Abstract: Acute renal ischemia/reperfusion (I/R) injury often occurs during kidney transplantation and other kidney surgeries, and the molecular mechanism involves oxidative stress. We hypothesized that ginsenoside Rg1 (Rg1), a saponin derived from ginseng, would protect the renal tissue against acute renal I/R injury by upregulating 5 ′ adenosine monophosphate-activated protein kinase α1 (AMPKα1) expression and inhibiting oxidative stress. The models of acute anoxia/reoxygenation (A/R) damage in normal rat kidney epithelial cell lines (NRK-52E) and acute renal I/R injury in mice were constructed. The results revealed that pretreatment with 25 μM Rg1 significantly increased NRK-52E viability, decreased lactate dehydrogenase (LDH) activity and apoptosis, suppressed reactive oxygen species generation and oxidative stress, stabilized mitochondrial membrane potential and reduced mitochondria permeability transition pore openness, decreased adenosine monophosphate/adenosine triphosphate ratio, and upregulated the expression of AMPKα1, cytochrome b-c1 complex subunit 2, NADH dehydrogenase (ubiquinone) 1 beta subcomplex subunit 8, and B-cell lymphoma 2, while downregulating BCL2-associated X protein expression. The effects of Rg1 pretreatment were similar to those of pAD/Flag-AMPKα1. After acute renal I/R injury, serum creatinine, blood urea nitrogen, LDH activity, and oxidative stress in renal tissue significantly increased. Rg1 pretreatment upregulated AMPKα1 expression, which protects against acute renal I/R injury by maintaining renal function homeostasis, inhibiting oxidative stress, and reducing apoptosis. Compound C, a specific inhibitor of AMPK, reversed the effects of Rg1. In summary, Rg1 pretreatment upregulated AMPKα1 expression, inhibited oxidative stress, maintained mitochondrial function, improved energy metabolism, reduced apoptosis, and ultimately protected renal tissue against acute renal I/R injury.

Abstract: Recent years have witnessed the success of encoder-decoder structure-based approaches in lung region segmentation of chest X-ray (CXR) images. However, accurate lung region segmentation is still challenging due to the following three issues: (1) inaccurate lung region segmentation boundaries, (2) existence of lesion-related artifacts (e.g., opacity and pneumonia), and (3) lack of the ability to utilize multiscale information. To address these issues, we propose an edge-assisted computing and mask attention based network (called EAM-Net), which consists of an encoder-decoder network, an edge-assisted computing module, and multiple mask attention modules. Based on the encoder-decoder structure, an edge-assisted computing module is first proposed, which integrates the feature maps of the shallow encoding layers for edge prediction, and uses the edge evidence map as a strong cue to guide the lung region segmentation, thereby refining the lung region segmentation boundaries. We further design a mask attention module after each decoding layer, which employs a mask attention operation to make the model focus on lung regions while suppressing the lesion-related artifacts. Besides, a multiscale aggregation loss is proposed to optimize EAM-Net. Extensive experiments on the JSRT, Shenzhen, and Montgomery datasets demonstrate that EAM-Net outperforms existing state-of-the-art lung region segmentation methods.

Abstract: The development of metallic fluorescent materials, π -conjugated molecular systems with high-efficiency generation of blue light, and new ways to fabricate metallic/organic luminescent materials plays an important role in the fields of large-scale flat panel displays and soft optical devices. Herein, different fluorescent films have been fabricated by low vacuum physical vapor deposition method (LVPVDM), including single/two-component films. Compared with raw materials, all films show novel fluorescent behaviors, which means potential application in the fields of multicolor luminescence and thickness-optical response sensors. Meanwhile, the speculation is demonstrated in many ways that the maximum emission ( λ e m m a x ) at 406 nm and 426 nm of pristine 2,6-naphthalenedicarboxylic acid (26NCA) is caused by the crystal structure, while the shoulder peak at 445 nm is caused by the self-structure of 26NCA molecule. Significantly, this speculation may afford new insight into the relationship between not only crystal structure and luminescence, but also molecular self-structure and luminescence, which means a new strategy to tune the fluorescent behaviors based on molecular self-structure by LVPVDM. Therefore, this work provides a facile way to fabricate single/multicomponent metallic/organic film materials with tunable blue luminescence properties, which have potential application in the fields of next generation of photofunctional materials.

