    Kurzfassung: Truck-lifting accidents are common in container-lifting operations. Previously, the operation sites are needed to arrange workers for observation and guidance. However, with the development of automated equipment in container terminals, an automated accident detection method is required to replace manual workers. Considering the development of vision detection and tracking algorithms, this study designed a vision-based truck-lifting prevention system. This system uses a camera to detect and track the movement of the truck wheel hub during the operation to determine whether the truck chassis is being lifted. The hardware device of this system is easy to install and has good versatility for most container-lifting equipment. The accident detection algorithm combines convolutional neural network detection, traditional image processing, and a multitarget tracking algorithm to calculate the displacement and posture information of the truck during the operation. The experiments show that the measurement accuracy of this system reaches 52 mm, and it can effectively distinguish the trajectories of different wheel hubs, meeting the requirements for detecting lifting accidents.

    Kurzfassung: NM23 expression is closely associated with hepatocellular carcinoma (HCC) recurrence, but the hereditary factors influencing NM23 levels are unknown. Using public database, the diagnostic value of NM23 in HCC was investigated. A total of 424 hepatitis B virus- (HBV-) related HCC patients were enrolled to perform a genome–wide association study for identifying candidate variants associated with NM23 expression level. Additionally, a logistic regression model, haplotypes, and survival analysis were performed in the subsequent analysis. We identified high NM23 expression levels that have a diagnostic accuracy in HCC tissues and had a poor recurrence-free survival in HBV-related HCC patients. Variants near Psoriasis susceptibility 1 candidate 1 ( PSORS1C1 ) and StAR related lipid transdomain containing 3 ( STARD3 ) are associated with NM23 expression. The PSORS1C1 haplotype TGCACA and the STARD3 haplotype GG have favorable cumulative effects on NM23 expression. Further, variants in PSORS1C1 were associated with either overall survival (rs556285588, rs3095301, and rs3131003) only or overall survival and recurrence-free survival (rs560052000 and rs541820233) both in HCC patients. Our findings suggested that variants at the PSORS1C1 and STARD3 loci play an important role in NM23 regulation. Moreover, variants in PSORS1C1 are potential biomarkers for the prediction of postoperative clinical outcomes in HBV-related HCC patients. Thus, variants in PSORS1C1 and STARD3 are associated with NM23 expression and clinical outcomes of HBV-related HCC patients, which may be regarded as potential biomarkers for this disease.

    Kurzfassung: The Xiashan landslide, which is classified as a typical basalt platform landslide, is the most massive landslide in Zhejiang Province, China. Once sliding occurs, it will pose a severe threat to the life and property of downstream residents and the nearby section of Hangzhou-Taizhou Expressway. On the basis of the geological conditions, present situation, and latest monitoring data of the landslide, this study finds that rainfall is the main influencing factor, and the creep mode is the main prediction mode of its subsequent deformation. The rainfall statistics of the landslide area in the past 30 years show that the rainfall and rainfall frequency in the landslide area display an increasing trend. The probability of heavy rain with rainfall intensity of 100–250 mm/day in the landslide area is very high. On this basis, combined with the numerical analysis method, a finite element model of the slope considering rainfall and groundwater conditions is constructed to analyze the causes and failure mechanism of this landslide comprehensively. Results indicate that the maximum tensile stress at the top of the trailing edge under the natural state is 5.10 MPa, which is very close to the saturated tensile strength of rock mass. Thus, tensile cracks are easily generated and developed, thereby causing the failure mode to be the hydraulic driving type. Also, with the increase in rainfall intensity, the slope plastic strain increases and the slope plastic zone develops and extends until it is completely penetrated. When the rainfall intensity is more than 200 mm/day, the slope safety factor is close to unity, and the slope approaches a failure condition. Therefore, the landslide should be controlled through water treatment and integrated with engineering measures.

