    Abstract: Forsythiae Fructus (FF), the fruit of Forsythia suspensa (Thunb.) Vahl. (Lianqiao), is one of the most fundamental herbs in Traditional Chinese Medicines (TCM), mainly due to its heat-clearing and detoxifying effects. There are two types of FF, the greenish fruits that start to ripen (GF) and the yellow fruits that are fully ripe (RF), called “Qingqiao” and “Laoqiao” referred to the Chinese Pharmacopoeia, respectively. It undergoes a complex series of changes during the maturation of FF. However, the clinical uses and preparation of phytopharmaceuticals of FF have not been distinguished to date. Moreover, there is limited information on the study of the difference in pharmacological activity between RF and GF. In this study, a rat model of bile duct ligation (BDL)-induced cholestasis was used to compare the differences in their effects. RF was found to have better results than GF in addressing toxic bile acids (BAs) accumulation and related pathological conditions caused by BDL. The underlying mechanism may be related to the interventions of gut microbiota. The results of the present study suggest that the better detoxifying effect of RF than GF may be indirectly exerted through the regulation of gut microbiota and thus the improvement of BAs metabolism.

    Abstract: Shrubs play a major role in maintaining ecosystem stability in the arid deserts of Central Asia. During the long-term adaptation to extreme arid environments, shrubs have developed special assimilative branches that replace leaves for photosynthesis. In this study, four dominant shrubs with assimilative branches, namely Haloxylon ammodendron , Haloxylon persicum , Calligonum mongolicum , and Ephedra przewalskii , were selected as the research objects, and the dry mass, total length, node number, and basal diameter of their assimilative branches and the average length of the first three nodes were carefully measured, and the allometric relationships among five traits of four species were systematically compared. The results indicated that: (1) Four desert shrubs have different assimilative branches traits. Compared with H. persicum and H. ammodendron , C. mongolicum and E. przewalskii have longer internodes and fewer nodes. The dry mass of H. ammodendron and the basal diameter of H. persicum were the smallest; (2) Significant allometric scaling relationships were found between dry mass, total length, basal diameter, and each trait of assimilative branches, all of which were significantly less than 1; (3) The scaling exponents of the allometric relationship between four traits and the dry mass of assimilative branches of H. persicum were greater or significantly greater than those of H. ammodendron . The scaling exponents of the relationships between the basal diameter, dry mass, and total length of E. przewalskii were higher than those of the other three shrubs. Therefore, although different species have adapted to drought and high temperatures by convergence, there was great variability in morphological characteristics of assimilative branches, as well as in the scaling exponents of relationships among traits. The results of this study will provide valuable insights into the ecological functions of assimilative branches and survival strategies of these shrubs to cope with aridity and drought in desert environments.

    Abstract: Aquatic gastropods are important integral components of the macroinvertebrate community in freshwater ecosystems and play critical roles in freshwater ecosystems by contributing to biodiversity, nutrient cycling, and water quality. However, the variation of aquatic gastropods’ community structure under the combined effects of warming and nutrient enrichment remains largely unknown. To investigate this question, we performed an outdoor mesocosm experiment examining the interaction of warming (a 4.5 °C increase in mean temperature above ambient conditions) and nutrient enrichment (phosphorus addition) on the aquatic gastropods’ community and dominant population (Bellamya aeruginosa). We analyzed the changes in community dynamics (abundance and biomass), size structure, and stoichiometric traits (only B. aeruginosa ). Results showed that phosphorus enrichment alone had a positive effect on the total abundance and biomass of gastropods, as well as the abundance and biomass of B. aeruginosa . Warming alone only produced a positive effect on total abundance. However, the combined effects of warming and phosphorus enrichment negatively affected the biomass and abundance of the whole gastropod community and the dominant gastropod population. The body mass of B. aeruginosa increased because of warming, whereas the body mass of the gastropod community negatively responded to warming. Phosphorus enrichment alone had no remarkable effects on body mass. The combined effects of warming and phosphorus enrichment negatively affected the whole community’s body mass but had no substantial effect on the body mass of B. aeruginosa . For body stoichiometric traits, warming or phosphorus enrichment alone produced positive effects on the nitrogen and phosphorus contents of B. aeruginosa . The combined effects caused adverse effects on the contents of the two elements. The effect of warming alone only decreased the ratio of nitrogen to phosphorus. Results suggested that the response levels in size structure between the gastropod community and the dominant population differed remarkably. Composition species shift was the main cause of the decrease in aquatic gastropods’ community size structure. The shift in species composition at the whole gastropod community level caused by warming and phosphorus enrichment may result in more complex and unpredicted consequences through cascade effects on the structure and function of freshwater ecosystems.

