    Abstract: Hashimoto’s thyroiditis (TH) is a risk factor for the occurrence of papillary thyroid carcinoma (PTC), which is considered to be the most common type of thyroid cancer. In recent years, the prevalence of PTC with TH has been increasing, but little is known about the genetic alteration in PTC with TH. This study analyzed the mutation spectrum and mutation signature of somatic single nucleotide variants (SNV) for 10 non-tumor and tumor pair tissues of PTC with TH using whole-exome sequencing. The ANK3 protein expression was evaluated by immunohistochemistry in PTC with TH and PTC samples. Moreover, the functional role of ANK3 in PTC cells was determined by CCK-8 proliferation assay, colony formation assays, cell cycle analysis, cell invasion and migration and in vivo study through overexpression assay. Our results showed three distinct mutational signatures and the C & gt;T/G & gt;A substitution was the most common type of SNV. Gene-set enrichment analysis showed that most of the significantly mutated genes were enriched in the regulation of actin cytoskeleton signaling. Moreover, NCOR2, BPTF, ANK3 , and PCSK5 were identified as the significantly mutated genes in PTC with TH, most of which have not been previously characterized. Unexpectedly, it was found that ANK3 was overexpressed in cytoplasm close to the membrane of PTC cells with TH and in almost all PTC cases, suggesting its role as a diagnostic marker of PTC. Ectopic expression of ANK3 suppressed invasion and migration, increased apoptosis of B-CPAP and TPC-1 cells. Moreover, our findings revealed that enhanced ANK3 expression inhibits growth of PTC cells both in vitro and in vivo . Ectopic expression of ANK3 significantly enhanced E-cadherin protein expression and inhibited PTC progression, at least in part, by suppression of epithelial-mesenchymal transition (EMT). Our study shows that ANK3 exerts an anti-oncogenic role in the development of PTC and might be an indolent maintainer of PTC.

    Abstract: The shortage of land resources restricts the sustainable development of agricultural production. Multiple cropping has been widely used in Southern China, but whether the continuous planting will cause a decline in soil quality and crop yield is unclear. To test whether multiple cropping could increase grain yield, we investigated the farmlands with different cultivation years (10–20 years, 20–40 years, and & gt;40 years). Results showed that tobacco-rice multiple cropping rotation significantly increased soil pH, nitrogen nutrient content, and grain yield, and it increased the richness of the bacterial community. The farmland with 20–40 years of cultivation has the highest soil organic carbon (SOC), ammonium nitrogen, and grain yield, but there is no significant difference in the diversity and structure of the bacterial community in farmlands with different cultivation years. The molecular ecological network indicated that the stability of the bacterial community decreased across the cultivation years, which may result in a decline of farmland yields in multiple cropping system & gt; 40 years. The Acidobacteria members as the keystone taxa (Zi ≥ 2.5 or Pi ≥ 0.62) appeared in the tobacco-rice multiple cropping rotation farmlands, and the highest abundance of Acidobacteria was found in the farmland with the highest SOC and ammonium nitrogen content, suggesting Acidobacteria Gp4, GP7, GP12 , and GP17 are important taxa involved in the soil carbon and nitrogen cycle. Therefore, in this study, the multiple cropping systems for 20 years will not reduce the crop production potential, but they cannot last for more than 40 years. This study provides insights for ensuring soil quality and enhancing sustainable agricultural production capacity.

    Abstract: As the core component of the automotive air conditioning cooling system, the automotive cooling fan directly affects the cooling performance of the entire system. Therefore, it is necessary to study the flow field and noise of the automotive cooling fan. This study focuses on the analysis off low field characteristics and noise generation in an automotive air conditioning systems, specifically related to an automobile cooling fan at different speeds. The flow field properties of the fan are examined through the distribution off low line trajectory, pressure distribution on the blade surface, and velocity distribution characteristics at various speeds. The analysis reveals that the maximum air velocity occurs at the point where the fan blade meets the wind guard, while the maximum pressure is observed at this point and at the edge where the fan blade cuts through the cavity. Furthermore, a monitoring point located 1 m in front of the fan is selected to study the average value of fan pneumatic noise pulsation and generate a noise cloud map, aiming to identify the main noise source of the fan. To better understand the noise distribution and variation at different speeds, various noise metrics are utilized, including noise Sound Pressure Level (SPL) spectrum, 1/3-octave SPL spectrum, and 1/3-octave A-weighted SPL spectrum. The noise characterization results indicate that the primary areas of fan noise generation are located at the tip of the leaf margin of the forward swept wing. This research provides valuable insights for the improvement and optimization of the cooling fan design, as well as subsequent noise reduction efforts.

