    Abstract: Organophosphate flame retardants (OPFRs) are widely distributed in the environment and their metabolites are observed in urine, but little is known regarding OPFRs in a broad-spectrum young population from newborns to those aged 18 years. Objectives Investigate urinary levels of OPFRs and OPFR metabolites in Taiwanese infants, young children, schoolchildren, and adolescents within the general population. Methods Different age groups of subjects (n=136) were recruited from southern Taiwan to detect 10 OPFR metabolites in urine samples. Associations between urinary OPFRs and their corresponding metabolites and potential health status were also examined. Results The mean level of urinary Σ 10 OPFR in this broad-spectrum young population is 2.25 μg/L (standard deviation (SD) of 1.91 μg/L). Σ 10 OPFR metabolites in urine are 3.25 ± 2.84, 3.06 ± 2.21, 1.75 ± 1.10, and 2.32 ± 2.29 μg/L in the age groups comprising of newborns, 1-5 year-olds, 6-10 year-olds, and 11-18 year-olds, respectively, and borderline significant differences were found in the different age groups ( p =0.125). The OPFR metabolites of TCEP, BCEP, DPHP, TBEP, DBEP, and BDCPP predominate in urine and comprise more than 90% of the total. TBEP was highly correlated with DBEP in this population (r=0.845, p & lt;0.001). The estimated daily intake (EDI) of Σ 5 OPFRs (TDCPP, TCEP, TBEP, TNBP, and TPHP) was 2,230, 461, 130, and 184 ng/kg bw/day for newborns, 1-5 yr children, 6-10 yr children, and 11-17 yr adolescents, respectively. The EDI of Σ 5 OPFRs for newborns was 4.83-17.2 times higher than the other age groups. Urinary OPFR metabolites are significantly correlated with birth length and chest circumference in newborns. Conclusion To our knowledge, this is the first investigation of urinary OPFR metabolite levels in a broad-spectrum young population. There tended to be higher exposure rates in both newborns and pre-schoolers, though little is known about their exposure levels or factors leading to exposure in the young population. Further studies should clarify the exposure levels and factor relationships.

    Abstract: The underlying mechanisms of thymocyte development and lineage determination remain incompletely understood, and the emerging evidences demonstrated that RNA binding proteins (RBPs) are deeply involved in governing T cell fate in thymus. Serine/arginine-rich splicing factor 1 (SRSF1), as a classical splicing factor, is a pivotal RBP for gene expression in various biological processes. Our recent study demonstrated that SRSF1 plays essential roles in the development of late thymocytes by modulating the T cell regulatory gene networks post-transcriptionally, which are critical in response to type I interferon signaling for supporting thymocyte maturation. Here, we report SRSF1 also contributes to the determination of the CD8 + T cell fate. By specific ablation of SRSF1 in CD4 + CD8 + double positive (DP) thymocytes, we found that SRSF1 deficiency impaired the maturation of late thymocytes and diminished the output of both CD4 + and CD8 + single positive T cells. Interestingly, the ratio of mature CD4 + to CD8 + cells was notably altered and more severe defects were exhibited in CD8 + lineage than those in CD4 + lineage, reflecting the specific function of SRSF1 in CD8 + T cell fate decision. Mechanistically, SRSF1-deficient cells downregulate their expression of Runx3 , which is a crucial transcriptional regulator in sustaining CD8 + single positive (SP) thymocyte development and lineage choice. Moreover, forced expression of Runx3 partially rectified the defects in SRSF1-deficient CD8 + thymocyte maturation. Thus, our data uncovered the previous unknown role of SRSF1 in establishment of CD8 + cell identity.

