    Abstract: Glucuronidation is a major metabolism process of detoxification for carcinogens, 4-(methylnitrosamino)-1-(3-pyridy)-1-butanone (NNK) and 1,2-dimethylhydrazine (DMH), of reactive oxygen species (ROS). However, intestinal E. coli    β -glucuronidase (e β G) has been considered pivotal to colorectal carcinogenesis. Specific inhibition of e β G may prevent reactivating the glucuronide-carcinogen and protect the intestine from ROS-mediated carcinogenesis. In order to develop specific e β G inhibitors, we found that 59 candidate compounds obtained from the initial virtual screening had high inhibition specificity against e β G but not human β G. In particular, we found that compounds 7145 and 4041 with naphthalenylidene-benzenesulfonamide (NYBS) are highly effective and selective to inhibit e β G activity. Compound 4041   ( I C 50 = 2.8  μ M) shows a higher inhibiting ability than compound 7145   ( I C 50 = 31.6  μ M) against e β G. Furthermore, the molecular docking analysis indicates that compound 4041 has two hydrophobic contacts to residues L361 and I363 in the bacterial loop, but 7145 has one contact to L361. Only compound 4041 can bind to key residue (E413) at active site of e β G via hydrogen-bonding interactions. These novel NYBS-based e β G specific inhibitors may provide as novel candidate compounds, which specifically inhibit e β G to reduce e β G-based carcinogenesis and intestinal injury.

    Abstract: Wharton’s jelly mesenchymal stem cells (WJMSCs) transfer healthy mitochondria to cells harboring a mitochondrial DNA (mtDNA) defect. Mitochondrial myopathy, encephalomyopathy, lactic acidosis, and stroke-like episodes (MELAS) is one of the major subgroups of mitochondrial diseases, caused by the mt.3243A 〉 G point mutation in the mitochondrial tRNALeu (UUR) gene. The specific aim of the study is to investigate whether WJMSCs exert therapeutic effect for mitochondrial dysfunction in cells of MELAS patient through donating healthy mitochondria. We herein demonstrate that WJMSCs transfer healthy mitochondria into rotenone-stressed fibroblasts of a MELAS patient, thereby eliminating mutation burden and rescuing mitochondrial functions. In the coculture system in vitro study, WJMSCs transferred healthy mitochondria to rotenone-stressed MELAS fibroblasts. By inhibiting actin polymerization to block tunneling nanotubes (TNTs), the WJMSC-conducted mitochondrial transfer was abrogated. After mitochondrial transfer, the mt.3243A 〉 G mutation burden of MELAS fibroblasts was reduced to an undetectable level, with long-term retention. Sequencing results confirmed that the transferred mitochondria were donated from WJMSCs. Furthermore, mitochondrial transfer of WJMSCs to MELAS fibroblasts improves mitochondrial functions and cellular performance, including protein translation of respiratory complexes, ROS overexpression, mitochondrial membrane potential, mitochondrial morphology and bioenergetics, cell proliferation, mitochondrion-dependent viability, and apoptotic resistance. This study demonstrates that WJMSCs exert bioenergetic therapeutic effects through mitochondrial transfer. This finding paves the way for the development of innovative treatments for MELAS and other mitochondrial diseases.

    Abstract: Breast cancer stem cells (CSCs) are highly tumorigenic and possess the capacity to self-renew. Recent studies indicated that pluripotent gene NANOG involves in regulating self-renewal of breast CSCs, and expression of NANOG is correlated with aggressiveness of poorly differentiated breast cancer. We initially confirmed that breast cancer MCF-7 cells expressed NANOG, and overexpression of NANOG enhanced the tumorigenicity of MCF-7 cells and promoted the self-renewal expansion of CD2 4 − / low CD44 + CSC subpopulation. In contrast, knockdown of NANOG significantly affected the growth of breast CSCs. Utilizing flow cytometry, we identified five cyclohexylmethyl flavonoids that can inhibit propagation of NANOG-positive cells in both breast cancer MCF-7 and MDA-MB231 cells. Among these flavonoids, ugonins J and K were found to be able to induce apoptosis in non-CSC populations and to reduce self-renewal growth of CD2 4 − / low CD44 + CSC population. Treatment with ugonin J significantly reduced the tumorigenicity of MCF-7 cells and efficiently suppressed formation of mammospheres. This suppression was possibly due to p53 activation and NANOG reduction as either addition of p53 inhibitor or overexpression of NANOG can counteract the suppressive effect of ugonin J. We therefore conclude that cyclohexylmethyl flavonoids can possibly be utilized to suppress the propagation of breast CSCs via reduction of NANOG.

