    Abstract: Urothelial Carcinoma of the bladder (BLCA) is the most prevalent cancer of the urinary system. In cancer patients, HRG fusion is linked to a poor prognosis. The prediction of HRG expression by imaging omics in BLCA has not yet been fully investigated. Methods HRG expression in BLCA and healthy adjoining tissues was primarily identified utilizing data sourced from The Cancer Genome Atlas (TCGA). Using Kaplan–Meier survival curves and Landmark analysis, the relationship between HRG expression, clinicopathological parameters, and overall survival (OS) was investigated. Additionally, gene set variation analysis (GSVA) was conducted and CIBERSORTx was used to investigate the relationship between HRG expression and immune cell infiltration. The Cancer Imaging Archive (TCIA) provided CT images that were used for prognostic analysis, radiomic feature extraction, and construction of the model, respectively. The HRG expression levels were predicted using the constructed and evaluated LR and SMV models. Results HRG expression was shown to be substantially lower in BLCA tumors as opposed to that observed in normal tissues (p & lt; 0.05). HRG expression had a close positive relationship with Eosinophils and a close negative relationship with B cells naive. The findings of the Landmark analysis illustrated that higher HRG was associated with improved patient survival at an early stage (P=0.048). The predictive performance of the two models, based on logistic regression analysis and support vector machine, was outstanding in the training and validation sets, yielding AUCs of 0.722 and 0.708, respectively, in the SVM model, and 0.727 and 0.662, respectively, in the LR.The models have good predictive efficiency. Conclusion HRG expression levels can have a significant impact on BLCA patients’ prognoses. The radiomic characteristics can successfully predict the pre-surgical HRG expression levels, based on CT- Image omics.

    Abstract: Introduction: Focused ultrasound ablation surgery (FUAS) has been emerging to treat a wide range of conditions non-invasively and effectively with promising therapeutic outcomes. The focusing capability of an ultrasound transducer (i.e., focus shift, beam distortion, and acoustic pressure at the focus) determines the ablation effects. However, the focus shift and focal beam distortion after ultrasound propagating through multi-layered heterogeneous viscoelastic biological tissues become significant and are found to deteriorate the performance of FUAS in clinics. Methods: To achieve an accurate and reliable focal field among patients with large variations in the anatomical structures and properties, a spherical cavity transducer with open ends and sub-wavelength focal size (Li et al., APL, 2013,102:204102) was applied here. Both experimental measurements and numerical simulations were performed to characterize the acoustic fields of the spherical cavity transducer in water, the multi-layered concentric cylindrical phantom, and the heterogeneous tissue model (an adult male pelvis enclosed by porcine skin, fat, and muscle) and then compared with those of a conventional concave transducer at the same electrical power output. Results: It is found that standing-wave focusing using the spherical cavity transducer results in much less focus shift (0.25λ vs . 1.67λ) along the transducer axis and focal beam distortion (−6 dB beam area of 0.71 mm 2 vs . 4.72 mm 2 in water and 2.55 mm 2 vs . 17.30 mm 2 in tissue) in the focal plane but higher pressure focusing gain (40.05 dB vs . 33.61 dB in tissue). Discussion: Such a highly accurate and reliable focal field is due to the excitation at an appropriate eigen-frequency of the spherical cavity with the varied media inside rather than the reverberation from the concave surface. Together with its sub-wavelength focal size, the spherical cavity transducer is technically advantageous in comparison to the concave one. The improved focusing capability would benefit ultrasound exposure for not only safer and more effective FUAS in clinics, but also broad acoustic applications.

    Abstract: Male infertility is a widespread health problem affecting approximately 6%–8% of the male population, and hypoxia may be a causative factor. In mammals, two types of hypoxia are known, including environmental and pathological hypoxia. Studies looking at the effects of hypoxia on male infertility have linked both types of hypoxia to poor sperm quality and pregnancy outcomes. Hypoxia damages testicular seminiferous tubule directly, leading to the disorder of seminiferous epithelium and shedding of spermatogenic cells. Hypoxia can also disrupt the balance between oxidative phosphorylation and glycolysis of spermatogenic cells, resulting in impaired self-renewal and differentiation of spermatogonia, and failure of meiosis. In addition, hypoxia disrupts the secretion of reproductive hormones, causing spermatogenic arrest and erectile dysfunction. The possible mechanisms involved in hypoxia on male reproductive toxicity mainly include excessive ROS mediated oxidative stress, HIF-1α mediated germ cell apoptosis and proliferation inhibition, systematic inflammation and epigenetic changes. In this review, we discuss the correlations between hypoxia and male infertility based on epidemiological, clinical and animal studies and enumerate the hypoxic factors causing male infertility in detail. Demonstration of the causal association between hypoxia and male infertility will provide more options for the treatment of male infertility

