    Abstract: To identify less invasive and easily applicable serum cytokine-derived biomarkers which contribute to the diagnostic utility and risk assessment ability of the prostate health index (PHI) based multivariable model in grey zone aggressive prostate cancer (AG PCa) early detection. Methods Serum 45 cytokines screening was performed in a small training cohort consisting of 10 sera by Luminex liquid array-based multiplexed immunoassays and identified TRAIL and IL-10 as new biomarkers for PHI diagnostic utility adjustment for further validation with a multivariable predictive model in a cohort including 79 aggressive prostate cancer patients and 209 benign prostatic hyperplasia or indolent PCa patients within the PSA grey zone. Results TRAIL and IL-10 were identified as potential serum biomarkers for AG PCa detection by the result of multi-cytokines screening in the univariate analysis, while multivariable logistic regression confirmed the AUC of the full risk predictive model (0.915) including tPSA, fPSA, PHI, TRAIL, and IL-10 was higher than various diagnostic strategies. DCA suggested a superior net benefit and indicated a good discriminative ability of the full risk model consistently with the result of the nomogram. Conclusion We suggest a significant advantage for the PHI-based multivariate combinations of serum TRAIL and IL-10 comparing to PHI or other serum-derived biomarkers alone in the detection and risk stratification of grey zone AG PCa.

    Abstract: In order to reveal the process and mechanism of gas flow in a low-permeability coal seam, a new multiple-relaxation-time lattice Boltzmann method (MRT-LBM) model of gas migration in coal micro/nanopores based on Langmuir monolayer adsorption theory, the slip boundary scheme and Bosanquet effective viscosity was established. The software MATLAB was used to carry out the simulation study of uniform pore gas flow based on the MRT-LBM model, and the results were compared and verified with the porous anodic alumina membrane gas flow experimental results. On this basis, the gas flow in coal pores with different micro/nanopore sizes considering adsorption was simulated. The results show that the dimensionless permeability coefficient increases with decreasing pore size under the same pressure, which reflects the subsequent enhancement of the microboundary constraint effect and reveals that the pore system becomes the main controlling factor of coal seam permeability within the coal matrix in the middle and late stages of coal seam gas extraction, while the role of the microboundary constraint effect needs to be considered. The gas adsorption layer weakens the pore gas flow capacity, but for pores with a radius greater than 16 nm, the apparent change in permeability caused by the adsorption layer is less than 5%, and the adsorption effect can be ignored. N 2 , CH 4, and CO 2 enter the transition flow regime under different pressure conditions; with gas extraction, the gas pressure decreases, and the difference in the gas flow characteristics of the three gases increases.

    Abstract: Strip coal pillars caused by strip mining are widely distributed in China. Over time, strip coal pillars are posing more potential hazards as their stability decreases; therefore, it becomes increasingly important to recover coal pillars without damaging the ground surface. In this paper, strip-filling and second-mining technique is adopted to probe into an optimal scheme for mine goafs. Construction waste was one type of the materials used for backfilling, which was taken to underground from the surface with a pumping system. Jisuo Coal Mine of Tengzhou, Shandong province was taken as the object of study. Through theoretical calculation and on-site survey, we set a re-mining scheme to control rock stratum stability, i.e., 8-m-wide backfilling for the first time and 4-m-wide backfilling for the second time; moreover, the ground pressure behavior and subsidence of working surface were monitored and a contour map of surface subsidence was thus mapped. In-situ practice indicated that the underground pressure from coal pillars was transmitted to the backfilling materials, the maximum normal stress of which was 5 MPa, so it was sufficient for the backfilling strip to support the weight of overlying strata; the maximum value of roof-to-floor convergence of backfilled mining face is 45 mm, the maximum surface subsidence 44mm, the maximum inclination value 0.65 mm/m, the maximum surface curvature 0.018 mm/m 2 , and the maximum horizontal deformation 0.16 mm/m. The filling body in the original goafs plays the role of “pier”. By the monitoring data of subsidence, its surface deformation value was less than that of buildings suffering from grade I damage, which verified the feasibility of strip-filling and second-mining scheme. The success in this technique provides reliable technical and theoretical support for mines with similar conditions.

