    Abstract: This study aims to differentiate preoperative Borrmann type IV gastric cancer (GC) from primary gastric lymphoma (PGL) by transfer learning radiomics nomogram (TLRN) with whole slide images of GC as source domain data. Materials and Methods This study retrospectively enrolled 438 patients with histopathologic diagnoses of Borrmann type IV GC and PGL. They received CT examinations from three hospitals. Quantitative transfer learning features were extracted by the proposed transfer learning radiopathomic network and used to construct transfer learning radiomics signatures (TLRS). A TLRN, which integrates TLRS, clinical factors, and CT subjective findings, was developed by multivariate logistic regression. The diagnostic TLRN performance was assessed by clinical usefulness in the independent validation set. Results The TLRN was built by TLRS and a high enhanced serosa sign, which showed good agreement by the calibration curve. The TLRN performance was superior to the clinical model and TLRS. Its areas under the curve (AUC) were 0.958 (95% confidence interval [CI], 0.883–0.991), 0.867 (95% CI, 0.794–0.922), and 0.921 (95% CI, 0.860–0.960) in the internal and two external validation cohorts, respectively. Decision curve analysis (DCA) showed that the TLRN was better than any other model. TLRN has potential generalization ability, as shown in the stratification analysis. Conclusions The proposed TLRN based on gastric WSIs may help preoperatively differentiate PGL from Borrmann type IV GC.     Abstract: Here provides a complementary treatment, acupressure at the Qiu acupoint, a novel acupoint, which potentially alleviates renal colic. Materials and methods 90 patients were included in this study. Acupressure-group patients ( n = 46) were administered acupressure at the Qiu acupoint following a preset protocol. Parecoxib sodium-group patients ( n = 44) were administered parecoxib sodium (40 mg) ( via the direct intravenous route). The visual analog scale (VAS) was used to evaluate pain intensity at baseline and at 1, 5, 10, 20, 30, and 120 min after initiating the intervention. Linear mixed effects model was performed to detect the rate of decrease of VAS per time and their covariant effect on the efficacy of acupressure. Results No significant statistical differences in baseline data and VAS scores were observed. The acupressure group obtained lower VAS scores at the 1st, 5th, 10th, and 20th minute than the parecoxib sodium group after initiating the intervention (mean: 4.33 vs. 7.61, mean difference (MD): 3.29, 95% CI: 0.23, 2.84; mean: 2.65 vs. 7.61, MD: 4.96, 95% CI: 4.44, 5.49; mean: 1.63 vs. 6.59, MD: 4.96, 95% CI: 4.48, 5.44; mean: 1.26 vs. 3.64 MD: 2.38, 95% CI: 1.87, 2.88; P & lt; 0.05). The markedly effective rate was similar between the two groups. The linear mixed effects model demonstrated that acupressure at the Qiu point was significantly faster than parecoxib sodium in decreasing VAS scores with an estimate of –2.05 (95% CI: –2.51, –1.59, p = 0.000), especially within 10 minutes with an estimate of 0.18 (95% CI: 0.12, 0.25, p = 0.000). Conclusion Acupressure at the Qiu acupoint is significantly faster than parecoxib sodium in decreasing VAS scores within 10 minutes.     Abstract: The rupture risk assessment of intracranial aneurysms (IAs) is clinically relevant. How to accurately assess the rupture risk of IAs remains a challenge in clinical decision-making. Purpose We aim to build an integrated model to improve the assessment of the rupture risk of IAs. Materials and Methods A total of 148 (39 ruptured and 109 unruptured) IA subjects were retrospectively computed with computational fluid dynamics (CFDs), and the integrated models were proposed by combining machine learning (ML) and deep learning (DL) algorithms. ML algorithms that include random forest (RF), k-nearest neighbor (KNN), XGBoost (XGB), support vector machine (SVM), and LightGBM were, respectively, adopted to classify ruptured and unruptured IAs. A Pointnet DL algorithm was applied to extract hemodynamic cloud features from the hemodynamic clouds obtained from CFD. Morphological variables and hemodynamic parameters along with the extracted hemodynamic cloud features were acted as the inputs to the classification models. The classification results with and without hemodynamic cloud features are computed and compared. Results Without consideration of hemodynamic cloud features, the classification accuracy of RF, KNN, XGB, SVM, and LightGBM was 0.824, 0.759, 0.839, 0.860, and 0.829, respectively, and the AUCs of them were 0.897, 0.584, 0.892, 0.925, and 0.890, respectively. With the consideration of hemodynamic cloud features, the accuracy successively increased to 0.908, 0.873, 0.900, 0.926, and 0.917. Meanwhile, the AUCs reached 0.952, 0.881, 0.950, 0.969, and 0.965 eventually. Adding consideration of hemodynamic cloud features, the SVM could perform best with the highest accuracy of 0.926 and AUC of 0.969, respectively. Conclusion The integrated model combining ML and DL algorithms could improve the classification of IAs. Adding consideration of hemodynamic cloud features could bring more accurate classification, and hemodynamic cloud features were important for the discrimination of ruptured IAs.

