    Abstract: As one of the important treatment methods for cancer patients, radiotherapy may lead to incidental irradiation of the heart, resulting in radiotherapy-induced heart disease (RIHD) arising many years after radiotherapy. While, there are few studies on early subclinical cardiac damage, which may be essential for the protection of late RIHD. To detect and predict RIHD and early subclinical cardiac damage induced by thoracic radiation therapy, based on two-dimensional speckle tracking echocardiography (2D STE) combined with multiple circulating biomarkers and accurate heart dosimetry. Methods and Analysis This is a monocentric prospective cohort study in which 104 patients treated for malignant tumors and with cardiac radiation exposure will be included. All participants will be followed for 12 months after radiotherapy. Echocardiography, 2D STE, and blood samples will be underwent at 5-time points (baseline; after completion of RT; 2, 6, and 12 months after RT). Left ventricular ejection fraction (LVEF); global longitudinal, radial, and circumferential strain; diastolic function parameters; creatine kinase (CK); creatine kinase isoenzyme (CK-MB); cardiac troponin T (cTnT); N-terminal pro-B-type natriuretic peptide (NT-proBNP) and hypersensitive C-reactive protein (hs-CRP) will be measured at baseline and every follow-up time. The incidence of major adverse cardiovascular events will be recorded. Discussion This study details the protocol and presents the primary limits and advantages of this single-center project. The inclusion of patients began in 2021, and the results are expected to be published in 2023. This study will be allowed to enhance knowledge on detection and prediction of early subclinical cardiac dysfunction induced by thoracic radiation therapy, based on two-dimensional speckle tracking echocardiography (2D STE) combined with circulating biomarkers and accurate heart dosimetry. Furthermore, we will evaluate risk factors of subtle cardiac damage and identify high-risk groups for early heart damage. Clinical Trial Registration ClinicalTrials.gov , identifier: NCT04443400.

    Abstract: The prognosis for endocrine-independent prostate carcinoma is still poor due to its highly metastatic feature. In the present work, TPX2 (the targeting protein for Xklp2), which is known as a micro-tubulin interacted protein, was identified as a novel coactivator of ETS-1, a transcription factor that plays a central role in mediating the metastasis of human malignancies. TPX2 enhanced the transcription factor activation of ETS-1 and increased the expression of ETS-1’s downstream metastasis-related genes, such as mmp3 or mmp9 , induced by HGF (hepatocyte growth factor), a typical agonist of the HGF/c-MET/ETS-1 pathway. The protein-interaction between TPX2 and ETS-1 was examined using immunoprecipitation (IP). TPX2 enhanced the accumulation of ETS-1 in the nuclear and the recruitment of its binding element (EST binding site, EBS) located in the promoter region of its downstream gene, mmp9 . Moreover, TPX2 enhanced the in vitro or in vivo invasion of a typical endocrine-independent prostate carcinoma cell line, PC-3. Therefore, TPX2 enhanced the activation of the HGF/ETS-1 pathway to enhance the invasion of endocrine-independent prostate carcinoma cells and thus it would be a promising target for prostate carcinoma treatment.

    Abstract: In densely populated coastal areas with sea-level rise (SLR), protecting the shorelines against erosion due to the wave impact is crucial. Along with many engineered structures like seawalls and breakwaters, there are also green structures like constructed oyster reefs (CORs) that can not only attenuate the incident waves but also grow and maintain pace with SLR. However, there is a lack of data and understanding of the long-term wave attenuation capacity of the living shoreline structures under SLR. In this study, we used the phase-resolving Boussinesq model, FUNWAVE-TVD, to examine the hydrodynamics including wave height and wave-induced currents around the CORs in the Gandys Beach living shoreline project area in the upper Delaware Bay, United States. Waves were measured at six locations (offshore to onshore, with and without CORs) in the Gandys Beach living shoreline project area for two winter months, during which four nor’easters occurred. We selected three cases that represent prevailing wind, wave, and tide conditions to examine the fine spatial and temporal changes in wave height and current velocity by the construction of the reefs. Wave heights and wave energy spectra generated from FUNWAVE-TVD were then validated with field observations. It is found that FUNWAVE-TVD is capable of simulating waves and associated hydrodynamic processes that interact with CORs. The model results show that wave attenuation rates vary with the incident wave properties and water depth, and wave-induced circulation patterns are affected by the CORs. The wave attenuation capacity of CORs over the next 100 years was simulated with the incorporation of the oyster reef optimal growth zone. Our study found that sustainable wave attenuation capacity can only be achieved when suitable habitat for COR is provided, thus it can vertically grow with SLR. Suitable habitat includes optimal intertidal inundation duration, current velocity for larval transport and settlement, on-reef oyster survival and growth, and other environmental conditions including salinity, temperature, and nutrient availability. Furthermore, the model results suggest that it would take CORs approximately 9 years after construction to reach and maintain the maximum wave attenuation capacity in sustainable living shorelines.

    Abstract: The distribution and composition of wetland habitats for wintering waterbirds are heavily influenced by water level fluctuations. Through polder construction and aquaculture activities, paddy fields and aquaculture ponds have decreased the lateral connectivity of water level fluctuations in the lake. However, the impacts of water level fluctuations on habitat suitability, which can be seen using high-resolution images analysis, often cannot separate disturbances caused by the paddy fields and aquaculture ponds, and it is difficult to capture the actual impact of water level fluctuations on wetland habitats. Based on remote sensing image data and hydrological data, we selected Caizi Lake as a study site and comparatively analyzed the changes in wintering waterbird habitats in a water level sequence under the two scenarios. Our work showed that paddy fields and aquaculture ponds should be considered as potential options for creating more suitable habitats for migratory waterbirds if combined with reasonable and effective management of the water level within the paddy fields and aquaculture ponds. The present study results could facilitate the management and sustainable utilization of Caizi Lake wetlands and provide support for creating small habitats by managing the water levels of paddy fields and aquaculture ponds.

    Abstract: Cancer has the highest mortality in humans worldwide, and the development of effective drugs remains a key issue. Traditional Chinese medicine Saussurea involucrata (SI) exhibits a series of effects, such as anti-cancer, but the action mechanisms are still unclear. Here, systems pharmacology was applied to reveal its anti-cancer mechanism. First, we screened the active compounds of SI. Then, the compound–target network, target–disease network, and target–pathway network were constructed. DAVID was applied for GOBP analysis and KEGG pathway enrichment analysis on cancer-related targets. Seven potential compounds and 187 targets were identified. The target–disease classification network showed that compounds mainly regulated proteins related to cancer, nervous system diseases, and cardiovascular system diseases. Also, SI anti-tumor effect mainly associated with the regulation of NO production, angiogenesis, MAPK, and PKB from GOBP enrichment. Additionally, KEGG pathway enrichment indicated that targets involved in anti-inflammatory action, inhibiting angiogenesis and anti-proliferation or inducing apoptosis. Experimental validation showed that four active compounds could inhibit cell proliferation and promote apoptosis in A549 (except for kaempferol), PC-3, and C6 cells. This study not only provides experimental evidence for further research on SI in cancer treatment but also promotes the development of potential drugs of SI in modern medicine.
