    Abstract: Backgrounds . Heme oxygenase-1 (HO-1) has been reported to play a regulatory role in osteoclastogenesis. Bone morphogenetic protein (BMP) pathways induce osteoblastic differentiation and bone remodeling. Aims . To identify serum levels of HO-1, BMP-7, and Runt related-transcription factor 2 (Runx2) in patients with rheumatoid arthritis (RA) and ankylosing spondylitis (AS) and to investigate the relationships between HO-1, BMP-7, Runx2, and other common biomarkers for bone metabolism. Results . Serum levels of HO-1 and BMP-7 were revealed to be significantly higher in patients with RA or AS than in healthy controls ( p 〈 0.0 1 ). In RA group, HO-1 was positively correlated with BMP-7, Runx2, and tartrate-resistant acid phosphatase-5b (TRAP-5b) ( p 〈 0.05 , resp.), BMP-7 was positively correlated with Runx2 and TRAP-5b ( p 〈 0.05 , resp.), and Runx2 was negatively correlated with N-terminal midfragment of osteocalcin (NMID) ( p 〈 0.05 ). In AS group, we observed identical correlation between HO-1 and BMP-7, but opposite correlations between BMP-7 and TRAP-5b and between Runx2 and NMID, when comparing with the RA cohort. Conclusion . Our findings suggest that HO-1 and BMP-7 are potential biomarkers for bone metabolism in patients with RA and AS. The different correlations between the bone markers point to distinct differences in bone remodeling pathways in the two types of arthritis.

    Abstract: Subchondral bone lesions, as the crucial inducement for accelerating cartilage degeneration, have been considered as the initiating factor and the potential therapeutic target of knee osteoarthritis (KOA). Acupotomy, the biomechanical therapy guided by traditional Chinese meridians theory, alleviates cartilage deterioration by correcting abnormal mechanics. Whether this mechanical effect of acupotomy inhibits KOA subchondral bone lesions is indistinct. This study aimed to investigate the effects of acupotomy on inhibiting subchondral bone resorption and to define the possible mechanism in immobilization-induced KOA rabbits. After KOA modeling, 8 groups of rabbits (4w/6w acupotomy, 4w/6w electroacupuncture, 4w/6w model, and 4w/6w control groups) received the indicated intervention for 3 weeks. Histological and bone histomorphometry analyses revealed that acupotomy prevented both cartilage surface erosion and subchondral bone loss. Further, acupotomy suppressed osteoclast activity and enhanced osteoblast activity in KOA subchondral bone, showing a significantly decreased expression of tartrate-resistant acid phosphatase (TRAP), matrix metalloproteinases-9 (MMP-9), and cathepsin K (Ctsk) and a significantly increased expression of osteocalcin (OCN); this regulation may be mediated by blocking the decrease in osteoprotegerin (OPG) and the increase in NF- κ B receptor activated protein ligand (RANKL). These findings indicated that acupotomy inhibited osteoclast activity and promoted osteoblast activity to ameliorate hyperactive subchondral bone resorption and cartilage degeneration in immobilization-induced KOA rabbits, which may be mediated by the OPG/RANKL signaling pathway. Taken together, our results indicate that acupotomy may have therapeutic potential in KOA by restoring the balance between bone formation and bone resorption to attenuate subchondral bone lesions.

    Abstract: The transition from surface mining to underground is a critical issue for metal mines. The commonly cited procedure cored by ultimate-pit-limit (UPL) methodology is restricted to maximize the profit from both surface and underground mining, due to the absence of the integration of the profit from either of them. Under the target for such maximization, this study proposes a new optimization approach, which directly relates the design of open-pit limit and underground stopes, by equalizing the marginal profit from either surface or underground mining. The variation of the crown pillar size is involved in this approach. The proposed approach is applied to the Dagushan iron mine, and results show the total profit increased from 3.79 billion CNYs (original design by conventional UPL methodology) to 4.17 billion CNYs (optimal design by the proposed approach), by 9.91%. Moreover, the marginal profit from surface and underground mining, as well as total profit, of all possible designs of surface-to-underground mining transition in Dagushan iron mine is calculated to validate the proposed approach. When the marginal profits satisfy the criterion of the proposed approach, the maximum value of the total profit appears, and this demonstrates the proposed approach is robust to maximize the total profit in surface-to-underground mining transition. This work contributes to existing literature studies primarily from practical aspect, by providing a unified approach to optimize the transition from surface to underground mining.

    Abstract: This paper proposed a new radar echo simulation method for ultra-low altitude targets in far-field conditions. Based on the electromagnetic (EM) scattering calculation of target and environment, combined with the weighted four-path model, the scattering data of target, environment, and multipath are obtained. The Range-Doppler ring partitioning method is used to determine the size of the minimum resolution units, and then the environment is divided into several scattering elements. By using the method of temporal decomposition, the wide-time pulse is decomposed into a plurality of narrow pulse signals, and the narrow pulses act on scattering elements with different distances and orientations in space. The total echo is obtained by a linear superposition of the responses of each scattering unit. In addition, the numerical results with different parameters, including signal bandwidths, target types, and target height, are simulated and analysed. The simulation results demonstrate that the proposed method can provide a better description of the scattering characteristics of sea-skimming targets in complex scenes in far-field conditions. Meanwhile, it can be applied to the detection and recognition of ultra-low altitude targets above the sea surface.

