Abstract: Uterine fibroids are one of the most common and yet understudied diseases in women mainly during their reproductive years. The study analyzed the efficacy and safety of oral administration of mifepristone combined with ultrasound-guided radiofrequency ablation in treating patients with uterine fibroids, as well as changes of inflammatory cytokines including procalcitonin (PCT), high-sensitivity C-reactive protein (hs-CRP), and tumor necrosis factor-alpha (TNF-α), adhesion molecules including monocyte chemotactic protein 1 (MCP-1) and soluble intercellular adhesion molecule 1 (sICAM-1), and growth factors including vascular endothelial growth factor (VEGF), epidermal growth factor (EGF), basic fibroblast growth factor (bFGF), transforming growth factor-β (TGF-β), and TGF-β receptor. A total of 130 patients who were admitted into our hospital for uterine fibroids from December 2016 to June 2018 were included, among which 65 women were given ultrasound-guided radiofrequency ablation alone (control group) and the remaining women were given oral administration of mifepristone combined with ultrasound-guided radiofrequency ablation (experimental group). Reduced volumes of uterus and fibroids were observed in both two groups after treatment ( P 〈 0.05 ). It was found that the experimental group exhibited higher total effective rate and smaller volumes of uterus and fibroids than the control group ( P 〈 0.05 ). Following treatment, the levels of MCP-1, sICAM-1, VEGF, EGF, bFGF, TGF-β, TGF-β receptor, PCT, hs-CRP, and TNF-α in the serum in the two groups were declined ( P 〈 0.05 ), while this decline was more significantly in the experimental group than the control group ( P 〈 0.05 ). Besides, there was no significant difference in the incidence rate of adverse reactions between the two groups ( χ 2 = 0.781 , P 〉 0.05 ). These results collectively indicate the high efficacy and safety of oral administration of mifepristone combined with ultrasound-guided radiofrequency ablation in treating patients with uterine fibroids, as this combination therapy could inhibit the expressions of inflammatory cytokines, adhesion molecules, and growth factors.

Abstract: Ultrasound imaging technology has the advantages of noninvasiveness, real-time, low price, and easy operation. It is one of the most used diagnostic tools for early detection and classification of premature ovarian failure. Although the rapid development of computer-aided diagnosis has provided a great help to the ultrasound diagnosis of premature ovarian failure, it still has many limitations and shortcomings, so this paper adopts transfer learning and feature fusion algorithms to improve the identification and prediction efficiency of premature ovarian failure. In this study, the POF group and the control group both adopted a unified scale. From the four aspects of sociological characteristics, past medical history, environmental factors, and living habits, a dedicated person asked and filled out the scale face to face. All patients participating in the experiment underwent ultrasound examinations. In this paper, the bottom-level feature fusion method is used to improve classification performance. The experiment uses 100 epochs. After each epoch training is completed, we used all the data and labels of the target domain to test. All experiments were performed five times, and the result is the average of five experiments. All the results of baseline and direct classification without migration use the average of five experimental results as the result. Migrating the features extracted by the InceptionV3 network has the best performance for predicting premature ovarian failure. Its classification accuracy is as high as 85.13%, and the F1 value is 0.78. The results show that the migration learning and feature fusion algorithms used in this paper can provide reliable predictive analysis and decision support for doctors in the diagnosis of premature ovarian failure.

Abstract: Background. The associations between hip/knee osteoarthritis (OA) and various psychiatry disorders, as well as the underlying genetic determinants, remain unclear. Methods. Based on the community-based prospective data of UK Biobank, we constructed two matched cohorts to assess the bidirectional associations between OA and five common psychiatry disorders. Then, we further examined the existence of overall genetic association for those disease pairs with demonstrated phenotypic association through polygenic risk score (PRS) prediction using individual-level genotyping data of UK Biobank and LD score regression (LDSC) analysis utilizing publicly available GWAS summary statistics. Last, also based on GWAS summary statistics, we performed enrichment analyses to pinpoint specific genetic determinants that might contribute to the observed overall genetic shares. Results. The phenotypic analyses revealed an elevated risk of hip/knee OA among individuals with any psychiatric disorders, compared to their matched unexposed individuals (hazard ratio   HR = 1.62 , 95% confidence interval (CI): 1.57-1.68), and vice versa (HR =1.93, 95% CI: 1.83-2.04). We further observed positive associations of knee OA with depression and stress-related disorder in the PRS analyses, which corroborated with the results of LDSC analyses ( r for genetic = 0.20 (95% CI: 0.15-0.25) and r for genetic = 0.29 (95% CI: 0.19-0.40), respectively). Using GWAS summary statistics, we identified several shared genes and pathways, particularly the biological process related to HDAC histones, between knee OA and depression/stress-related disorder. Conclusions. Our study demonstrated a bidirectional association between OA and multiple psychiatric disorders, and the findings of shared genetic architectures between knee OA and depression/stress-related disorder provided possible targets for further mechanistic exploration and intervention development.

