    Abstract: WHO 2/3 glioma is a common intracranial tumor that seriously affects the quality of life and survival time of patients. Previous studies have shown that the tricarboxylic acid (TCA) cycle is closely related to the occurrence and development of glioma, while recent studies have shown that cuproptosis, a novel programmed death pathway, is closely related to the inhibition of the TCA cycle. In our study, eight of ten cuproptosis-related genes (CRGs) were found to be differentially expressed between normal and WHO 2/3 glioma tissues. Through the LASSO algorithm, the cuproptosis-associated risk signatures (CARSs) were constructed, which can effectively predict the prognosis of WHO 2/3 glioma patients and are closely related to clinicopathological features. We analyzed the relationship between risk score and immune cell infiltration through Xcell, ssGSEA, TIMER database, and immune checkpoint molecules. In addition, the relationship between risk score and chemotherapeutic drug sensitivity was also investigated. The prognosis-related independent risk factors FDX1 and CDKN2A identified from CARSs are considered potential prognostic biomarkers for WHO 2/3 glioma. The clinical prognosis model based on cuproptosis is expected to provide an effective reference for the diagnosis and treatment of clinical WHO 2/3 glioma patients.

    Abstract: Temperature and water potentials are considered the most critical environmental factors in seed germinability and subsequent seedling establishment. The thermal and water requirements for germination are species-specific and vary with the environment in which seeds mature from the maternal plants. Pedicularis kansuensis is a root hemiparasitic weed that grows extensively in the Qinghai–Tibet Plateau’s degraded grasslands and has seriously harmed the grasslands ecosystem and its utilization. Information about temperatures and water thresholds in P . kansuensis seed germination among different populations is useful to predicting and managing the weed’s distribution in degraded grasslands. The present study evaluated the effects of temperature and water potentials on P . kansuensis seed germination in cool and warm habitats, based on thermal time and hydrotime models. The results indicate that seeds from cool habitats have a higher base temperature than those from warm habitats, while there is no detectable difference in optimum and ceiling temperatures between habitats. Seed germination in response to water potential differed among the five studied populations. There was a negative correlation between the seed populations’ base water potential for 50% ( Ψ b(50) ) germination and their hydrotime constant ( θ H ). The thermal time and hydrotime models were good predictors of five populations’ germination time in response to temperature and water potentials. Consequently, future studies should consider the effects of maternal environmental conditions on seed germination when seeking effective strategies for controlling hemiparasitic weeds in alpine regions.

    Abstract: The epoxy resin is the main solid insulating material for electrical equipment. PD (partial discharge) is an important factor leading to epoxy resin aging or failure. Fault detection technology based on audible acoustic imaging has the advantages of real-time monitoring, visibility, and flexibility and is gradually being used in the field of electrical equipment insulation state monitoring. However, the audible voiceprint features of epoxy resins with typical PD defects are not clear, which is not conducive to effective detection and fault diagnosis. Therefore, in this study, the study of audible voiceprint features of epoxy resins with a metal burr, pollution, metal foreign matters, and surface discharge defects was carried out. The discharge audible signals were obtained, and their time and frequency domain features were analyzed. The results show that the discharge audible signals of four PD defects can reflect the occurrence of PD simultaneously with pulse current signals. But the characteristics are obviously different in the time domain, which are embodied in listening perception, persistence, amplitude, and phase. In the frequency domain, the frequency spectra of discharge audible signals of four defects are mainly 5–10 and 13–24 kHz. Furthermore, the three feature vectors of frequency spectrum complexity, high-frequency energy factor, and maximum frequency point are extracted, which can well distinguish the discharge audible signals of four PD defects.

