Abstract: Introduction: Rheumatoid arthritis is an autoimmune disease characterized by chronic joint inflammation. Methotrexate is one of the most effective drugs for rheumatoid arthritis, but the adverse reactions caused by oral methotrexate greatly limit its clinical application. Transdermal drug delivery system is an ideal alternative to oral methotrexate by absorbing drugs into the human body through the skin. However, methotrexate in the existing methotrexate microneedles is mostly used alone, and there are few reports of combined use with other anti-inflammatory drugs. Methods: In this study, glycyrrhizic acid was first modified onto carbon dots, and then methotrexate was loaded to construct a nano-drug delivery system with fluorescence and dual anti-inflammatory effects. Then hyaluronic acid was combined with nano-drug delivery system to prepare biodegradable soluble microneedles for transdermal drug delivery of rheumatoid arthritis. The prepared nano-drug delivery system was characterized by transmission electron microscopy, fluorescence spectroscopy, laser nanoparticle size analyzer, ultraviolet-visible absorption spectroscopy, Fourier transform infrared spectroscopy, differential scanning calorimeter and nuclear magnetic resonance spectrometer. The results showed that glycyrrhizic acid and methotrexate were successfully loaded on carbon dots, and the drug loading of methotrexate was 49.09%. The inflammatory cell model was constructed by lipopolysaccharide-induced RAW264.7 cells. In vitro cell experiments were used to explore the inhibitory effect of the constructed nano-drug delivery system on the secretion of inflammatory factors by macrophages and the cell imaging ability. The drug loading, skin penetration ability, in vitro transdermal delivery and in vivo dissolution characteristics of the prepared microneedles were investigated. The rat model of rheumatoid arthritis was induced by Freund's complete adjuvant. Results: The results of in vivo animal experiments showed that the soluble microneedles of the nano drug delivery system designed and prepared in this study could significantly inhibit the secretion of pro-inflammatory cytokines and had a significant therapeutic effect on arthritis. Discussion: The prepared glycyrrhizic acid-carbon dots-methotrexate soluble microneedle provides a feasible solution for the treatment of Rheumatoid arthritis.

Abstract: In this work, silica aerogel was modified by 9,10-dihydro-9-oxa-10-phosphaphenanthrene-1-oxide (DOPO). Then DOPO-immobilized silica aerogel nanoparticles were used as a flame retardant to prepare flame-retardant polyurethane foams. Microscale combustion calorimeter and cone calorimeter tests were employed to evaluate the flame retardancy of polyurethane foams. It was found that both the heat release rate and the total heat release of the composites were reduced with the incorporation of DOPO immobilized silica aerogel. It is speculated that the DOPO-immobilized silica aerogel nanoparticles can inhibit the degradation of polyurethane and catalyze the formation of carbonaceous carbon on the surface.

Abstract: Strain Hardening Cementitious Composites (SHCC) are materials exhibiting high tensile ductility with the formation of multiple cracks. Since the mechanical properties of SHCC members are governed by the interfacial characteristics between fibers and matrix, understanding the mechanism of single fiber pullout behavior is crucial for SHCC material design. Existing model was set up based on Polyvinyl Alcohol (PVA) fibers, making it inapplicable to other kinds of synthetic fibers those exhibit divergent pullout behaviors. As a result, the simulated curves cannot agree well with the tested results of SHCC made with other fibers or hybrid fibers. In this study, a unified single fiber pullout model was proposed to take divergent kinds of fiber pullout behavior into account. Five parameters were defined to describe the single fiber pullout behavior, where the frictional strength during the pullout stage and the blocking length of fiber under pulley force were for the first time introduced. For verification purpose, fiber-pullout samples with Polyethylene terephthalate (PET) and PVA fibers were tested and the results agree well with the simulated curves from proposed model. The stochasticity of each parameters were then analyzed and described by normal distribution, through which fibers with various random pullout behaviors can be incorporated into a larger scale modelling. Therefore, the fiber-bridging constitutive law for a single crack was calculated and compared with the single crack test results, which confirmed again the validity of the proposed model.

Abstract: Rectal cancer (RC) is a globally prevalent malignant tumor, presenting significant challenges in its management and treatment. Currently, magnetic resonance imaging (MRI) offers superior soft tissue contrast and radiation-free effects for RC patients, making it the most widely used and effective detection method. In early screening, radiologists rely on patients’ medical radiology characteristics and their extensive clinical experience for diagnosis. However, diagnostic accuracy may be hindered by factors such as limited expertise, visual fatigue, and image clarity issues, resulting in misdiagnosis or missed diagnosis. Moreover, the distribution of surrounding organs in RC is extensive with some organs having similar shapes to the tumor but unclear boundaries; these complexities greatly impede doctors’ ability to diagnose RC accurately. With recent advancements in artificial intelligence, machine learning techniques like deep learning (DL) have demonstrated immense potential and broad prospects in medical image analysis. The emergence of this approach has significantly enhanced research capabilities in medical image classification, detection, and segmentation fields with particular emphasis on medical image segmentation. This review aims to discuss the developmental process of DL segmentation algorithms along with their application progress in lesion segmentation from MRI images of RC to provide theoretical guidance and support for further advancements in this field.

