    Notes: Our studies observed that, consistent with the literature, ischemic/hypoxic insults increased the expression of voltage-gated potassium channel (Kv) 1.2 potassium channel as well as elevating the endogenous level of vascular endothelial growth factor (VEGF) in neurons of adult rat brain following middle cerebral artery occlusion and in SH-SY5Y cells after hypoxia and glucose deprivation. Concomitantly, we also observed that ischemic injury increased the tyrosine phosphorylation of Kv 1.2 in in vivo and in vitro; the introduction of exogenous VEGF could attenuate cell death in in vitro models. Furthermore, we found that the protective effect of VEGF is mediated through its up-regulative actions on the tyrosine phosphorylation of Kv 1.2, which in turn has a direct influence on cell viability after ischemic insult. In substantiation of this result, we used anti-sense methodology to suppress the expression of endogenous VEGF, which significantly inhibited the tyrosine phosphorylation of Kv 1.2 and increased cell death elicited by ischemic/hypoxic injury. Finally, the enhancement of the tyrosine phosphorylation of the channel by VEGF in neuronal cells was significantly attenuated in the presence of wortmannin, an inhibitor of phosphatidylinositol 3-kinase (PI3-K), or genestin, an inhibitor of tyrosine kinase, thus suggesting that the phosphorylation of Kv 1.2 induced by VEGF is mechanistically linked to the PI3-K pathway.

    Notes: An orthorhombic crystal form of trp repressor (aporepressor plus L-tryptophan ligand) was solved by molecular replacement, refined to 1.65 Å resolution, and compared to the structure of the repressor in trigonal crystals. Even though these two crystal forms of repressor were grown under identical conditions, the refined structures have distinctly different conformations of the DNA-binding domains. Unlike the repressor/aporepressor structural transition, the conformational shift is not caused by the binding or loss of the L-tryptophan ligand. We conclude that while L-tryptophan binding is essential for forming a specific complex with trp operator DNA, the corepressor ligand does not lock the repressor into a single conformation that is complementary to the operator. This flexibility may be required by the various binding modes proposed for trp repressor in its search for and adherence to its three different operator sites.

    Notes: A series of novel symmetrical and asymmetrical 2,5-disubstituted-1,3,4-oxadiazoles and their intermediates were synthesized. A convenient procedure for preparation of aromatic nitrile and ester intermediates is reported. The insecticidal activities of oxadiazoles against Dropsophila melanogaster were recorded, and analysis of structure-activity relationships showed that the highest occupied molecular orbital energy (EH) was the main factor to affect the bioactivities.

    Notes: Liposomes, which mimic biomembranes, were used as a pseudostationary phase in capillary zone electrophoresis. The decrease in the mobility of an analyte owing to the presence of liposomes reflected interaction between the analyte and the liposomes. Equations were derived to calculate the specific capacity factor Ks (the capacity factor, K′, normalized to the liposome concentration 1 M) from the migration times and to estimate the difference in free energy, Δ(ΔG0), of the weak analyte/liposome interactions. The order of Ks values for the drugs tested was aspirin 〈 salicylic acid 〈 warfarin 〈 sulfasalazine. The peptide TyrGlySerThrProGlyCysCys interacted more strongly with the liposomes (Ks = 10.1 M-1) than did TyrGlySerThrProGlySerSer (Ks = 9.1 M-1). These results were similar to those obtained earlier by immobilized liposome chromatography.