    Abstract: Purpose. To study the therapeutic effect of Quyu (QY) Shengxin (SX) decoction (QYSXD) in mice with dextran sulfate sodium- (DSS-) induced ulcerative colitis and to investigate the effects of QYSXD on the regulation of the receptor-interacting protein kinase 1 (RIP1)/receptor-interacting protein kinase 3 (RIP3)/nucleotide-binding oligomerization domain-like receptor family pyrin domain protein 3 (NLRP3) signaling pathway. Method. Thirty-six mice were randomly divided into the following 6 groups: the experimental group (QYSX group), the model group (DSS group), the positive control group (5-aminosalicylic acid (5-ASA) group), the control group, the first component group (QY group), and the second component group (SX group). Each group included 6 mice. Ulcerative colitis (UC) was induced in the mice by providing 3.5% DSS in drinking water. The mice were weighed every day to evaluate the disease activity index (DAI). After 7 days, the mice were sacrificed, and colonic tissues were obtained for colon length measurement. The morphological changes in the colon and the pathological scores of the mice in each group were observed by hematoxylin-eosin (HE) staining. The messenger ribonucleic acid (mRNA) and protein expression levels of RIP1, RIP3, dynamin-related protein 1 (Drp1), NLRP3, cysteinyl aspartate specific proteinase-1 (caspase-1), interleukin (IL)-1β, and IL-18 in the colon tissues of the mice in each group were detected and compared by real-time quantitative reverse transcription PCR (RT-qPCR) and enzyme-linked immunosorbent assay (ELISA). The levels of RIP1, RIP3, NLRP3, IL-1β, and IL-8 in the colonic mucosa were detected by ELISA. Western blotting was used to compare the protein expression of Drp1, caspase-1, mitochondrial fission protein 1 (FIS1), and mitophagy-associated protein light chain 3a/b (LC3a/b) among groups. The levels of reactive oxygen species (ROS) in the colonic mucosal cells were compared by immunofluorescence. Results. Compared with those in the DSS group, the mice with DSS-induced colitis in the QYSX group exhibited clearly higher body weights ( P 〈 0.05 ) and DAI scores ( P 〈 0.05 ). The colon lengths of the mice in the QYSX group were longer than those in the DSS group ( P 〈 0.05 ), and the pathological score of the QYSX group was lower than that of the DSS group ( P 〈 0.05 ). The RIP1, RIP3, Drp1, IL-1β, IL-18, and caspase-1 mRNA levels in the QYSX, 5-ASA, SX, and QY groups were significantly lower than those in the DSS group ( P 〈 0.05 ), but there were no differences between the QYSX group and the 5-ASA group. The levels of RIP1, RIP3, NLRP3, IL-1β, and IL-18 in the QYSX group were lower than those in the DSS group ( P 〈 0.01 ). The levels of Drp1, caspase-1, FIS1, and LC3a/b in the QYSX group and the 5-ASA group were lower than those in the DSS group ( P 〈 0.05 ). The levels of ROS in the colonic mucosal cells in the QYSX, 5-ASA, and QY groups were lower than those in the DSS group ( P 〈 0.05 ). Conclusion. QYSXD has certain therapeutic effects on DSS-induced colitis in mice and may be as effective as 5-ASA. QY and SX decoctions also have certain effects on colitis; however, these decoctions are not as beneficial as QYSXD. QYSXD may ameliorate colitis by inhibiting the expression of RIP1/RIP3/NLRP3 pathway-related proteins and reversing mitochondrial dysfunction to control inflammation.

    Abstract: Since the antennas on UAVs may have slight vibrations or the interference source is in the state of rapid movement in practice, the interference suppression performance and robustness of the traditional methods may suffer a decline. In this paper, we propose a flexible asymmetric null widening technique, which allows flexible adjustment of the null width to accommodate the variation of the interference source. This method has a good effect of spreading zero trap on the two-dimensional array and can effectively reduce the waste of degrees of freedom. Firstly, the flexible asymmetric null widening method is extended to two-dimensional arrays to accommodate 2D array antennas of UAVs. Secondly, when the SMI algorithm is applied in adaptive beamforming, the desired signal appears in sampling snapshots or using data samples, resulting in a model mismatch. To solve the model mismatch problem of UAV antenna arrays, this paper applies a sparsity-based interference plus noise covariance matrix reconstruction technique. Finally, for the application scenario that the UAV may receive signals from multiple directions, we apply the linear constrained minimum variance criterion (LCMV) to achieve the main beam gain formation in multiple directions. The simulation results show that we can generate a wide null and adjust the null width asymmetrically. The results also show that the model mismatch problem is avoided, and the performance of the adaptive beamforming is almost optimal. For the UAV antenna, we also implemented multiple beams to receive multiple signals .

    Abstract: This study proposes to apply the combination of two 90-degree prisms and a holographic optical element to an image-dividing element, divide an image into three through the virtual image projection system, and accurately project the images to 160 cm behind the windshield. In order to distinguish the left image, middle image, and right image at the horizontal direction, the 90-degree prism is first utilized for horizontally deflecting the light, a holographic optical element is then used for presenting the vertical deflection of the image, and finally, the images are horizontally arranged as a real image on the diffuser. An image-dividing element is eventually integrated to the virtual image projection system. Then, the divided images are made as a virtual image which is projected to 160 cm behind the windshield to combine with the street scene. The horizontal angle of view of an eyebox is increased from 2.51° to 7.22°, and the size of the eyebox is 31.68 cm × 12.48 cm, allowing a wide-view laser-based head-up display with the large-angle beam-splitting function being successfully designed. It would not affect the field of view on the road, and this system could reduce the space and be arranged easily in a vehicle. Therefore, this proposed system is suitable for the aftermarket.