Abstract: The horizontal well multistage hydraulic fracturing technology is the most effective way to exploit shale gas resources. Compared with conventional reservoir fracturing, the flowback rate of a fracturing fluid in a shale reservoir is extremely low, and a large amount of fracturing fluid remains in the formation. Therefore, the research on the mechanism of shale reservoir fracturing fluid flowback process will contribute to laying a theoretical foundation for improving the effect of the innovation for increasing output of shale gas wells. Based on the shale in the Sichuan Basin, this study first describes basic experiments on physical properties such as the porosity, permeability, mineral composition, wettability, and microstructure. The physical properties of shale reservoirs were also analyzed, which laid the foundation for subsequent modeling. Second, CMG software is used to establish a numerical model that fits the characteristics of the flowback process. The effect of reservoir properties, fracturing parameters, drainage–production system, chemical permeability on gas and water production in the flowback process and their mechanisms are also analyzed. According to most numerical simulation results, the lower cumulative gas production will be with the higher cumulative water production which means the higher flowback rate. The pursuit of only a high flowback rate is not advisable, and the development of the drainage–production system requires reasonable control of the fracturing fluid flowback rate. This study provides a theoretical basis for the optimization of shale gas drainage–production system after hydraulic fracturing.

Abstract: Current targeted pulmonary arterial hypertension (PAH) therapies have improved lung hemodynamics, cardiac function, and quality of life; however, none of these have reversed the ongoing remodeling of blood vessels. Considering notopterol, a linear furocoumarin extracted from the root of traditional Chinese medicine Qiang-Huo ( Notopterygium incisum ), had shown the antiproliferative and anti-inflammatory properties in previous studies, we hypothesized that it could play a role in ameliorating PAH. Methods In vivo , we conducted monocrotaline (MCT) induced PAH rats and treated them with notopterol for 3 weeks. Then, the rats were examined by echocardiography and RV catheterization. The heart and lung specimens were harvested for the detection of gross examination, histological examination and expression of inflammatory molecules. In vitro , human pulmonary arterial smooth muscle cells (HPASMCs) were treated with notopterol after hypoxia; then, cell proliferation was assessed by cell counting kit-8 and Edu assay, and cell migration was detected by wound healing assays. Results We found that notopterol improved mortality rate and RV function while reducing right ventricular systolic pressure in MCT-induced PAH rats. Furthermore, notopterol reduced right ventricular hypertrophy and fibrosis, and it also eased pulmonary vascular remodeling and MCT-induced muscularization. In addition, notopterol attenuated the pro-inflammatory factor (IL-1β, IL-6) and PCNA in the lungs of PAH rats. For the cultured HPASMCs subjected to hypoxia, we found that notopterol can inhibit the proliferation and migration of HPASMCs. Conclusion Our studies show that notopterol exerts anti-inflammatory and anti-proliferative effects in the pulmonary arteries, which may contribute to prevention of PAH.

Abstract: The paper proposed a novel framework for efficient simulation of crack propagation in brittle materials. In the present work, the phase field represents the sharp crack surface with a diffuse fracture zone and captures the crack path implicitly. The partial differential equations of the phase field models are solved with physics informed neural networks (PINN) by minimizing the variational energy. We introduce to the PINN-based phase field model the degradation function that decouples the phase-field and physical length scales, whereby reducing the mesh density for resolving diffuse fracture zones. The numerical results demonstrate the accuracy and efficiency of the proposed algorithm.

Abstract: Transcranial alternating current stimulation (tACS) applies a sinusoidal oscillating current to modulate intrinsic oscillatory activity. Relevant studies of tACS have indicated that tACS can increase spontaneous brain activity in the occipital area. However, few studies have compared the effects of tACS with different current intensities on spontaneous brain activity in the occipital region. In this study, 10-Hz tACS was delivered to the occipital region at different current intensities (i.e., 1 and 2 mA). We investigated the effect of the tACS on both eyes-open and eyes-closed resting-state electroencephalography (EEG). A total of 20 subjects and fifteen subjects were recruited to participate in the 1-mA tACS experiment and the 2-mA tACS experiment, respectively. Ten subjects participated in both experiments. The experimental results demonstrated that both 1-mA tACS and 2-mA tACS could increase occipital resting-state EEG activities. For the eyes-open condition, alpha activity elicited by 2-mA tACS increased significantly greater than that elicited by 1-mA tACS, while 1-mA tACS could produce greater alpha activity compared to 2 mA for the eyes-closed condition. These results suggested that the optimal current intensity might be different for the eyes-open and eyes-closed resting-state conditions, laying a foundation for the subsequent study of occipital tACS on task-state EEG activities.

Abstract: Obsessive Compulsive Drinking Scale (OCDS) was established and introduced to measure the craving for alcohol and the severity of alcohol dependence. However, the Chinese version of OCDS is still unavailable and has not been validated in the Chinese population. We tended to translate and validate the OCDS in Chinese. We translated original OCDS into Chinese through bi-direction translations and tested the reliability and validity. We found that Chinese OCDS had high internal consistency and good test-retest reliability. The Chinese OCDS also presented good internal structure to reflect the severity of alcohol dependence. The Chinese OCDS could be used in clinical studies and research among the Chinese population.
