    Abstract: Deep ocean blue holes possess steep physicochemical gradients, especially low dissolved oxygen concentration, which shape the extraordinary microbial communities. However, the environmental responses of microorganisms with different lifestyles and knowledge of culturable microorganisms in the blue holes are still unknown. Here, we investigated the bacterial community structure with different lifestyles of the world’s deepest blue hole - the Yongle Blue Hole (YBH) in the South China Sea using both culture-dependent and -independent methods. YBH can be divided by oxygen content into an oxic zone, a suboxic zone and two anoxic zones. The abundance of bacteria, archaea, genes dsrB and soxB were all higher in the free-living (FL) lifestyle than in the particle-associated (PA) lifestyle, yet the diversity and richness of PA bacteria were higher than that of FL bacteria. More Gammaproteobacteria and less Alphaproteobacteria , Chloroflexi and Nitrospinae were observed within the FL fraction than within the PA fraction. The relative abundance of sulfur-oxidizing bacteria (SOB) was dominant between 100-140 m (anoxic zone I) in YBH, with a maximum of 90.0% (140 m FL fraction). The SOB in YBH were mainly colorless sulfur bacteria and purple non-sulfur bacteria, of which Thiomicrorhabdus and Sulfurimonas were the main representatives. In addition, a total of 294 bacterial strains were isolated on a variety of media and culture conditions, and 22.2% (18/81) of anaerobic strains were identified as potential novel species. Our study reveals a distinction between FL and PA bacteria in YBH. It contributes to further understanding of the bacterial community in deep ocean blue holes, and provides bacterial resources for subsequent studies on their adaptation to extreme marine environments.

    Abstract: Papillary renal cell carcinoma (pRCC) is the largest histologic subtype of non-clear-cell RCC. To date, there is no reliable nomogram to predict the prognosis of patients with pRCC after nephrectomy. We aimed to first establish an effective nomogram to predict the overall survival (OS) of patients with pRCC after nephrectomy. Methods A total of 3,528 eligible patients with pRCC after nephrectomy were identified from the Surveillance, Epidemiology, and End Results (SEER) database between 2010 and 2015. The patients were randomized into the training cohort ( n = 2,472) and the validation cohort ( n = 1,056) at a 7:3 ratio. In total, 122 real-world samples from our institute (titled the AHMU-pRCC cohort) were used as the external validation cohort. Univariate and subsequent multivariate Cox regression analyses were conducted to identify OS-related prognostic factors, which were further used to establish a prognostic nomogram for predicting 1-, 3-, and 5-year OS probabilities. The performance of the nomogram was evaluated by using the concordance index (C-index), receiver operating characteristic curve (ROC), calibration plot, and decision curve analysis (DCA). Results Multivariate Cox analysis showed that age, race, marital status, TNM stage, tumor size, and surgery were significant OS-related prognostic factors. A prognostic model consisting of these clinical parameters was developed and virtualized by a nomogram. High C-index and area under the ROC curve (AUC) values of the nomogram at 1, 3, and 5 years were found in the training, validation, and AHMU-pRCC cohorts. The calibration plot and DCA also showed that the nomogram had a satisfactory clinical application value. A risk classification system was established to risk-stratify patients with pRCC. Conclusion Based on a large cohort from the public SEER database, a reliable nomogram predicting the OS of patients with pRCC after nephrectomy was constructed, which could optimize the survival assessment and clinical treatment.

    Abstract: Micro-structured targets can be employed to enhance the coupling of laser energy to the high energy density plasma. Here we report on experimental measurement of enhanced proton beam energy from laser-driven micro-wire array (MWA) targets along the backward direction. An ultra-intense ( ∼ 2 × 10 20 W / c m 2 ) laser pulse of ∼   40   f s pulse duration interacts with the MWA structure and induces large population of energetic electrons. The enhanced sheath field efficiently accelerates protons both transversely and longitudinally. The spectrometers record proton cut-off energy of around 16   M e V and temperature 813 k e V along the backward direction, which is 20 % − 60 % higher than that of a flat target under commensurate laser conditions. Comparison with particle-in-cell simulations suggests that the enhancement originates from the increased temperature and population of the hot electrons within the micro-wires. These measurements provide a direct probe of the high energy density plasma condition in laser-driven solid targets and a useful benchmark for further studies on laser-driven micro-structured targets.

    Abstract: Schizophrenia patients with auditory verbal hallucinations (AVHs) are diseased groups of serious psychosis with still unknown etiology. The aim of this research was to identify the neurophysiological correlates of auditory verbal hallucinations. Revealing the neural correlates of auditory hallucination is not merely of great clinical significance, but it is also quite essential to study the pathophysiological correlates of schizophrenia. In this study, 25 Schizophrenia patients with AVHs (AVHs group, 23.2 ± 5.35 years), 52 Schizophrenia patients without AVHs (non-AVHs group, 25.79 ± 5.63 years) and 28 healthy subjects (NC group, 26.14 ± 5.45 years) were enrolled. Dynamic functional connectivity was studied with a sliding-window method and functional connectivity states were then obtained with the k-means clustering algorithm in the three groups. We found that schizophrenia patients with AVHs were characterized by significant decreased static functional connectivity and enhanced variability of dynamic functional connectivity (non-parametric permutation test, Bonferroni correction, p & lt; 0.05). In addition, the AVHs group also demonstrated increased number of brain states, suggesting brain dynamics enhanced in these patients compared with the non-AVHs group. Our findings suggested that there were abnormalities in the connection of brain language regions in auditory verbal hallucinations. It appears that the interruption of connectivity from the language region might be critical to the pathological basis of AVHs.

