    Abstract: Objective . The aim of this study was to investigate the expression of Prominin-1 (CD133) in cancer cells and its potential value as a prognostic indicator of survival in patients with non-small cell lung cancer (NSCLC). Methods . Cancerous tissues and matched normal tissues adjacent to the carcinoma from 239 NSCLC patients were obtained immediately after surgery. Immunohistochemistry of tissue microarrays was used to characterize the expression of CD133 in NSCLC and adjacent tissues. The correlation of CD133 expression with clinical characteristics and prognosis was determined by statistical analysis. Results . CD133 protein expression levels in both the cytoplasm and nucleus were significantly higher in NSCLC tissues compared with corresponding peritumoral tissue ( P 〈 0.05 ). CD133 expression in the nucleus of NSCLC cells was related to tumor diameter ( P = 0.027 ), tumor differentiation ( P 〈 0.001 ), and TNM stage ( P = 0.007 ). Kaplan-Meier survival and Cox regression analyses revealed that high CD133 expression in the nucleus was an independent predictor of poor prognosis of NSCLC, as was high cytoplasmic CD133 expression ( P 〈 0.001 ). Conclusion . Our findings provide the first evidence that high expression of CD133 in both the nucleus and cytoplasm is associated with poor prognosis in NSCLC.

    Abstract: Recent studies demonstrated high expression of lysosome-associated membrane protein 3 (LAMP3) in a variety of malignancies including esophageal squamous cell carcinoma, gastrointestinal cancer, breast cancer, and cervical cancer and its involvement in several biological activities of tumor cells. However, the expression of LAMP3 and its value in oral squamous cell carcinoma (OSCC) remain unclear. In this study, we examined the expression of LAMP3 in OSCC tissue samples and investigated the relationship between LAMP3 and clinical characteristics of patients with OSCC. We examined mRNA and protein levels of LAMP3 in OSCC tissues and neighboring normal tissues using quantitative real-time polymerase chain reaction and immunohistochemistry analyses, respectively. Both the mRNA and protein levels of LAMP3 were significantly higher in OSCC tissues than in adjacent normal tissues. Chi-square analysis showed that the high LAMP3 expression was notably linked to the degree of tumor differentiation and advanced TNM stage. Univariate and multivariate analyses showed that the high LAMP3 expression was an independent prognostic marker in OSCC. Our results suggest that LAMP3 might act as a potential anticancer target and a prognostic marker in patients with OSCC.

    Abstract: A spectral deferred correction method is presented for the initial value problems of fractional differential equations (FDEs) with Caputo derivative. This method is constructed based on the residual function and the error equation deduced from Volterra integral equations equivalent to the FDEs. The proposed method allows that one can use a relatively few nodes to obtain the high accuracy numerical solutions of FDEs without the penalty of a huge computational cost due to the nonlocality of Caputo derivative. Finally, preliminary numerical experiments are given to verify the efficiency and accuracy of this method.

    Abstract: Background. Receptor expressed in lymphoid tissues-like 2 (RELL2), which is a member of RELT family, is closely associated with the plasma membrane and acts as a modulator for RELT signaling. Overexpression of RELL2 induces the activation of MAPK14/p38 cascade and apoptosis. However, whether RELL2 contributes to cancers remains unclear. Here, we examined its role in cancer patient prognosis and various tumors. Methods. We used several bioinformatics methods, specifically gene set enrichment analysis (GSEA), ScanNeo, and ESTIMATE, to analyze the CCLE dataset, GTEx dataset, and TCGA dataset. We investigated the possible association of RELL2 with the microsatellite instability (MSI) of various tumors, tumor mutational burden (TMB), immune checkpoint, immune neoantigens, immune microenvironment, and patient prognosis. Result. RELL2 is highly expressed in cancer compared with normal tissues. RELL2 expression is linked with worse progression-free interval and overall survival in numerous cancers. In most cancers, high RELL2 expression was related to a poor prognosis. RELL2 expression was significantly associated with the tumor microenvironment, MSI, and TMB. RELL2 expression is strongly associated with phenotypes that are of major clinical significance, particularly those associated with immune neoantigens and the expression profiles of immune checkpoint genes in pan-cancer. RELL2 expression strongly linked with the expressions of methyltransferases and DNA repair genes. It also significantly correlated with multiple signaling pathways through gene set enrichment analysis. Conclusion. RELL2 may be a prognostic biomarker in pan-cancer and may have an important function in tumorigenesis and progression.