    Description: Quantitative proteomics can be used as a screening tool for identification of differentially expressed proteins as potential biomarkers for cancers. Here, we comparatively analyzed the proteome profiles of ovarian cancer tissues and normal ovarian epithelial tissues. Using the high-throughput proteomic technology of isobaric tags for relative and absolute quantitation (iTRAQ)-coupled with two-dimensional-liquid chromatography-tandem mass spectrometry, 1259 unique proteins were identified. Of those, 205 were potentially differentially expressed between ovarian cancer and normal ovarian tissues. Several of the potentially differentially expressed proteins were validated by Western blotting and real-time quantitative RT-PCR analyses. Furthermore, up-regulation of KRT8, PPA1, IDH2, and S100A11 were validated in ovarian tissue microarrays by immunohistochemistry. Silencing of S100A11 expression suppressed the migration and invasion properties of ovarian cancer cells in vitro. Our study represents the successful application of iTRAQ technology to an investigation of ovarian cancer. Many of the potentially differentially expressed proteins identified had not been linked to ovarian cancer before, and provide valuable novel insights into the underlying mechanisms of carcinogenesis in human ovarian cancer.     Description: The great achievement of the development of intensive in agriculture in China can be partly attributed to substantial increases in mineral-nutrient application. However, whereas farmers tend to apply high levels of nitrogen (N) and phosphorus (P) application of potassium (K) has been neglected. A greater understanding of the relationship between maize ( Zea mays L.) grain yield and K-application rate is thus required to provide an improved rationale for K fertilization for farmers in the various agro-ecological regions of China. In this study, a total of 2765 farmers' survey data and 3124 on-farm experiments across major maize agro-ecological regions in China were collected and evaluated for farmers' K-management status and to determine grain-yield response to K application. Nationally, the average K-application rate on farms was 26 kg K ha –1 and varied from 0 to 158 kg K ha –1 , with a coefficient of variation of 107%, but the applied K-fertilizer rates were not related to grain yield. Maize grain yields at recommended K rates increased by 14.0%, 14.7%, 19.4%, and 4.3% in Northeast China, North China Plain, Southwest China, and Northwest China, respectively, compared to zero K fertilization (K 0 ). Increased yield due to K fertilization (IY max , difference between maximum yield across all treatments and K 0 -treatment yield for each experiment) averaged 1.4 t ha –1 but varied widely in different agro-ecological regions. Soil extractable K (NH 4 OAc-K) and intercounty variation resulted in large variation in IY max in agro-ecological regions, as did other factors, such as use of particular maize hybrids, soil types, or years in different regions.

    Description: Bioactivity-guided isolation of the rare gorgonian Muricella sibogae ( Nutting ) yielded the two new eunicellin diterpenes sibogin A and B ( 1 and 2 ), the three new 9,10-secosteroids sibogol A–C ( 6 – 8 ), together with the three known eunicellin diterpenes 3 – 5 and the five known 9,10-secosteroids 9 – 13 . Their structures were established by extensive spectral analysis (1D- and 2D-NMR, IR, and MS). The cytotoxicity of the isolates 1 – 13 was evaluated in vitro against the selected tumor cell lines P388 and BEL-7402. All the compounds showed only weak activity against P388 cell lines, with an inhibition rate ranging from 10 to 60% at a concentration of 50 μg/ml, whereas the were inactive against BEL-7402 cell lines.

    Description: Prophenoloxidase (PPO) is an essential enzyme in insect innate immunity because of its role in humoral defense. In this study, we have cloned a full-length cDNA of Antheraea pernyi prophenoloxidase (ApPPO) with an open-reading frame encoding 683 amino acids, and the deduced amino acid sequence of ApPPO exhibited a high similarity with those of lepidoptera. The expression of ApPPO was inducible so that the mRNA level was significantly upregulated in the microbial challenged tissues, including fat body, hemocytes, and midgut. To better investigate the enzymatic and immunological properties of ApPPO, recombinant ApPPO (rApPPO) was produced in Escherichia coli . Several functional verification experiments were performed after studying the enzymatic properties. It was found that rApPPO could be stimulated by the microbial challenged larvae hemolymph and then killed bacteria in the radial diffusion assay. Furthermore, rApPPO also induced the transcription of cecropins after injected into the larvae 24 h later.

    Description: Inflammatory processes are considered to play an important role in the progression of neurodegenerative changes in Alzheimer's disease (AD). A number of studies have reported inflammatory processes are highly correlated with cognitive deficits in AD-like mice. Transplantation of neural stem cells (NSCs) has been considered as a potential new therapy for the treatment of AD because of its effects in improving cognitive ability. However, NSCs have not been evaluated for their protective effects against inflammatory changes in AD. Here, we injected NSCs into amyloid precursor protein (APP)/PS1 transgenic mice to analyse cognitive function and to measure glial fibrillary acidic protein (GFAP), ionized calcium binding adaptor molecule-1 (Iba-1) and toll-like receptors 4(TLR4) activation. We also quantified TLR-4 pathway-related agents, Aβ concentration and the levels of pro-inflammatory mediators. Our results showed that in NSC-injected APP/PS1 mice, activation of GFAP, Iba-1, TLR4 and TLR4 pathway-related agents (MyD88, TRIF, P38 MAPK, NF-κB P65) were significantly decreased with decreased expression of pro-inflammatory mediators (IL-1, IL-6, TNF-α and PGE2). These changes were associated with the amelioration of cognitive deficits, but no difference was found in Aβ concentration. Our results provide novel evidence that NSCs transplantation in APP/PS1 mice significantly improved cognitive deficits and was accompanied by the attenuation of inflammatory injury via suppression of glial and TLR4-mediated inflammatory pathway activation. Our data indicate that these pathways may potentially be important therapeutic targets to prevent or delay AD. This article is protected by copyright. All rights reserved.
