    Abstract: Antiphospholipid syndrome (APS) is a multisystem disorder characterized by thrombosis and/or recurrent fetal loss. This clinical phenotype heterogeneity may result in differences in response to treatment and prognosis. In this study, we aimed to identify primary thrombotic APS (TAPS) from primary obstetric APS (OAPS) using urine proteomics as a non-invasive method. Only patients with primary APS were enrolled in this study from 2016 to 2018 at a single clinical center in Shanghai. Urine samples from 15 patients with TAPS, 9 patients with OAPS, and 15 healthy controls (HCs) were collected and analyzed using isobaric tags for relative and absolute quantification (iTRAQ) labeling combined with liquid chromatography-tandem mass spectrometry analysis to identify differentially expressed proteins. Cluster analysis of urine proteomics identified differentiated proteins among the TAPS, OAPS, and HC groups. Urinary proteins were enriched in cytokine and cytokine receptor pathways. Representative secreted cytokines screened out (fold change & gt;1.20, or & lt;0.83, p & lt;0.05) in these differentiated proteins were measured by enzyme-linked immunosorbent assay in a validation cohort. The results showed that the levels of C-X-C motif chemokine ligand 12 (CXCL12) were higher in the urine of patients with TAPS than in those with OAPS ( p =0.035), while the levels of platelet-derived growth factor subunit B (PDGFB) were lower in patients with TAPS than in those with OAPS ( p =0.041). In addition, correlation analysis showed that CXCL12 levels were positively correlated with immunoglobulin G anti-β2-glycoprotein I antibody ( r =0.617, p =0.016). Our results demonstrated that urinary CXCL12 and PDGFB might serve as potential non-invasive markers to differentiate primary TAPS from primary OAPS.

    Abstract: Demand from consumers for small quantities of live swimming crab Portunus trituberculatus is rising with the development of e-commerce. However, it is challenging to keep P. trituberculatus alive post-capture. In this study, a transport bag containing oxygen and seawater (24 ppt) was used to investigate the survival and metabolic changes of P. trituberculatus during transport. The results showed that more than 80% P. trituberculatus could survive at least 24 h in the transport bag. The ability of the crabs to survive may be attributed to adaptive metabolism, as suggested by the switch from an aerobic to an anaerobic pathway for energy generation and a decline in amino acid metabolism, nucleic acid metabolism, and osmoregulation. Our findings suggest that the transport bag could effectively extend the post-capture survival time of P. trituberculatus . Metabolic adaptation – especially energy homeostasis – is crucial for crab survival during transport. Our study provides a promising method for the transport of live P. trituberculatus .

    Abstract: The Hongniu–Hongshan Cu skarn deposit is located in the central part of the Zhongdian porphyry and skarn Cu belt in southwestern China. Various elements, including Al, Ti, Li, K, Na, Ca, Fe, and Ge, have been completed by using scanning electron microscopy–cathodoluminescence (SEM-CL) and laser ablation inductively coupled plasma-mass spectrometry (LA-ICP-MS) on quartz phenocrysts from the Hongniu–Hongshan porphyry and skarn Cu deposit. Three quartz generations were identified in the porphyritic granite based on the CL textures and trace element signatures. Samples of the first-generation quartz (Qtz1) contain dark gray luminescent cores assaying 22–85 ppm Ti, 58–129 ppm Al, 4–18 ppm Li, and 0.43–40 ppm Fe. The Ti-in quartz geothermometer indicates crystallization temperatures of 593–664°C for Qtz1. Samples of the second-generation quartz (Qtz2) are characterized by clear bright gray oscillatory overgrowths with medium Ti concentrations of 51–70 ppm with elevated and variable amounts of Al, Li, and Fe, and low K. The Ti-in quartz geothermometer indicates crystallization temperatures of 672–706°C. Samples of the third-generation quartz (Qtz3) contain narrow light gray rims assaying 56–93 ppm Ti, 80–101 ppm Al, 11–19 ppm Li, 1.42–17 ppm K, and 2–25 ppm Fe. The Qtz3 crystallised crystallized at higher temperatures of 706–799°C. Our study indicates that the quartz phenocryst in the Hongniu–Hongshan porphyry may have undergone two mixing episodes involving a second hotter magma. Before the first resorption, average Ti concentration in the quartz phenocryst cores was 24 ppm, and Ti of the bright band halo at the edge of the crystal core is 56 ppm; the maximum temperature difference is 109°C higher than that of the pre-resorption. Furthermore, the Ti concentration is 75 ppm at the edge of the quartz phenocryst before resorption. After resorption, the average Ti increased to 81 ppm at maximum temperature difference of 54°C higher than that of pre-resorption. Moreover, on the basis of quartz composition collected from 14 different deposits and our new dataset, we propose that covariations of Ge/Al ratio can be used to effectively discriminate magmatic quartz and hydrothermal quartz. Magmatic quartz has a Ge/Al ratio of & lt;0.013, and the hydrothermal quartz has a ratio of & gt;0.013.

