    Abstract: Fibroblast growth factor-inducible molecule 14 (Fn14) plays a principal role in triggering tubular damage during septic acute kidney injury (AKI). Here, we explore the mechanism underlying Fn14 deregulation in septic AKI. We identify Fn14 as a bona fide target of miR-19a, which directly binds to 3 ′ UTR of Fn14 for repression independent of cylindromatosis (CYLD), the deubiquitinase (DUB) downstream of miR-19a, and thereby antagonizes the LPS-induced tubular cell apoptosis. Genetic ablation of Fn14, but not of CYLD, abolishes the ability of miR-19a to antagonize the tubular apoptosis by lipopolysaccharide (LPS). In mice, systemic delivery of miR-19a confers protection against septic AKI. Our findings implicate that miR-19a may serve as a promising therapeutic candidate in the prevention of septic AKI.

    Abstract: To widen the application of nanofibers web in the field of medical health materials, a new Ag/amino-terminated hyperbranched polymer (HBP)/polyacrylonitrile (PAN) nanofiber web with excellent antimicrobial activity and filtration property was produced with Ag/HBP dispersion solution and PAN nanofiber. Ag/HBP dispersion solution was prepared with HBP as reducer and stabilizer, and Ag/HBP/PAN nanofiber was prepared by modifying electrospun PAN nanofiber with Ag/HBP aqueous solution. The characterization results showed that spherical Ag nanoparticles were prepared and they had a narrow distribution in HBP aqueous solution. The results of Ag/HBP/PAN nanofiber characterized with SEM and EDS showed that the content of silver nanoparticles on the surface of PAN nanofiber was on the increase when the treating temperature rose. The bacterial reduction rates of HBP-treated PAN nanofiber against S. aureus and E. coli were about 89%, while those of the Ag/HBP/PAN nanofiber against S. aureus and E. coli were 99.9% and 99.96%, respectively, due to the cooperative effects from the amino groups in HBP and Ag nanoparticles. Moreover, the small pores and high porosity in Ag/HBP/PAN nanofiber web resulted in high filtration efficiency (99.9%) for removing smaller particles (0.1  μ m~0.7  μ m), which was much higher than that of the gauze mask.

    Abstract: Background. To investigate the role of gray matter (GM) volume in the identification of HIV-positive patients with HIV-associated neurocognitive impairment (HAND) using a machine learning approach from normal healthy controls. Methods. Twenty-seven HIV-infected patients and 14 healthy controls were enrolled in our study. Each set of BRAVO images was postprocessed using DPARSF3.1 to coregister all brains on the MNI template, and volume extraction of 90 brain regions was performed using custom-designed code. The machine learning method was performed using PRoNTo2.1.1 toolbox. The differences in brain volume between the HAND and non-HAND groups were analyzed. Results. GM volume effectively distinguished HIV-positive patients from healthy subjects with an AUC equals to 0.73. The sensitivity, specificity, and accuracy of the established classification were 85.19%, 42.86%, and 70.73%, respectively. GM volume value of the top ten brain regions was related to digit symbols, trail making test, digit span, vocabulary fluency, stroop C time, stroop CW time, CD4, and neuropsychological group. Conclusions. A machine learning approach facilitates early diagnosis of HAND in HIV patients by MRI-based GM volume measurement.

    Abstract: A new method for the design of elliptical beam reflector antenna is presented in this paper. By means of the basic principles of ring-focus antenna, a circularly symmetric feed and two specially shaped reflectors are used to form an elliptical beam antenna. Firstly, the design process of this ring-focus elliptical beam antenna is studied in detail. Transition function is defined and used in the design process. Then, combining the needs of practical engineering, a ring-focus elliptical beam reflector antenna is manufactured and tested. The gain at center frequency (12 GHz) is 37.7 dBi with an aperture efficiency of 74.6%. 3 dB beam-width in φ = 0 ° and φ = 9 0 ° plane is 2 . 6 ° and 1 . 4 ° , respectively. Ratio of the elliptical beam (ratio of 3 dB beam-width in φ = 0 ° and φ = 9 0 ° plane) is 2.6 / 1.4 = 1.85 , substantially equal to designed ratio 2. Simulating and testing results match well, which testify the effectiveness of this design method.

