    Abstract: Three new humulane-type sesquiterpenoids, penirolide A ( 1 ), penirolide B ( 2 ), and 10-acetyl-phomanoxide ( 3 ), together with three known compounds aurasperone A ( 4 ), pughiinin A ( 5 ), and cyclo( l -Leu- l -Phe) ( 6 ) were isolated from the endophytic fungus Penicillium sp . derived from the leaves of Carica papaya L. Their structures including their absolute configurations were determined based on the analysis of NMR and HRESIMS spectra, NMR chemical shifts, and ECD calculations. Compounds 2 , 3 , 5 , and 6 significantly inhibited glucagon-induced hepatic glucose production, with EC 50 values of 33.3, 36.1, 18.8, and 32.1 μM, respectively. Further study revealed that compounds 2 , 3 , 5 , and 6 inhibited hepatic glucose production by suppression of glucagon-induced cAMP accumulation.

    Abstract: Honeybees ( Apis mellifera ) can be exposed via numerous potential pathways to ambient nanoparticles (NPs), including rare earth oxide (REO) NPs that are increasingly used and released into the environment. Gut microorganisms are pivotal in mediating honeybee health, but how REO NPs may affect honeybee health and gut microbiota remains poorly understood. To address this knowledge gap, honeybees were fed pollen and sucrose syrup containing 0, 1, 10, 100, and 1000mgkg −1 of nano-La 2 O 3 for 12days. Nano-La 2 O 3 exerted detrimental effects on honeybee physiology, as reflected by dose-dependent adverse effects of nano-La 2 O 3 on survival, pollen consumption, and body weight ( p & lt;0.05). Nano-La 2 O 3 caused the dysbiosis of honeybee gut bacterial communities, as evidenced by the change of gut bacterial community composition, the enrichment of pathogenic Serratia and Frischella , and the alteration of digestion-related taxa Bombella ( p & lt;0.05). There were significant correlations between honeybee physiological parameters and the relative abundances of pathogenic Serratia and Frischella ( p & lt;0.05), underscoring linkages between honeybee health and gut bacterial communities. Taken together, this study demonstrates that nano-La 2 O 3 can cause detrimental effects on honeybee health, potentially by disordering gut bacterial communities. This study thus reveals a previously overlooked effect of nano-La 2 O 3 on the ecologically and economically important honeybee species Apis mellifera .

    Abstract: Vespa velutina nigrithorax and Vespa velutina auraria are two subspecies of Vespa velutina Lepeletier. V. velutina preys managed honey bees, other pollinators, and insects. However, the Vespa midgut microbiota of three forms, namely queens, workers, and males have not been reported, thus the objective of this study was to analyze the midgut bacterial diversity of the three forms of V. v. nigrithorax and V. v. auraria . Our results showed that Proteobacteria , Firmicutes , Bacteroidetes , Tenericutes , and Actinobacteria were the most abundant phyla, and Lactobacillus (17.21%) and Sphingomonas (11.39%) were the most abundant genera in the midgut of V. v. nigrithorax and V. v. auraria . We found that the midgut bacterial compositions of the V. velutina males were special, in terms of richness and diversity of bacterial communities, as well as the content of lactic acid bacteria. By comparing the gut bacterial compositions of Vespa from different regions (Japan, South Korea, Italy, and China), it was discovered that the gut bacterial compositions were very similar at the phylum and class level, and Gammaproteobacteria, Bacilli, and Alphaproteobacteria were the most abundant classes of bacteria and consistent in the genus Vespa . Besides, though Vespa from different regions had quite different gut bacterial communities at the genus level, Lactobacillus and other lactic acid bacteria were abundant and played important roles in protection and metabolism in V. velutina collected from different regions. This is the first report of midgut bacterial diversity of three forms queens, workers, and males of V. velutina . Our findings provide insight that Proteobacteria and Firmicutes (especially Lactobacillus and other lactic acid bacteria) are consistent and may play important roles in the genus Vespa . The understanding of the microbiome in the midgut of Vespa and the discovery of the vital bacteria would provide useful information to design pest biological control agents. Thus, the significance of this study is to provide a basis for the study of the relationship between gut microbiota and physiology and health of Vespa , as well as the control of Vespa .

    Abstract: In this work, a dispersible graphene-based material with a characteristic of aggregation-induced emission (AIE) was prepared by wet chemical reduction of graphene oxide (GO). During the GO reduction process, a conjugated molecule TPEP containing tetraphenylethylene (TPE) and pyrene was employed as a stabilizer because of the π –π interactions and the wrapping effect. The as-prepared rGO-TPEP not only has good dispersion in solution but also processes the AIE feature. Its fluorescence intensity is 2.23 times higher than that of TPEP at the same condition. The unique optical properties and AIE effect enable the rGO-TPEP as a chemical sensor for highly sensitive explosive detection in aggregated state and solid state. In the aggregated state, trace 2,4-dinitrotoluene (DNT) can be detected by the rGO-TPEP even when the concentration is as low as 0.91 ppm, and the quenching constant is as high as 2.47 × 104 M -1 .

