    Abstract: Pancreatic cancer is one of the most malignant gastrointestinal tumors, and it is of great significance to explore the molecular mechanism of its progression and find new biological therapeutic targets. CIRBP is a cold-induced protein that plays a key role in many physiological and pathological processes, but its role in pancreatic cancer is still unclear. The expression of CIRBP in pancreatic cancer tissues was slightly lower than that in normal tissues, and the high expression of CIRBP was beneficial to survival. At the same time, immunohistochemical detection showed that the expression level of CIRBP in the cytoplasm of cancer tissues was significantly lower than that of adjacent tissues; survival curve analysis showed that pancreatic cancer patients with high nuclear CIRBP expression had a longer overall survival period. RIP results showed that CIRBP antibody significantly enriched p53 RNA, indicating that it could directly bind to p53. Cold treatment of pancreatic cancer cells significantly induced the expression of CIRBP, DPP4, NOX1, and FTH1 and inhibited the expression of p53 and GPX4. Cold induction enhanced the accumulation of Fe2+ in cells, promoted the generation of ROS, and inhibited the expression of GSH-Px. Therefore, cold induction promotes the process of ferroptosis by inducing the expression of CIRBP and then regulating key factors such as p53 and GPX4. In addition, cold induction significantly inhibited the proliferation of pancreatic cancer cells and induced cell apoptosis, but after the addition of ferroptosis inhibitor, cell proliferation and apoptosis did not change significantly. Therefore, CIRBP acts as a tumor suppressor gene in pancreatic cancer and induces ferroptosis through the p53/GPX4 pathway to inhibit cell growth, which may be an important target for the diagnosis and treatment of pancreatic cancer.

    Abstract: Background . The transcriptional networks of Cyr61 and its function in cell injury are poorly understood. The present study depicted the lncRNA and mRNA profiles and the involvement in angiotensin II-induced injury after Cyr61 knockdown mediated by CRISPR/Cas9 in HEK293T cells. Methods . HEK293T cells were cultured, and Cyr61 knockdown was achieved by transfection of the CRISPR/Cas9 KO plasmid. lncRNA and mRNA microarrays were used to identify differentially expressed genes (DEGs). Gene ontology (GO) and the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analyses were performed to determine biofunctions and signaling pathways. RT-PCR was used to validate the microarray results. Cells were divided into four groups: control, Cyr61 knockdown, angiotensin II (Ang II) without Cyr61 knockdown, and Ang II with Cyr61 knockdown. CCK8, western blotting, and flow cytometry analysis were carried out to dissect cellular function. Results . A total of 23184 lncRNAs and 28264 mRNAs were normalized. 26 lncRNAs and 212 mRNAs were upregulated, and 74 lncRNAs and 233 mRNAs were downregulated after Cyr61 knockdown. Analysis of cellular components, molecular functions, biological processes, and regulatory pathways associated with the differentially expressed mRNAs revealed down stream mechanisms of the Cyr61 gene. The differentially expressed genes were affected for small cell lung cancer, axon guidance, Fc gamma R-mediated phagocytosis, MAPK signaling pathway, focal adhesion, insulin resistance, and metabolic pathways. In addition, Cyr61 expression was increased in accordance with induction of cell cycle arrest and apoptosis and inhibition of cell proliferation induced by Ang II. Knockdown of Cyr61 in HEK293T cells promoted cell cycle procession, decreased apoptosis, and promoted cell proliferation. Conclusions . The Cyr61 gene is involved in Ang II-induced injury in HEK293T cells. Functional mechanisms of the differentially expressed lncRNAs and mRNAs as well as identification of metabolic pathways will provide new therapeutic targets for Cyr61-realated diseases.

    Abstract: With the recent development of deep convolutional neural network (CNN), remote sensing for ship detection methods has achieved enormous progress. However, current methods focus on the whole ships and fail on the component’s detection of a ship. To detect ships from remote-sensing images in a more refined way, we employ the inherent relationship between ships and their critical parts to establish a multilevel structure and propose a novel framework to improve the performance in identifying the multilevel objects. Our framework, named the dual detector network (DD-Net), consists of two carefully designed detectors, one for ships (the ship detector) and the other for their critical parts (the critical part detector), for detecting the critical parts in a coarse-to-fine manner. The ship detector offers detection results of the ship, based on which the critical part detector detects small critical parts inside each ship region. The framework is trained in an end-to-end way by optimizing the multitask loss. Due to the lack of publicly available datasets for critical part detection, we build a new dataset named RS-Ship with 1015 remote-sensing images and 2856 annotations. Experiments on the HRSC2016 dataset and the RS-Ship dataset show that our method performs well in the detection of ships and critical parts.