    Abstract: Objective: Butyrate, a short-chain fatty acid (SCFA) produced by the intestinal microbiota, plays a protective role in cardiovascular diseases (CVDs), but the mechanisms involved in this process remain unelucidated. We aimed to explore the effect of butyrate on myocardial ischemia/reperfusion (I/R) injury through the gut-brain neural circuit. Methods: Rats were randomly divided into four groups: sham group (sham), I/R group (I/R), I/R+ butyrate group (butyrate), and I/R+ butyrate+ vagotomy group (vagotomy). The rats were treated with sodium butyrate for 4 weeks, and the gut-brain neural circuit was investigated by subdiaphragmatic vagotomy. Results: Butyrate treatment significantly reduced the infarct size and decreased the expression of creatine kinase (CK), creatine kinase myocardial isoenzyme (CK-MB), and lactate dehydrogenase (LDH) compared with the values found for the I/R group. In addition, the I/R-induced increases in inflammation, oxidative stress, and apoptosis were attenuated by butyrate. However, the above-mentioned protective effects were diminished by subdiaphragmatic vagotomy. The RNA sequencing results also revealed that the butyrate-induced protective changes at the cardiac transcription level were reversed by vagotomy. An analysis of the heart rate variability (HRV) and the detection of norepinephrine (NE) showed that butyrate significantly inhibited the I/R-induced autonomic imbalance, but this inhibition was not observed in the vagotomy group. Butyrate treatment also suppressed the neural activity of the paraventricular nucleus (PVN) and superior cervical ganglion (SCG), and both of these effects were lost after vagotomy. Conclusions: Butyrate treatment significantly improves myocardial I/R injury via a gut-brain neural circuit, and this cardioprotective effect is likely mediated by suppression of the sympathetic nervous system.

    Abstract: Ischemic stroke is an acute cerebrovascular disease characterized by sudden interruption of blood flow in a certain part of the brain, leading to serious disability and death. At present, treatment methods for ischemic stroke are limited to thrombolysis or thrombus removal, but the treatment window is very narrow. However, recovery of cerebral blood circulation further causes cerebral ischemia/reperfusion injury (CIRI). The endoplasmic reticulum (ER) plays an important role in protein secretion, membrane protein folding, transportation, and maintenance of intracellular calcium homeostasis. Endoplasmic reticulum stress (ERS) plays a crucial role in cerebral ischemia pathophysiology. Mild ERS helps improve cell tolerance and restore cell homeostasis; however, excessive or long-term ERS causes apoptotic pathway activation. Specifically, the protein kinase R-like endoplasmic reticulum kinase (PERK), activating transcription factor 6 (ATF6), and inositol-requiring enzyme 1 (IRE1) pathways are significantly activated following initiation of the unfolded protein response (UPR). CIRI-induced apoptosis leads to nerve cell death, which ultimately aggravates neurological deficits in patients. Therefore, it is necessary and important to comprehensively explore the mechanism of ERS in CIRI to identify methods for preserving brain cells and neuronal function after ischemia.

    Abstract: With the widespread use of renewable energy worldwide, the impact of its randomness and volatility on the grid is increasing. To promote the consumption of renewable energy, the traditional grid is being transformed into a complex grid with integrated source–grid–load–storage. Since the complex grid has the characteristics of source–grid–load–storage interaction, the traditional grid investment decision method will no longer be applicable. First, this study proposes the unilateral indexes of source, grid, load, and storage in complex grids and the interactive indexes considering grid–source interaction, load–grid interaction, source–load interaction, source–storage interaction, load–storage interaction, and grid–storage interaction are proposed to establish the investment decision system. Then, a hesitance fuzzy linguistic term set combined with regret theory is used to calculate the specific values of the subjectivity index, taking into full consideration the regret avoidance and loss avoidance psychology of investors. In order to comprehensively consider the index preference of investors and the objectivity of weight assignment, a combined weighting method based on the analytic network process (ANP) and entropy weight method (EWM) is obtained according to the game theory method. Finally, using a grid in a region of southwest China as an example, the results demonstrate that the construction order obtained in this study can prioritize the projects with the largest comprehensive benefits while considering the subjective preferences of decision-makers and the objectivity of the indexes.