    Abstract: Background . Low free triiodothyronine (fT3) levels are generally associated with poor prognosis in patients with heart diseases, but this is controversial and there is a lack of data about ST-elevation myocardial infarction (STEMI) in Chinese patients. Objective . To assess the association between fT3 levels and the prognosis of patients with STEMI. Methods . This was a prospective observational study of 699 consecutive patients with STEMI treated at the Xinqiao Hospital between January 1, 2013, and December 31, 2014. The patients were divided into the low fT3 (fT3 〈 3.1 pmol/L; n = 179 , 27.5%) and normal fT3 (fT3 ≥ 3.1 pmol/L; n = 473 , 72.5%) groups according to fT3 levels at admission. Patients were followed up at 1, 3, 6, and 12 months for all-cause death and major adverse cardiac events (MACE). Results . During the 1-year follow-up, there were 70 all-cause deaths (39.1%) in the low fT3 group and 40 (8.5%) in the normal fT3 group ( P 〈 0.001 ). MACE occurred in 105 patients (58.7%) in the low fT3 group and 74 (15.6%) in the normal fT3 group ( P 〈 0.001 ). Multivariate Cox proportional hazards regression analysis indicated that fT3 levels were independently associated with 30-day and 1-year all-cause death [30-day: hazard ratio (HR) = 0.702, 95% confidence interval (95% CI): 0.501–0.983, P = 0.04 ; 1-year: HR = 0.557, 95% CI: 0.411–0.755, P 〈 0.001 ] and MACE (30-day: HR = 0.719, 95% CI: 0.528–0.979, P = 0.036 ; 1-year: HR = 0.557, 95% CI: 0.445–0.698, P 〈 0.001 ). Conclusion. Low fT3 levels were strongly associated with poor prognosis in patients with STEMI. Measurement of fT3 levels may be a valuable and simple way to identify high-risk STEMI patients.

    Abstract: This work is aimed at presenting a new numerical scheme for COVID-19 epidemic model based on Atangana-Baleanu fractional order derivative in Caputo sense (ABC) to investigate the vaccine efficiency. Our construction of the model is based on the classical SEIR, four compartmental models with an additional compartment V of vaccinated people extending it SEIRV model, for the transmission as well as an effort to cure this infectious disease. The point of disease-free equilibrium is calculated, and the stability analysis of the equilibrium point using the reproduction number is performed. The endemic equilibrium’s existence and uniqueness are investigated. For the solution of the nonlinear system presented in the model at different fractional orders, a new numerical scheme based on modified Simpson’s 1/3 method is developed. Convergence and stability of the numerical scheme are thoroughly analyzed. We attempted to develop an epidemiological model presenting the COVID-19 dynamics in Italy. The proposed model’s dynamics are graphically interpreted to observe the effect of vaccination by altering the vaccination rate.

    Abstract: Hazardous rock refers to an unstable rock block that is cut by weak structural planes and gradually separates from the slope. Hazardous rock generally collapses rapidly, and at present, it is challenging to effectively identify the separation degree of the rock and accurately predict its sudden failure. In this study, focusing on a hazardous rock with tilt behavior, a microelectromechanical system (MEMS) acceleration sensor is used in combination with the calculation principle of the included angle of the space vector to establish a microtilt angle monitoring method. A physical model test is designed, in which a thermally sensitive material (with heat-sensitive strength) is adopted as the weak structural plane of the hazardous block, and the change in the tilt angle during the process of block instability is monitored at a sampling frequency of 1000 Hz. The test results show that the accelerated evolution of the tilt angle is a precursor to hazardous rock failure. In the rapid acceleration stage, the reciprocal of the tilt angle rate is approximately linear with time, and a correlation equation is obtained. Assuming that the change rate of the tilt angle is approximately infinite, the failure time of hazardous rock can be predicted using the correlation equation. In addition, the effectiveness of the instability prediction method based on microtilt angle monitoring is verified by analyzing the long-term monitoring data of hazardous rock.

