    Abstract: Liver biomarkers and metabolic associated fatty liver disease (MAFLD) have been shown to be associated with cardiovascular disease (CVD). However, there is limited evidence on CVD subtypes [myocardial infarction (MI), ischemic stroke (IS), and intracerebral hemorrhage (ICH)], especially in the Chinese population. We examined these associations overall, by genetic predisposition to non-alcoholic fatty liver disease (NAFLD), and by lifestyle risk factors. Approach and results This is a nested case-control study of CVD (10,298 cases and 5,388 controls) within the China Kadoorie Biobank. Cox regression was used to estimate adjusted hazard ratios (HRs) for CVD associated with liver biomarkers and MAFLD and by stratum of genetic risk and a combined high-risk lifestyle score. For liver enzymes, there were positive associations with MI and IS, but no associations with ICH or carotid plaque. There were positive associations of NAFLD with risks of MI, IS, and ICH (HR 1.43 [95% CI 1.30–1.57], 1.25 [1.16–1.35] , and 1.12 [1.02–1.23]) as well as carotid plaque (odds ratio 2.36 [1.12–4.96] ). The associations of NAFLD with CVD and carotid plaque were stronger among individuals with a high genetic risk (ICH: p -interaction & lt; 0.05), while the associations with stroke were stronger among those with a favorable lifestyle ( p -interaction & lt; 0.05). The results for MAFLD mirrored those for NAFLD. Conclusion In Chinese adults, liver biomarkers and MAFLD were associated with risk of CVD, with different magnitudes of associations by CVD subtypes. Genetic predisposition to NAFLD and lifestyle factors modified the associations of fatty liver with stroke.

    Abstract: Colorectal cancer (CRC) is one of the most common malignancies and its incidence and mortality are increasing yearly. 5-Fluorouracil (5-FU) has long been used as a standard first-line treatment for CRC patients. Although 5-FU-based chemotherapy is effective for advanced CRC, the consequent resistance remains a key problem and causes the poor prognosis of CRC patients. Thus, there is an urgent need to identify new biomarkers to predict the response to 5-FU-based chemotherapy. Methods CRC samples were retrieved from Gene Expression Omnibus (GEO) and The Cancer Genome Atlas (TCGA). The immune-related genes were retrieved from the ImmPort database. Single-cell sequencing results from colorectal cancer were obtained by the ArrayExpress database. 5-FU resistance-related genes were filtered and validated by R packages. ESTIMATE algorithms were used to assess the tumor microenvironment (TME). KEGG and GO analysis were performed to explore the biological signaling pathway for resistant-response patients and sensitive-response patients in the tumor microenvironment. pRRophetic algorithms were used to predict 5-FU sensitivity. GSEA and GSVA analysis was performed to excavate the biological signaling pathway of the RBP7 gene. Results Nine immune-related genes were identified to be associated with 5-FU resistance and poor disease-free survival (DFS) of CRC patients and the signature of these genes was developed in a DFS-prognostic model. Four immune-related genes were determined to be associated with 5-FU resistance and overall survival (OS) of CRC patients. The signature of these genes was developed an OS-prognostic model. ESTIMATE scores showed a significant difference between 5-FU resistant and 5-FU sensitive CRC patients. Resistant-response patients and sensitive-response patients to 5-FU based chemotherapy showed different GO and KEGG enrichment on the tumor microenvironment. RBP7, as a tumor immune microenvironment (TIME) related gene, was found to have the potential of predicting chemotherapy resistance and poor prognosis of CRC patients. GSEA analysis showed multiple signaling differences between the high and low expression of RBP7 in CRC patients. Hypoxia and TNFα signaling via NFκB gene sets were significantly different between chemotherapy resistant (RBP7 High ) and chemotherapy sensitive (RBP7 Low ) patients. Single-cell RNA-seq suggested RBP7 was centrally distributed in endothelial stalk cells, endothelial tip cells, and myeloid cells. Conclusions Immune-related genes will hopefully be potential prognostic biomarkers to predict chemotherapy resistance for CRC. RBP7 may function as a tumor microenvironment regulator to induce 5-FU resistance, thereby affecting the prognosis of CRC patients.

