    Abstract: Eight Gram-negative, aerobic, motile with paired polar flagella and rod-shaped bacteria were isolated from six tobacco fields in Yunnan, PR China. 16S rRNA gene sequence analysis revealed that all the strains belonged to the genus Ralstonia . Among them, strain 22TCCZM03-6 had an identical 16S rRNA sequence to that of R. wenshanensis 56D2 T , and the other strains were closely related to R. pickettii DSM 6297 T (98.34–99.86%), R. wenshanensis 56D2 T (98.70–99.64%), and R. insidiosa CCUG 46789 T (97.34–98.56%). Genome sequencing yielded sizes ranging from 5.17 to 5.72 Mb, with overall G + C contents of 63.3–64.1%. Pairwise genome comparisons showed that strain 22TCCZM03-6 shared average nucleotide identity (ANI) and digital DNA–DNA hybridization (dDDH) values above the species cut-off with R. wenshanensis 56D2 T , suggesting that strain 22TCCZM03-6 is a special strain of the R. wenshanensis . Five strains, including 21MJYT02-10 T , 21LDWP02-16, 22TCJT01-1, 22TCCZM01-4, and 22TCJT01-2, had ANI values & gt;95% and dDDH values & gt;70% when compared with each other. These five strains had ANI values of 73.32–94.17% and dDDH of 22.0–55.20% with the type strains of the genus Ralstonia individually, supporting these five strains as a novel species in the genus Ralstonia . In addition, strains 21YRMH01-3 T and 21MJYT02-11 T represent two independent species. They both had ANI and dDDH values below the thresholds for species delineation when compared with the type species of the genus Ralstonia . In strains 21YRMH01-3 T and 21MJYT02-10 T , the main fatty acids were summed features 3, 8, and C 16:0 ; however, strain 21MJYT02-11 T contained C 16:0 , cyclo-C 17:0 , and summed features 3 as major fatty acids. The main polar lipids, including diphosphatidylglycerol, phosphatidylglycerol, and phosphatidylethanolamine, were identified from strains 21YRMH01-3 T , 21MJYT02-10 T , and 21MJYT02-11 T . The ubiquinones Q-7 and Q-8 were also detected in these strains, with Q-8 being the predominant quinone. Based on the above data, we propose that the eight strains represent one known species and three novel species in the genus Ralstonia , for which the names Ralstonia chuxiongensis sp. nov., Ralstonia mojiangensis sp. nov., and Ralstonia soli sp. nov. are proposed. The type strains are 21YRMH01-3 T (=GDMCC 1.3534 T  = JCM 35818 T ), 21MJYT02-10 T (=GDMCC 1.3531 T  = JCM 35816 T ), and 21MJYT02-11 T (=GDMCC 1.3532 T  = JCM 35817 T ), respectively.

    Abstract: Sphingosine-1-phosphate (S1P) is a signaling lipid involved in many biological processes, including inflammatory and immune regulatory responses. The study aimed to determine whether admission S1P levels are associated with disease severity and prognosis after spontaneous intracerebral hemorrhage (ICH). Methods Data of 134 patients with spontaneous ICH and 120 healthy controls were obtained from Biological Resource Sample Database of Intracerebral Hemorrhage at the First Affiliated Hospital of Zhengzhou University. Plasma S1P levels were measured. Regression analyses were used to analyze the association between S1P levels and admission and 90-day modified Rankin scale (mRS) scores. Receiver operating characteristic (ROC) curves assessed the predictive value of S1P levels for ICH severity and prognosis. Results Patients with ICH exhibited elevated plasma S1P levels compared to the control group (median 286.95 vs. 239.80 ng/mL, p & lt; 0.001). When divided patients into mild-to-moderate and severe groups according to their mRS scores both at admission and discharge, S1P levels were significantly elevated in the severe group compared to the mild-to-moderate group (admission 259.30 vs. 300.54, p & lt; 0.001; 90-day 275.24 vs. 303.25, p & lt; 0.001). The patients were divided into three groups with different concentration gradients, which showed significant statistical differences in admission mRS scores (3 vs. 4 vs. 5, p & lt; 0.001), 90-day mRS scores (2.5 vs. 3 vs. 4, p & lt; 0.001), consciousness disorders (45.5% vs. 68.2% vs. 69.6%, p = 0.033), ICU admission (29.5% vs. 59.1% vs. 89.1%, p & lt; 0.001), surgery (15.9% vs. 47.7% vs. 82.6%, p & lt; 0.001), intraventricular hemorrhages (27.3% vs. 61.4% vs. 65.2%, p & lt; 0.001) and pulmonary infection (25% vs. 47.7% vs. 84.8%, p & lt; 0.001). Multivariate analysis displayed that S1P level was an independent risk factor for disease severity (OR = 1.037, 95% CI = 1.020–1.054, p & lt; 0.001) and prognosis (OR = 1.018, 95% CI = 1.006–1.030, p = 0.003). ROC curves revealed a predictive value of S1P levels with an area under the curve of 0.7952 (95% CI = 0.7144–0.8759, p & lt; 0.001) for disease severity and 0.7105 (95% CI = 0.6227–0.7983, p & lt; 0.001) for prognosis. Conclusion Higher admission S1P is associated with worse initial disease severity and 90-day functional outcomes in intracerebral hemorrhage.