Abstract: A unique orthogonal crosslinker-induced hydrogel, whose mechanical strength can be tunable by the crosslinker topology upon thermal treatment, is described herein. The crosslinker containing cationic moieties and crosslinkable styrenyl groups was employed for the preparation of orthogonally crosslinked hydrogels having ionic and covalent characteristics. The manipulation of the orthogonal crosslinkers topology and the ionic bond strength between cationic and anionic moieties facilitated the control of the mechanical properties. Short-term temporal modulation of hydrogel moduli, a key factor of the substrates for cell development, was demonstrated and could provide dynamic microenvironment for biological process. In addition, on-demand control of the elastic properties of the hydrogels by application of a thermal stimulus provides new avenues to regulate cell growth. Furthermore, the orthogonality of the crosslinkers allowed molecular functionalization of a wide range of molecules of interest to the hydrogels by thiol-ene Michael addition (nucleophilic addition of sulfhydryl and carbon-carbon double bonds) in a friendly manner as demonstrated in our work.

Abstract: The influences of hot air drying (AD), medium- and short-wave infrared drying (IR), instant controlled pressure drop drying (DIC), and vacuum freeze drying (FD) on cell wall polysaccharide modification were studied, and the relationship between the modifications and texture properties was analyzed. The results showed that the DIC treated apple chips exhibited the highest crispness (92) and excellent honeycomb-like structure among all the dried samples, whereas the FD dried apple chips had low crispness (10), the minimum hardness (17.4 N), and the highest volume ratio (0.76) and rehydration ratio (7.55). Remarkable decreases in the contents of total galacturonic acid and the amounts of water extractable pectin (WEP) were found in all the dried apple chips as compared with the fresh materials. The highest retention of WEP fraction (102.7 mg/g AIR) was observed in the FD dried apple chips, which may lead to a low structural rigidity and may be partially responsible for the lower hardness of the FD apple chips. In addition, the crispness of the apple chips obtained by DIC treatment, as well as AD and IR at 90°C, was higher than that of the samples obtained from the other drying processes, which might be due to the severe degradation of pectic polysaccharides, considering the results of the amounts of pectic fractions, the molar mass distribution, and concentrations of the WEP fractions. Overall, the data suggested that the modifications of pectic polysaccharides of apple chips, including the amount of the pectic fractions and their structural characteristics and the extent of degradation, significantly affect the texture of apple chips.

Abstract: Adenosine receptors A1 (A1AR) and A2a (A2aAR) play an important role in regulating glutamate uptake to avoid glutamate accumulation that causes excitotoxicity in the brain; however, the precise mechanism of the effects of A1AR and A2aAR is unclear. Herein, we report that expression of the A1AR protein in the astrocyte membrane and the level of intracellular glutamate were decreased, while expression of the A2aR protein was elevated in cells exposed to oxygen-glucose deprivation (OGD) conditions. Coimmunoprecipitation (Co-IP) experiments showed that A1AR interacts with A2aAR under OGD conditions. The activation of A1AR and inactivation of A2aAR by 2-chloro-N6-cyclopentyladenosine (CCPA) and SCH58251, respectively, partly reversed OGD-mediated glutamate uptake dysfunction, elevated EAAT2, and PPAR γ protein levels, and suppressed the expression of Ying Yang 1 (YY1). Both the silencing of YY1 and the activation of PPAR γ upregulated EAAT2 expression. Moreover, YY1 silencing elevated the PPAR γ level under both normal and OGD conditions. Histone deacetylase (HDAC)1 was found to interact with YY1, and HDAC1 silencing improved PPAR γ promoter activity. Taken together, our findings suggest that A1AR-A2aAR heteromers regulate EAAT2 expression and glutamate uptake through the YY1-mediated recruitment of HDAC1 to the PPAR γ promoter region.