    Abstract: Abnormal vascular smooth muscle cell (VSMC) proliferation has an important role in the pathogenesis of both atherosclerosis restenosis and hypertension. Vascular endothelial growth factor (VEGF) has been shown to stimulate VSMC proliferation. In addition, angiogenesis is one of the hallmarks of cancerous growth. VEGF is the key modulator for the initial stages of angiogenesis that acts through the endothelial-specific receptor tyrosine kinases (VEGFRs). VEGFR-2 blockage is a good approach for suppression of angiogenesis. In order to discover novel VEGFR-2 TK inhibitors, we have designed and synthesized three new series of pyridine-containing compounds. The new compounds were all screened against a panel of three cell lines (HepG-2, HCT-116, and MCF-7). Promising results encouraged us to additionally evaluate the most active members for their in vitro VEGFR-2 inhibitory effect. Compound 7a, which is the most potent candidate, revealed a significant increase in caspase-3 level by 7.80-fold when compared to the control. In addition, Bax and Bcl-2 concentration levels showed an increase in the proapoptotic protein Bax (261.4 Pg/ml) and a decrease of the antiapoptotic protein Bcl-2 (1.25 Pg/ml) compared to the untreated cells. Furthermore, compound 7a arrested the cell cycle in the G2/M phase with induction of apoptosis. The immunomodulatory effect of compound 7a, the most active member, showed a reduction in TNF-α by 87%. Also, compound 7a caused a potent inhibitory effect on smooth muscle proliferation. Docking studies were also performed to get better insights into the possible binding mode of the target compounds with VEGFR-2 active sites.

    Abstract: Purpose. By comparing the performance of different models between artificial intelligence (AI) and doctors, we aim to evaluate and identify the optimal model for future usage of AI. Methods. A total of 500 fundus images of glaucoma and 500 fundus images of normal eyes were collected and randomly divided into five groups, with each group corresponding to one round. The AI system provided diagnostic suggestions for each image. Four doctors provided diagnoses without the assistance of the AI in the first round and with the assistance of the AI in the second and third rounds. In the fourth round, doctor B and doctor D made diagnoses with the help of the AI and the other two doctors without the help of the AI. In the last round, doctor A and doctor B made diagnoses with the help of AI and the other two doctors without the help of the AI. Results. Doctor A, doctor B, and doctor D had a higher accuracy in the diagnosis of glaucoma with the assistance of AI in the second ( p = 0.036 , p = 0.003 , and p ≤ 0.000 ) and the third round ( p = 0.021 , p ≤ 0.000 , and p ≤ 0.000 ) than in the first round. The accuracy of at least one doctor was higher than that of AI in the second and third rounds, in spite of no detectable significance ( p = 0.283 , p = 0.727 , p = 0.344 , and p = 0.508 ). The four doctors’ overall accuracy ( p = 0.004 and p ≤ 0.000 ) and sensitivity ( p = 0.006 and p ≤ 0.000 ) as a whole were significantly improved in the second and third rounds. Conclusions. This “Doctor + AI” model can clarify the role of doctors and AI in medical responsibility and ensure the safety of patients, and importantly, this model shows great potential and application prospects.

    Abstract: Porcine reproductive and respiratory syndrome (PRRS), which causes reproductive failure in sows and respiratory symptoms in piglets, poses a significant threat to the global pig industry. PRRS virus (PRRSV) variants continue to emerge and spread among pigs. NADC34-like PRRSV has been imported into China in recent years and has shown potential as an endemic strain, which is of great concern. In this study, a NADC34-like PRRSV, named HLJ13 strain, was isolated from a farm where pigs experienced respiratory symptoms and abortions. Genomic analysis revealed that the HLJ13 strain was a potential recombinant of NADC34-like and NADC30-like strains, and the restriction fragment length polymorphism of HLJ13 was a novel pattern that was not yet listed. In the PRRSV HLJ13-inoculated group, the piglets showed mild clinical symptoms, such as persistent fever, and showed histopathological lesions in the lungs, and the virus was detectable at 3 and 7 days postinoculation in anal and nasal swabs, respectively. Recombination analysis revealed that interlineage recombinant events were detected in 8 out of 27 Chinese NADC34-like PRRSVs. Phylogenetic analysis showed that Chinese NADC34-like PRRSVs were distributed in two clades of lineage 1, and Chinese NADC34-like PRRSVs showed different N-glycosylation modifications in glycoproteins, especially in GP3 and GP5. These findings shed light on the genomic characteristics and pathogenicity of the NADC34-like PRRSV in China.

    Abstract: Fracture is the main form of marine structural failure due to the action of waves and oceans. It is necessary to monitor the initiation and extension of cracks in a timely and accurate manner, and the stress intensity factor (SIF) is an important indicator for analysing the propagation of cracks. Based on existing research, this paper combines the calculation method of the SIF based on a single strain gauge (SSG) and maximum crack opening displacement (CMOD) and proposes a method based on the Kalman filter (KF). Numerical simulation and experimental verifications of this method have been carried out. The results show that the method can be used for a variety of configurations of precracked specimens. The KF-based method obtains more accurate results than the SSG-based and CMOD-based methods, and the relative error (RE) is not greater than 1.2%.