    Abstract: Coronary microembolization (CME) commonly develops as a complication after percutaneous coronary intervention (PCI), and associated inflammation is a leading driver of myocardial damage. Cardiomyocyte loss in the context of ischemic myocardial disease has been linked to inflammatory pyroptotic cell death. Additionally, miR-200a-3p dysregulation has been linked to myocardial ischemia-reperfusion and many other pathological conditions. However, how miR-200a-3p impacts cardiomyocyte pyroptosis in the context of CME remains to be assessed. Herein, a rat model of CME was established via the injection of microembolic spheres into the left ventricle. When myocardial tissue samples from these rats were analyzed, miR-200a-3p levels were markedly decreased, whereas thioredoxin-interacting protein (TXNIP) levels were increased. The ability of miR-200a-3p to directly target TXNIP and to control its expression was confirmed via dual-luciferase reporter assay. Adeno-associated virus serotype 9-pre-miR-200a-3p (AAV-miR-200a-3p) construct transfection was then employed as a means of upregulating this miRNA in CME model rats. Subsequent assays, including echocardiography, enzyme-linked immunosorbent assays (ELISAs), hematoxylin-eosin (H & amp;E) staining, hematoxylin-basic fuchsin-picric acid (HBFP) staining, TdT-mediated dUTP nick-end labeling (TUNEL) staining, immunofluorescence staining, quantitative real-time polymerase chain reaction (qRT-PCR), and Western blotting revealed that miR-200a-3p overexpression inhibited cardiomyocyte pyroptosis and alleviated CME-induced myocardial injury by inhibiting the TXNIP/NOD-like receptor family pyrin domain-containing 3 (NLRP3) pathway. The ability of miR-200a-3p to protect against CME-induced myocardial injury thus highlights a novel approach to preventing or treating such myocardial damage in clinical settings.

    Abstract: To identify pollutant origins and their potential sources from either long-range transported (LRT) or local emissions, the Boron (B) concentrations and the B isotopic compositions (δ 11 B) in the water-soluble fraction of aerosols were determined. These aerosols were collected from an islet offshore in northeastern Taiwan, Peng Chia Yu (PCY), a non-residential islet, from January 1998 to March 2000. This islet may be influenced by various pollutants or chemical transport during monsoon periods, but suffers minimal human perturbation locally. The B in the specimens falls to 0.3–1.63 ng m −3 during the SW monsoon seasons when compared to 0.46–2.56 ng m −3 in the NE monsoon. However, the δ 11 B results show no clear variations in both monsoon seasons (10.7–24.3‰), regardless of differences in air mass origin. A two end-member mixing scenario is proposed to explain our observations using the obtained δ 11 B and 1/[B] results. The ocean endmember is characterized by high 1/[B] and high δ 11 B; while other endmember is from continental endmember or anthropogenic contributions that are characterized by of 1/[B] and lowδ 11 B (δ 11 B & lt; 10‰). Based on these chemical and isotopic results, we found aerosol emissions in northern Taiwan are characterized as low B with nearly constant δ 11 B, when compared with long-range transported continental endmembers derived from nearby regions. This study provides preliminary B and δ 11 B levels in aerosols derived from LRT/local emissions and discusses potential monsoonal effects on aerosols offshore of NE Taiwan.

    Abstract: Boron (B) and B isotopic compositions (δ 11 B) in biogenic carbonates are useful proxies for pH reconstruction in the ocean. However, high-resolution archives are scarce due to associated sampling and analytical difficulty. In this study, a modern long-lived massive coral skeleton ( Porites lobata ) from Lanyu Islet off southeast Taiwan was drilled and used for high-resolution major/trace element analyses, including trace elements B and δ 11 B, as well as oxygen and carbon isotopes, to investigate the associated environmental changes during 1991–1997. To avoid complicated biological influence, the top-most tissue layer was excluded in this study. The coralline records show a clear temporal trend in metal/Ca-based sea surface temperatures (SSTs) on annual and monthly timescales. In particular, the Mg/Ca-SSTs, the most sensitive temperature proxy at the site, show a significant warming trend (+0.2°C year −1 ) during the study period. On the other hand, subtle changes in the annual δ 11 B record were identified, corresponding to ~0.2 pH unit, which is comparable with other coral records in the Pacific, e.g., the South China Sea (SCS), Guam Island, Flinders, and Arlington Reef, as well as the in-situ seawater pH measurement at Hawaii station. This corresponds to an acidification rate of ~0.25 pH unit 100 year −1 , similar to other coralline data, in-situ pH/pCO 2 measurement, or model predictions, and emphasizes the importance of ocean acidification due to anthropogenic activities. Combined with the Mg/Ca-SST, the intra-annual data show a clear seasonal cycle with higher pH in winter, consistent with the pCO 2 at the oceanic surface. These chemical and isotopic results in corals conclude that marine biogenic carbonates are informative for oceanic pH reconstruction and can provide new insights into the relationships between climate changes and environmental responses on the coast of Taiwan.

