    Abstract: Auxin response factors (ARFs) are transcription factors that play important roles in plants. ARF2 is a member of the ARF family and participates in many plant growth and developmental processes. However, the role of ARF2 in strawberry fruit quality remains unclear. In this study, FveARF2 was isolated from the woodland strawberry ‘Ruegen’ using reverse transcription-polymerase chain reaction (RT-PCR), which showed that FveARF2 expression levels were higher in the stem than in other organs of the ‘Ruegen’ strawberry. Moreover, FaARF2 was higher in the white fruit stage of cultivated strawberry fruit than in other stage. Subcellular localization analysis showed that FveARF2 is located in the nucleus, while transcriptional activation assays showed that FveARF2 inhibited transcription in yeast. Silencing FveARF2 in cultivated strawberry fruit revealed earlier coloration and higher soluble solid, sugar, and anthocyanin content in the transgenic fruit than in the control fruit, overexpression of FveARF2 in strawberry fruit delayed ripening and lower soluble solid, sugar, and anthocyanin content compared to the control fruit. Gene expression analysis indicated that the transcription levels of the fruit ripening genes FaSUT1 , FaOMT , and FaCHS increased in FveARF2-RNAi fruit and decreased in FveARF2-OE fruit, when compared with the control. Furthermore, yeast one-hybrid (Y1H) and GUS activity experiments showed that FveARF2 can directly bind to the AuxRE (TGTCTC) element in the FaSUT1 , FaOMT , and FaCHS promoters in vitro and in vivo . Potassium ion supplementation improved the quality of strawberry fruit, while silencing FveARF2 increased potassium ion content in transgenic fruit. The Y1H and GUS activity experiments also confirmed that FveARF2 could directly bind to the promoter of FveKT12 , a potassium transporter gene, and inhibited its expression. Taken together, we found that FveARF2 can negatively regulate strawberry fruit ripening and quality, which provides new insight for further study of the molecular mechanism of strawberry fruit ripening.

    Abstract: Nowadays, diarrheic shellfish toxicity (DSP) toxin and microplastics (MPs) are commonly found in coastal waters worldwide. Due to their widespread use, their persistence and toxicity, they may induce adverse effects on Mytilus coruscus . However, the underlying toxic mechanisms of DSP and MPs on M. coruscus remain unclear. This study explored the physiological index and transcriptome change of the digestive gland of adult M. coruscus exposed for 3 days to polystyrene (PS) MPs (0.2 mg/L, 90-100 μm) and Prorocentrum lima alone or in combination. The results showed that the CAT activity and MDA content significantly increased, respiration rate and feeding rate significantly decreased. The combination of MPs and P. lima caused more structural damage to the rough surface endoplasmic reticulum and mitochondria in the digestive glands of M. coruscus . The transcriptome analysis showed that 485 and 220 genes were up- and down-regulated, respectively, after exposure to P. lima ; 1,989 up-regulated DEGs and 1,098 down-regulated DEGs were identified after exposure to MP treatment, and 1,004 up-regulated DEGs and 664 down-regulated DEGs were identified after exposure to the combination of P. lima and MPs. The DEGs were mainly enriched in the lysosome, mRNA surveillance pathway, carbon metabolism, the mTOR signaling pathway, the complement and coagulation cascades, and the TNF signaling pathway. The MP, P. lima exposure mainly induced the expression of RNA-binding protein musashi, serine/arginine repetitive matrix protein 1, low affinity immunoglobulin epsilon Fc receptor, toll-like receptor 2, caspase 7, calmodulin, E3 ubiquitin-protein ligase, serine/threonine-protein kinase PRP4, glutathione S-transferase, and heat shock 70 kDa protein. MPs and P. lima poison mainly influence the expression of RNA transport, immune related gene, apoptosis, signal related gene, and antioxidant gene change. The combination of MPs and P. lima has a synergistic toxic effect. This study provides a new insights into its physiological and molecular responses of M. coruscus to MPs and P. lima toxic exposure.

    Abstract: Understanding the traits related to species colonization and invasion, is a key question for both pest management and evolution. One of the key components is flight, which has been measured for a number of insect species through radar and tethered flight mill systems, but a general understanding of insect flight at a community level is lacking. In this study, we used flight mill experiments to quantify flight abilities of moth species, and simulation experiments to study which moths in mainland China have the potential for cross-island dispersal. We found that moths from superfamily Geometroidea (family Geometridae) have the weakest flight ability among the seven Lepidoptera superfamilies, which is characterized by the shortest longest single flight (LSF), the shortest time corresponding to the longest single flight ( T LSF ) ( timecorrespondingtothelongestsingleflight ), the lowest total distance flown (TDF), and the lowest average speed during the flight ( V TDF ). Surprisingly, the family Pyralidae (superfamily Pyraloidea ) has the highest flight endurance of all 186 species of 12 families in this study, which is unexpected, given its small size and morphological traits yet it shows the longest LSF and T LSF . The comparison between species common to mainland and islands shows that flight distance (LSF) may be more important for species spread than flight speed. The results of mainland-island simulations show that when P (LSF & gt;CD) (the proportion of individuals whose LSF is greater than the closest distance (CD) between mainland and island to the total number of individuals in the population) is less than 0.004, it is difficult for moth species to disperse to across islands without relying on external factors such as airflow. Over extended periods, with the immigration of species with strong flight abilities, islands are more likely to recruit species with stronger flight abilities.

