Kurzfassung: This research was performed to observe the effect of GTS-21 on Collagen Induced Arthritis (CIA). CIA model was used and after the onset of arthritis, the rats were divided into three groups based on their clinical symptoms score. Two groups were intraperitoneally (IP) injected daily with GTS-21 (1 mg/kg, 2.5 mg/kg) for a week, whereas phosphate buffered saline (PBS) was used for the control group. Cytokine titers, radiological, and histological examinations were performed at different time points after treatment with GTS-21. Compared with those of the control, the levels of TNF- α , IL-1, and IL-6 in the serum were significantly reduced after GTS-21 management. In addition, radiological results show that bone degradation was inhibited as well. Moreover, the hematoxylin and eosin (H & E) staining indicated that the histological score was significantly alleviated in the therapeutic group. Tartrate-resistant acid phosphatase (TRAP) stain-positive cells were also detected in the destruction of the articular cartilage, which was significantly reduced compared with the control group. This study provides the first evidence on the effect of GTS-21 as a potential treatment for RA.

Kurzfassung: As a new efficient rock-breaking technology, harmonic impact drilling technology has received great attention, but the research on its rock-breaking mechanism is insufficient, which limits its development. Based on the theory of vibration, a simplified model of high-frequency harmonic vibration impact system is established in this paper. The numerical model was solved by Matlab and the motion equations of drill bit and rock at different stages of motion are obtained, respectively. Based on the factor analysis of the mathematical model, the dynamic characteristics of the impact system under harmonic excitation are studied. Finally, numerical simulations are carried out to further analyze the drilling effect of harmonic impact drilling and verify the correctness of the simplified model. The results show that when the excitation frequency equals the resonance frequency of rock, the vibration displacement of rock reaches the peak value. The drilling speed is greatly increased by harmonic impact drilling compared with conventional drilling.

Kurzfassung: A novel vector with high gene delivery efficiency and special cell-targeting ability was developed using a good strategy that utilized low-molecular-weight polyethylenimine (PEI; molecular weight: 600 KDa [PEI600]) crosslinked to β -cyclodextrin ( β -CyD) via a facile synthetic route. Fibroblast growth factor receptors (FGFRs) are highly expressed in a variety of human cancer cells and are potential targets for cancer therapy. In this paper, CY11 peptides, which have been proven to combine especially with FGFRs on cell membranes were coupled to PEI- β -CyD using N -succinimidyl-3-(2-pyridyldithio) propionate as a linker. The ratios of PEI600, β -CyD, and peptide were calculated based on proton integral values obtained from the 1 H-NMR spectra of the resulting products. Electron microscope observations showed that CY11-PEI- β -CyD can efficiently condense plasmid DNA (pDNA) into nanoparticles of about 200 nm, and MTT assays suggested the decreased toxicity of the polymer. Experiments on gene delivery efficiency in vitro showed that CY11-PEI- β -CyD/pDNA polyplexes had significantly greater transgene activities than PEI- β -CyD/pDNA in the COS-7 and HepG2 cells, which positively expressed FGFR, whereas no such effect was observed in the PC-3 cells, which negatively expressed FGFR. Our current research indicated that the synthesized nonviral vector shows improved gene delivery efficiency and targeting specificity in FGFR-positive cells.

Kurzfassung: The pseudopotential lattice Boltzmann method (LBM) with a tunable surface tension term is applied to study a droplet impact on a moving thin film. The Re effects of dimensionless parameters on the upstream and downstream crown evolution are studied, including Reynolds number ( Re ), Weber number ( We ), liquid film thickness, and horizontal velocity of the liquid film. The movement of the liquid film causes the asymmetry development of the upstream and downstream crown. Both the instability of upstream and downstream crowns increases with the increase of Re and We , and the upstream crown becomes more prone to break up. And a critical value of film thickness exists with the height of the upstream and downstream liquid crowns reaches the maximum value. And the velocity of liquid film restrains the development of the height of the upstream and downstream crowns, but it promotes the growth of the crown radius.
