    Abstract: Low-temperature stress (LTS) drastically affects vegetative and reproductive growth in fruit crops leading to a gross reduction in the yield and loss in product quality. Among the fruit crops, temperate fruits, during the period of evolution, have developed the mechanism of tolerance, i.e., adaptive capability to chilling and freezing when exposed to LTS. However, tropical and sub-tropical fruit crops are most vulnerable to LTS. As a result, fruit crops respond to LTS by inducing the expression of LTS related genes, which is for climatic acclimatization. The activation of the stress-responsive gene leads to changes in physiological and biochemical mechanisms such as photosynthesis, chlorophyll biosynthesis, respiration, membrane composition changes, alteration in protein synthesis, increased antioxidant activity, altered levels of metabolites, and signaling pathways that enhance their tolerance/resistance and alleviate the damage caused due to LTS and chilling injury. The gene induction mechanism has been investigated extensively in the model crop Arabidopsis and several winter kinds of cereal. The ICE1 (inducer of C-repeat binding factor expression 1) and the CBF (C-repeat binding factor) transcriptional cascade are involved in transcriptional control. The functions of various CBFs and aquaporin genes were well studied in crop plants and their role in multiple stresses including cold stresses is deciphered. In addition, tissue nutrients and plant growth regulators like ABA, ethylene, jasmonic acid etc., also play a significant role in alleviating the LTS and chilling injury in fruit crops. However, these physiological, biochemical and molecular understanding of LTS tolerance/resistance are restricted to few of the temperate and tropical fruit crops. Therefore, a better understanding of cold tolerance’s underlying physio-biochemical and molecular components in fruit crops is required under open and simulated LTS. The understanding of LTS tolerance/resistance mechanism will lay the foundation for tailoring the novel fruit genotypes for successful crop production under erratic weather conditions.

    Abstract: The present study deals with an account of the primary productivity and trophic status of Rewalsar, Kuntbhyog, and Prashar lakes of Western Himalayas. Seasonal variations in gross primary productivity (GPP) (g C m –2 day –1 ), net primary productivity (NPP, respiration (R), and the NPP/R ratio have been studied and compared with hydrobiological factors. Among various factors, temperature, light, nutrients, and chlorophyll a were accountable for high productivity. Nygaard’s trophic state indices were calculated. GPP showed a positive relationship with water temperature, penetration of light, nitrate, and with phosphate. The extensive range of the compound index observed in Rewalsar (12.00–20.00) and Kuntbhyog lakes (12.50–18.00) confirmed their eutrophic nature. Based on the trophic classification of waters and on the basis of productivity, Nygaard’s trophic state indices, and richness of nutrients, Rewalsar and Kuntbhyog lakes could be categorized as eutrophic, whereas Prashar Lake, as oligo-eutrophic. Based on the presence, absence, frequency of appearance, and abundance of different organisms recorded during the present investigation, bioindicators of trophic status have been determined.

    Abstract: Wheat is one of the major staple cereal food crops in India. However, most of the wheat-growing areas experience several biotic and abiotic stresses, resulting in poor quality grains and reduced yield. To ensure food security for the growing population in India, there is a compelling need to explore the untapped genetic diversity available in gene banks for the development of stress-resistant/tolerant cultivars. The improvement of any crop lies in exploring and harnessing the genetic diversity available in its genetic resources in the form of cultivated varieties, landraces, wild relatives, and related genera. A huge collection of wheat genetic resources is conserved in various gene banks across the globe. Molecular and phenotypic characterization followed by documentation of conserved genetic resources is a prerequisite for germplasm utilization in crop improvement. The National Genebank of India has an extensive and diverse collection of wheat germplasm, comprising Indian wheat landraces, primitive cultivars, breeding lines, and collection from other countries. The conserved germplasm can contribute immensely to the development of wheat cultivars with high levels of biotic and abiotic stress tolerance. Breeding wheat varieties that can give high yields under different stress environments has not made much headway due to high genotypes and environmental interaction, non-availability of truly resistant/tolerant germplasm, and non-availability of reliable markers linked with the QTL having a significant impact on resistance/tolerance. The development of new breeding technologies like genomic selection (GS), which takes into account the G × E interaction, will facilitate crop improvement through enhanced climate resilience, by combining biotic and abiotic stress resistance/tolerance and maximizing yield potential. In this review article, we have summarized different constraints being faced by Indian wheat-breeding programs, challenges in addressing biotic and abiotic stresses, and improving quality and nutrition. Efforts have been made to highlight the wealth of Indian wheat genetic resources available in our National Genebank and their evaluation for the identification of trait-specific germplasm. Promising genotypes to develop varieties of important targeted traits and the development of different genomics resources have also been highlighted.