    Abstract: The current study assessed the ecological–health risks of potentially toxic arsenic (As) and antimony (Sb) in the vegetable Centella asiatica , topsoils, and mangrove sediments sampled from Peninsular Malaysia. The As concentrations ranged from 0.21 to 4.33, 0.18 to 1.83, and 1.32 to 20.8 mg/kg dry weight, for the leaves, stems, and roots of the vegetable, respectively. The ranges of Sb concentrations were 0.31–0.62, 0.12–0.35, and 0.64–1.61 mg/kg dry weight, for leaves, stems, and roots of the vegetable, respectively. The children’s target hazard quotient (THQ) values indicated no non-carcinogenic risks of As and Sb in both leaves and stems, although children’s THQ values were higher than those in adults. The calculated values of estimated weekly intake were lower than established provisional tolerable weekly intake of As and Sb for both children and adult consumers. The carcinogenic risk (CR) values of As for children’s intake of leaves and stems of vegetables showed more public concern than those of adults. The levels of Sb and As in the topsoils were generally higher (although not significantly) than those in the mangrove sediments, resulting in a higher geoaccumulation index, contamination factor and ecological risk, hazard index, THQ, and CR values. This indicated that the anthropogenic sources of Sb and As originated from the land-based activities before reaching the mangrove near the coast. The CR of As signifies a dire need for comprehensive ecological–health risks exposure studies, as dietary intake involves more than just vegetable consumption. Therefore, risk management for As and Sb in Malaysia is highly recommended. The present findings of the ecological–health risks of As and Sb based on 2010–2012 samples can be used as an important baseline for future reference and comparison.

    Abstract: The South China Sea (SCS) is the largest marginal sea in the western Pacific, fed by large Asian rivers, characterized by substantial volumes of sediment derived from surrounding Asian continents and islands. This study utilizes the rare earth elements (REEs), Sr isotopes ( 87 Sr/ 86 Sr), and Nd isotopes (Ɛ Nd ) in detritus fractions of MD05-2901 sediments in northwestern SCS, to evaluate the relative contribution among the Mekong River, the Red River, the Pearl River, and rivers in SW Taiwan over the last 32 ka. This deep-sea core is located off the coast of central Vietnam, an upwelling region with only minor continental inputs, and this study is important for a better understanding of the main sources of terrigenous material and their temporal variations in the region. In addition, potential particle exchange is examined using the same proxies in the Fe-Mn oxyhydroxide fractions. The obtained records are systematically compared with the available climatic archives in the region. The REEs in the Fe-Mn oxyhydroxides support the argument of water mass signature separation in bulk sediments by chemical leaching procedures. These 87 Sr/ 86 Sr in the Fe-Mn oxyhydroxides phase fall in a range between 0.709262 and 0.709313, deviating significantly from the modern seawater. Ɛ Nd values vary & gt;3 Ɛ , being more radiogenic in the Fe-Mn oxyhydroxides, where the lowest value occurred in the Bølling/Allerød (B/A) and higher values occurred during the Younger Dryas (YD). The latter implies increased contribution of loess dusts or Taiwanese rivers under dry/cold climatic conditions, compared with the situation in modern times. The Red River and the Mekong River together may have contributed & gt;60% during the Last Glacial Maximum (LGM) and YD. In contrast, the Pearl River components have increased sharply to ~60% during the warm B/A. These results demonstrate that the MD05-2901 sediments sensitively record changes in weathering intensity on land and transport pathways in the coastal regions in the Holocene.

