    Abstract: Cryopyrin-associated periodic syndrome (CAPS) comprises a group of disorders characterized by recurrent bouts of systemic inflammation related to overactivation of inflammasome. So far, neither large cases of the correlation between genotype and phenotype nor treatment strategies have been clearly stated in China. Here, we studied the clinical and genetic characteristics and their correlation from 30 CAPS patients in China. We identified the pathogenesis for novel mutations by activating NLRP3 inflammasome for peripheral cells with ATP plus LPS, compared characteristics with other case series, and analyzed treatment outcomes of these patients. The patients harbored 19 substitutions in NLRP3 , and 8 of them were novel mutations. Among these novel mutations, percentages of severe musculoskeletal, ophthalmologic, and neurological symptoms were higher compared with other case serials. The correlation of phenotypes and their variants seemed different in our cases, such as T350M, S333G/I/R, and F311V (somatic mosaicism). Ten patients received Canakinumab treatment, which proved effective at alleviating musculoskeletal, neurological, auditory, visual manifestations, fever, and rash for 10–20 months follow-up. Patients treated with prednisolone or prednisolone plus thalidomide or methotrexate, tocilizumab, TNF inhibiting agents, and sirolimus achieved only partial remission. Importantly, we firstly identified somatic mosaicism mutation of F311V, which was severe. Our study extended the spectrum of genotype and phenotype and characteristics of their correlations and provided detailed responses to different treatment strategies. These data provide guidance for future diagnosis and management for CAPS.

    Abstract: Carbon storage is an important component of ecosystem services. Under climate warming and human activities, land use/land cover (LULC) have been undergoing tremendous change, leading to spatio-temporal variations in carbon storage. Based on seven series of LULC data and combined with carbon module of Integrated Valuation of Ecosystem Services and Tradeoffs (InVEST) model, spatial-temporal changes in LULC types and carbon storage were analyzed and estimated for Heilongjiang Province, Northeast China. Results show varied carbon storage among different types of LULC. Forest and cropland are the dominant LULC types in Heilongjiang Province, Northeast China, accounting for 46–49% (20.90 × 10 4 –22.12 × 10 4 km 2 ) and 30–37% (13.56 × 10 4 –16.70 × 10 4 km 2 ) of the total area. Areal extents of forest, grassland, and unused land declined from 1980 to 2015, by 1.22, 0.84, and 1.11 × 10 4 km 2 , respectively; while those of cropland and construction land expanded, by 3.14 and 0.08 × 10 4 km 2 , respectively. From 1980 to 2015, carbon storage displayed consistent change trends with those of LULC types: carbon storage of forest, grassland, and unused land decreased by 236.22, 116.61 and 21.82 × 10 6  Mg C, respectively; and those of cropland and construction land increased by 414.65 and 0.99 × 10 6  Mg C, respectively. The total carbon storage in the study region was 6,863.06 × 10 6 –6,907.64 × 10 6  Mg C, for which the forest, cropland, and grassland were the major contributor (6,778.75 × 10 6 –6,840.57 × 10 6  Mg C). Due to the conversion of large extents of forest, grassland, and unused land to cropland, which facilitated the formation of carbon sinks and thus enlarged the carbon storage by 45.36 × 10 6  Mg C from 1980 to 2015. Frequent forest fires, urban expansion, farmland reclamation, and engineering construction were the important factors of changes in the LULC, accelerating permafrost degradation and leading to obvious changes in the total carbon storage in the Heilongjiang Province, Northeast China. Therefore, the estimation of carbon storage in different LULC types can provide important data support and have important implications for evaluation of ecosystem services and carbon cycle.

    Abstract: Autosomal dominant hyper-IgE syndrome (AD-HIES) is a rare inherited primary immunodeficient disease (PIDs), which is caused by STAT3 gene mutations. Previous studies indicated a defective Toll-like receptor (TLR) 9-induced B cell response in AD-HIES patients, including proliferation, and IgG production. However, the other TLRs-mediated B cell responses in AD-HIES patients were not fully elucidated. In this study, we systematically studied the B cell response to TLRs signaling pathways in AD-HIES patients, including proliferation, activation, apoptosis, cytokine, and immunoglobulin production. Our results showed that the TLRs-induced B cell proliferation and activation was significantly impaired in AD-HIES patients. Besides, AD-HIES patients had defects in TLRs-induced B cell class switch, as well as IgG/IgM secretion and IL-10 production in B cells. Taken together, we first systematically reported the deficiency of TLRs driven B cell response in AD-HIES patients, which help to have a better understanding of the pathology of AD-HIES.

    Abstract: Globally, liver cancer poses a serious threat to human health and quality of life. Despite numerous studies on the microbial composition of the gut in hepatocellular carcinoma (HCC), little is known about the interactions of the gut microbiota and metabolites and their role in HCC. This study examined the composition of the gut microbiota and serum metabolic profiles in 68 patients with HCC, 33 patients with liver cirrhosis (LC), and 34 healthy individuals (NC) using a combination of metagenome sequencing and liquid chromatography−mass spectrometry (LC−MS). The composition of the serum metabolites and the structure of the intestinal microbiota were found to be significantly altered in HCC patients compared to non-HCC patients. LEfSe and metabolic pathway enrichment analysis were used to identify two key species ( Odoribacter splanchnicus and Ruminococcus bicirculans ) and five key metabolites (ouabain, taurochenodeoxycholic acid, glycochenodeoxycholate, theophylline, and xanthine) associated with HCC, which then were combined to create panels for HCC diagnosis. The study discovered that the diagnostic performance of the metabolome was superior to that of the microbiome, and a panel comprised of key species and key metabolites outperformed alpha-fetoprotein (AFP) in terms of diagnostic value. Spearman’s rank correlation test was used to determine the relationship between the intestinal flora and serum metabolites and their impact on hepatocarcinogenesis and progression. A random forest model was used to assess the diagnostic performance of the different histologies alone and in combination. In summary, this study describes the characteristics of HCC patients’ intestinal flora and serum metabolism, demonstrates that HCC is caused by the interaction of intestinal flora and serum metabolites, and suggests that two key species and five key metabolites may be potential markers for the diagnosis of HCC.