    Abstract: In sheep industry, hypothermia caused by insufficient brown adipose tissue (BAT) deposits is one of the major causes of lamb deaths. Enhancing the formation and function of BAT in neonatal lamb increases thermogenesis and hence reduces economic losses. The aim of the present study was to explore the effect and mechanism of melatonin on sheep brown adipocyte formation and function. Sheep brown adipocyte precursor cells (SBACs) isolated from perirenal BAT were treated with melatonin (1 and 10 nM). The SBACs subjected to melatonin exhibited a decreased proliferation ability, accompanied by down-regulated proliferating cell nuclear antigen, cyclin D1, and CDK4 protein contents in a melatonin dose-dependent manner. Melatonin promoted brown adipocyte formation and induced the expression of brown adipogenic markers, including uncoupling protein 1 and PR domain-containing 16 during differentiation of SBAC. Moreover, the AMP-activated protein kinase α1 (AMPKα1) activity was positively correlated with brown adipocyte formation potential. Importantly, melatonin effectively activated AMPKα1. Furthermore, promotional effects of melatonin were abolished by AMPKα1 knockout, suggesting the involvement of AMPKα1 in this process. Collectively, these results suggested that melatonin enhanced brown adipocyte formation in SBACs in vitro through activation of AMPKα1.

    Abstract: The brain compensation mechanism in postoperative delirium (POD) has not been reported. We uncovered the mechanism by exploring the association between POD and glioma grades, and the relationship between preoperative brain structural and functional compensation with POD in patients with frontal glioma. Methods A total of 335 adult patients with glioma were included. The multivariable analysis examined the association between tumor grade and POD. Then, 20 patients with left frontal lobe glioma who had presurgical structural and functional MRI data and Montreal Cognitive Assessment (MoCA) in this cohort were analyzed. We measured the gray matter volume (GMV) and functional connectivity (FC) in patients with ( n = 8) and without ( n = 12) POD and healthy controls (HCs, n = 29) to detect the correlation between the structural and functional alteration and POD. Results The incidence of POD was 37.3%. Multivariable regression revealed that high-grade glioma had approximately six times the odds of POD. Neuroimaging data showed that compared with HC, the patients with left frontal lobe glioma showed significantly increased GMV of the right dorsal lateral prefrontal cortex (DLPFC) in the non-POD group and decreased GMV of right DLPFC in the POD group, and the POD group exhibited significantly decreased FC of right DLPFC, and the non-POD group showed the increasing tendency. Partial correlation analysis showed that GMV in contralesional DLPFC were positively correlated with preoperative neurocognition, and the GMV and FC in contralesional DLPFC were negatively correlated with POD. Conclusions Our findings suggested that insufficient compensation for injured brain regions involving cognition might be more vulnerable to suffering from POD.

    Abstract: Central nervous system (CNS) infection is one of the most serious complications after neurosurgery. Traditional clinical methods are difficult to diagnose the pathogen of intracranial infection. Due to recent advances in genomic approaches, especially sequencing technologies, metagenomic next-generation sequencing (mNGS) has been applied in many research and clinical settings. Case presentation Here, we report a case of CNS infection with Acinetobacter baumannii in a 15-year-old woman, who previously underwent surgery for recurrence of ependymoma in the fourth ventricle. On the eleventh postoperative day, the patient had a high fever and leukocytosis in the cerebrospinal fluid (CSF). mNGS using CSF rapidly and accurately identified the causative pathogen as A. baumannii with carbapenem resistance genes blaOXA-23 and blaOXA-51 , which were confirmed by subsequent culture and susceptibility tests within 5 days. During the disease, mNGS, culture, and drug susceptibility testing were continued to monitor changes in pathogenic bacteria and adjust medication. At present, there are no case reports on to the use of mNGS for detecting pathogens in postoperative infection with ependymoma and guide medication. Conclusion mNGS has great advantages in pathogen identification and even pathogen resistance prediction. Multiple mNGS examinations during the course of the disease play an important role in the dynamic monitoring of pathogens.

    Abstract: Data concerning the epidemiology of sepsis in critically ill post-craniotomy patients are scarce. This study aimed to assess the incidence, risk factors, and outcomes of sepsis in this population. Methods This was a single-center prospective cohort study. Post-craniotomy patients admitted to the intensive care unit (ICU) were screened daily for the presence of infection and sepsis. Results Of the 900 included patients, 300 developed sepsis. The cumulative incidence of sepsis was 33.3% [95% confidence interval (CI), 30.2–36.4%]. Advanced age, male, hypertension, trauma, postoperative intracranial complications, and lower Glasgow Coma Scale (GCS) on the first postoperative day were independent risk factors of sepsis. Septic patients had higher hospital mortality (13.7 vs. 8.3%, P = 0.012), longer ICU length of stay (LOS) (14 vs. 4 days, P & lt; 0.001), longer hospital LOS (31 vs. 19 days, P & lt; 0.001), and higher total medical cost (CNY 138,394 vs. 75,918, P & lt; 0.001) than patients without sepsis. Conclusion Sepsis is a frequent complication in critically ill post-craniotomy patients. Advanced age, male, hypertension, trauma, postoperative intracranial complications, and lower GCS on the first postoperative day were independent risk factors of sepsis.