    Abstract: At present, the energy loss (EL) mechanism of intracranial aneurysm (IA) rupture is explored based on the global EL calculated by Bernoulli equation, but the details of EL are still unclear. This study aimed to explore the temporal and spatial characteristics of EL of IAs and reveal its mechanism. A novel method for calculating the EL of IAs based on dissipation function (DF) was proposed. DF was derived from the differential form of the energy equation and reflected the irreversible conversion from mechanical energy to internal energy caused by the friction between the fluid micelles. Eight sidewall IAs located at the posterior communicating segment of the internal carotid artery were collected; the three-dimensional (3D) geometric models of IAs were established employing image segmentation and 3D reconstruction. Computational fluid dynamics was applied to obtain hemodynamic parameters of IAs. The temporal and spatial characteristics of EL of IAs were achieved utilizing our proposed method. The simulation results indicated that EL occurred mainly in the boundary layer and the region adjacent to high-velocity inflow jet, EL increased rapidly during cardiac systole and reached its maximum at end-systolic phase and then decreased gradually during diastole until the end of cardiac cycle. The proposed method achieved some improvements over the traditional Bernoulli equation–based method by acquiring the temporal and spatial characteristics of EL, and it could provide insights into the EL of IAs and contribute to further rupture mechanism investigation.

    Abstract: It is of potential clinical value to improve the accuracy of Alzheimer's disease (AD) recognition using structural MRI. We proposed a reparametrized convolutional neural network (Re-CNN) to discriminate AD from NC by applying morphological metrics and deep semantic features. The deep semantic features were extracted through Re-CNN on structural MRI. Considering the high redundancy in deep semantic features, we constrained the similarity of the features and retained the most distinguishing features utilizing the reparametrized module. The Re-CNN model was trained in an end-to-end manner on structural MRI from the ADNI dataset and tested on structural MRI from the AIBL dataset. Our proposed model achieves better performance over some existing structural MRI-based AD recognition models. The experimental results show that morphological metrics along with the constrained deep semantic features can relatively improve AD recognition performance.     Abstract: Urolithin A (UroA) is one of the primary intestinal metabolites of ellagitannins, showing translational potential as a nutritional intervention in humans. Mounting evidence suggests that fructose consumption contributes to the progression of chronic kidney disease (CKD) that manifests in hyperuricemic nephropathy, renal inflammation, and tubulointerstitial injury. Here, we investigated the efficacy of UroA in alleviating fructose-induced hyperuricemic nephropathy in mice. Uric acid-exposed human kidney-2 (HK-2) cells were utilized for in vitro mechanism validation. Histopathological staining, immunoblotting, and transmission electron microscope were performed for the mechanistic investigations. Our results revealed that UroA ameliorated fructose-induced hyperuricemic nephropathy in mice. The histopathologic assessment showed that UroA attenuated tubular hypertrophy and dilation, glomerular basement membrane thickening, and collagen deposition in the kidney of fructose-fed mice. Mechanistically, UroA treatment impaired STING-NLRP3 activation, resulting in reduced production of proinflammatory cytokines IL-1β, IL-6, and TNF-α. Notably, UroA exhibited a scavenging effect against reactive oxygen species (ROS) and restored fructose-impaired PINK1/Parkin-mediated mitophagy in nephropathic mice. Furthermore, the inhibitory effect of UroA in STING-NLRP3 activation was impaired after Parkin gene silencing in HK-2 cells. Together, this study suggests that UroA alleviates fructose-induced hyperuricemic nephropathy by promoting Parkin-dependent mitophagy, thereby suppressing STING-NLRP3 axis-mediated inflammatory response. Thus, dietary supplementation with UroA or ellagitannins-rich foods may serve as a promising intervention to prevent CKD progression.