    Abstract: Globally, between one quarter and one-third of total grains produced each year are lost during storage mainly through infestation of insect pests. Among the available control options such as chemical and physical techniques, fumigation with aluminum phosphide (AlP) is so far considered the best control strategy against storage insect pests. However, these insect pests are now developing resistance against AIP due to its indiscriminate use due to non-availability of any effective alternative control option. Resistance to AIP among storage insect pests is increasing, and its inhalation has shown adverse effects on animals and human beings. Nanotechnology has opened up a wide range of opportunities in various fields such as agriculture (pesticides, fertilizers, etc.), pharmaceuticals, and electronics. One of the applications of nanotechnology is the usage of nanomaterial-based insecticide formulations for mitigating field and storage insect pests. Several formulations, namely, nanoemulsions, nanosuspensions, controlled release formulations, and solid-based nanopesticides, have been developed with different modes of action and application. The major advantage is their small size which helps in proper spreading on the pest surface, and thus, better action than conventional pesticides is achieved. Besides their minute size, these have no or reduced harmful effects on non-target species. Nanopesticides can therefore provide green and efficient alternatives for the management of insect pests of field and storage. However, an outcry against the utilization of nano-based pesticides is also revealed. It is considered by some that nano-insecticides may also have hazardous effects on humans as well as on the environment. Due to limited available data, nanopesticides have become a double-edged weapon. Therefore, nanomaterials need to be evaluated extensively for their large-scale adoption. In this article, we reviewed the nanoformulations that are developed and have proved effective against the insect pests under postharvest storage of grains.

    Abstract: Multiple myeloma (MM) is an incurable plasma cell malignancy with dose-limiting toxicities and inter-individual variation in response/resistance to the standard-of-care/primary drugs, proteasome inhibitors (PIs), and immunomodulatory derivatives (IMiDs). Although newer therapeutic options are potentially highly efficacious, their costs outweigh the effectiveness. Previously, we have established that clofazimine (CLF) activates peroxisome proliferator-activated receptor-γ, synergizes with primary therapies, and targets cancer stem-like cells (CSCs) in drug-resistant chronic myeloid leukemia (CML) patients. In this study, we used a panel of human myeloma cell lines as in vitro model systems representing drug-sensitive, innate/refractory, and clonally-derived acquired/relapsed PI- and cereblon (CRBN)-negative IMiD-resistant myeloma and bone marrow-derived CD138+ primary myeloma cells obtained from patients as ex vivo models to demonstrate that CLF shows significant cytotoxicity against drug-resistant myeloma as single-agent and in combination with PIs and IMiDs. Next, using genome-wide transcriptome analysis (RNA-sequencing), single-cell proteomics (CyTOF; Cytometry by time-of-flight), and ingenuity pathway analysis (IPA), we identified novel pathways associated with CLF efficacy, including induction of ER stress, autophagy, mitochondrial dysfunction, oxidative phosphorylation, enhancement of downstream cascade of p65-NFkB-IRF4-Myc downregulation, and ROS-dependent apoptotic cell death in myeloma. Further, we also showed that CLF is effective in killing rare refractory subclones like side populations that have been referred to as myeloma stem-like cells. Since CLF is an FDA-approved drug and also on WHO’s list of safe and effective essential medicines, it has strong potential to be rapidly re-purposed as a safe and cost-effective anti-myeloma drug.

    Abstract: During the second wave of the COVID-19 pandemic in India, the Ministry of Ayush conducted a community study to provide therapeutic care to patients with asymptomatic, mild, and moderate COVID-19 in home isolation based on the empirical evidence generated on the efficacy of AYUSH-64 in COVID-19. Objective To document disease characteristics, care-seeking behavior, and outcomes in patients with asymptomatic, mild, or moderate COVID-19 in home isolation who used AYUSH-64 for COVID-19. Methods Cross-sectional analysis of the data generated through a community study conducted in India from 08 May to 31 August 2021 was performed to study the disease characteristics, care-seeking behavior during home isolation, clinical outcomes, adverse events, and the association between various risk factors and clinical recovery during the study period. The data were collected through semi-structured questionnaires, available in electronic data collection format at the baseline, 7, 14, and 21 days. A logistic regression was performed to explore the relationship between relevant variables and clinical recovery. Results Data from 64,642 participants were analyzed for baseline assessment, and final analysis was done for 49,770 participants. The mean age of the enrolled participants was 38.8 ± 11.7 years, and 8.4% had co-morbidities. AYUSH-64 was utilized as an add-on to the standard care by 58.3% of participants. Comparable clinical outcomes were observed in participants utilizing AYUSH-64 either as a standalone or as an add-on to standard care, in terms of clinical recovery, disease progression, the requirement for oxygen supplementation, hospitalization, ICU admission, and need for ventilator support. Younger age, having no co-morbidities or substance abuse, and having been vaccinated were associated with early clinical recovery than those who were older and not vaccinated. Conclusions The study findings suggest that AYUSH-64 use, either standalone or as an adjunct to standard care, in asymptomatic, mild, or moderate COVID-19 is associated with good clinical outcomes. Ayush services and interventions can be effectively integrated into the mainstream public health architecture to serve public health goals.