    Abstract: Early screening of clinically significant prostate cancer (csPCa) may offer opportunities in revolutionizing the survival benefits of this lethal disease. We sought to introduce a modified prostate health index density (mPHI) model using imaging indicators and to compare its diagnostic performance for early detection of occult onset csPCa within the prostate-specific antigen (PSA) gray zone with that of PHI and PHID. Methods and Participation Between August 2020 and January 2022, a training cohort of 278 patients (total PSA 4.0–10.0 ng/ml) who were scheduled for a prostate biopsy were prospectively recruited. PHI and PHID were compared with mPHI ( LD TRD × APD × TPV × PHI ) for the diagnosis performance in identifying csPCa. Pathology outcomes from systematic prostate biopsies were considered the gold standard. Results This model was tested in a training cohort consisting of 73 csPCa, 14 non-clinically significant prostate cancer(non-csPCa), and 191 benign prostatic hyperplasia (BPH) samples. In the univariate analysis for the PSA gray zone cohort, for overall PCa, the AUC of mPHI (0.856) was higher than PHI (0.774) and PHID (0.835). For csPCa, the AUC of mPHI (0.859) also surpassed PHI (0.787) and PHID (0.825). For detection of csPCa, compared with lower specificities from PHI and PHID, mPHI performed the highest specificity (76.5%), by sparing 60.0% of unnecessary biopsies at the cost of missing 11 cases of csPCa. The mPHI outperformed PHI and PHID for overall PCa detection. In terms of csPCa, mPHI exceeds diagnostic performance with a better net benefit in decision curve analysis (DCA) compared with PHI or PHID. Conclusions We have developed a modified PHI density (mPHI) model that can sensitively distinguish early-stage csPCa patients within the PSA gray zone.     Abstract: Graphitic carbon nitride (g-C 3 N 4 ) has shown to be a promising photocatalyst that, however, suffers from strong charge recombination and poor conductivity, while MXenes have shown to be perfect cocatalysts for the photocatalytic process but show poor stability. In this study, we successfully constructed 2D/2D heterojunctions of Fe-C 3 N 4 /Ti 3 C 2 for the photocatalytic degradation of antibiotics. In this study, multilayer Ti 3 C 2 was obtained by etching Ti 3 AlC 2 , and then Fe-C 3 N 4 /Ti 3 C 2 photocatalyst was prepared by the one-pot microwave method and high-temperature calcination method. The synthesized samples were characterized by XRD, SEM, TEM, XPS, TGA, BET, DRS, PL, and other means. The photocatalytic degradation of tetracycline hydrochloride by Fe-C 3 N 4 /Ti 3 C 2 was in accordance with the first-order reaction kinetics model, and the apparent rate constant k was 2.83, 2.06, and 1.77 times that of g-C 3 N 4 , Fe-C 3 N 4 , and g-C 3 N 4 /Ti 3 C 2 , respectively. Through the mechanism study, it was shown that the most active species in the reaction system was • O 2 − , while h + and •OH had a relatively lower effect on the degradation system.

    Abstract: Unconsciousness is a kind of brain activity that occurs below the level of consciousness, and the masked priming paradigm is a classic paradigm to study unconscious perceptual processing. With the deepening of unconscious perception research, different researchers mostly use different experimental materials and different masked priming paradigms in a single experiment but not for the comprehensive analysis of the unconscious information processing mechanism itself. Thus, the purpose of this study is to conduct a comprehensive analysis through a cross-experimental paradigm, cross-experimental materials, and cross-experimental purposes. We used activation likelihood estimation to test functional magnetic resonance imaging studies, involving 361 subjects, 124 foci in eight studies representing direct comparison of unconscious processing with baseline, and 115 foci in 10 studies representing direct comparison of unconscious priming effects. In the comparison of unconscious processing and baseline, clusters formed in the left superior parietal gyrus, the right insular gyrus, and the right inferior frontal gyrus (IFG) triangular part after correcting for familywise error (FWE). In the comparison of priming effects, clusters formed in only the right IFG triangular part after correcting for FWE. Here, we found that ventral and dorsal pathways jointly regulate unconscious perceptual processes, but only the ventral pathway is involved in the regulation of unconscious priming effects. The IFG triangular part is involved in the regulation of unconscious perceptual processing and unconscious priming effects and may be an important brain area in unconscious information processing. These preliminary data provide conditions for further study of the neural correlation of unconscious information processing.

    Abstract: Ferroptosis is a form of non-apoptotic and iron-dependent cell death originally identified in cancer cells. Recently, emerging evidence showed that ferroptosis-targeting therapy could be a novel promising anti-tumour treatment. However, systematic analyses of ferroptosis-related genes for the prognosis of non-small cell lung cancer (NSCLC) and the development of antitumor drugs exploiting the ferroptosis process remain rare. This study aimed to identify genes related to ferroptosis and NSCLC and to initially screen lead compounds that induce ferroptosis in tumor cells. We downloaded mRNA expression profiles and NSCLC clinical data from The Cancer Genome Atlas database to explore the prognostic role of ferroptosis-related genes. Four prognosis-associated ferroptosis-related genes were screened using univariate Cox regression analysis and the lasso Cox regression analysis, which could divide patients with NSCLC into high- and low-risk groups. Then, based on differentially expressed risk- and ferroptosis-related genes, the negatively correlated lead compound flufenamic acid (FFA) was screened through the Connective Map database. This project confirmed that FFA induced ferroptosis in A549 cells and inhibited growth and migration in a dose-dependent manner through CCK-8, scratch, and immunofluorescence assays. In conclusion, targeting ferroptosis might be a therapeutic alternative for NSCLC.