    Abstract: Stigma exsertion rate (SER) is an index of outcrossing ability in rice and is a key trait of male sterile lines (MSLs) in hybrid rice. In this study, it was found that the maintainer lines carrying gs3 and gs3 / gw8 showed higher SER. Single-segment substitution lines (SSSLs) carrying gs3 , gw5 , GW7 or gw8 genes for grain shape and gene pyramiding lines were used to reveal the relationship between grain shape and SER. The results showed that the grain shape regulatory genes had pleiotropic effects on SER. The SERs were affected by grain shapes including grain length, grain width and the ratio of length to width (RLW) not only in low SER background, but also in high SER background. The coefficients of determination (R 2 ) between grain length and SER, grain width and SER, and grain RLW and SER were 0.78, 0.72, and 0.91 respectively. The grain RLW was the most important parameter affecting SER, and a larger grain RLW was beneficial to stigma exsertion. The pyramiding line PL- gs3 / GW7 / gw8 showed the largest grain RLW and the highest SER, which will be a fine breeding resource. Further research showed that the grain shape regulatory genes had pleiotropic effects on stigma shape, although the R 2 values between grain shape and stigma shape, and stigma shape and SER were lower. Our results demonstrate that grain shape is a factor affecting SER in rice, in part by affecting stigma shape. This finding will be helpful for breeding MSLs with high SER in hybrid rice.

    Abstract: Approximately 75% of Chinese hypertensive patients have elevated homocysteine (Hcy). Its implication in risk assessment and prevention of the first stroke remains an important clinical and public health question. Methods This study was based on a community cohort recruited from 2016 to 2018 in the rural China. To maximize cost efficiency, we used a nested case-control design, including 3,533 first stroke cases and 3,533 controls matched for age ±1 years, sex, and village. Individual associations of tHcy and traditional risk factors with the first stroke were examined, and their population-attributable risks (PARs) were estimated. Results There was a significant dose-response association between first stroke and total Hcy (tHcy) levels, with adjusted odds ratios of 1.11 (95% CI: 0.97, 1.26) for tHcy 10–15 μmol/L and 1.44 (1.22, 1.69) for tHcy ≥ 15 μmol/L, all compared to tHcy & lt; 10 μmol/L. A similar trend was found for ischemic and hemorrhagic stroke. tHcy and systolic blood pressure (SBP) were independently and additively associated with the risk of first stroke (tHcy: 1.06 [1.02, 1.1]; SBP: 1.13 [1.1, 1.16] ; P -interaction, 0.889). Among the ten main risk factors examined, the top two contributors to the first stroke were SBP and tHcy, with PARs of 25.73 and 11.24%, respectively. Conclusions Elevated tHcy is the second most important contributor and acts additively with SBP to increase the risk of the first stroke. This finding underscores the importance of screening and treating elevated tHcy along with traditional risk factors to further reduce the burden of the first stroke in the high-risk populations.

    Abstract: This study aimed to establish and validate a prognostic model based on magnetic resonance imaging and clinical features to predict the survival time of patients with glioblastoma multiforme (GBM). Methods In this study, a convolutional denoising autoencoder (DAE) network combined with the loss function of the Cox proportional hazard regression model was used to extract features for survival prediction. In addition, the Kaplan–Meier curve, the Schoenfeld residual analysis, the time-dependent receiver operating characteristic curve, the nomogram, and the calibration curve were performed to assess the survival prediction ability. Results The concordance index (C-index) of the survival prediction model, which combines the DAE and the Cox proportional hazard regression model, reached 0.78 in the training set, 0.75 in the validation set, and 0.74 in the test set. Patients were divided into high- and low-risk groups based on the median prognostic index (PI). Kaplan–Meier curve was used for survival analysis ( p = & lt; 2e-16 in the training set, p = 3e-04 in the validation set, and p = 0.007 in the test set), which showed that the survival probability of different groups was significantly different, and the PI of the network played an influential role in the prediction of survival probability. In the residual verification of the PI, the fitting curve of the scatter plot was roughly parallel to the x -axis, and the p -value of the test was 0.11, proving that the PI and survival time were independent of each other and the survival prediction ability of the PI was less affected than survival time. The areas under the curve of the training set were 0.843, 0.871, 0.903, and 0.941; those of the validation set were 0.687, 0.895, 1.000, and 0.967; and those of the test set were 0.757, 0.852, 0.683, and 0.898. Conclusion The survival prediction model, which combines the DAE and the Cox proportional hazard regression model, can effectively predict the prognosis of patients with GBM.

    Abstract: Non-alcoholic fatty liver disease (NAFLD) is a chronic progressive liver disease with increasing prevalence. Lipophagy is a type of programmed cell death that plays an essential role in maintaining the body’s balance of fatty acid metabolism. However, the livers of NAFLD patients are abnormally dysregulated in lipophagy. mTORC1 is a critical negative regulator of lipophagy, which has been confirmed to participate in the process of lipophagy through various complex mechanisms. Therefore, targeting mTORC1 to restore failed autophagy may be an effective therapeutic strategy for NAFLD. This article reviews the main pathways through which mTORC1 participates in the formation of lipophagy and the intervention effect of mTORC1-regulated lipophagy in NAFLD, providing new therapeutic strategies for the prevention and treatment of NAFLD in the future.