    Abstract: An eight-week experiment was conducted to evaluate the effects of eight light intensities (0, 0.83, 1.61, 8.17, 15.89, 31.38, 63.69, and 124.31 μmol m -2 s -1 ) on the survival, growth, antioxidant, molting-related and apoptosis-related gene expression of juvenile swimming crab Portunus trituberculatus . The results showed that the survival rate of crabs was the highest under the light intensities of 8.17 and 15.89 μmol m -2 s -1 . Under this light intensity, the crabs had the lowest hemolymph glucose (GLU) but the highest triglyceride (TG) content. The crabs in these treatments also had the highest total antioxidant capacity (T-AOC), superoxide dismutase (SOD), catalase (CAT) activities, and the lowest malondialdehyde (MDA) content. The growth performance (including weight gain and specific growth rate) and molting frequency of crabs increased with increasing light intensity. The improved growth performance and molting of 8.17 and 15.89 μmol m -2 s -1 groups were also accompanied by up-regulated of the retinoid-X receptor ( rxr ), ecdysone receptor ( ecr ), nuclear receptor E75 ( e75 ) gene expression, and down-regulated molt-inhibiting hormone ( mih ) gene expression. In addition, suboptimal light intensity (0, 0.83, 63.69, and 124.31 μmol m -2 s -1 ) significantly up-regulated the expression of apoptosis-related genes including c-Jun N-terminal kinase ( jnk ), tumor suppressor p53 ( p53 ), and B-cell lymphoma-2 ( bcl-2 ). Taken together, the suitable light intensity for the juvenile P. trituberculatus was estimated to be 9.96 μmol m -2 s -1 .

    Abstract: Mold spoilage of dairy products such as yogurt is a concern in dairy industry. Not only does it lead to substantial food waste, economic losses, and even brand image damage, but it may also cause public health concern due to the potential production of mycotoxin. Good hygiene practices are necessary to prevent contamination, but contamination may nevertheless occur at the production site and, not least, at the site of the consumer. In recent years, there has been a growing interest from consumers for “clean label” food products, which are natural, less-processed, and free of added, chemical preservatives, and a wish for shelf lives of considerable length in order to minimize food waste. This has sparked an interest in using lactic acid bacteria (LAB) or their metabolites as biopreservatives as a way to limit the growth of spoilage organisms in dairy products. A range of compounds produced by LAB with potential antifungal activity have been described as contributing factors to the inhibitory effect of LAB. More recently, growth inhibition effects caused by specific competitive exclusion have been elucidated. It has also become clear that the sensitivity toward both individual antifungal compounds and competition mechanisms differ among molds. In this review, the main spoilage molds encountered in dairy products are introduced, and an overview of the antifungal activity of LAB against different spoilage molds is presented including the main antifungal compounds derived from LAB cultures and the sensitivity of the spoilage molds observed toward these compounds. The recent findings of the role of competitive exclusion with emphasis on manganese depletion and the possible implications of this for biopreservation are described. Finally, some of the knowledge gaps, future challenges, and trends in the application of LAB biopreservation in dairy products are discussed.

    Abstract: Fungal spoilage of dairy products is a major concern due to food waste and economical losses, some fungal metabolites may furthermore have adverse effects on human health. The use of lactic acid bacteria (LAB) is emerging as a potential clean label alternative to chemical preservatives. Here, our aim was to characterize the growth potential at three storage temperatures (5, 16, and 25°C) of a panel of molds (four Mucor and nine Penicillium strains) isolated from dairy products, then investigate the susceptibility of the molds toward 12 LAB cultures. Fungal cell growth and morphology in malt extract broth was monitored using oCelloScope at 25°C for 24 h. Mucor plumbeus 01180036 was the fastest growing and Penicillium roqueforti ISI4 ( P. roqueforti ISI4) the slowest of the tested molds. On yogurt-agar plates, all molds grew at 5, 16, and 25°C in a temperature-dependent manner with Mucor strains growing faster than Penicillium strains regardless of temperature. The sensitivity toward 12 LAB cultures was tested using high-throughput overlay method and here all the molds except P. roqueforti ISI4 were strongly inhibited. The antifungal action of these LAB was confirmed when spotting mold spores on agar plates containing live cells of the LAB strains. However, if cells were removed from the fermentates, the inhibitory effects decreased markedly. The antifungal effects of volatiles tested in a plate-on-plate system without direct contact between mold and LAB culture media were modest. Some LAB binary combinations improved the antifungal activity against the growth of several molds beyond that of single cultures in yogurt serum. The role of competitive exclusion due to manganese depletion was examined as a possible antifungal mechanism for six Penicillium and two Mucor strains. It was shown that this mechanism was a major inhibition factor for the molds tested apart from the non-inhibited P. roqueforti ISI4 since addition of manganese with increasing concentrations of up to 0.1 mM resulted in partly or fully restored mold growth in yogurt. These findings help to understand the parameters influencing the mold spoilage of dairy products and the interactions between the contaminating strains, substrate, and bioprotective LAB cultures.