    Abstract: Poor response to immune checkpoint inhibitors (ICIs) has been observed in most triple-negative breast cancer (TNBC) cases (around 80%). Our aim was to investigate the status of mismatch repair (MMR), microsatellite instability (MSI), programmed death-ligand 1 (PD-L1), and lymphocyte-activation gene 3 (LAG-3) in TNBC. Methods A total of 74 TNBC samples were retrospectively analyzed. MMR and MSI were evaluated by immunohistochemistry (IHC) and polymerase chain reaction (PCR) using Promega 1.2 and NCI panels, respectively. PD-L1, LAG-3, and CD8 expression was assessed by IHC. Results None of the cases demonstrated deficient MMR (dMMR) or MSI. In total, 43/74 cases (58.1%) were PD-L1+, including 1 tumor PD-L1+, 25 tumor-infiltrating lymphocytes (TILs) PD-L1+, and 17 cases involving concurrence of tumor and TIL PD-L1+. The rate of TIL PD-L1+ was remarkably higher than that of tumor PD-L1+ (P & lt;0.001). We identified 20 LAG-3+ cases (27.0%, 20/74), all of which were PD-L1+. Co-expression of PD-L1 and LAG-3 was noted in 46.5% (20/43) of the PD-L1+ population. In the LAG-3+ subtype (co-expression of PD-L1 and LAG-3), high correlation between TILs PD-L1+ and LAG-3+ was observed (P & lt;0.01). A high frequency of CD8+ (98.6%, 73/74) was observed. Conclusion dMMR/MSI characteristics may not be a practical predictive marker for ICIs in TNBC. PD-L1+ is more common in TILs than in tumors. In the PD-L1+ population, approximately half of the cases showed LAG-3 co-expression. For patients with a poor response to PD-1(L1) mono ICI, dual blockade of PD-1(L1) and LAG-3 may be a viable option for the management of TNBC.

    Abstract: Humans have a natural ability to understand the emotions and feelings of others, whether one actually witnesses the situation of another, perceives it from a photograph, reads about it in a fiction book, or merely imagines it. This is the phenomenon of empathy, which requires us to mentally represent external information to experience the emotions of others. Studies have shown that individuals with high empathy have high anterior insula and adjacent frontal operculum activation when they are aware of negative emotions in others. As a negative emotion, disgust processing involves insula coupling. What are the neurophysiological characteristics for regulating the levels of empathy and disgust? To answer this question, we collected electroencephalogram microstates (EEG-ms) of 196 college students at rest and used the Disgust Scale and Interpersonal Reactivity Index. The results showed that: (1) there was a significant positive correlation between empathy and disgust sensitivity; (2) the empathy score and the intensity of transition possibility between EEG-ms C and D were significantly positively correlated; and (3) the connection strength between the transition possibility of EEG-ms C and D could adjust the relationship between the disgust sensitivity score and the empathy score. This study provides new neurophysiological characteristics for an understanding of the regulate relationship between empathy and disgust and provides a new perspective on emotion and attention.

    Abstract: B cell expansion with NF-κB and T cell anergy (BENTA) is a rare primary immunodeficiency disorder caused by gain-of-function (GOF) mutations in the CARD11 gene. Affected patients present with persistent B cell lymphocytosis in early childhood paired with lymphadenopathy and splenomegaly. Until now only six activating mutations from 14 patients have been reported in CARD11 . Here we report a patient from China with polyclonal B cell lymphocytosis and frequent infections in early life. A heterozygous mutation (c.377G & gt;A, G126D) in exon 5 of CARD11 gene (NM_032415) was identified by whole exome sequencing. In vitro functional studies showed that the G126D mutation is associated with increased expression of CARD11 and NF-κB activation in Hela cells. Flow cytometry analysis indicated NK cell activity and CD107a degranulation of the patient were decreased. RNA sequencing analysis showed that a number of genes in NF-κB pathway increased while those involved in NK cell activity and degranulation were down-regulated. In summary, our work identified a de novo germline GOF mutation in CARD11 with functional evidence of BENTA.

    Abstract: Lower diabetes prevalence and cardiovascular mortality have been observed in residents at a higher altitude. Leptin is associated with incident diabetes and adverse cardiovascular outcomes, and our aim was to investigate the association of living altitude with serum leptin concentrations. Methods Two cross-sectional surveys were used in this study, including native populations living at Tibet (high altitude) and Beijing (low altitude). A propensity score was conducted for matching age and body mass index (BMI) between native women at high and low altitude. Pearson’s correlation analysis was performed to evaluate the correlation of leptin with other variables. Results A total of 1414 native women were included in this study, including 594 at high altitude and 820 at low altitude. The serum leptin concentrations of native women living at high altitude were 13.74 ± 11.03 ng/ml, which was significantly lower than that of native women living at low altitude (20.90 ± 12.91 ng/ml). After matching age and BMI, women living at the high altitude still had lower serum leptin concentrations. After adjusting for the potential confounding factors, the correlation coefficient between Ln (leptin) and BMI of women at high altitude was significantly lower than that of women at low altitude (0.228 versus 0.559; P & lt; 0.0001). The serum leptin concentrations of each BMI subgroup ( & lt;18.5, 18.5 to & lt;25, 25 to & lt;30, ≥ 30 kg/m 2 ) in women at high altitude were lower than that in women at low altitude. Conclusions Serum leptin concentrations were significantly decreased in native women living at high altitude, and living altitude may alter the correlation of BMI and leptin. The findings of our study support that residents at high altitude have a protective effect with regards to improving cardiovascular and metabolic outcomes.