    Kurzfassung: Background. Colorectal cancer liver metastasis (CRLM) is a high degree of malignancy with rapid disease progression and has a poor prognosis. Both serum apolipoprotein A-I (ApoA-I) and neutrophil-to-lymphocyte ratio (NLR) play key roles in anti-inflammation and antitumor. This study is aimed at evaluating the implication of serum ApoA-I level in combination with NLR in the prognosis of CRLM. Methods. We retrospectively analyzed the serum ApoA-I level and NLR in 237 patients with CRLM. Cox regression analyses were used to identify the independent prognostic significance of these indicators. Kaplan-Meier method and Log-rank test were applied to compute overall survival (OS). Both the ApoA-I and NLR were divided into three levels, according to their medians. A risk-stratified prediction model was established to evaluate the prognosis of patients with CRLM. The ROC curve AUC values were applied to evaluate the capability of the model. Results. Higher levels of ApoA-I and lower NLR were strongly associated with prolonged OS (Log-rank test, P 〈 0.05 ). The patients were then grouped into three queues according to the ApoA-I level and NLR. There was a crucial diversity in the OS ( P 〈 0.001 ) between the high-risk ( ApoA − I ≤ 1.03  g/L and NLR 〉 3.24 ), medium-risk ( ApoA − I 〉 1.03   g/L or NLR ≤ 3.24 ) and low-risk groups ( ApoA − I 〉 1.03   g/L and NLR ≤ 3.24 ). The AUC value of the prediction model ( AUC = 0.623 , 95% CI: 0.557-0.639, P = 0.001 ) was higher than other individual indicators (including ApoA-I, NLR, cT classification, and cN classification). Additionally, the association of the prediction model and cTN classification ( AUC = 0.715 , 95% CI: 0.606-0.708, P 〈 0.001 ) was better than the model and cTN classification alone. Conclusion. The combination of ApoA-I level and NLR could be a prognostic indicator for CRLM.

    Kurzfassung: Background . Hepatocellular carcinoma (HCC) is a highly aggressive malignancy. Traditional Chinese Medicine (TCM), with the characteristics of syndrome differentiation, plays an important role in the comprehensive treatment of HCC. This study aims to develop a nonnegative matrix factorization- (NMF-) based feature selection approach (NMFBFS) to identify potential clinical symptoms for HCC patient stratification. Methods . The NMFBFS approach consisted of three major steps. Firstly, statistics-based preliminary feature screening was designed to detect and remove irrelevant symptoms. Secondly, NMF was employed to infer redundant symptoms. Based on NMF-derived basis matrix, we defined a novel similarity measurement of intersymptoms . Finally, we converted each group of redundant symptoms to a new single feature so that the dimension was further reduced. Results . Based on a clinical dataset consisting of 407 patient samples of HCC with 57 symptoms, NMFBFS approach detected 8 irrelevant symptoms and then identified 16 redundant symptoms within 6 groups. Finally, an optimal feature subset with 39 clinical features was generated after compressing the redundant symptoms by groups. The validation of classification performance shows that these 39 features obviously improve the prediction accuracy of HCC patients. Conclusions . Compared with other methods, NMFBFS has obvious advantages in identifying important clinical features of HCC.

    Kurzfassung: Objective. The endocrine function of bone in energy metabolism may be mediated by the osteocalcin (OC). We examined the association between OC and energy metabolism among Chinese postmenopausal women. Design and Setting. A cross-sectional cohort study enrolling 1635 participants was conducted using data from the Peking Vertebral Fracture study. Partial correlation analysis was performed to explore the correlation of OC, parathyroid hormone (PTH), or 25-hydroxyvitamin D (25(OH)D) with glycemic and lipid metabolic parameters. A logistic regression model was used to investigate the association of OC, PTH, or 25(OH)D with the prevalence of diabetes and dyslipidemia. Results. Serum levels of OC, PTH, and 25(OH)D were all positively correlated with serum cholesterol levels, whereas only OC was negatively associated with serum glucose level. In the logistic regression model, both OC and PTH were negatively associated with the prevalence of diabetes (odds ratio [OR], 95% confidence interval [95% CI] : 0.967, 0.948–0.986 for OC and 0.986, 0.978–0.994 for PTH). No significant association was found between 25(OH)D and diabetes. Both OC and 25(OH)D, rather than PTH, were associated with abnormalities of high cholesterol levels, such as hypercholesterolemia and high LDL-C levels. Further classifying the population based on the median value of OC and PTH, low OC and low PTH subgroup had the highest OR, 95% CI for diabetes (1.873, 1.287–2.737) and the lowest OR, 95% CI for hypercholesterolemia (0.472, 0.324–0.688) and for high LDL-C (0.538, 0.376–0.771). Conclusion. Among Chinese postmenopausal women, a lower serum level of OC was associated with a higher prevalence of diabetes and lower serum cholesterol levels, and a low PTH concentration could magnify these associations.