    Abstract: Background . Long noncoding RNAs (lncRNAs) were previously found to be closely related to the pathogenesis of diabetes. Objectives . To reveal the differentially expressed lncRNAs and messenger RNAs (mRNAs) involved in type 2 diabetes mellitus (T2DM) and latent autoimmune diabetes in adults (LADA) and predict the lncRNA target genes to derive their expression profiles for the diagnosis of T2DM and LADA and their differential diagnosis. Methods . Twelve venous blood samples were collected from T2DM patients, LADA patients, and nondiseased subjects to obtain total RNAs. After removing rRNA from total RNAs to establish the desired library for sequencing, quality control and quantification analyses were carried out. The fragments per kilobase of exon model per million reads mapped (FPKM) of lncRNAs were calculated to construct the gene expression profiles of lncRNAs and mRNAs. Fold changes (fold change: 2.0) and p values ( p value: 0.05, FPKM ≥ 0.1) were calculated, and then differentially expressed lncRNAs and mRNAs were screened. To obtain the significantly coexpressed lncRNA-mRNA pairs, the lncRNA target genes were deduced according to the association between adjacent sites. Results . Compared to nondiseased controls, 68,763 versus 28,523 lncRNAs and 133 versus 1035 mRNAs were significantly upregulated and significantly downregulated, respectively, in T2DM patients. For LADA patients, 68,748 versus 28,538 lncRNAs and 219 versus 805 mRNAs were significantly upregulated and significantly downregulated, respectively, relative to nondiseased controls. Compared to T2DM patients, 74,207 versus 23,079 lncRNAs and 349 versus 137 mRNAs were significantly upregulated and significantly downregulated, respectively, in LADA patients. Based on the correlation analysis, seven lncRNA-mRNA pairs (BTG2, A2M, HECTD4, MBTPS1, DBH, FLVCR1, and NCBP2) were significantly coexpressed, and two lncRNAs (ENST00000608916 and ENST00000436373) were newly discovered. Conclusion . Significant differences in lncRNA expression were discovered among the three groups. Furthermore, after predicting lncRNA expression profiles, GO/KEGG pathway analysis could deduce the target gene function.

    Abstract: In order to study the effect of mineral composition on the physical properties of lateritic clays, the mineral compositions of four kinds of lateritic clay from Guangxi province in China were quantitatively analyzed by means of DTA, XRD, SEM, XRF and total chemical element analysis. On this basis, the variation law of macroscopic physical properties of lateritic clay was analyzed from the perspective of mineralogy. The results show that kaolinite and goethite are the main minerals of lateritic clays from Guangxi in China. The mineral compositions of these four samples are quantitatively and accurately analyzed by Bogue’s method. According to the study on mineral compositions of soil samples, the physical properties of lateritic clay are related to its clay minerals and cemented substances. The liquid limit (LL) or plastic limit (PL) increases with the increase of the ability and content of clay minerals to adsorb water. The lateritic clay is dominated by kaolinite minerals with low adsorption capacity, so the observed boundary water content (LL and PL) of high value of lateritic clays can be explained by the contribution of “inert water” in soil pores. The iron-bonded cement minerals (the form of existence is goethite) in lateritic clay have great influence on the liquid and plastic limits, which decrease linearly with the increase of goethite, whereas the goethite has relatively smaller effect on shrinkage characteristics. It is believed that the shrinkage characteristics of lateritic clay may also be affected by other factors.

    Abstract: In the present study, the hypoglycemic, hypolipidemic, and antioxidative effects of metformin (MET) combined with Jiang Tang Xiao Ke (JTXK) granule derived from the “Di Huang Tang” were evaluated in mice with type 2 diabetes mellitus (DM) induced by high-fat diet/streptozotocin. DM mice were orally treated with MET (0.19 g/kg) either alone or combined with different doses (1.75, 3.5, or 7 g/kg) of JTXK for 4 weeks. Results showed that the serum and hepatic glucose, lipids, and oxidative stress levels were elevated in DM mice, when compared with the normal mice. MET treatment decreased FBG and serum glucagon levels of DM mice. Combination treatment with MET and JTXK 3.5 g/kg increased the hypoglycemia and insulin sensitivity at 4 weeks when compared with the DM mice treated with MET alone. However, neither MET nor MET/JTXK treatment could completely reverse the hyperglycemia in DM mice. JTXK enhanced the serum triglyceride (TG) and hepatic lipid-lowering effect of MET in a dose-dependent manner in DM mice. JTXK 1.75 and 3.5 g/kg improved the hepatoprotective effect of MET in DM mice. Synergistic effect of combination treatment with MET and JTXK on antioxidant stress was also found in DM mice compared with MET alone.