    Abstract: Suicidal ideation increases precipitously in patients with depression, contributing to the risk of suicidal attempts. Despite the recent advancement in transcranial magnetic stimulation, its effectiveness in depression disorder and its wide acceptance, the network mechanisms of the clinical response to suicidal ideation in major depressive disorder remain unclear. Independent component analysis for neuroimaging data allows the identification of functional network connectivity which may help to explore the neural basis of suicidal ideation in major depressive disorder. Resting-state functional magnetic resonance imaging data and clinical scales were collected from 30 participants (15 major depressive patients with suicidal ideation and 15 healthy subjects). Individual target-transcranial magnetic stimulation (IT-TMS) was then used to decrease the subgenual anterior cingulate cortex activity through the left dorsolateral prefrontal cortex. Thirty days post IT-TMS therapy, seven of 15 patients (46.67%) met suicidal remission criteria, and 12 patients (80.00%) met depression remission criteria. We found that IT-TMS could restore the abnormal functional network connectivity between default mode network and precuneus network, left executive control network and sensory-motor network. Furthermore, the changes in functional network connectivity between the default mode network and precuneus network were associated with suicidal ideation, and depressive symptoms were related to connectivity between left executive control network and sensory-motor network. These findings illustrate that IT-TMS is an effective protocol for the accurate restoration of impaired brain networks, which is consistent with clinical symptoms.

    Abstract: Preeclampsia is a heterogeneous and complex disease with its pathogenesis mechanism not fully elucidated. A certain subset of patients with preeclampsia exhibit disturbances in lipid metabolism before clinical symptoms. Moreover, there is a tendency for preeclampsia to run in families. Whether genetic factors play a role in abnormal lipid metabolism during the incidence of preeclampsia has not been well investigated. Methods Preeclampsia patients ( n = 110) and healthy age- and gravidity-matched pregnant women ( n = 110) were enrolled in this study. Peripheral blood specimens were used for genomic analysis ( n = 10/group) or laboratory validation ( n = 100/group). We retrospectively obtained the baseline clinical characteristics of 68 preeclampsia patients and 107 controls in early pregnancy (12–14 gestational weeks). Correlation analyses between differential genes and baseline lipid profiles were performed to identify candidate genes. In vitro and in vivo gain-of-function models were constructed with lentivirus and adeno-associated virus systems, respectively, to investigate the role of candidate genes in regulating lipid metabolism and the development of preeclampsia. Results We observed that preeclampsia patients exhibited significantly elevated plasma TC ( P = 0.037) and TG ( P & lt; 0.001) levels and increased body mass index ( P = 0.006) before the disease onset. Within the region of 27 differential copy number variations, six genes potentially connected with lipid metabolism were identified. The aberrant copies of APOBEC3A , APOBEC3A_B , BTNL3 , and LMF1 between preeclampsia patients and controls were verified by quantitative polymerase chain reaction. Especially, APOBEC3A showed a significant positive correlation with TC ( P & lt; 0.001) and LDL ( P = 0.048) in early pregnancy. Then, our in vitro data revealed that overexpression of APOBEC3A disrupted lipid metabolism in HepG2 cells and affected both cholesterol and fatty acid metabolisms. Finally, in vivo study in a hepatic-specific overexpressed APOBEC3A mouse model revealed abnormal parameters related to lipid metabolism. Pregnant mice of the same model at the end of pregnancy showed changes related to preeclampsia-like symptoms, such as increases in sFlt-1 levels and sFlt-1/PLGF ratios in the placenta and decreases in fetal weight. Conclusion Our findings established a new link between genetics and lipid metabolism in the pathogenesis of preeclampsia and could contribute to a better understanding of the molecular mechanisms of preeclampsia.

    Abstract: This study aimed to evaluate the effect of perioperative transcutaneous electrical acupoint stimulation (TEAS) on postoperative cognitive dysfunction (POCD) in older patients with lumbar spine surgery. Methods Older patients (aged 60–80 years old) receiving lumbar spine surgery under general anesthesia were randomly divided into group A, 3-day intervention group; group B, 7-day intervention group; control group C, sham TEAS group, selected “Baihui” (GV 20) and “Dazhui” (GV 14) point was intervened once 30 min before operation with “HANS” transcutaneous electrical stimulation device, and then once a day after operation for 30 min each time. The primary outcome was the incidence of postoperative cognitive impairment assessed by the use of the Mini Mental Rating Scale (MMSE), patients developed POCD according to the Z score method. The secondary outcome was serum interleukin-6 (IL-6), tumor Necrosis factor α (TNF-α), neuron-specific enolase (NSE), and S100β protein levels. Results Three days after surgery, the incidence of POCD in groups A((22.4%)) and B ((18.3%)) were lower than those in group C ((42.9%)) ( P & lt; 0.05). There was no significant difference between groups A and B ( P & gt; 0.05). Seven days after surgery, the incidence of POCD in group B (18.3%) was lower than that in groups A (26.5%) and B (42.9%), and the comparison between groups B and C was statistically significant ( P & lt; 0.05). On the 3rd and 7th days after surgery, the levels of IL-6, TNF-α, NSE, and S100β in the two TEAS groups were lower than those in the sham TEAS group ( P & lt; 0.01), but higher than the preoperative levels in the three groups ( P & lt; 0.01). Conclusion It seems that Perioperative TEAS intervention could reduce the level of inflammatory factors IL-6, TNF-α in the blood of older patients with lumbar spine surgery, and reduce the incidence of POCD.