    Abstract: Cisplatin chemotherapy causes myelosuppression and often limits treatment duration and dose escalation in patients. Novel approaches to circumvent or lessen myelotoxicity may improve clinical outcome and quality of life in these patients. Chlorella sorokiniana (CS) is a freshwater unicellular green alga and exhibits encouraging efficacy in immunomodulation and anticancer in preclinical studies . However, the efficacy of CS on chemoprotection remains unclear. We report here, for the first time, that CS extract (CSE) could protect normal myeloid cells and PBMCs from cisplatin toxicity. Also, cisplatin-induced apoptosis in HL-60 cells was rescued through reservation of mitochondrial function, inhibition of cytochrome c release to cytosol, and suppression of caspase and PARP activation. Intriguingly, cotreatment of CSE attenuated cisplatin-evoked hypocellularity of bone marrow in mice. Furthermore, we observed the enhancement of CSF-GM activity in bone marrow and spleen in mice administered CSE and cisplatin, along with increased CD11b levels in spleen. In conclusion, we uncovered a novel mechanism of CSE on myeloprotection, whereby potentially supports the use of CSE as a chemoprotector against cisplatin-induced bone marrow toxicity. Further clinical investigation of CSE in combination with cisplatin is warranted.

    Abstract: Whether BDNF protein and BDNF mRNA expression of the medial prefrontal cortex (mPFC; cingulated cortex area 1 (Cg1), prelimbic cortex (PrL), and infralimbic cortex (IL)), amygdala, and hippocampus (CA1, CA2, CA3, and dentate gyrus (DG)) was involved in fear of posttraumatic stress disorder (PTSD) during the situational reminder of traumatic memory remains uncertain. Footshock rats experienced an inescapable footshock (3 mA, 10 s), and later we have measured fear behavior for 2 min in the footshock environment on the situational reminder phase. In the final retrieval of situational reminder, BDNF protein and mRNA levels were measured. The results showed that higher BDNF expression occurred in the Cg1, PrL, and amygdala. Lower BDNF expression occurred in the IL, CA1, CA2, CA3, and DG. BDNF mRNA levels were higher in the mPFC and amygdala but lower in the hippocampus. The neural connection analysis showed that BDNF protein and BDNF mRNA exhibited weak connections among the mPFC, amygdala, and hippocampus during situational reminders. The present data did not support the previous viewpoint in neuroimaging research that the mPFC and hippocampus revealed hypoactivity and the amygdala exhibited hyperactivity for PTSD symptoms. These findings should be discussed with the previous evidence and provide clinical implications for PTSD.

    Abstract: Although innovative technologies for somatic cell reprogramming and transdifferentiation provide new strategies for the research of translational medicine, including disease modeling, drug screening, artificial organ development, and cell therapy, recipient safety remains a concern due to the use of exogenous transcription factors during induction. To resolve this problem, new induction approaches containing clinically applicable small molecules have been explored. Small molecule epigenetic modulators such as DNA methylation writer inhibitors, histone methylation writer inhibitors, histone acylation reader inhibitors, and histone acetylation eraser inhibitors could overcome epigenetic barriers during cell fate conversion. In the past few years, significant progress has been made in reprogramming and transdifferentiation of somatic cells with small molecule approaches. In the present review, we systematically discuss recent achievements of pure chemical reprogramming and transdifferentiation.

    Abstract: The resistivity index is an important parameter for determining the rock saturation index. However, the saturation index changes greatly in unconventional reservoirs, which leads to oil saturation estimation with great difficulty. Hence, we try to establish the relationship between the resistivity index and log data. Firstly, a novel model of estimating the resistivity index with T 2 time was derived based on fractal theory, the relationship between nuclear magnetic resonance (NMR) T 2 spectrum and capillary pressure curve ( T 2 - P c ), and Archie formula. It regards the logarithm of the resistivity index as the dependent variable, with T 2 time and T 2 time when water saturation is 100% as the independent variables. Second, 17 cores were drilled, and T 2 spectrum and the relationship between the resistivity index and water saturation ( I r - S w ) were jointly measured. Next, the experimental results were substituted into the established model to get the model parameters via the multivariate statistics regression method. Then, the experimental data engaged and not engaged in modeling were used to test the established model. The average relative errors of estimated resistivity indices and experimental results are smaller than 8%, and those of the regressed saturation index are smaller than 5%. Finally, the established model was applied in log data processing and interpretation with good effects. It thus proves that the method of the estimating resistivity index with T 2 time is reliable, which provides a novel solution for determining rock electrical parameter of unconventional reservoirs.