    Abstract: SARS-Cov-2 caused the COVID-19 pandemic worldwide. ADAM17 functions as a disintegrin and transmembrane metalloproteinase domain protein involved in the regulation of SARS-CoV-2 receptor ACE2. However, its impact on cancer patients infected with COVID-19 and its correlation with immune cell infiltration is unclear. This study compared ADAM17 expression between normal and tumor tissues based on GEPIA. The correlations between ADAM17 expression and immune cell infiltration and immunomodulators were investigated. Besides, treated drugs for targeting ADAM17 were searched in the TISDB database. We found that ADAM17 was highly conserved in many species and was mainly expressed in lung, brain, female tissues, bone marrow and lymphoid tissues. It was also highly expressed in respiratory epithelial cells of rhinitis and bronchus. ADAM17 expression in tumors was higher than that in several paired normal tissues and was negatively correlated with the prognosis of patients with malignant tumors. Interestingly, ADAM17 expression significantly correlated with immunomodulators and immune cell infiltration in normal and tumor tissues. Moreover, eight small molecules targeting ADAM17 only demonstrate therapeutic significance. These findings imply important implications for ADAM17 in cancer patients infected with COVID-19 and provide new clues for development strategy of anti-COVID-19.

    Abstract: Carbonate buried hill madeexploration breakthroughs recently in the offshore Bohai Bay Basin, China, but the plane distribution of the buried hill reservoirs are unclear due to the highly heterogeneous. Taking the CFD2 oilfield as an example, based on core, thin section, seismic, and well logging data, the characteristics of Carbonate buried hill reservoirs in the study area were clarified, the formation mechanism of the reservoirs was discussed, and the development model of the reservoir was established. The results show that the reservoirs are mainly fractured-dissolved reservoirs, and the formation of the reservoirs is mainly related to structural fractures and fluid dissolution along the fractures. The NWW-trending structural fractures were formed under the control of the Indosinian compression, and the NEE-trending structural fractures were formed under the control of the Yanshanian strike-slip transpression. Dolomite is more brittle than limestone and is the main lithology for forming effective fractures. Structural fractures provide favorable channels for atmospheric water dissolution. The C and O isotope values reveal that at least two stages of dissolution have occurred in the study area which are supergene karstification and burial karstification. A model of the fractured-dissolved reservoir under the control of “structure-lithology-fluid” was established. This model highlights that the structural fractures formed by tectonic activities are crucial to reservoir development, and lithology is the internal factor controlling reservoir distribution. Dolomite exhibits the compressive strength of only half of the limestone, and it is the dominant lithology for reservoir development. The dissolution of atmospheric water in the two stages along the fractures greatly improved the physical properties of the reservoirs, and it is the guarantee for the development of effective reservoirs.

    Abstract: Microvascular invasion (MVI) is highly associated with poor prognosis in patients with liver cancer. Predicting MVI before surgery is helpful for surgeons to better make surgical plan. In this study, we aim at establishing a nomogram to preoperatively predict the occurrence of microvascular invasion in liver cancer. Method A total of 405 patients with postoperative pathological reports who underwent curative hepatocellular carcinoma resection in the Third Affiliated Hospital of Sun Yat-sen University from 2013 to 2015 were collected in this study. Among these patients, 290 were randomly assigned to the development group while others were assigned to the validation group. The MVI predictive factors were selected by Lasso regression analysis. Nomogram was established to preoperatively predict the MVI risk in HCC based on these predictive factors. The discrimination, calibration, and effectiveness of nomogram were evaluated by internal validation. Results Lasso regression analysis revealed that discomfort of right upper abdomen, vascular invasion, lymph node metastases, unclear tumor boundary, tumor necrosis, tumor size, higher alkaline phosphatase were predictive MVI factors in HCC. The nomogram was established with the value of AUROC 0.757 (0.716–0.809) and 0.768 (0.703–0.814) in the development and the validation groups. Well-fitted calibration was in both development and validation groups. Decision curve analysis confirmed that the predictive model provided more benefit than treat all or none patients. The predictive model demonstrated sensitivity of 58.7%, specificity of 80.7% at the cut-off value of 0.312. Conclusion Nomogram was established for predicting preoperative risk of MVI in HCC. Better treatment plans can be formulated according to the predicted results.