    Abstract: Objective. The objective is to observe the synergistic and attenuating effect of electroacupuncture (EA) on aconitine (ACO) in improving heart failure (HF) and to explore its underlying mechanism for calcium regulation. Methods. Twenty-four male Sprague-Dawley rats were randomly divided into four groups: normal control (NC) (n = 6), HF(n = 6), ACO (n = 6), and ACO + EA (n = 6). The maximum rates of left ventricular pressure rising and declining (±dp/dtmax), arrhythmia, the left ventricular systolic pressure (LVSP), ejection fraction (LVEF), and fractional shortening (LVFS) were measured by physiological recorder and ultrasound, respectively. Protein expressions of sarcoplasmic/endoplasmic reticulum Ca2+ ATPase (SERCA2a), phospholamban (PLB), and Na+-Ca2+ exchange (NCX1) in the left ventricle tissue were detected by fluorescence immunoblotting. Results. Compared with the NC group, LVSP, ±dp/dtmax, LVEF, and LVFS were decreased in the HF group; compared with the HF group, LVSP, ±dp/dtmax, LVEF, and LVFS were significantly increased in the ACO + EA group. Compared with the ACO group, the incidence and the degree of arrhythmia were significantly reduced in the ACO + EA group. Compared with the NC group, the activity of SERCA2a was decreased, and the expression of PLB and NCX1 was enhanced in the HF group; compared with the HF group and ACO group, the activity of SERCA2a was increased, and the expression of PLB and NCX1 was significantly attenuated in the ACO + EA group. Conclusions. EA plays a synergistic and attenuated role in ACO improving HF, and the mechanism may be related to the enhancement of the SERCA2a activity and the decrease of the expression of PLB and NCX1 in cardiomyocytes.

    Abstract: Background. To evaluate the associations between long-term oral Chinese herbal medicines (CHMs) and recurrence and metastasis (R & M) in patients with stage II and III colorectal cancer (CRC). Furthermore, we aimed to determine the correlation between different syndrome patterns and prognosis and summarized the regularities among CHMs prescriptions, providing reference for clinical practice. Methods. An ambispective cohort study was conducted. All CRC patients who sought evaluation and treatment at Xiyuan Hospital and Beijing Cancer Hospital from August 2014 to August 2016 were included. In this study, “whether patients voluntarily take CHMs” was taken as the exposure factor, and the exposure degree was “the duration of CHM use.” Stratification was performed according to the duration of TCM use to determine the relationship with R & M of CRC. The primary outcome was disease-free survival. Patients who had R & M of CRC after taking CHMs for ≥6 months were defined as “worst patients.” R software was used for statistical analysis. The Kaplan–Meier method and Cox regression analysis were used to determine the prognosis. IBM SPSS was used to model a priori association rules; drug use rules were analyzed on this basis. Results. A total of 186 patients with stage II and III CRC after radical resection were enrolled. All patients reached the study endpoint by August 2021. The difference in disease-free survival between the two groups was most significant when the cutoff value for CHMs was 18 months ( P = 0.0012 ). Multivariate analysis showed that 18 CHMs were independent protective factors for R & M of CRC ( P = 0.001 , HR = 0.20, 95% CI = 0.08–0.53). The ratio of Pi (spleen) and Shen (kidney) deficiency in the worst cases was higher than patients without R & M ( P = 0.018 ). Sijunzi and Liuwei Dihuang decoctions were the most frequently used prescriptions in the anti-R & M phase. Conclusion. CHMs complying with the “Jianpi Bushen” principle may attenuate the risk of R & M in patients with stage II and III CRC. Pi (spleen) and Shen (kidney) deficiency in patients receiving TCM intervention for the first time within 6 months of radical resection may be associated with a higher CRC R & M rate. Further research is warranted to validate these findings and elucidate underlying biological mechanisms.
