    Abstract: Objective . Type 2 deiodinase (Dio2) is an enzyme responsible for the conversion of T4 to T3. The Thr92Ala polymorphism has been shown related to an increased risk for developing type 2 diabetes mellitus (T2DM). The aim of this study is to assess the association between this polymorphism and glycemic control in T2DM patients as marked by the HbA1C levels. Design and Methods. The terms “rs225014,” “thr92ala,” “T92A,” or “dio2 a/g” were used to search for eligible studies in the PubMed, Embase, and Cochrane databases and Google Scholar. A systematic review and meta-analysis of studies including both polymorphism testing and glycated hemoglobin (HbA1C) assays were performed. Results . Four studies were selected, totaling 2190 subjects. The pooled mean difference of the studies was 0.48% (95% CI, 0.18–0.77%), indicating that type 2 diabetics homozygous for the Dio2 Thr92Ala polymorphism had higher HbA1C levels. Conclusions . Homozygosity for the Dio2 Thr92Ala polymorphism is associated with higher HbA1C levels in T2DM patients. To confirm this conclusion, more studies of larger populations are needed.

    Abstract: This letter reports a piezoelectric vibration energy harvester which energy conversion efficiency is significantly improved by arraying piezoelectric sheets on cantilever beams, and the operation frequency band is widened by applying two-segment cantilever beams. A prototype is developed and tested. In this case, two group piezoelectric arrays are combined on the cantilever beams with the optimum load resistance. The total output power remains above 6.54 mW within the operation frequency band ranges from 27.5 Hz to 37.5 Hz when the generator is under an acceleration of 0.7 g and reaches two power peaks: 20.5 mW at 29.2 Hz and 12.95 mW at 35.4 Hz.

    Abstract: Black-box fuzzing is a testing technique to find both known and unknown vulnerabilities in software. When applying black-box fuzzing to smart devices, the main idea is to take a smart device as a black box and provide random input through a network-based interface, such as a Web interface. Due to the diversity of Web interface implementations and complex data format, a blind mutation of the message makes the message unable to pass the verification of the device component. Therefore, each Web interface needs a unique fuzzer, which precisely defines a message format of the target interface, a state maintenance method, the field positions to be mutated, and a specific input mutation method. At the time of writing, a fuzzer is completely developed by a security engineer. To save human labor, we present PDFuzzerGen, a tool to automatically synthesize complex black-box fuzzers for smart devices. PDFuzzerGen generates multiple fuzzing policies by analyzing raw messages and then synthesizes fuzzers based on policies. PDFuzzerGen requires no human intervention and can be applied to a wide range of smart devices. Furthermore, the generated fuzzers can expose bugs and flaws that rest deep in smart devices. PDFuzzerGen was evaluated to generate fuzzers for 19 different smart devices from 6 vendors. It has found 14 previously unknown vulnerabilities, 5 of which were confirmed and disclosed by the China National Vulnerability Database (CNVD) and 2 of which were confirmed and disclosed by Common Vulnerabilities and Exposures (CVE). The generated fuzzers outperform some manually crafted fuzzers on a few metrics, including the vulnerability detection rate and time cost of a newly developed fuzzer, which demonstrates the effectiveness and efficiency of PDFuzzerGen.

    Abstract: Arginase is a ubiquitous enzyme in the urea cycle (UC) that hydrolyzes L-arginine to urea and L-ornithine. Two mammalian arginase isoforms, arginase1 (ARG1) and arginase2 (ARG2), play a vital role in the regulation of β-cell functions, insulin resistance (IR), and vascular complications via modulating L-arginine metabolism, nitric oxide (NO) production, and inflammatory responses as well as oxidative stress. Basic and clinical studies reveal that abnormal alterations of arginase expression and activity are strongly associated with the onset and development of diabetes mellitus (DM) and its complications. As a result, targeting arginase may be a novel and promising approach for DM treatment. An increasing number of arginase inhibitors, including chemical and natural inhibitors, have been developed and shown to protect against the development of DM and its complications. In this review, we discuss the fundamental features of arginase. Next, the regulatory roles and underlying mechanisms of arginase in the pathogenesis and progression of DM and its complications are explored. Furthermore, we review the development and discuss the challenges of arginase inhibitors in treating DM and its related pathologies.

    Abstract: TrialDesign. Patients undergoing laminoplasty and laminectomy often experience severe postoperative pain. Local infiltration analgesia during spine surgery significantly reduces postoperative pain, which only upholds for a short time. Whether methylprednisolone and local anaesthetics are better than local anaesthetics alone in postoperative analgesia is yet to be determined. The primary aim of this research was the postoperative evaluation of efficacy and safety of methylprednisolone when used as an adjunct to local anaesthesia, ropivacaine, before wound closure after surgical procedures, laminoplasty or laminectomy. Methods. 132 patients were divided with a ratio of 1 : 1 into methylprednisolone-ropivacaine and ropivacaine alone groups. Every 30 ml of local infiltration solution consisted of 15 ml of 1% ropivacaine with 14 ml of saline along with 1 ml of 40 mg methylprednisolone and 15 ml of 1% ropivacaine with 15 ml of saline in methylprednisolone-ropivacaine group and ropivacaine group, respectively. The standardization of the study solution depended on the number of levels involved in surgery. Primary outcome was the 48-hour cumulative sufentanil demand. Results. Demographic characters and surgical variables among the groups were identical. The average 48-hour cumulative sufentanil demand was 32.5 ± 20.6 μg in the methylprednisolone-ropivacaine group and 50.9 ± 27.2 μg in the ropivacaine group ( p 〈 0.001 ). The estimated median time of demand of the first analgesia via patient-controlled analgesia (PCA) pump was 2.5 hours and 2 hours in the methylprednisolone-ropivacaine group and the ropivacaine group, respectively (hazard ratio (HR) was 0.53, with 95% Cl 0.33 to 0.87 and Log-rank of p = 0.0019 ). Conclusion. The infiltration of methylprednisolone as adjunct ropivacaine before wound closure is a safe and efficient strategy for pain management following laminoplasty or laminectomy.