    Abstract: Real world evidence (RWE) and real-world data (RWD) are drawing ever-increasing attention in the pharmaceutical industry and drug regulatory authorities (DRAs) all over the world due to their paramount role in supporting drug development and regulatory decision making. However, there is little systematic documentary analysis about how RWE was integrated for the use by the DRAs in evaluating new treatment approaches and monitoring post-market safety. This study aimed to analyze and discuss the integration of RWE into regulatory decision-making process from the perspective of DRAs. Different development strategies to develop and adopt RWE by the DRAs in the US, Europe, and China were reviewed and compared, and the challenges encountered were discussed. It was found that different strategies on development of RWE were applied by FDA, EMA, and NMPA. The extent to which RWE was adopted in China was relatively limited compared to that in the US and EU, which was highly related to the national pharmaceutical environment and development stages. A better understanding of the overall goals, inputs, activities, outputs, and outcomes in developing RWE will help inform actions to harness RWD and leverage RWE for better health care decisions.

    Abstract: Objectives: Clínica Universidad de Navarra-Body Adiposity Estimator (CUN-BAE) is considered to be a more accurate indicator of body fat estimation. We aimed to investigate the association of CUN-BAE with the risk of type 2 diabetes mellitus (T2DM) and to compare the strength of the association between CUN-BAE, body mass index (BMI), waist circumference (WC), waist-to-height ratio (WHtR) and T2DM. Methods: The data were obtained from the annual health checkup database of residents in Xinzheng, China. From January 2011 to December 2021, 80,555 subjects aged ≥45 years met the inclusion criteria. Cox proportional hazard regression models were used to estimate the hazard ratios (HRs) and 95% confidence intervals (CIs) for CUN-BAE, BMI, WC, and WHtR in T2DM. Results: During a mean follow-up of 6.26 years, T2DM occurred in 12,967 subjects. The multivariable-adjusted HRs (95% CIs) of T2DM (highest vs. reference group) were 1.994 (1.811–2.196) for CUN-BAE, 1.751 (1.665–1.842) for WC, 1.715 (1.631–1.804) for WHtR, and 1.510 (1.436–1.588) for BMI, respectively. In addition, the risk of T2DM increased with baseline CUN-BAE (HR: 1.374; 95% CI: 1.328, 1.421), WC (HR: 1.236; 95% CI: 1.215, 1.256), WHtR (HR: 1.228; 95% CI: 1.208, 1.248), and BMI (HR: 1.175; 95% CI: 1.156, 1.195). Conclusion: Compared to BMI, WC or WHtR, CUN-BAE may more adequately reflect the adverse effects of adiposity on the risk of T2DM.

    Abstract: The impact of ablation parameters on acute tissue lesion formation after pulmonary vein isolation (PVI) has not been sufficiently evaluated in patients with atrial fibrillation. Radiofrequency ablation lesion can be visualized by late gadolinium enhancement cardiac magnetic resonance (LGE-CMR). We sought to quantitatively analyze the relationship between ablation parameter and tissue lesion following PVI at different segments of pulmonary vein (PV) using LGE-CMR. Methods Twenty-one patients with atrial fibrillation who underwent PVI procedure were retrospectively enrolled. All patients underwent LGE-CMR examination within 3 days after radiofrequency ablation. Ablation parameters during PVI were documented, including lesion size index (LSI), force–time integral (FTI), power, contact force, temperature, and time of duration. The ablation point was projected onto 3-dimensional (3D) left atrial shell constructed base on LGE-CMR and corresponding image intensity ratio (IIR) was calculated on the same shell. A tissue lesion point was defined when the LGE-CMR IIR was & gt; 1.2. Results In total, 1,759 ablation points were analyzed. The ablation parameters and IIRs for each PV segment were significantly different ( P & lt; 0.0001). IIRs corresponding to ablation points at posterior of PV tended to be higher than those at non-posterior of PV when similar ablation parameters were applied during ablation. LSI was a better predictor of tissue lesion existence following PVI than FTI, contact force, power, temperature, and duration time at non-posterior wall of PV. The IIR showed positive correlation with LSI at non-posterior wall of PV (non-posterior of right PV, r = 0.13, P = 0.001, non-posterior of left PV, r = 0.26, P & lt; 0.0001). Conclusion When similar ablation parameters were applied during PVI, the posterior wall of PV had more severe tissue lesion than non-posterior wall of PV. Therefore, it was reasonable to decrease ablation energy at posterior wall of PV. Moreover, LSI was a better index to reflect tissue lesion quality following PVI at non-posterior of PV.