Abstract: Objective. By comparing the differences in microcirculatory responses of the heart and lung meridians induced by moxibustion on these two meridians, respectively, this study aimed to investigate the specificity for site-to-site association on body surface between different meridians. Methods. Eighty healthy adults were enrolled and divided into the lung meridian intervention group and heart meridian intervention group in a ratio of 1 : 1. Three-channel laser Doppler flowmetry was used to monitor microcirculatory responses for the heart and lung meridians. Primary outcome was change of blood perfusion units (PU) of three measurement sites along the two meridians. Results. In the lung meridian intervention group, following moxibustion performed at LU5 of the lung meridian, PU in the distal site of the lung meridian increased significantly. By contrast, the PU of HT3 in the heart meridian, which was nearest to the moxibustion site, did not change significantly. PU in the distal site of the heart meridian declined. Meanwhile, significant difference in PU change was detected between the distal site of the lung meridian and the other two control sites of the heart meridians during moxibustion and postmoxibustion. Alternatively, similar to the results of the lung meridian intervention group, the specificity of microcirculatory response between the heart and lung meridians was observed in the heart meridian intervention group. Conclusions. For the heart and lung meridians, the effect of moxibustion-induced microcirculatory response may be more related to meridian routes than the specific distance between two sites located at different meridians, thereby supporting possible specificity for site-to-site association on the body surface between these two meridians. Nevertheless, given that only two meridians and limited measurement sites were compared, all current findings are not sufficiently robust. Further research should be conducted to investigate more meridians and measurement sites.

Abstract: The lack of effective treatment for liver cirrhosis and hepatocellular carcinomas imposes serious challenges to the healthcare system. Here, we investigated the efficacy and mechanism of liquiritigenin involved in preventing or retarding the progression of liver diseases in a rat model with chronic carbon tetrachloride (CCl4) exposure. Sprague Dawley rats were given CCl4 and lliquiritigenin alone or simultaneously for 8 weeks before liver was harvested to check histological changes by Hematoxylin and Eosin (H & E) staining, apoptosis by TUNEL assay, ROS by dihydroethidium staining, antioxidant enzyme activities and malondialdehyde using specific kits, and gene expression by quantitative real-time PCR and western blot. Chronic CCl4 exposure caused profound changes in liver histology with extensive hepatocyte death (necrosis and apoptosis), fat accumulation, and infiltration of inflammatory cells, accompanied by depressed activities of antioxidant enzymes, increased oxidative stress, elevated expression of inflammation and fibrotic genes, and downregulation of PGC-1 α , ND1, and Bcl-x in rat liver. All these changes were abolished or alleviated by lliquiritigenin. The results demonstrated that liquiritigenin is effective in protecting liver from injury or treating chronic liver diseases. The modulation of PGC-1 α and its downstream genes might play a critical role in relieving CCl4-induced hepatic pathogenesis by liquiritigenin.

Abstract: Astilbin, a flavonoid compound, was isolated from the rhizome of Smilax glabra Roxb. (with red cross-section) grown in Guizhou Province, China. We accessed its effect and potential mechanism on attenuation of the inflammatory response in CFA-induced AA rats. Our results showed that daily oral administration of astilbin at 5.3 mg/kg reduced joint damage in the hind paw of AA rats. Accordingly, astilbin exhibited remarkable inhibitory effects on TNF- α , IL-1 β , and IL-6 mRNA expression. Significant decrease of serum cytokine levels of TNF- α , IL-1 β , and IL-6 was also observed in astilbin-treated AA rats compared to the vehicle-treated AA rats. The reduced expression of these cytokines was associated with protein activity suppression of three key molecular targets in the pathogenesis of RA, including IKK β , NF- κ B p65 subunit, and TLR adaptor MyD88. Furthermore, the therapeutic effects of astilbin on the inhibition of cytokines production as well as the reduction of inflammatory response in AA rats are close to a commonly used antirheumatic drug, leflunomide. Collectively, our data suggest that the action mechanism of astilbin, as an anti-inflammatory agent for RA treatment, is associated with modulating the production of proinflammatory cytokines and inhibiting the expression of key elements in NF- κ B signaling pathway mediated by TLR.