    Abstract: Background . C3d is a product of both the classic and the alternative complement cascades; however, few studies have addressed the role of C3d in renal biopsies and its relationship with long-term graft survival rate is not very clear. Methods . 94 patients with biopsy-proven acute rejection episodes were included in the study. We investigated the associations between histological findings, clinical examinations, and outcome. Results . The overall prevalence for C4dPTC and C3dPTC was 42.6% and 29.8%. There was a significant association between C3dPTC and C4dPTC ( P 〈 0.001 ). C3dPTC and C4dPTC were related with histological types ( P = 0.024 and P 〈 0.001 , resp.). The long-term survival rate for C4dPTC positive transplants was lower than that of C4dPTC negative transplants, but it was not statistic significant in our study ( P = 0.150 ). The survival rate of C3dPTC positive group was much lower than the negative group ( P = 0.014 ). Patients with double positives for C4dPTC and C3dPTC exhibited the lowest survival rate significantly different from those of the C3dPTC only and C4dPTC only groups ( P = 0.01 and P = 0.0037 ). Conclusions . This longitudinal cohort study has demonstrated that C3d deposition in the PTC was closely related to renal dysfunction and pathological changes.

    Abstract: With the development of the economy, people’s living standards continue to improve, and the management awareness of their own health and safety has gradually increased. The human body usually has a long process of transition from being healthy to the appearance of disease, and people’s lifestyle and daily behavior will affect the health of the body to a large extent. This paper studies the national sports AI health management service system based on edge computing. This paper organically combines edge computing, artificial intelligence, and health management service systems. Through detection devices with smart sensors and smart health systems, a comprehensive health management service platform for the masses is constructed to promote the development of sports health management services. Upgrade and transform to make it more in line with the modern intelligent environment. The advancement of modern technology has gradually transformed my country’s traditional health record management service to an intelligent electronic health record management system. From the experimental data, it can be seen that the filing rate and pass rate of residents’ electronic files have reached 88.275% and 87.435%, but the utilization rate of health files is only 31.685%. The establishment of a supporting health management service system is precisely to effectively increase the utilization rate of health files, so that users can truly enjoy intelligent, professional, and real-time health services.

    Abstract: Human pluripotent stem cell (hPSC-) derived cardiomyocytes have potential applications in drug discovery, toxicity testing, developmental studies, and regenerative medicine. Before these cells can be reliably utilized, characterization of their functionality is required to establish their similarity to native cardiomyocytes. We tracked fluorescent beads embedded in 4.4–99.7 kPa polyacrylamide hydrogels beneath contracting neonatal rat cardiomyocytes and cardiomyocytes generated from hPSCs via growth-factor-induced directed differentiation to measure contractile output in response to changes in substrate mechanics. Contraction stress was determined using traction force microscopy, and morphology was characterized by immunocytochemistry for α-actinin and subsequent image analysis. We found that contraction stress of all types of cardiomyocytes increased with substrate stiffness. This effect was not linked to beating rate or morphology. We demonstrated that hPSC-derived cardiomyocyte contractility responded appropriately to isoprenaline and remained stable in culture over a period of 2 months. This study demonstrates that hPSC-derived cardiomyocytes have appropriate functional responses to substrate stiffness and to a pharmaceutical agent, which motivates their use in further applications such as drug evaluation and cardiac therapies.

    Abstract: The compressor geometric variable system is vital for aeroengines, as it affects their performance and design. To monitor the compressor geometric variable system states and detect anomalies in real time, a t -step forecasting method based on the MAE (masked autoencoders) model was proposed in this article. Unlike previous studies that used simulated or lab-generated data, we use actual flight data recorded by the aircraft data acquisition system to make our results more realistic. Through our experimental efforts, the feasibility of forecasting the compressor geometric variable system based on the MAE model is verified. That is not only the first application of transformer models with a masked pretraining mechanism in time series forecasts but also taking the lead in exploring the possibility of this key system forecast. We also test the generalizability of our method across different types of aeroengines. Finally, to make our theories more reasonable and convincing, experiments on different aeroengine states, including the transition state and the steady state, are carried out.

    Abstract: The decision-making models that are able to deal with complex and dynamic urban intersections are critical for the development of autonomous vehicles. A key challenge in operating autonomous vehicles robustly is to accurately detect the trajectories of other participants and to consider safety and efficiency concurrently into interactions between vehicles. In this work, we propose an approach for developing a tactical decision-making model for vehicles which is capable of predicting the trajectories of incoming vehicles and employs the conflict resolution theory to model vehicle interactions. The proposed algorithm can help autonomous vehicles cross intersections safely. Firstly, Gaussian process regression models were trained with the data collected at intersections using subgrade sensors and a retrofit autonomous vehicle to predict the trajectories of incoming vehicles. Then, we proposed a multiobjective optimization problem (MOP) decision-making model based on efficient conflict resolution theory at intersections. After that, a nondominated sorting genetic algorithm (NSGA-II) and deep deterministic policy gradient (DDPG) are employed to select the optimal motions in comparison with each other. Finally, a simulation and verification platform was built based on Matlab/Simulink and PreScan. The reliability and effectiveness of the tactical decision-making model was verified by simulations. The results indicate that DDPG is more reliable and effective than NSGA-II to solve the MOP model, which provides a theoretical basis for the in-depth study of decision-making in a complex and uncertain intersection environment.