    Kurzfassung: Background. SARS-CoV-2 induces apoptosis and amplifies the immune response by continuously stressing the endoplasmic reticulum (ER) after invading cells. This study aimed to establish a protein-metabolic pathway associated with ER dysfunction based on the invasion mechanism of SARS-CoV-2. Methods. This study included 17 healthy people and 46 COVID-19 patients, including 38 mild patients and 8 severe patients. Proteomics and metabolomics were measured in the patient plasma collected at admission and one week after admission. The patients were further divided into the aggravation and remission groups based on disease progression within one week of admission. Results. Cross-sectional comparison showed that endoplasmic reticulum molecular chaperone-binding immunoglobulin protein (ERC-BiP), angiotensinogen (AGT), ceramide acid (Cer), and C-reactive protein (CRP) levels were significantly increased in COVID-19 patients, while the sphingomyelin (SM) level was significantly decreased P   〈   0.05 . In addition, longitudinal comparative analysis found that the temporal fold changes of ERC-BiP, AGT, Cer, CRP, and SM were significantly different between the patients in the aggravation and remission groups P   〈   0.05 . ERC-BiP, AGT, and Cer levels were significantly increased in aggravation patients, while SM was significantly decreased P   〈   0.05 . Meanwhile, ERC-BiP was significantly correlated with AGT (r = 0.439; P   〈   0.001 ). Conclusions. ERC-BiP can be used as a core index to reflect the degree of ER stress in COVID-19 patients, which is of great value for evaluating the functional state of cells. A functional pathway for AGT/ERC-BiP/glycolysis can directly assess the activation of unfolded protein reactions. The ERC-BiP pathway is closer to the intracellular replication pathway of SARS-CoV-2 and may help in the development of predictive protocols for COVID-19 exacerbation.

    Kurzfassung: To establish a high-efficiency system of isolated microspore culture for different barley genotypes, we investigated the effects of nitrogen sources and concentrations on callus induction and plant regeneration in different barley genotypes. The results showed that the organic nitrogen sources greatly increased the callus induction, and the great reduction of total nitrogen sources would significantly decrease the callus induction. And the further optimization experiments revealed that the increasing of organic nitrogen sources was much important in callus induction while it seemed different in plant regeneration. Based on the great effects of organic nitrogen on callus induction, the medium of N6-ANO1/4-2000 might be the best choice for the microspore culture system. In addition, the phylogenetic analysis indicated that there were clear differences of genetic backgrounds among these barley genotypes, and it also suggested that this medium for microspore culture had widespread utilization in different barley genotypes.

    Kurzfassung: Accumulated evidence indicates that miR-455-5p functions as tumor suppressor in the progression of various cancers. However, the mechanism through which miR-455-5p influences the tumorigenesis of human prostate cancer (PCa) remains undetermined. In this study, reanalysis of data obtained from the Memorial Sloan Kettering Cancer Center showed that miR-455-5p can be used as biomarker for PCa diagnosis and predictor of poor prognosis. Functional assays indicated that miR-455-5p overexpression could suppress cellular proliferation, inhibit tumor growth, and trigger apoptosis by activating and cleaving caspase 3. We experimentally verified that miR-455-5p negatively regulated the C–C motif chemokine receptor 5 (CCR5). Overall, our data demonstrate that miR-455-5p suppressed PCa cellular proliferation and induced cell apoptosis by downregulating CCR5. Thus, miR-455-5p may be considered a new therapeutic strategy for PCa.