    Abstract: Home gardening is an indigenous practice of cultivation that has effectively adapted to local ecological conditions over generations. This study examined the effects of disturbance and garden size on biodiversity to develop a better understanding of vegetation cover and its role in livelihood and provision of forest management in the Vindhyan highlands. Data were collected from 60 gardens which were classified into large ( & gt; 650 m 2 ), medium (400–650 m 2 ), and small ( & lt; 400 m 2 ), based on size and disturbance gradients viz., high, medium, and low. A total of 133 species from 50 families were recorded, in which trees (47.4%) were dominant followed by shrubs (18%) and herbs (16.5%). With respect to disturbance, the highest number of tree species (39) were found at low disturbance (LD) followed by 33 species in medium disturbance (MD) and 32 species in high disturbhance (HD). The total mean richness of species was greater at LD (20.3 ± 2.3) and lowest at HD (18.5 ± 2.2). Tree density was significantly ( P ≤ 0.05) higher at LD (293.75 ± 16.1 individual ha –1 ) as compared to MD (221 ± 11.5 individual ha –1 ) and HD (210 ± 10.3 individual ha –1 ). However, the results for shrubs and herbs density were considerably different, where shrubs density was highest at HD (70 ± 6.9 individual per 1,000 m 2 ) and lowest at LD (62.5 ± 5.8 individual per 1,000 m 2 ), while the maximum density of herbs was recorded at MD (466.25 ± 29.8 individual per 100 m 2 ) and minimum at LD (370 ± 21.4 individual per 100 m 2 ). The summed dominance ratio indicated frequent use of garden plants in bio-fencing, vegetables, ornamental, and ethnomedicine. Diversity ( P & lt; 0.01) and species richness ( P & lt; 0.05) showed a significant positive correlation with garden size. The Principal Component Analysis (PCA) showed that the first component (PC1) accounted for 28.6% of variance, whereas the second explained 21.9% of variance in both disturbance and garden size with a cumulative variance of 50.5%. These components depicted the positive association with HD (14.34), SDiv (13.91), TCD (12.47), and HDiv (12.09). We concluded that the diversity of home gardens changed with disturbance, which crucially served as a refuge for native tree species in a degraded landscape. This pattern highlighted the importance of home gardens for plant biodiversity conservation and local livelihood, which must be a viable option for regeneration of deforested dry tropics, while also reducing the burden on dry tropical forest regions.

    Abstract: Wheat ( Triticum aestivum L .) is a staple food crop for the global human population, and thus wheat breeders are consistently working to enhance its yield worldwide. In this study, we utilized a sub-set of Indian wheat mini core germplasm to underpin the genetic architecture for seed shape-associated traits. The wheat mini core subset (125 accessions) was genotyped using 35K SNP array and evaluated for grain shape traits such as grain length (GL), grain width (GW), grain length, width ratio (GLWR), and thousand grain weight (TGW) across the seven different environments (E 1 , E 2 , E 3 , E 4 , E 5 , E 5 , E 6 , and E 7 ). Marker-trait associations were determined using a multi-locus random-SNP-effect Mixed Linear Model (mrMLM) program. A total of 160 non-redundant quantitative trait nucleotides (QTNs) were identified for four grain shape traits using two or more GWAS models. Among these 160 QTNs, 27, 36, 38, and 35 QTNs were associated for GL, GW, GLWR, and TGW respectively while 24 QTNs were associated with more than one trait. Of these 160 QTNs, 73 were detected in two or more environments and were considered reliable QTLs for the respective traits. A total of 135 associated QTNs were annotated and located within the genes, including ABC transporter, Cytochrome450, Thioredoxin_M-type, and hypothetical proteins. Furthermore, the expression pattern of annotated QTNs demonstrated that only 122 were differentially expressed, suggesting these could potentially be related to seed development. The genomic regions/candidate genes for grain size traits identified in the present study represent valuable genomic resources that can potentially be utilized in the markers-assisted breeding programs to develop high-yielding varieties.

    Abstract: Accurate information about aerosol particle size distribution and its variation under different meteorological conditions are essential for reducing uncertainties related to aerosol-cloud-climate interaction processes. New particle formation (NPF) and the coagulation significantly affect the aerosol size distribution. Here we study the monthly and seasonal variability of aerosol particle size distribution at Delhi from December 2011 to January 2013. Analysis of aerosol particle size distribution using WRAS-GRIMM reveals that aerosol particle number concentration is highest during the post monsoon season owing to the effect of transported crop residue and biomass burning aerosols. Diurnal variations in number concentration show a bimodal pattern with two Aitken mode peaks in all the seasons. Monthly volume size distribution also shows bi-modal distribution with distinct coarse and fine modes. NPF events are observed less frequently in Delhi. Out of 222 days of WRAS data, only 17 NPF events have been observed, with higher NPF frequency during summer season. Growth rate of the nucleation mode of NPF events vary in the range 1.88–21.66 nm/h with a mean value of ∼8.45 ± 5.73 nm/h. It is found that during NPF events the Aitken and nucleation mode particles contribute more to the number concentration. Simultaneous measurement of UV flux and particulate matter (PM 10 and PM 2.5 ) have also been done along with particle number size distribution measurement to understand the possible mechanisms for NPF events over the study location.