    Abstract: Participation in mass sports is one of the most efficient strategies for people to attain physical and mental health in China. Prosocial behavior has a positive effect on social development. This study developed a conceptual model with mass sports activity as the independent variable, prosocial behavior as the dependent variable, and flow trait and subjective wellbeing as the mediating variables. Methods Participants ( N = 351) completed an online survey. Mass sports activity, flow trait, subjective wellbeing, and prosocial behavior were measured using the physical activity rank scale-3 (PARS-3), short dispositional flow scale (SDFS), index of wellbeing (IWB), and prosocial tendencies measure (PTM), respectively. Descriptive statistics compared differences between sports population (PARS-3, ≥ 36) and non-sports population (PARS-3, & lt;36). Mediation effect was analyzed using the PROCESS (Template, Model 6). Results Sports population scored significantly higher (all P ≤ 0.05) on SDFS, IWB, and PTM than non-sports population. Participation in mass sports stimulated flow trait and thus improved prosocial behavior, with a mediation effect value of 0.061 (95% CI , 0.028–0.104), which accounted for 30.18% of the total effect. Participation in mass sports enhanced subjective wellbeing and thus improved prosocial behavior, with a mediation effect value of 0.044 (95% CI , 0.007–0.090), which accounted for 21.96% of the total effect. Flow trait and subjective wellbeing mediated the relationship between mass sports activity and prosocial behavior in a sequential manner, with a mediation effect value of 0.059 (95% CI , 0.035–0.090), which accounted for 29.23% of the total effect. Conclusion The preliminary results of the mediation model validated the hypothesized sequential links between mass sports activity, flow trait, subjective wellbeing, and prosocial behavior. Greater participation in mass sports increases the likelihood of prosocial behavior.

    Abstract: The conserved Dis1/Stu2/XMAP215 microtubule association proteins (MAPs) family plays an important role in microtubule dynamics, nucleation, and kinetochore-microtubule attachments. However, function of Dis1/Stu2/XMAP215 homolog in plant pathogenic fungi has not been determined. Here, we identified and investigated the Dis1/Stu2/XMAP215 homolog (FGSG_10528) in Fusarium graminearum ( Fg Stu2p). Co-localization experiment and co-immunoprecipitation (Co-IP) assay demonstrated that Fg Stu2p is a microtubule associated protein. Besides, Fg Stu2 could also interact with Fg γ-tubulin and presumed Fg Ndc80, which suggested that the Fg Stu2 gene might associate with microtubule nucleation and kinetochore-microtubule attachments like Dis1/Stu2/XMAP215 homologs in other species. Moreover, the Fg Stu2 promoter replacement mutants ( Fg Stu2-Si mutants) produced twisted hyphae and decreased growth rate. Microscope examination further showed that the microtubule polymerization was reduced in Fg Stu2-Si mutants, which could account for the aberrant morphology. Although the microtubule polymerization was affected in Fg Stu2-Si mutants, the Fg Stu2-Si mutants didn’t show highly increased sensitivity to anti-microtubule fungicide carbendazim (methyl benzimidazol-2-ylcarbamate [MBC]). In addition, the Fg Stu2-Si mutants exhibited curved conidia, decreased number of conidial production, blocked ability of perithecia production, decreased pathogenicity and deoxynivalenol (DON) production. Taken together, these results indicate that the Fg Stu2 gene plays a crucial role in vegetative growth, morphology, sexual reproduction, asexual reproduction, virulence and deoxynivalenol (DON) production of F. graminearum , which brings new insights into the functions of Dis1/Stu2/XMAP215 homolog in plant pathogenic fungi.