    Abstract: The objective of this study was to apply a machine learning method to evaluate the risk factors associated with serious adverse events (SAEs) and predict the occurrence of SAEs in cancer inpatients using antineoplastic drugs. A retrospective review of the medical records of 499 patients diagnosed with cancer admitted between January 1 and December 31, 2017, was performed. First, the Global Trigger Tool (GTT) was used to actively monitor adverse drug events (ADEs) and SAEs caused by antineoplastic drugs and take the number of positive triggers as an intermediate variable. Subsequently, risk factors with statistical significance were selected by univariate analysis and least absolute shrinkage and selection operator (LASSO) analysis. Finally, using the risk factors after the LASSO analysis as covariates, a nomogram based on a logistic model, extreme gradient boosting (XGBoost), categorical boosting (CatBoost), adaptive boosting (AdaBoost), light-gradient-boosting machine (LightGBM), random forest (RF), gradient-boosting decision tree (GBDT), decision tree (DT), and ensemble model based on seven algorithms were used to establish the prediction models. A series of indicators such as the area under the ROC curve (AUROC) and the area under the PR curve (AUPR) was used to evaluate the model performance. A total of 94 SAE patients were identified in our samples. Risk factors of SAEs were the number of triggers, length of stay, age, number of combined drugs, ADEs occurred in previous chemotherapy, and sex. In the test cohort, a nomogram based on the logistic model owns the AUROC of 0.799 and owns the AUPR of 0.527. The GBDT has the best predicting abilities (AUROC = 0.832 and AUPR = 0.557) among the eight machine learning models and was better than the nomogram and was chosen to establish the prediction webpage. This study provides a novel method to accurately predict SAE occurrence in cancer inpatients.

    Abstract: Background: The Zhuyu pill (ZYP), composed of Coptis chinensis Franch. and Tetradium ruticarpum (A. Jussieu) T. G. Hartley, is an effective traditional Chinese medicine with potential anti-cholestatic effects. However, the underlying mechanisms of ZYP remain unknown. Objective: To investigate the mechanism underlying the interventional effect of ZYP on mRNA-seq analysis in cholestasis rat models. Materials and methods: This study tested the effects of a low-dose (0.6 g/kg) and high-dose (1.2 g/kg) of ZYP on a cholestasis rat model induced by α-naphthyl-isothiocyanate (ANIT, 50 mg/kg). Serum biochemistry and histopathology results were used to evaluate the therapeutic effect of ZYP, and mRNA-Seq analysis was performed and verified using real-time fluorescence quantitative PCR (qRT-PCR). GO, KEGG, and GSEA analyses were integrated to identify the mechanism by which ZYP impacted cholestatic rats. Results: ZYP was shown to significantly improve abnormal changes in the biochemical blood indexes and liver histopathology of cholestasis rats and regulate pathways related to bile and lipid metabolism, including fatty acid metabolism, retinol metabolism, and steroid hormone biosynthesis, to alleviate inflammation, cholestasis, and lipid metabolism disorders. Relative expression of the essential genes Cyp2a1, Ephx2, Acox2, Cyp1a2, Cyp2c11, and Sult2a1 was verified by qRT-PCR and showed the same trend as mRNA-seq analysis. Conclusion: ZYP has a significant anti-cholestatic effect by regulating bile metabolism and lipid metabolism related pathways. These findings indicate that ZYP is a novel and promising prospect for treating cholestasis.

    Abstract: This study aimed to explore possible associations between molecular subtypes and site of distant metastasis in advanced breast cancer (ABC). Methods 3577 ABC patients were selected from 21 hospitals of seven geographic regions in China from 2012-2014. A questionnaire was designed to collect medical information regarding demographic characteristics, risk factors, molecular subtype, recurrence/metastasis information, and disease-free survival (DFS). The cancers were classified into Luminal A, Luminal B, HER2-enriched and Triple Negative subtypes. Chi-square test and multivariate Cox proportional hazard models were performed to explore the associations between molecular subtypes and distant metastasis sites. Results A total of 2393 cases with molecular subtypes information were finally examined. Patients with Luminal A (51.1%) and Luminal B (44.7%) were most prone to bone metastasis, whereas liver metastasis was more frequently observed in HER2-enriched ABC patients (29.1%).The cumulative recurrence and metastasis rates of ABC patients at 36 months of DFS were the most significant within molecular types, of which Triple Negative was the highest (82.7%), while that of Luminal A was the lowest (58.4%). In the adjusted Cox regression analysis, Luminal B, HER2-enriched and Triple Negative subtypes increased the risk of visceral metastasis by 23%, 46% and 87% respectively. In addition, Triple Negative patients had a higher probability of brain metastasis (HR 3.07, 95% CI: 1.04-9.07). Conclusion Molecular subtypes can predict the preferential sites of distant metastasis, emphasizing that these associations were of great help in choices for surveillance, developing appropriate screening and cancer management strategies for follow-up and personalized therapy in ABC patients.

    Abstract: Depression often triggers addictive behaviors such as Internet addiction. In this network analysis study, we assessed the association between Internet addiction and residual depressive symptoms in patients suffering from clinically stable recurrent depressive disorder (depression hereafter). Materials and methods In total, 1,267 depressed patients were included. Internet addiction and residual depressive symptoms were measured using the Internet Addiction Test (IAT) and the two-item Patient Health Questionnaire (PHQ-2), respectively. Central symptoms and bridge symptoms were identified via centrality indices. Network stability was examined using the case-dropping procedure. Results The prevalence of IA within this sample was 27.2% (95% CI: 24.7–29.6%) based on the IAT cutoff of 50. IAT15 (“Preoccupation with the Internet”), IAT13 (“Snap or act annoyed if bothered without being online”) and IAT2 (“Neglect chores to spend more time online”) were the most central nodes in the network model. Additionally, bridge symptoms included the node PHQ1 (“Anhedonia”), followed by PHQ2 (“Sad mood”) and IAT3 (“Prefer the excitement online to the time with others”). There was no gender difference in the network structure. Conclusion Both key central and bridge symptoms found in the network analysis could be potentially targeted in prevention and treatment for depressed patients with comorbid Internet addiction and residual depressive symptoms.
