deposition and lipid peroxidation production. The OFOP and OFOR groups significantly increased the polyunsaturated fatty acid content furthermore to improve nutritional value of muscle compared to the OFO group. Intervention with L. pentosus significantly influenced muscle flavor. OFOP and OFOR groups significantly upregulated expression of partial growth factor-related genes while downregulating expression of antioxidant-related genes. Consequently, the intervention of L. pentosus decreased lipid deposition and improved the nutritional value and flavor of grouper muscle, and it was recommended as a probiotic to prevent the deterioration of aquatic product quality.

    Abstract: Immune checkpoint blockade (ICB) has been explored as a therapeutic strategy to recover the antitumor immune activities against endometrial cancer (EC) escaping from immune surveillance. Increasing evidence has indicated that microsatellite instability (MSI) is a promising biomarker to stratify patients for the ICB therapy. However, even in patients with MSI-High (MSI-H) endometrial cancers, PD-L1 inhibitors, avelumab, and durvalumab have shown only 27% of response rates. Therefore, there is an urgent need to discover new biomarkers for a predictive response to ICB therapy. In this study, we demonstrated that the immune cytolytic activity (CYT) index was significantly correlated with the development and response to immunotherapy in EC. The data showed that higher CYT was significantly associated with better clinical outcome, more antitumor infiltrating immune cells, fewer somatic copy number alterations, but a higher TMB (Tumor mutational burden) status. Furthermore, CYT-high EC was notably relevant to the high expression of various immune checkpoint molecules and showed more effective responses to ICB treatment. Taken together, this study provided new insights into the connection between diverse genetic events and the immune microenvironment in EC and indicated that the CYT status might be a promising biomarker to stratify patients with EC for ICB therapy.

    Abstract: Corneal neovascularization (CoNV) in response to chemical burns is a leading cause of vision impairment. Although glutamine metabolism plays a crucial role in macrophage polarization, its regulatory effect on macrophages involved in chemical burn-induced corneal injury is not known. Here, we elucidated the connection between the reprogramming of glutamine metabolism in macrophages and the development of alkali burn-induced CoNV. Glutaminase 1 (GLS1) expression was upregulated in the mouse corneas damaged with alkali burns and was primarily located in F4/80-positive macrophages. Treatment with a selective oral GLS1 inhibitor, CB-839 (telaglenastat), significantly decreased the distribution of polarized M2 macrophages in the alkali-injured corneas and suppressed the development of CoNV. In vitro studies further demonstrated that glutamine deprivation or CB-839 treatment inhibited the proliferation, adhesion, and M2 polarization of bone marrow-derived macrophages (BMDMs) from C57BL/6J mice. CB-839 treatment markedly attenuated the secretion of proangiogenic factors, including vascular endothelial growth factor-A (VEGF-A) and platelet-derived growth factor-BB (PDGF-BB) from interleukin-4- (IL-4-) regulated M2 macrophages. Our findings revealed that GLS1 inhibition or glutamine deprivation prevented alkali-induced CoNV by inhibiting the infiltration and M2 polarization of macrophages. This work suggests that pharmacological GLS1 inhibition is a feasible and effective treatment strategy for chemical burn-related CoNV in humans.

    Abstract: Vibrio alginolyticus is responsible for significant economic losses in the fish industry. It is urgent to develop a vaccine against V. alginolyticus. In this study, a recombinant Lactococcus lactis expressing the VscO protein of V. alginolyticus (Lc-pMG36e-vscO) is constructed. The Lc-pMG36e-vscO has good hereditary stability. The immune responses in pearl gentian grouper (Epinephelus fuscoguttatus (♀) × E. lanceolatu (♂)) vaccinated with Lc-pMG36e-vscO are evaluated. The titers of specific antibody in the immunized grouper displayed an upward and then downward tendency during the total experiments and reached the peak value of 9.6 at the fourth week after booster immunization. The expression levels of IL-1β, IL-16, TNF-α, and MHC-1α gene in the Lc-pMG36e-vscO group were significantly upregulated after vaccination, indicating that the Lc-pMG36e-vscO could induce an effective and durable immune response. In vivo challenge of the Lc-pMG36e-vscO-immunized fish with V. alginolyticus showed a relative survival percentage of 68%. These results indicated that Lc-pMG36e-vscO could be promising as a candidate oral vaccine against V. alginolyticus.

    Abstract: The tellurite 60TeO2-20PbO-(20-x)ZnO-xBaF2 (x = 1, 3, 5, 6, 7, 9, and 11) series glass samples were prepared. The samples’ SEM, XRD, DSC, density, refractive index, and absorption spectra were tested. A self-made optical platform was used to measure and calibrate the refractive index. The relationship between glass composition and crystallinity was discussed. The effects of density, molar refractive index, and metal standard value on the refractive index of glass samples were studied. The results showed that TPZBF glass has a high refractive index, and the refractive index increases with the increase in BaF2 content.