    Abstract: Polo-like kinase 4 (PLK4), a key regulator of centriole biogenesis, is frequently overexpressed in cancer cells. However, roles and the mechanism of PLK4 in the leukemiagenesis of acute myeloid leukemia (AML) remain unclear. In this study, the PLK4 inhibitor Centrinone and the shRNA knockdown were used to investigate roles and the mechanism of PLK4 in the leukemiagenesis of AML. Our results indicated that Centrinone inhibited the proliferation of AML cells in a dose- and time-dependent manner via reduced the expression of PLK4 both in the protein and mRNA levels. Moreover, colony formation assay revealed that Centrinone reduced the number and the size of the AML colonies. Centrinone induced AML cell apoptosis by increasing the activation of Caspase-3/poly ADP-ribose polymerase (PARP). Notably, Centrinone caused the G2/M phase cell cycle arrest by decreasing the expression of cell cycle-related proteins such as Cyclin A2, Cyclin B1, and Cyclin-dependent kinase 1 (CDK1). Consistent with above results, knockdown the expression of PLK4 also inhibited cell proliferation and colony formation, induced cell apoptosis, and caused G2/M phase cell cycle arrest without affecting cell differentiation. All in all, this study suggested that PLK4 inhibited the progression of AML in vitro , and these results herein may provide clues in roles of PLK4 in the leukemiagenesis of AML.

    Abstract: ARTEMIN (ARTN), one of the glial-cell derived neurotrophic factor family of ligands, has been reported to be associated with a number of human malignancies. In this study, the enhanced expression of ARTN in colorectal carcinoma (CRC) was observed; the expression of ARTN positively correlated with lymph node metastases and advanced tumor stages and predicted poor prognosis. Forced expression of ARTN in CRC cells enhanced oncogenic behavior, mesenchymal phenotype, stem cell-like properties and tumor growth and metastasis in a xenograft model. These functions were conversely inhibited by depletion of endogenous ARTN. Forced expression of ARTN reduced the sensitivity of CRC cells to 5-FU treatment; and 5-FU resistant CRC cells harbored enhanced expression of ARTN. The oncogenic functions of ARTN were demonstrated to be mediated by p44/42 MAP kinase dependent expression of CDH2 (CADHERIN 2, also known as N-CADHERIN). Inhibition of p44/42 MAP kinase activity or siRNA mediated depletion of endogenous CDH2 reduced the enhanced oncogenicity and chemoresistance consequent to forced expression of ARTN induced cell functions; and forced expression of CDH2 rescued the reduced mesenchymal properties and resistance to 5-FU after ARTN depletion. In conclusion, ARTN may be of prognostic and theranostic utility in CRC.

    Abstract: The potential toxicity of flupyradifurone (FPF) to honey bees has been a subject of controversy in recent years. Understanding the effect of pesticides on nurse bees is important because the fitness of nurse bees is critical for in-hive activities, such as larval survival and performing hive maintenance. In order to evaluate the acute oral toxicity of flupyradifurone on nurse bees, flupyradifurone at five different concentrations was selected to feed both larvae and nurse bees. Our results showed that nurse bees were more sensitive to flupyradifurone than larvae (LD 50 of the acute oral toxicity of flupyradifurone was 17.72 μg a.i./larva and 3.368 μg a.i./nurse bee). In addition, the apoptotic rates of neurons in mushroom bodies of nurse bees were significantly induced by flupyradifurone at sublethal concentrations (8 mg/L, 20 mg/L, and 50 mg/L) and the median lethal concentration LC 50 (125 mg/L). The expression of immune-related genes ( Hsp90 , Toll-8/Tollo , and defensin ) was significantly changed in exposed nurse bees at the field-realistic concentration of flupyradifurone. However, three detoxifying enzyme genes ( CYP9Q1 , -2, and -3) were not affected by pesticide exposure. Our data suggest that although flupyradifurone had a relatively lower acute oral toxicity than many other common pesticides, exposures to the field-realistic and other sublethal concentrations of flupyradifurone still have cytotoxicity and immune-responsive effects on nurse bees. Therefore, flupyradifurone should be considered for its application in crops.

    Abstract: Tigecycline serves as a last-resort antimicrobial agent against severe infections caused by multidrug-resistant bacteria. Tet(X) and its numerous variants encoding flavin-dependent monooxygenase can confer resistance to tigecycline, with tet (X4) being the most prevalent variant. This study aims to investigate the prevalence and characterize tigecycline resistance gene tet (X) in E. coli isolates from various origins in Yangzhou, China, to provide insights into tet (X) dissemination in this region. In 2022, we tested the presence of tet (X) in 618 E. coli isolates collected from diverse sources, including patients, pig-related samples, chicken-related samples, and vegetables in Yangzhou, China. The antimicrobial susceptibility of tet (X)-positive E. coli isolates was conducted using the agar dilution method or the broth microdilution method. Whole genome sequencing was performed on tet (X)-positive strains using Illumina and Oxford Nanopore platforms. Four isolates from pig or pork samples carried tet (X4) and exhibited resistance to multiple antimicrobial agents, including tigecycline. They were classified as ST542, ST10, ST761, and ST48, respectively. The tet (X4) gene was located on IncFIA8-IncHI1/ST17 (n=2), IncFIA18-IncFIB(K)-IncX1 (n=1), and IncX1 (n=1) plasmids, respectively. These tet (X4)-carrying plasmids exhibited high similarity to other tet (X4)-bearing plasmids with the same incompatible types found in diverse sources in China. They shared related genetic environments of tet (X4) associated with IS CR2 , as observed in the first identified tet (X4)-bearing plasmid p47EC. In conclusion, although a low prevalence (0.65%) of tet (X) in E. coli strains was observed in this study, the horizontal transfer of tet (X4) among E. coli isolates mediated by pandemic plasmids and the mobile element IS CR2 raises great concerns. Thus, heightened surveillance and immediate action are imperative to curb this clinically significant resistance gene and preserve the efficacy of tigecycline.
