    Abstract: The aim of this study was to investigate whether uric acid (UA) might exert neuroprotection via activating the nuclear factor erythroid 2-related factor 2 (Nrf2) pathway and regulating neurotrophic factors in the cerebral cortices after transient focal cerebral ischemia/reperfusion (FCI/R) in rats. UA was intravenously injected through the tail vein (16 mg/kg) 30 min after the onset of reperfusion in rats subjected to middle cerebral artery occlusion for 2 h. Neurological deficit score was performed to analyze neurological function at 24 h after reperfusion. Terminal deoxynucleotidyl transferase-mediated dNTP nick end labeling (TUNEL) staining and hematoxylin and eosin (HE) staining were used to detect histological injury of the cerebral cortex. Malondialdehyde (MDA), the carbonyl groups, and 8-hydroxyl-2 ′ -deoxyguanosine (8-OHdG) levels were employed to evaluate oxidative stress. Nrf2 and its downstream antioxidant protein, heme oxygenase- (HO-) 1,were detected by western blot. Nrf2 DNA-binding activity was observed using an ELISA-based measurement. Expressions of BDNF and NGF were analyzed by immunohistochemistry. Our results showed that UA treatment significantly suppressed FCI/R-induced oxidative stress, accompanied by attenuating neuronal damage, which subsequently decreased the infarct volume and neurological deficit. Further, the treatment of UA activated Nrf2 signaling pathway and upregulated BDNF and NGF expression levels. Interestingly, the aforementioned effects of UA were markedly inhibited by administration of brusatol, an inhibitor of Nrf2. Taken together, the antioxidant and neuroprotective effects afforded by UA treatment involved the modulation of Nrf2-mediated oxidative stress and regulation of BDNF and NGF expression levels. Thus, UA treatment could be of interest to prevent FCI/R injury.

    Abstract: During the fruit ripening period, the orchard temperature difference between day and night has a substantial impact on the fruit quality, and it is not easy to be controlled like that of in greenhouse, this paper designed the smart orchard Internet of Things (IoT), through the fuzzy PID (Proportion-Integration-Differentiation) algorithm to control the water spraying to adjust the temperature difference Δ T between day and night in the orchard, intelligently influenced the orchard energy conversion process, and promoted the fruit sugar accumulation. Experiments show that the maximum Δ T can reach 20°C, the intelligent energy regulation based on the IoT has a good effect, and a scheme to intervene the microclimate in farmland (quite different from the greenhouse environment) is provided.

    Abstract: In order to explore the changes of intestinal flora and serum levels of relevant substances in patients with gastric cancer before and after surgery with carbon nanoparticle laparoscopy, a total of 180 patients with early distal gastric cancer who adopted laparoscopic radical gastrectomy for distal gastric cancer in the general surgery department of TCM Hospital of Shi Jia Zhuang City from January 2018 to January 2020 were selected and randomly divided into two groups: traditional laparoscopic operation (control group) and carbon nanoparticle laparoscopic operation (experimental group) were adopted for treatment for the two groups, respectively. Postoperative evaluation included the difference between the two groups in the operative time, the efficiency of intraoperative lymph node dissection, and the number of lymph node detection. The adverse reactions, changes of intestinal flora before and after surgery in the two groups, and the serum levels of epidermal growth factor receptor (EGFR), interleukin-32 (IL-32), and gastrin 17 were evaluated. In the experimental group, the success rate of carbon nanoparticle tracer black staining reached 100%, and the operation time of the experimental group was significantly shorter than that of the control group ( P 〈 0.05 ). The lymph node detection rate of the experimental group was higher than that of the control group ( P 〈 0.05 ), but there was no significant difference in the lymph node metastasis rate between the two groups ( P 〉 0.05 ). The sentinel lymph node sensitivity of the experimental group reached 92.3%, and the specificity, accuracy, and positive and negative prediction rates reached 100%; the experimental group patients were with an obviously higher incidence of level I-II gastrointestinal reaction ( P 〈 0.05 ). Postoperative increases in Bifidobacteria and Lactobacillus were observed in both groups, while decreases in Enterococcus and Escherichia coli were observed in both groups ( P 〈 0.05 ). Moreover, the degree of increase and decrease in the experimental group was greater than that in the control group ( P 〈 0.05 ). The serum levels of EGFR, IL-32, and gastrin 17 in the two groups were significantly lower than those in the control group on 3 d, 7 d, and 15 d after surgery ( P 〈 0.05 ). In the radical gastrectomy for distal gastric cancer, carbon nanoparticle laparoscopy was not only helpful for the localization of small tumors but also for the thorough dissection of lymph nodes after the surgery, and the postoperative adverse reactions of carbon nanoparticle laparoscopy were also less, which was of great significance for the improvement of intestinal flora and the reduction of serum levels of EGFR, IL-32, and gastrin 17 in gastric cancer patients.