Abstract: Based on the mathematical model of the mass moment aerospace vehicles (MMAV), a coupled nonlinear dynamical system is established by rational simplification. The flight control system of MMAV is designed via utilizing nonlinear predictive control (NPC) approach. Aiming at the parameters of NPC is generally used the trial-and-error method to optimize and design, a novel kind of NPC parameters optimization strategy based on ant colony genetic algorithm (ACGA) is proposed in this paper. The method for setting NPC parameters with ACA in which the routes of ants are optimized by the genetic algorithm (GA) is derived. And then, a detailed realized process of this method is also presented. Furthermore, this optimization algorithm of the NPC parameters is applied to the flight control system of MMAV. The simulation results show that the system not only meets the demands of time-response specifications but also has excellent robustness.

Abstract: Reactive oxygen species (ROS) plays an essential role in the development of cancer. Here, we chose ROS-related miRNAs for consensus clustering analysis and ROS score construction. We find that ROS is extremely associated with prognosis, tumor immune microenvironment (TIME), gene mutations, N6-methyladenosine (m6A) methylation, and chemotherapy sensitivity in hepatocellular carcinoma (HCC). Mechanistically, ROS may affect the prognosis of HCC patients in numerous ways. Moreover, miR-210-3p and miR-106a-5p significantly increased the ROS level and stagnated cell cycle at G2/M in HCC; the results were more obvious in cells after ionizing radiation (IR). Finally, the two miRNAs suppressed cell proliferation, migration, and invasion and promoted apoptosis in huh7 and smmc7721 cells. It indicated that ROS might affect the prognosis of HCC patients through immune response and increase the sensitivity of HCC patients to radiotherapy and chemotherapy.

Abstract: Subarachnoid hemorrhage (SAH) refers to vascular brain injury mainly from a ruptured aneurysm, which has a high lifetime risk and imposes a substantial burden on patients, families, and society. Previous studies on SAH mainly focused on neurons in gray matter (GM). However, according to literature reports in recent years, in-depth research on the mechanism of white matter (WM) is of great significance to injury and recovery after SAH. In terms of functional recovery after SAH, all kinds of cells in the central nervous system (CNS) should be protected. In other words, it is necessary to protect not only GM but also WM, not only neurons but also glial cells and axons, and not only for the lesion itself but also for the prevention and treatment of remote damage. Clarifying the mechanism of white matter injury (WMI) and repair after SAH is of great importance. Therefore, this present review systematically summarizes the current research on WMI after SAH, which might provide therapeutic targets for treatment after SAH.

Abstract: Objectives . To evaluate CNN models’ performance of identifying the clinical images of basal cell carcinoma (BCC) and seborrheic keratosis (SK) and to compare their performance with that of dermatologists. Methods . We constructed a Chinese skin diseases dataset which includes 1456 BCC and 1843 SK clinical images and the corresponding medical history. We evaluated the performance using four mainstream CNN structures and transfer learning techniques. We explored the interpretability of the CNN model and compared its performance with that of 21 dermatologists. Results . The fine-tuned InceptionResNetV2 achieved the best performance, with an accuracy and area under the curve of 0.855 and 0.919, respectively. Further experimental results suggested that the CNN model was not only interpretable but also had a performance comparable to that of dermatologists. Conclusions . This study is the first on the assistant diagnosis of BCC and SK based on the proposed dataset. The promising results suggested that CNN model’s performance was comparable to that of expert dermatologists.