    Abstract: The dry leaf of Apocynum venetum tea extracts (AVTEs) belonging to the Apocynaceae family is a traditional Chinese medicine. The aim of this study is to identify the bioactive components of AVTE and analyse its antioxidant and anticancer activity in vitro. Method . Flavones and polyphenols in AVTE were determined by high-performance liquid chromatography (HPLC) assay. The scavenging capacity of tea extracts to 1,1-diphenyl-2-picrylhydrazyl (DPPH); 2,2′-azino- bis -(3-ethylbenzthiazoline-6-sulfonic acid) diammonium salt (ABTS); hydroxyl (OH); and superoxide anion-free radicals were investigated by spectrophotometry. We also detailed the cytotoxicity assay of AVTE (50, 100, and 200  μ g/mL) to human embryonic kidney 293T cells, the protective effect of AVTE on 293T cells induced by hydrogen peroxide (0.3 mmol/L), and the anticancer effect against the human hepatoma HepG2 cells via 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT) assay. We investigated the antioxidative effects of AVTE in human embryonic kidney 293T cells and the anticancer mechanism in HepG2 human hepatoma cells via quantitative real-time reverse transcription-polymerase chain reaction (RT-qPCR) assay. Results . HPLC analysis showed that AVTEs contain neochlorogenic acid, chlorogenic acid, rutin, isoquercetin, isochlorogenic acid B, astragalin, isochlorogenic acid C, rosmarinic acid, quercetin, and trans-cinnamic acid. These extracts have high antioxidant activity and dose-dependent relation through free radical scavenging experiments. The cell viability of 293T cells treated with hydrogen peroxide (0.3 mmol/L) was significantly lower than that of normal cells, and the cell viability of oxidatively stressed 293T cells after AVTE (50, 100, and 200  μ g/mL) treatment was significantly improved ( P 〈 0.05 ). Moreover, cytotoxicity experiments showed that the survival rate of 293T cells was over 90%, but the proliferation of HepG2 cells was significantly inhibited in a dose-dependent manner by AVTE. Furthermore, cytoprotective effects in 293T cells were induced via upregulation of glutathione peroxidase (GSH-Px), GSH, superoxide dismutase (SOD), and catalase (CAT) antioxidant-related factors, as well as apoptosis in HepG2 cells was induced via upregulation of caspase-3, caspase-9, p21, and p53 apoptosis-associated factors, as assessed via mRNA expression levels after treatment with AVTE, which were consistent with the results of antioxidant gene detections. As a conclusion, AVTE appears to be an effectively functional drink, due to its rich functional components and antioxidant and anticancer activities.

    Abstract: GRO β (CXCL2) is a chemokine produced by endotoxin-treated macrophages that mediates inflammation and tumor development. However, little is known about GRO β expression in gastrointestinal stromal tumors (GIST) or the relationship between GRO β expression and clinical attributes of GIST. GRO β expression was examined via immunohistochemical staining of 173 GIST samples using tissue microarray. The relationship between GRO β expression and relevant patient and tumor characteristics was assessed, using chi-square tests. Univariate and multivariate analysis was carried out using the Cox regression method. High GRO β cytoplasm staining was detected in 56 (32.4%) specimens; high GRO β nuclear staining was detected in 64 (37.0%) specimens. High GRO β cytoplasm staining was significantly associated with patients’ age ( P = 0.043 ) and tumor location ( P = 0.014 ), while high GRO β nucleus staining was significantly associated with mitotic index ( P = 0.034 ), tumor location ( P = 0.049 ), and AFIP-Miettinen risk classification ( P = 0.048 ). Kaplan-Meier survival curves showed GIST patients with low GRO β cytoplasm expression ( P = 0.023 ) and mitotic index 〈 6 per 50 HPFs ( P = 0.026 ) to have a more favorable prognosis. These findings indicate that GRO β expression correlates with malignant GIST phenotypes and could be an unfavorable prognostic marker in patients with GIST.