Abstract: Purpose . To evaluate the role of SPARC in the antiproliferation effect of MMC on human Tenon’s fibroblasts (HTF). Method . Sixteen PACG patients aged 59 ± 10 years (31–72 years), including 6 males and 10 females, were recruited. Tenon tissue was harvested during filtering surgery. Cell density was evaluated after MMC application with different concentrations and application times, by which the optimized MMC application modality was determined. MMC, si-SPARC, or SPARC protein was used when needed to evaluate the cell densities under different conditions, by which the role of SPARC in MMC-mediated antifibrotic process was identified. Results . Considering that the cell densities, as well as SPARC expression on mRNA and protein levels, are relatively stable when the MMC concentration is higher than 0.02% and exposure time longer than 90 s, we chose the MMC application pattern with 0.02% and 90 s as an optimized pattern for the downstream work. Compared to control, the si-SPARC and MMC downregulated the SPARC protein by 91% ( P 〈 0.01 ) and 65% ( P 〈 0.01 ) and mRNA by 96% ( P 〈 0.01 ) and 64% ( P 〈 0.01 ), respectively. MMC decreases the cell densities by 53.50% compared to control. si-SPARC + MMC dramatically deceased the cell density no matter compared to the control group ( P 〈 0.01 ) or MMC group ( P 〈 0.01 ); correspondingly, the relative collagen gel area in the MMC + si-SPARC group was higher than that in the MMC group or si-SPARC group ( P 〈 0.05 ). The reactive oxygen species expression in the MMC + si-SPARC group is higher than that in the MMC group ( P 〈 0.05 ). Conclusion . This study demonstrates that in HTF, (1) MMC downregulates the expression of SPARC in protein and mRNA levels; (2) SPARC depletion has synergistic effect on the antifibrotic effect of MMC; and (3) reactive oxygen species are the possible mediator in the antifibrotic effect of MMC and si-SPARC.

Abstract: In this paper, we are concerned with a class of quaternion-valued stochastic neural networks with time-varying delays. Firstly, we cannot explicitly decompose the quaternion-valued stochastic systems into equivalent real-valued stochastic systems; by using the Banach fixed point theorem and stochastic analysis techniques, we obtain some sufficient conditions for the existence of square-mean pseudo almost periodic solutions for this class of neural networks. Then, by constructing an appropriate Lyapunov functional and stochastic analysis techniques, we can also obtain sufficient conditions for square-mean exponential stability of the considered neural networks. All of these results are new. Finally, two examples are given to illustrate the effectiveness and feasibility of our main results.

Abstract: The knee osteoarthritis is a common joint disease that causes pain and inconvenience. Clinically, patients with knee osteoarthritis often have response points on the gastrocnemius. Gastrocnemius plays an essential role in stabilizing joints and changing gait and pace, which also has a close relationship with the knee joint. The objective of this study is to determine changes in the tibiofemoral joint after medial and lateral gastrocnemius injury. Rabbits were divided into a medial gastrocnemius injury group, a lateral gastrocnemius injury group, and a control group with two intervals: 6 and 8 weeks after modeling of the semisevered gastrocnemius. The gastrocnemius was weighed and sectioned for histology. The joint space and subchondral bone were observed using X-ray and microcomputed tomography. The cartilage was observed histologically using Safranin O fast green and Masson and immunohistochemically using antibodies to collagen type II, matrix metalloproteinase 13, and integrin beta1. Results showed muscle fiber atrophy, and fibrotic changes occurred after gastrocnemius semidissociation. After gastrocnemius injury, the femoral condyle of the tibiofemoral joint produced abnormal sclerosis and bone degeneration. The pathological changes of cartilage included disordered or reduced cell alignment, cartilage matrix loss, and collagen loss due to decreased collagen type II and increased matrix metalloproteinase 13 activity. The increase of integrin beta1 in the injured group may be related to mechanical conduction process. The results suggest that gastrocnemius injury is an essential factor in tibiofemoral arthritis.