    Abstract: To clarify the effects of acute hyperglycemia on the responses of choroidal structural components and vascularity index during light modulation in healthy participants using techniques including image binarization and artificial intelligence (AI) segmentation based on swept-source optical coherence tomography (SS-OCT). Methods Twenty-four eyes of 24 healthy participants were imaged at different stages after ambient light, 40 min of dark adaptation, and 5 min of light adaptation in two imaging sessions: control and after receiving 75 g of oral glucose solution. The choroidal structural parameters, including luminal volume (LV), stromal volume (SV), total choroidal volume (TCV), and choroidal vascularity index (CVI) within a 6 mm area were determined using a custom algorithm based on image binarization and AI segmentation of SS-OCT. These measurements were compared among the conditions after adjusting for axial length, age to identify the differences. Results In the dark, CVI decreased (-0.36 ± 0.09%) significantly in acute hyperglycemia compared to the control condition. During the transition to ambient light, there was an increasing trend in the choroidal parameters compared with the control experiment. However, only TCV (0.38 ± 0.17 mm 3 ) and LV (0.27 ± 0.10 mm 3 ) showed a significant increase at the time point of 5 min after ambient light. Conclusion Analysis of choroidal structural parameters and CVI based on SS-OCT images is a potentially powerful method to objectively reflect subtle changes in neurovascular coupling between the choroid and photoreceptor during dark adaptation.

    Abstract: Background: Tumors frequently evade immune surveillance through multiple pathways to escape T cell recognition and destruction. Previous studies indicated that lipid metabolism alteration could affect the anti-tumor immunity of cancer cells. Nonetheless, the studies that investigated lipid metabolism-related gene for cancer immunotherapy are still few. Materials and methods: By mining the TCGA database, we screened out carnitine palmitoyltransferase-2 (CPT2), a key enzyme in the fatty acid β-oxidation (FAO) process associated with anti-tumor immunity. We then analyzed the gene expression and clinicopathological features of CPT2 using open-source platforms and databases. Molecular proteins interacting with CPT2 were also identified using web interaction tools. Subsequently, the relationship between CPT2 and survival was analyzed in cancer patients. Results: Our study revealed that CPT2 played a vital role in tumor microenvironment and immune response signaling pathways. We have also demonstrated that increased CPT2 gene expression could enhance the level of tumor immune cell infiltration. Furthermore, high CPT2 expression positively related with overall survival associated with immunotherapy. CPT2 expression was also associated with the prognosis of human cancers, suggesting that CPT2 may be a potential biomarker for predicting the efficacy of cancer immunotherapy. Conclusion: To the best of our knowledge, the relationship between CPT2 and tumor immune microenvironment was first proposed in this study. Therefore, further studies on CPT2 may provide new insights into the development of effective cancer immunotherapy.

    Abstract: Microbial community may systematically promote the development of fermentation process of foods. Traditional fermentation is a spontaneous natural process that determines a unique nutritional characteristic of crab paste of Portunus trituberculatus , However, rare information is available regarding the development pattern and metabolic role of bacterial community during the fermentation of crab paste. Here, using a 16S rRNA gene amplicon sequencing technology, we investigated dynamics of bacterial community and its relationship with metabolites during the fermentation of crab paste. The results showed that bacterial community changed dynamically with the fermentation of crab paste which highlighted by consistently decreased α-diversity and overwhelming dominance of Vibrio at the later days of fermentation. Vibrio had a positive correlation with trimethylamine, hypoxanthine, formate, and alanine while a negative correlation with inosine and adenosine diphosphate. In contrast, most of other bacterial indicators had a reverse correlation with these metabolites. Moreover, Vibrio presented an improved function potential in the formation of the significantly increased metabolites. These findings demonstrate that the inexorable rise of Vibrio not only drives the indicator OTUs turnover in the bacterial community but also has incriminated the quality of crab paste from fresh to perished.