    Abstract: Mesenchymal stem cells (MSCs) can affect the microenvironment of a wound and thereby accelerate wound healing. Wnt proteins act as key mediators of skin development and participate in the formation of skin appendages such as hair. The mechanisms of action of MSCs and Wnt proteins on skin wounds are largely unknown. Here, we prepared a Wnt7a-containing conditioned medium (Wnt-CM) from the supernatant of cultured human umbilical cord-MSCs (UC-MSCs) overexpressing Wnt7a in order to examine the effects of this CM on cutaneous healing. Our results revealed that Wnt-CM can accelerate wound closure and induce regeneration of hair follicles. Meanwhile, Wnt-CM enhanced expression of extracellular matrix (ECM) components and cell migration of fibroblasts but inhibited the migratory ability and expression of K6 and K16 in keratinocytes by enhancing expression of c-Myc. However, we found that the CM of fibroblasts treated with Wnt-CM ( H F Wnt-CM -CM) can also promote wound repair and keratinocyte migration; but there was no increase in the number of hair follicles of regeneration. These data indicate that Wnt7a and UC-MSCs have synergistic effects: they can accelerate wound repair and induce hair regeneration via cellular communication in the wound microenvironment. Thus, this study opens up new avenues of research on the mechanisms underlying wound repair.

    Abstract: Ship detection is one of the fundamental tasks in computer vision. In recent years, the methods based on convolutional neural networks have made great progress. However, improvement of ship detection in aerial images is limited by large-scale variation, aspect ratio, and dense distribution. In this paper, a Critical and Align Feature Constructing Network (CAFC-Net) which is an end-to-end single-stage rotation detector is proposed to improve ship detection accuracy. The framework is formed by three modules: a Biased Attention Module (BAM), a Feature Alignment Module (FAM), and a Distinctive Detection Module (DDM). Specifically, the BAM extracts biased critical features for classification and regression. With the extracted biased regression features, the FAM generates high-quality anchor boxes. Through a novel Alignment Convolution, convolutional features can be aligned according to anchor boxes. The DDM produces orientation-sensitive feature and reconstructs orientation-invariant features to alleviate inconsistency between classification and localization accuracy. Extensive experiments on two remote sensing datasets HRS2016 and self-built ship datasets show the state-of-the-art performance of our detector.

    Abstract: Objective. To evaluate the efficacy of traditional Chinese herbal ChuanXiong Ding Tong herbal formula granule (CXDT-HFG) for migraine patients with “the Syndrome of Liver Wind and Blood Stasis.” Methods. 150 migraine patients were recruited and assigned randomly in a double-blind, placebo-controlled study to receive CXDT-HFG ( n = 99 ) plus necessary analgesics, or placebo ( n = 51 ) plus necessary analgesics for 16 weeks (12 weeks’ intervention and 4 weeks’ follow up). Outcome measures included migraine days, frequency of migraine attacks, analgesics consumption for acute treatment, and the proportion of responders as well as the visual analogue scale (VAS) scores and intensity for pain. Results. Compared with the placebo group, the CXDT-HFG group showed significant reduction in migraine days and attacks frequency at week 12 and follow-up period ( P 〈 0.05 ) as well as in the reduction of VAS scores at follow-up period.There was significant difference in the proportion of responders between the two groups at follow-up period ( P = 0.014 ). However there were no significant differences between the two groups in analgesics consumption ( P 〉 0.05 ) . Conclusion. CXDT-HFG was more effective than placebo in decreasing days of migraine attacks, frequency, VAS scores, and relieving pain intensity for migraine patients.

    Abstract: This study was aimed at exploring common oncogenic genes and pathways both in osteosarcoma and Ewing’s sarcoma. Microarray data were obtained from the Gene Expression Omnibus (GEO) database. Differentially expressed genes (DEGs) were identified using the limma package. Then, protein-protein interaction (PPI) networks were constructed and hub genes were identified. Furthermore, functional enrichment analysis was analyzed. The expression of common oncogenic genes was validated in 38 osteosarcoma and 17 Ewing’s sarcoma tissues by RT-qPCR and western blot compared to normal tissues. 201 genes were differentially expressed. There were 121 nodes and 232 edges of the PPI network. Among 12 hub genes, hub genes FN1, COL1A1, and COL1A2 may involve in the development of osteosarcoma and Ewing’s sarcoma. And they were reduced to expression both in osteosarcoma and Ewing’s sarcoma tissues at mRNA and protein levels compared to normal tissues. Knockdown of FN1, COL1A1, and COL1A2 enhanced the cell proliferation and migration of U2OS under the restriction of cisplatin. Our findings revealed the common oncogenic genes such as FN1, COL1A1, and COL1A2, which may act as antioncogene by enhancing cisplatin sensitivity in osteosarcoma cells, and pathways were both in osteosarcoma and Ewing’s sarcoma.