    Abstract: Mud volcano is one of the most important conduits for deep seated materials to migrate upward in sedimentary basins, convergent margins, and subduction zones. Understanding their temporal and spatial characteristics and variations provides us the important information on fluid sources and chemical compositions at depth. Mud volcano Lei-Gong-Huo (MV LGH) is a unique mud volcano, which is located on the mélange formation lying on the andesitic volcanic arc. Fluids emitted from 46 mud pools in MV LGH in eastern Taiwan were sampled and their major trace constitutes as well as H, O, and Sr isotopes ( 87 Sr/ 86 Sr and δ 88 Sr) were measured. Major constitutes of the fluids are Cl − , Na, and Ca. Compared with seawater, LGH fluids have lower Cl − , δD, δ 18 O, Na/Cl, K/Cl, and Mg/Cl but higher Ca/Cl ratios, indicating water–rock interaction of igneous rock and the ancient seawater at the source region. This interpretation is further supported by Sr isotopes, which show low value of 87 Sr/ 86 Sr ratio down to 0.70708. The result of spatial distribution showing strong negative correlation between Na and Ca concentration as well as Ca and 87 Sr/ 86 Sr ratios indicates that two end-member mixing is the major chemical characteristic. The fluids interacting with igneous rock carry high Ca, high δ 88 Sr, low Na, and low 87 Sr/ 86 Sr ratio, while those interacting with sedimentary rock carry low Ca, low δ 88 Sr, high Na, and high 87 Sr/ 86 Sr ratio. The source from the igneous region dominates the eastern and southeastern parts of the mud pools while sedimentary source dominates the western and northwestern parts. Most mud pools show mixing behavior between the two sources. Some of the sedimentary-dominated mud pools reveal existence of residual ancient water as indicated by 87 Sr/ 86 Sr. The major factor to fractionate the stable Sr isotopes in LGH waters is the source lithology. In summary, fluids emitted by mud pools in LGH originate from two sources, which are water–rock interactions of igneous rock with the ancient seawater from the east and sedimentary rock from the west at depth, resulting from the complex geologic background of mélange formation.

    Abstract: Acute exacerbation of chronic obstructive pulmonary disease (AECOPD) is associated with high mortality rates. Viral and bacterial coinfection is the primary cause of AECOPD. How coinfection with these microbes influences host inflammatory response and the gut microbiota composition is not entirely understood. Methods We developed a mouse model of AECOPD by cigarette smoke exposure and sequential infection with influenza H1N1 virus and non-typeable Haemophilus influenzae (NTHi). Viral and bacterial titer was determined using MDCK cells and chocolate agar plates, respectively. The levels of cytokines, adhesion molecules, and inflammatory cells in the lungs were measured using Bio-Plex and flow cytometry assays. Gut microbiota was analyzed using 16S rRNA gene sequencing. Correlations between cytokines and gut microbiota were determined using Spearman’s rank correlation coefficient test. Results Coinfection with H1N1 and NTHi resulted in more severe lung injury, higher mortality, declined lung function in COPD mice. H1N1 enhanced NTHi growth in the lungs, but NTHi had no effect on H1N1. In addition, coinfection increased the levels of cytokines and adhesion molecules, as well as immune cells including total and M1 macrophages, neutrophils, monocytes, NK cells, and CD4 + T cells. In contrast, alveolar macrophages were depleted. Furthermore, coinfection caused a decline in the diversity of gut bacteria. Muribaculaceae , Lactobacillus , Akkermansia , Lachnospiraceae , and Rikenella were further found to be negatively correlated with cytokine levels, whereas Bacteroides was positively correlated. Conclusion Coinfection with H1N1 and NTHi causes a deterioration in COPD mice due to increased lung inflammation, which is correlated with dysbiosis of the gut microbiota.