    Abstract: Helicobacter pylori causes gastric infections in more than half of the world's population. The bacterium's survival in the stomach is mediated by the abundant production of urease to enable acid acclimation. In this study, our transcriptomic analysis demonstrated that the expression of urease structural proteins, UreA and UreB, is induced by the autoinducer AI-2 in H. pylori . We also found that the orphan response regulator HP1021 is downregulated by AI-2, resulting in the induction of urease expression. HP1021 represses the expression of urease by directly binding to the promoter region of ureAB , ranging from −47 to +3 with respect to the transcriptional start site. The study findings suggest that quorum sensing via AI-2 enhances acid acclimation when bacterial density increases, and might enable bacterial dispersal to other sites when entering gastric acid.

    Abstract: The recent World Endoscopy Organization (WEO) guidelines now recognize precursor lesions of colorectal cancer (CRC) as legitimate screening targets. However, an optimal screening method for detecting advanced adenoma (AA), a significant precursor lesion, remains elusive. Methods We employed five machine learning methods, using clinical and laboratory data, to develop and validate a diagnostic model for identifying patients with AA (569 AAs vs. 3228 controls with normal colonoscopy). The best-performing model was selected based on sensitivity and specificity assessments. Its performance in recognizing adenoma-carcinoma sequence was evaluated in line with guidelines, and adjustable thresholds were established. For comparison, the Fecal Occult Blood Test (FOBT) was also selected. Results The XGBoost model demonstrated superior performance in identifying AA, with a sensitivity of 70.8% and a specificity of 83.4%. It successfully detected 42.7% of non-advanced adenoma (NAA) and 80.1% of CRC. The model-transformed risk assessment scale provided diagnostic performance at different positivity thresholds. Compared to FOBT, the XGBoost model better identified AA and NAA, however, was less effective in CRC. Conclusion The XGBoost model, compared to FOBT, offers improved accuracy in identifying AA patients. While it may not meet the recommendations of some organizations, it provides value for individuals who are unable to use FOBT for various reasons.

    Abstract: There is a growing global concern regarding the rise of antimicrobial resistance among Ureaplasma spp. isolates. However, studies on the antimicrobial susceptibility profiles, resistance mechanisms, and clonality of Ureaplasma spp. clinical isolates are still limited and cover only some geographic regions. Firstly, Ureaplasma species from the urogenital tracts of patients in Shanghai, China, were isolated by using the culture medium (A8 and 10B broth), and identified the genotype by polymerase chain reaction (PCR). Secondly, the antimicrobial susceptibility tests were determined by using broth microdilution assay. Then, the resistance genetic determinants to fluoroquinolones (FQs), macrolides, and tetracyclines were investigated through PCR/DNA sequencing. Finally, the molecular epidemiology of Ureaplasma species was studied by multilocus sequence typing (MLST). Among 258 isolates, Ureaplasma parvum (UPA) and Ureaplasma urealyticum (UUR) were found in 226 (87.60%) and 32 (12.40%) isolates, respectively. The minimum inhibitory concentrations (MICs) of 258 Ureaplasma spp. strains ranged from 0.015 to 64μg/ml for all 11 kinds of antimicrobials. Regardless of species, the isolates were most sensitive to AZI (1.94%), JOS (3.49%), and CLA (4.23%). Among them, there were 39 (15.12%) multidrug-resistant (MDR) strains, including 32 UPA isolates. The resistance rates of UPA to CIP (91.59%), and ROX (36.28%) were significantly higher than those of UUR. Twenty six FQ-resistant isolates had amino acid substitutions in gyrA and in parC (Ser83Leu). Mutations were detected in genes encoding ribosomal proteins L4 (Thr84Ile) and L22 (Ser81Pro) in macrolide-resistant isolates. Tet (M) was found in four UPA isolates. These mutations were mainly found in UPA isolates. Sequence type 1 (ST1) was the predominant ST, which contained 18 isolates. In conclusion, this study showed a higher resistance rate (especially to ROX and CIP), higher substitution rate, and higher MDR rate among UPA strains. The most active antimicrobial agents were AZI, JOS, and CLA. Identifying UPA or UUR in clinical isolates could help clinicians to choose appropriate drugs for treatment. The main resistance mechanisms may involve gene substitution of Ser83Leu in parC and Ser81Pro in L22. ST1 was the predominant ST of Ureaplasma isolates with MDR to FQs and macrolides in Shanghai, China.