    Abstract: Gliomas are the most common type of primary brain tumor, yet the prognosis for glioma patients remains poor. Mutations in the promoter region of the telomerase reverse transcriptase gene (TERTp) are associated with diagnosis and poor prognosis in gliomas. Here, we developed a precise and rapid Sanger sequencing assay to screen or TERTp mutations. We established the Sanger sequencing approach for the detection of TERTp mutations based on human glioma cell lines U251 and assessed the analytical validation by determining the accuracy, sensitivity, precision, and specificity. In our study, we verified the accuracy of Sanger sequencing by the real-time polymerase chain reaction method. Our data showed that TERTp mutations were detected at an analytical sensitivity of 10% per mutant. The precision and specificity validation also showed the desired results. In total, 147 glioma patients were investigated for TERTp mutations, and of each patient, clinical data and molecular characteristics were analyzed. We found that anaplastic oligodendroglioma had the highest frequency of TERTp mutations (66.7%). No differences in TERTp mutation frequency were observed between frozen tissue specimens and formalin-fixed and paraffin-embedded tissue. TERTp mutations were associated with older patients (≥45 years), whereas isocitrate dehydrogenase (IDH) mutations were inclined to a younger age ( 〈 45 years), frontal location, and pathologic stage II-III patients. IDH mutations were significantly associated with O6-methylguanine-DNA methyltransferase (MGMT) methylation ( P = 0.003 ) and lower Ki-67 protein expression ( P = 0.011 ). Moreover, MGMT methylation was enriched in IDH-mutant/TERTp-mutant gliomas, and Ki-67 protein expression was the highest in the IDH-wild type/TERTp-mutant group. Taken together, the findings of this study indicate the establishment of a rapid, precise, and practical Sanger sequencing technique for TERTp mutations in gliomas that may show promising results in clinical applications.

    Abstract: For the nondeterministic factors of an aeroengine blisk, including both factors with sufficient and insufficient statistical data, based on the dynamic substructural method of determinate analysis, the extremum response surface method of probabilistic analysis, and the interval method of nonprobabilistic analysis, a methodology called the probabilistic and nonprobabilistic hybrid reliability analysis based on dynamic substructural extremum response surface decoupling method (P-NP-HRA-DS-ERSDM) is proposed. The model includes random variables and interval variables to determine the interval failure probability and the interval reliability index. The extremum response surface function and its flow chart of mixed reliability analysis are given. The interval analysis is embedded in the most likely failure point in the iterative process. The probabilistic analysis and nonprobabilistic analysis are investigated alternately. Tuned and mistuned blisks are studied in a complicated environment, and the results are compared with the Monte Carlo method (MCM) and the multilevel nested algorithm (MLNA) to verify that the hybrid model can better handle reliability problems concurrently containing random variables and interval variables; meanwhile, it manifests that the computational efficiency of this method is superior and more reasonable for analysing and designing a mistuned blisk. Therefore, this methodology has very important practical significance.

    Abstract: Naoxintong capsule (NXTC) was a famous patent medicine of Traditional Chinese Medicine (TCM) to treat cerebrovascular diseases in China. An LC-MS/MS method was developed for simultaneous determination of 11 major ingredients (paeoniflorin, ecdysterone, amygdalin, mulberroside A, caffeic acid, ferulic acid, salvianolic acid B, astragaloside IV, formononetin, cryptotanshinone, and tanshinone IIA) in NXTC in rat plasma. All analytes were separated on an Eclipse plus C 18 column using a gradient mobile phase system of acetonitrile-0.1% formic acid aqueous solution. The lower limits of quantification of 11 ingredients were between 0.075 and 10 ng mL −1 . The precision was less than 15% and the accuracies were between 85% and 115%. The results showed that caffeic acid, ferulic acid, formononetin, cryptotanshinone, and tanshinone IIA could be detected after oral administration of NXTC. The validated method was successfully applied to pharmacokinetic study of the caffeic acid, ferulic acid, formononetin, cryptotanshinone, and tanshinone IIA in rats after oral administration of NXTC at single and triple dose.

    Abstract: Under the influence of COVID-19, online office and online education has ushered in a golden period of development. The teaching quality of online education has been a controversial issue. Our study takes online course teaching quality assessment as the starting point, explores the influencing factors of online course quality assessment with online courses as the research object, and analyzes the latest research proposal for an online course quality index. To make the online course quality assessment more intelligent, we propose an online course quality assessment method based on a fuzzy neural network. The method uses fuzzy rules as the baseline and adds a TSK perception mechanism to expand the perception domain of the fuzzy neural network and improve the course quality index prediction accuracy. At the input side of the fuzzy neural network, we preclassify the online course data into four parts, and each part of the data represents a different assessment domain. Due to the large data cost, we expanded the collective amount of data using data augmentation methods. In addition, we parse the structure of the fuzzy neural network hierarchy and introduce the construction and role of the TSK perception mechanism in the fuzzy rules. An optimal learning strategy is proposed in the fuzzy neural network training. Finally, in the experimental session, we verify the effectiveness of data augmentation and explore the distribution of course quality assessment weights. In the comparison of the model prediction results with the actual assessment results, our method achieves an excellent matching rate, which proves the high efficiency of our method in the online course quality assessment system.

    Abstract: Hepatocellular carcinoma (HCC), the most common type of primary liver cancer, is an aggressive tumor with a high mortality rate because of the limited systemic and locoregional treatment modalities. The development and progression of HCC depend on epigenetic changes that result in the activation or inhibition of some signaling pathways. The mTOR signaling pathway is essential for many pathophysiological processes and is considered a major regulator of cancer. Increasing evidence has shown that epigenetics plays a key role in HCC biology by regulating the mTOR signaling pathway. Therefore, epigenetic regulation through the mTOR signaling pathway to diagnose and treat HCC will become a very promising strategy.
